Role of Krebs von den Lungen-6 (KL-6) in Assessing Hypersensitivity Pneumonitis

  • Background: Hypersensitivity pneumonitis (HP) is an increasingly recognized form of diffuse parenchymal lung disease. Krebs von den Lungen-6 (KL-6) is now classified as a human MUC1 mucin protein, and regenerating type II pneumocytes are the primary cellular source of KL-6/MUC1 in the affected lungs of patients with interstitial lung diseases (ILD). Serum KL-6/MUC1 levels have been demonstrated to be useful for the evaluation of various ILD. To determine the role of circulating KL-6 in evaluating the disease activity and management of HP. Methods: An observational cross-sectional study was conducted on 51 patients with HP and 20 healthy controls. Serum KL-6 levels were measured in both groups. Patients were further assessed based on chest high-resolution computed tomography (HRCT), pulmonary function test, 6-minute walk test, echocardiography, bronchioalveolar lavage, and/or transbronchial biopsy. Patients were divided into the fibrotic and non-fibrotic groups according to the HRCT findings. Results: The median serum KL-6 levels were significantly higher in HP patients as compared to the control group. The median serum KL-6 levels were found to be higher in the non-fibrotic HP group (1,900 IU/mL) as compared to the fibrotic group (1,200 IU/mL). There was a significant inverse correlation between serum KL-6 serum level and the dose of steroids as well as the duration of steroid therapy. Conclusion: The presence of higher KL-6 levels in the non-fibrotic HP group implies its enhanced production by regenerating pneumocytes in response to alveolar injury. The significant association between serum KL-6 levels and the dose and the duration of steroid therapy emphasizes the significant role of steroids in the stabilization of the disease.

  • Klotho (KL) is a single transmembrane protein composed of KL1 and KL2 repeats possessing ${\beta}$-glucuronidase activity and maintains calcium homeostasis in physiological state. It has been implicated in pigs that calcium is important for the establishment and maintenance of pregnancy, and our previous study has shown that transient receptor potential vanilloid type 6 (TRPV6), a calcium ion transporter, is predominantly expressed in the uterine endometrium during pregnancy in pigs. However, expression and function of KL in the uterine endometrium has not been determined in pigs. Thus, the present study determined expression and regulation of KL in the uterine endometrium during the estrous cycle and pregnancy in pigs. Real-time RT-PCR analysis showed that levels of KL mRNA decreased between Days 12 to 15 of the estrous cycle, and its expression showed a biphasic manner during pregnancy. KL mRNA was expressed in conceptuses and in chorioallantoic tissues during pregnancy. Explant culture study showed that expression levels of KL were not affected by treatment of steroid hormones or interleukin-1beta during the implantation period. Furthermore, levels of KL mRNA in the uterine endometrium from gilts carrying somatic cell nuclear transfer (SCNT)-derived embryos were significantly lower than those from gilts carrying natural mating-derived embryos on Day 12 of pregnancy. These results exhibited that KL was expressed at the maternal-conceptus interface in a pregnancy status- and stage-specific manner, and its expression was affected by SCNT procedure, suggesting that KL may play an important role in the establishment and maintenance of pregnancy in pigs.

Effect of Heat Treatment and Antibiotics on the Growth of Cellulomonas sp. KL-6 (Cellulomonas sp. KL-6의 증식에 미치는 열처리 및 항생물질의 효과)

  • For producing single cell protein from the agricultural waste, heat treatment and antibiotics on the growth of Cellulomonas sp. KL-6, isolated in rotting leaf and the adjacent soil mixture, were examined. The organism was able to grow until 5 min. at $65^{\circ}C$, 1 min. at $75^{\circ}C$ and 1/4 min. at $85^{\circ}C$ in gradually rising temperatures. It can be Seen that preheating the suspension at $48^{\circ}C$ results in a marked decrease in heat resistance. On heating at temperature of $55^{\circ}C$ for 30 min., strain KL-6 was more resisted in the 0.1 M phosphate buffer when such substrates as casamino acid (1%), yeast extract (1%) or xylose (5%) were added to it whereas this organism was appeared weaker resistances in 0.1 M phosphate buffer when cysteine (0.03 M), sodium citrate (1%) or casein (1%) were in fused into it. Test strain was susceptible to penicillin-G $(1.563\;{\mu}g/ml)$ and ampicillin $(3.125\;{\mu}g/ml)$, but the organism was resisted to kanamycin $(>200\;{\mu}g/ml)$. The treatment of strain KL-6 with sodium dodecyl sulfate (SDS) resulted in the elimination of R-plasmid from the host strain and the elimination rate with SDS $(10{\sim}30\;{\mu}g/ml)$ was about $9.2{\sim}31.2%$, respectively.

Isolation and physiological characteristics of cellulolytic bacteria (섬유소 분해세균의 분리 및 생리적인 특성)

  • Three hundred and one cellulolytic bacterial were isolated from the 148 screening sources such as decomposed wood, soil, compost and leaf mold. Among them, strain KL-6 was found to have the highest of cellulase activity, and identified as species belonged to the genus Cellulomonas. Strain KL-6 was decompose up to 90% of the filter paper (whatman No. 1) substrate within 50 hours, and showed the colony halo formation (11 cm). The activities of CMCase (67 unit/ml), FPase (70 unit/ml) and ${\beta}-glucosidase$ (0.68 unit/ml) were obtained when this strain was cultured for 50 hrs at $30^{\circ}C$. Glucose was not found in detectable amounts at the FP medium. The optimum composition of nutrient medium for the cell growth by strain KL-6 was sucrose 0.5%, yeast extract 0.1%, $(NH_4)_2HPO_4\;0.1%$, $K_2HPO_4\;0.1%$, $MgSO_4{\cdot}7H_2O\;0.01%$, $CaCl_2\; 0.01%$, NaCl 0.6%, $CaCO_3\;0.1%$ and the optimum pH and temperature were 7.0 and $30^{\circ}C$, respectively.

  • Two vibrio sp. strains were isolated from disease catfish(silurus asotus). The present isolates were identified as Vibrio ordalii based on their biological and biochemical characteristics ; they were positive for acid production from glucose, Iactose, maltose, sucrose and salicine, while negative for arabinose, galactose, inocitol and xylose. They are named KL-1 and KL-2, KL-1 and KL-2 strains were similar to physiological characteristics ; growth was observed at pH 5 to 10 and in 0% to 6.0% NaCl. Two strains did not growth at a concentration above 7.0% NaCl and pH10. This bacterium was infected into health catfish hypodermically. Such injection was found to induce haemorrhagic ulcers very similar to those observed in naturally infected fish. At 24h post-infection, the red spot developed around the injection site and grew bigger to from a red sport area. At 120h post-infection, the muscle ncerosis was extended near the ventral fin. The seventy percent lethal dosage was appeared to water temperature at $25^{\circ}C$. Two strains were tested for drug senistiveity by plate method. KL-1 and KL-2 strains were sensitive to GM. K, N, S and SxT, but resistant to CF, L and VA.

Fermentation Aspects of Fruit-Vegetable Juice by Mixed Cultures of Lactic Acid Bacteria Isolated from Kimchi and Yeast (김치 젖산균과 효모의 혼합배양 방법에 의한 과채류즙의 발효양상)

  • Fermented beverage using lactic acid bacteria isolated from kimchi was investigated. Lactic acid bacteria KL 1, KD 6, KL 4 strains from kimchi, or obtained Lactobacillus acidophilus, Lactobacillus plantarum, Leuconostoc mesenteroides with and without yeast(Saccharomyces cerevisiae) were inoculated in fruit vegetable juice for single and mixed culture fermentation. During the fermentation by bacterial strain and yeast for 1~3 days at 30oC, various fermentation behaviors were observed. The growth rate of mixed culture of KL 1 and yeast was higher than that of single culture by KL 1 alone during the fermentation. The amount of organic acid produced by the mixed culture fermentation of KL 1 and yeast was 0.82%(3 day) or 0.58%(1 day) and with the final pH of 3.3(3 day) or 4.2(1 day). These mixed culture systems of isolated strains or other bacterial strains had almost similar results of growth rate and acid production. Among several bacterial strains, KL 1 was suitable for the mixed culture fermentation with yeast in terms of desirable fermentation behavior and organoleptical quality. The selected strain, KL 1 was identified as Leuconostoc spp. through the series of tests on carbohydrate fermentation and biochemical characteristics.

Kraft Lignin Biodegradation by Dysgonomonas sp. WJDL-Y1, a New Anaerobic Bacterial Strain Isolated from Sludge of a Pulp and Paper MillS

  • Wastewater containing kraft lignin (KL) discharged from pulp and paper industries could cause serious environmental contamination. Appropriate effluent treatment is required to reduce the pollution. Investigations on anaerobic bacteria capable of degrading KL are beneficial to both lignin removal and biofuel regeneration from the effluent. In this paper, an anaerobic strain capable of degrading KL was isolated from the sludge of a pulp and paper mill and identified as Dysgonomonas sp. WJDL-Y1 by 16S rRNA analysis. Optimum conditions for KL degradation by strain WJDL-Y1 were obtained at initial pH of 6.8, C:N ratio of 6 and temperature of 33℃, based on statistical analyses by response surface methodology. For a 1.2 g/l KL solution, a COD removal rate of 20.7% concomitant with biomass increase of 17.6% was achieved after 4 days of incubation under the optimum conditions. After the treatment by strain WJDL-Y1, KL was modified and degraded.

Applicability of Serum Krebs von den lungen-6 as a Biological Exposure Index for Workers Exposed to Indium (인듐 노출 근로자를 위한 생물학적 노출지표로써 혈청 Krebs von den lungen-6의 활용가능성)

  • Objectives: Although several cases of lung diseases caused by indium have been reported in Japan, the United States and China, South Korea, which is estimated to have been the world's largest consumer of indium, has not yet established a criteria for the diagnosis of lung diseases caused by indium exposure. In this study, we tried to determine the applicability of the Krebs von den lungen-6, which has been widely recognized for its use with interstitial lung disease in Japan, as a biological exposure index for indium. Methods: Methods: The analysis of indium in serum was conducted by inductively coupled plasma mass spectrometry and the analysis of KL-6 in serum was carried out using enzyme-linked immunosorbent assay kit. Results: The indium levels in serum were distributed from below the detection limit to a peak of $125.78{\mu}g/L$, and the values of the KL-6 were distributed from 104.5 U/mL to 2162.2 U/mL. The serum indium and KL-6 showed good correlation ($R^2$=0.389,pfortrend=0.000) and smoking did not affect the KL-6. Conclusions: The usefulness of KL-6 as a specific biomarker for interstitial lung disease has been recognized. In addition, it is expected that effective prevention of health problems can be achieved by determining the lung-damage progress at an early stage according to individual susceptibility.

Production of Single Cell Protein on Rice Straw and Their Utilities (섬유소를 이용한 단세포단백질의 생산 및 그 이용)

  • Experiments were carried out to find out the optimal condition of SCP produced by Cellulomonas sp. KL-6 and to evaluate nutritional value for the protein of this organism Intracellular- and extracellular proteins produced by this strain were estimated to be nearly maximum, $266\;{\mu}g/m{\ell}\;and\;37\;{\mu}g/m{\ell}$, in the medium containing 0.001% of thiamin after 5 days cultivation. When used rice straw as carbon source for the cell growth of this organism after crusing them by cutting mill, and treating them with 1.0% of NaOH and 10.0% of $NH_4OH\;at\;80^{\circ}C$ for 30 minutes and neutralizing continuatively them with 85% of $H_3PO_4$, SCP production rates were very increased to $1.63\;g/{\ell}$ (NaOH) and $1.47\;g/{\ell}$ (NH4OH), respectively than $0.5\;g/{\ell}$ produced in untreated rice straw. We compared their amino acids patterns with that of FAO provisional patterns. Amino acids content of strain KL-6 was excellents. However. when intended these cell mass to use in practical animal feeding test it would be advisable that destruction or lysis of cell wall should be done.

Production of Cellulase from Cellulomonas sp. KL-6 (Cellulomonas sp. KL-6에 의한 섬유소 분해효소의 생산)

  • Among the cellulases by Cellulomonas sp. KL-6. CMCase and filter paperase, which were produced as the out enzymes of cell, had been much produced, but very small amounts of ${\beta}-glucosidase $, the enzyme of which is cell bound form, was produced by this organism. The optimal culture times for CMCase and filter paperase productions were 5 days, while that of ${\beta}-glucosidase$ was 4 days. When this strain was cultured under the optimal medium for enzyme production, CMCase, FPase and ${\beta}-glucosidase$ were $82\;units/m{\ell},\;80\;units/m{\ell}\;and\;1.2\;units/m{\ell}$, respectively. Thus these results were showed to increase enzyme productivities as about $60{\sim}70%$ than those produced in basal medium. $CaCO_3$ injected to the medium as the ratio of 0.1% was not only enhanced cellulase activities but also effective as acid neutralizing agent. The production effects of lignase and lactase by this bacterium in filter paper medium was not appeared.
