Taurine Transporter Activity in the Human Colon Carcinoma cell Line(HT-29) is Decreased during Cell Differentiation (인체 소장상피세포주(HT-29)의 분화단계에 따른 타우린수송체 활성의 변화)

  • Previous studies on the effect of age on intestinal taurine transport in animals have invariably shown a decline in the activity of the transport system with increasing age. In the present study changes in taurine transporter activity were observed during cell differentiation in the human colon carcinoma cell line HT-29 This cell line exhibits various enterocytic characteristics when differentiated and therefore has frequently been used to study the characteristcs and regulation of nutrient and drug absorption in the small intestine at the cellular level. Pre-treatment of the cells with $\beta$-alanine(10mM) reduced the taurine transport activity to 33% of the value for the control cells(p<0.05) which implies that taurine and $\beta$-alanine share a common $\beta$-amino acid transport system for their celluar uptake in the HI-29 was continued until 21 days post seeding. Kinetic studies of the taurine transporter were conducted in the HT-29 cell line with varying taurine concentration(5-60$\mu$M) in the uptake medium Both Vmax and the Michaelis-Menten constant(Km) of taurine transporter were decreased as differentiation of the HT-29 cell line was progressed ; Vmax of the taurine transporter in cells incubated for 4, 14 and 21 days post seeding was 2.79$\pm$3.4m 16.89$\pm$1.74, and 0.85$\pm$0.08 and 0.32$\pm$0.01nmol.mg protein-1 .30min-1 respectively(p<0.001) and Km was 42.3$\pm$3.4, 16.89$\pm$1.74, and 11.2$\pm$3.0$\mu$M respectively (p<0.01) These results indicate that the activity of sodium dependent active taurine transport system in the HT-29 cell line is decreased as confluent cells are differentiated. This phenomenon in cell culture system corresponds well with the earlier observation of lower intestinal taurine transport activity in suckling rats compared to that in adult animals although direct relationship of cell differentiation with in vivo aging process needs further verification.

  • This experiment was conducted to investigate the characteristics of alkaline protease from Aspergillus fumigatus which was isolated from soil as a superior strain for the production of the alkaline protease. The optimum temperature for enzyme activity was $50^{\circ}C$ and optimum pH was 9.0. The enzyme was stable at pH 8.0 to 10.0 and thermal inactivation was shown $30^{\circ}C$. The activity of the enzyme was increased by the addition of $Mn^{++},\;Cu^{++},\;Ba^{++},\;Mg^{++},\;$wheras it was inhibitied by $K^+,\;Fe^{+++},\;Ag^+,\;Pb^{++},\;Na^+,\;Ca^{++},\;Hg^+,\;Zn^{++}$. EDTA. 2, 4-DNP, ${\varepsilon}-amino$ caproic acid did not show inhibitory effect on the proteolytic activity of alkaline protease but P-chloromercuribenzoic acid inhibited the enzyme activity, indicating that reactive sulfhydryl group is required for the enzymatic activity. The reaction of this enzyme followed typical Michael-Menten Kinetics with the Km value of $8.33{\times}10^{-4}mole/{\ell}$ with the Vmax of $47.62{\mu}g/min$. This enzyme had stronger proteolytic activity than trypsin on substrate such as casin and hemoglibin.

Comparison of the Symmetry of Buttock Pressure during Simulated Driving between Heathy Adults and Patients with Stroke

  • Purpose: Driving is an important activity that is affected by various motor and cognitive deficits after stroke. On the other hand, there is no standard screening tool to evaluate the sitting asymmetry during driving, which is the stereotyped postural characteristic observed in patients with stroke. Therefore, this study compared the buttock pressure during simulated driving between healthy adults and patients with stroke. Methods: Ten post-stroke patients and ten healthy subjects participated in the experiment. The participants experienced simulated driving of 6.1 km during approximately 5 minutes for adaptation. The driving scenario consisted of 3.5 km urban traffic conditions, 10 km straight highway, and 7 km curved or hilly rural roads. Force sensitive application (FSA) was used to analyze the distribution of the buttock pressure on the driver's seat. The symmetry index (SI) was determined using the average buttock pressure of each side of the buttock. The closer SI is to zero, the higher the symmetry of buttock pressure. Results: These studies showed that the SI of healthy subjects was significantly closer to zero than that of the stroke patients. Conclusion: The buttock pressure of the stroke patients showed more asymmetry than that of the healthy subjects during simulated driving. Therefore, a therapeutic approach is needed for symmetrical sitting to improve the driving performance.

Production and Characteristics of Environment-Friendly Antimicrobial Substance by Pseudomonas aeruginosa EL-KM (Pseudomonas aeruginosa EL-KM에 의한 환경친화적 항균물질의 생산과 특성)

  • An antimicrobial substance-producing microorganism was isolated from soil samples. Based of the taxonomic characteristics of its morphological, cultural, physiological properties and 16s rRNA sequence alignment, this microorganism was identified as Pseudomonas aeruginosa, and we named Pseudomonas aeruginosa EL-KM. The optimal culture condition for production of antimicrobial substance was 1% mannitol, 0.4% yeast extract, 0.5% Nacl, 0.2% $K_2SO_4$, 100$\mu$M $MgSO_4$.$7H_2O$, 10$\mu$M $CaCl_2$.$2H_2O$, 1$\mu$M $FeSO_4$.$7H_2O$, 1$\mu$M $MnSO_4$.$4-5H_2O$, initial pH 7 and 200 rpm at 3$0^{\circ}C$. The purification of the antimicrbial substance was performed by silica gel column chromatographys, and fraction with TLC $R_f$ 0.77 value represented good antimicrobial activity. The crude antimicrbial substance was stable within a pH range of 3-10 and temperature range of 4$^{\circ}C$-121$^{\circ}C$ autoclaving. This crude antibacterial substance acted as bacteriolytic agent against Vibrio cholerae non-Ol ATCC 25872, and also exhibited excellent properties, when the substance was demonstrated against many other gram-positive, gram-negative bacteria, yeast and fungi.

Liver Protective Effects of Jageum-Jung in Alcohol-induced liver injury mice model (알코올 유발 간 손상 마우스 모델에서 자금정의 간 보호 효과)

  • Objectives : This study investigated the hepatoprotective effects effects of Jageum-jung extract on alcohol-induced liver disease mice model. Methods : Alcoholic liver disease was induced by Ethanol in C57/BL6 male mice, which were fed Lieber-DeCarli liquid diet containing ethanol. Jageum-jung (100,200 and 300 mg/kg bw/day) were orally administered daily in the alcoholic fatty liver disease mice for 16 days. Results : The results indicate that Jageum-jung promotes hepatoprotective effects by significantly reducing aspartate transaminase (AST) and alanine transaminase (ALT) levels as indicators of liver damage in the serum. Furthermore, Jageum-jung decreased accumulation of triglyceride and total cholesterol, increased levels of superoxide dismutase (SOD) and glutathione (GSH) in the serum of the alcoholic fatty liver disease mice model. Additionally, it improved the serum alcohol dehydrogenase (ADH) activity. Conclusions : This study confirmed the anti-oxidative and hangover elimination effects of Jageum-jung extract, and suggests the possibility of using Jageum-jung to treat alcholic liver disease.

Differential Effects of Transforming Growth Factor-β on NKG2D Ligands Expression and NK Cell-mediated Immune Responses in Primary and Metastatic Colon Cancer (원발성 및 전이성 대장암에서 TGF-beta가 NKG2D 리간드 발현과 NK 세포 매개 면역반응에 미치는 영향)

  • Transforming growth factor-β (TGF-β) is a multifunctional cytokine that affects not only the survival and growth of cancer cells but also the activity of immune cells. Although it has been generally accepted that cancer cell-derived TGF-β could promote the survival and growth of early cancer cells and have immunosuppressive roles, it has been known that TGF-β has differential effects according to the type or stage of cancer cells. Therefore, it is hard to clearly define its role in cancer progression and immune responses. This study investigated the effects of TGF-β signaling on the expression of five NKG2D ligands and the NK cell-mediated anticancer immune response in the primary colon cancer cell line KM12C and its two metastatic cell lines, KM12SM and KM12L4A. At the surface protein level, exogenous TGF-β decreased the expression of MICA, MICB, ULBP1, and ULBP2, and galunisertib increased the expression of MICA, MIAB, ULBP1, ULBP2, and ULBP3 in KM12C. However, KM12SM and KM12L4A showed no significant changes in the expression of NKG2DLs after treatment with TGF-β or galunisertib. TGF-β signaling inhibition via galunisertib improved the NK cell-mediated anticancer immune response against KM12C but did not show a significant response to KM12SM and KM12L4A. Therefore, the suppression of TGF-β signaling could improve the NK cell-mediated anticancer immune response against KM12C. However, an increase in NKG2DLs expression and an enhanced NK cell-mediated cancer immune response is hard to expect due to the alteration of TGF-β signaling in KM12SM and KM12L4A.

A Study on the Change of Magma Activity from 2002 to 2009 at Mt. Baekdusan using Surface Displacement (지표변위를 활용한 백두산의 2002-2009년 마그마 활동 양상 변화 연구)

  • There have been a number of observed precursors of volcanic activities- such as volcanic earthquake, surface inflation, specific volcanic gas emission, temperature of hot spring- at Mt. Baekdusan since 2002. We identified the increase of the volume of magma chamber beneath Mt. Baekdusan as we observed an inflation trend of vertical and horizontal surface displacement around Cheonji caldera lake by using precise leveling data from 2002 to 2009. The surface displacement trend changed to deflation in 2010, and the trend changed to inflation again after a while. Utilizing the data of inflated surface (46.33 mm) on the northern slope of Mt. Baekdusan from 2002 to 2003, we calculated the volume change of magma chamber beneath the Mt. Baekdusan. The volume change was about 0.008 $km^3$ ($7.7-8.0{\times}10^6m^3$) from 2002 to 2003. It indicated that a new magma (0.008 $km^3$) injected to the magma chamber 5 km below Mt. Baekdusan.

  • Mount Fuji is the focus of intense study because of its potential hazard signaled by seismic, geologic and historical activity. Based on extensive seismic data recorded in the vicinity of Mt. Fuji, coda quality factor ($Q_c^{-1}$) using a single scattering model hypothesis, and intrinsic and scattering quality factor $(Q_i^{-1}$ and $Q_s^{-1})$ using the Multiple Lapse Time Window Analysis (MLTW) method was measured. To focus the study on the magmatic structure below Mt. Fuji, to the data were separated into two groups: a near-Fuji region of rays traversing an area with radius 5 km around the summit (R < 5 km), and a far-Fuji region of rays beyond a radius of 20 km around the summit (R > 20 km). The results of the study have a small error range due to the large data sample, showing that all $Q^{-1}$ values in near-Fuji area are greater than those of far-Fuji area, and $Q_i^{-1}$ for both the near and far-Fuji area is higher than $Q_s^{-1}$ at high frequencies. The $Q_i^{-1}$ values of the near-Fuji area are lower than those of the other volcanic areas considered, while values of $Q_s^{-1}$ are not. The low $Q_i^{-1}$ for the volcanic region of near-Fuji suggests that the magmatic activity, or percent of partial melt, at Mt. Fuji is not as active as hot spot volcanoes such as Kilauea, Hawaii.

Purification and Properties of Homoseine Dehydrogenases in Canavalia Iineata Cotyledons (해녀콩(Canavalia lineata) 자엽에서 Homoserine Dehydrogenase의 정제 및 특성)

  • Two forms of homo serine dehydrogenase have been isolated from 8-day-old cotyledons of Canavalin lineata by a heat denaturation, ammonium sulfate fractionation, DEAE-8ephacel ion exchange and Sephacryl 8-300 gel filtration chromatographies, and Pro cion red dye, Cibacron blue dye and Resource Q column chromatographies. The molecular weights of T -form (threonine-sensitive) and K-form(threonine- insensitive) were estimated to 230 kD and 135 kD, respectively. In the presence of 10 mM threonine, the activity of T-form was inhibited with almost 70%, but that of K-form was not at all. The Km values tor homo serine of T- and Kform were 1.6 mM and 0.3 mM, respectively. The Km values for NAD of T- and K-form were 2.34 mM and 0.03 mM, respectively. And Km values for NADP of two isozymes were the same as 0.01 mM. The activities of T- and K-form were markedly stimulated up to 4.9and 2.8-fold, respectively, by 400 mM KCI. The partial purified(gel filtration) enzymes(Tform and K-form) can be reversibly converted.verted.

Purification and Properties of Branched Chain Amino Acid Arminotransferase from Fasciola hepatica (간질(Fasciola hepatica)의 Branched Chain 아미노산 Aminotransferase의 정제 및 성상)

  • The distribution and Properties of branched chain amino acid aminotransferase (EC 2.6. 1.42) was investigated in adult Fasciola hepatica. Fascicla hepatica was fractionated by differential centrifugation into nuclear, mitochondrial and cytosolic fractions. The activity of branched chain amino acid aminotransferase was measured by the method of Ichihara and Koyama (1966) . Isozyme patterns of this enzyule was also examined by DEAE-cellulose column chromatography. The results obtained were as follows; 1. The activity in homogenate was found to be 12.69 units/g wet tissue. The activity of this enzyme was relatively high compared with those in rat tissues. 2. The distribution of branched chain amino acid aminotransferase in the subcellular organelles showed that 87.8% of the activity was in cytosolic, 10.9% in mitochondrial and 1.3% was in nuclear fraction. 3. Cytosolic fraction of Fasciola hepatica contained Enzyme I, but not Enzyme II and III, of branched chain amino acid aminotransferase. Ensyme I was eluted by 50mM phosphate buffier from DEAE-cellulose column and catalyzed the transamination of all three branched chain amino acids. 4. The Enzyme I was purified about 22-folds increase in specific activity after chromatography on DEAE-cellulose. 5. The best substrate among three amino acids (leucine, isoleucine and valise) was L-isoleucine. 6. The optimal temperature of Enzyme I was $45^{\circ}C$ and the optimal pH was 8.2. 7. The Km value for leucine of Enzyme I was 4.17 mM. 8. The Km values for a-ketoglutarate and pyridoxal phosphate of Enzyme I were 0.41mM and $4.76{\times}10^{-3}{\;}mM$, respectively.
