분지아미노산 생합성 과정에 관여하는 첫 번째 효소인 acetolactate synthase (ALS)를 대상으로 수행할 수 있도록 고효율 검색방법(High Throughput Screening, HTS)을 개발하였고, 이를 이용하여 식물특이적 효소 저해제로 알려진 107개의 기존 화합물 중에서 새로운 ALS 저해 화합물을 선발하였다. 기존의 방법과 비교할 경우 한사람이 1회 수행한다고 하면 8 배 효율이지만 연속적으로 수행한다고 할 경우 1/10 이하의 양, 동일한 재료의 적용, 측정 결과의 계산, enzyme kinetics 등을 감안하면 최소 100 배 이상의 효과를 얻을 수 있다. 새로운 ALS 저해제로 탐색된 화학물질은, ammooxyacetic acid, azelaic acid, citric acid, cyanuric fluoride, glyoxylic acid, itaconic acid, malonic acid, niclosamid, oxalic acid, 2-oxoglutaric acid, suramin 등이었다. 앞으로 이들을 기본 구조로 하여 신규 ALS 저해 제초제의 개발을 위한 유도체의 합성에 이용되었으면 한다.
The n -octanol/water partition coefficient (Pow) is one of the most important parameters employed for estimating a chemiral's environmental fate and toxicity. The shake-flask method, one direct experimental method, i.1 prone to experimental artifacts for highly hydrophobic compounds. Thus, a valid method for direct determination of the Pow of highly hydrophobic compounds is needed. The slow -stirring method has been demonstrated to provide reliable log Pow data to log Pow greater than 5. This study was performed to evaluate the accuracy of slow- stirring experiment for determination of log Pow, particularly for highly hydrophobic compounds. 1, 2, 3, 4-tetrachlorobenzene, hexachlorobezene, 2, 2', 3, 3', 5, 5', 6, 6'-octachlorobiphenyl, decachlorobiphenyl, and p, p'-DDT (4.5$\times$0.02, 5.41$\times$0.06, 7.26$\times$0.04, 7.87$\times$0.10, and 6.03$\times$0.06, respectively. The octanol/water partition coefficient by the slow-stirring method were very similar to the literature values. These results indicate that the slow- stirring method allows for reliable determination of log Pow of highly hydrophobic chemicals.

We examined the pressure effects on petroleum pitch synthesis by using open and closed reaction systems. The pressure effects that occur during the pitch synthesis were investigated in three pressure systems: a closed system of high pressure and two open systems under either an atmosphere or vacuum. A thermal reaction in the closed system led to the high product yield of a pitch by suppressing the release of light components in pyrolysis fuel oil. Atmospheric treatment mainly enhanced the polymerization degree of the pitch via condensation and a polymerization reaction. Vacuum treatment results in a softening point increase due to the removal of components with low molecular weights. To utilize such characteristic effects of system pressure during pitch preparations, we proposed a method for synthesizing cost-competitive pitch precursors for carbon materials. The first step is to increase product yield by using a closed system; the second step is to increase the degree of polymerization toward the desired molecular distribution, followed by the use of vacuum treatment to adjust softening points. Thus, we obtained an experimental quinoline insolubles-free pitch of product yield over 45% with softening points of approximately $130^{\circ}C$. The proposed method shows the possibility to prepare cost-competitive pitch precursors for carbon materials by enhancing product yield and other properties.

Coke aggregates and carbon artifacts were produced to investigate the interactions of coke and pitch during the kneading process. In addition, the kneading ratio of the coke and binder pitch for the coke aggregates was controlled to identify the formation of voids and pores during carbonization at $900^{\circ}C$. Experiments and thermogravimetric analysis revealed that carbon yields were improved over the theoretical yield calculated by the weight loss of the coke and binder pitch; the improvement was due to the binding interactions between the coke particles and binder pitch by the kneading process. The true, apparent, and bulk densities fluctuated according to the kneading ratio. This study confirmed that an excessive or insufficient kneading ratio decreases the density with degradation of the packing characteristics. The porosity analysis indicated that formation of voids and pores by the binder pitch increased the porosity after carbonization. Image analysis confirmed that the kneading ratio affected the formation of the coke domains and the voids and pores, which revealed the relations among the carbon yields, density, and porosity.
나노구조를 갖는 중공구형(CNS) 및 중공반구형(NBC) 다공성 탄소 담체에 각각 약 1.0 wt%의 구리를 담지시켜 두 종류의 촉매를 제조하였고 과산화수소수에 의한 페놀의 수산화 반응에서 촉매의 성능을 두 종류의 서로 다른 용매(물, 아세토니트릴)에 대하여 비교 분석하였다. 촉매에 담지된 구리의 양은 EDS 분석으로 확인하였고 비표면적, 기공 부피, 기공 분포도 등을 비교 분석하였다. 두 종류의 촉매에서 모두 아세토니트릴보다 물에서 더 높은 전환율과 과산화수소 유효도 및 카테콜과 하이드로퀴논의 생성율을 얻을 수 있었고, 물을 용매로 사용했을 때 1.0 Cu/CNS 촉매가 1.0 Cu/NBC 촉매보다 50% 이상의 전환율과 과산화수소 유효도를 보였다.
This study was conducted to investigate the potential embryo-fetal toxicity and toxicokinetics of the antimalarial agent artesunate (ARTS) in Sprague-Dawley rats. Pregnant rats were administered ARTS daily from gestational day 6~15 via oral gavage, at test doses of 0, 2, 4, or 8 mg/kg (22 females per group). The fetuses were examined for external, visceral, and skeletal abnormalities on gestational day 20. With regard to the dams, there were no deaths, treatment-related clinical signs, changes in body weight, or food intake in any of the treatment groups. There were no treatment-related gross findings at necropsy in any treatment group. In the 8 mg/kg group, there was a decrease in gravid uterine weight and in the weight of female fetuses. There was also an increase in fetal deaths (primarily late resorptions) and an increase in post-implantation losses (37%) at 8 mg/kg. An increase in the incidence of visceral and skeletal variations at 4 and 8 mg/kg was observed. These defects included minor changes in the appearance of the kidney and thymus, as well as absent ribs or thoracic vertebrae. Toxicokinetics were assessed in a parallel study, using 4 mated females per group. Using liquid chromatography-mass spectrometry (LC-MS) analysis, the concentration of ARTS and its metabolite dihydroartemisinin (DHA) were quantified in plasma from rats on gestational days 5, 6, 10, and 15. Amniotic fluid was assayed for ARTS and DHA on gestational day 15. There was evidence of rapid conversion of ARTS to the metabolite DHA in maternal plasma, since ARTS could not be consistently detected in plasma at the three doses tested. ARTS and DHA were not detected in amniotic fluid at gestational day 15, indicating limited placental transfer of the two agents. The embryo-fetal no-observable-adverse-effect level (NOAEL) of the test item was considered to be 8 mg/kg/day for dams, and 2 mg/kg/day for embryo-fetal development.
