Enhancement of Biocontrol Activity of Antagonistic Chryseobacterium Strain KJ1R5 by Adding Carbon Sources against Phytophthora capsici

  • Carbon utilization by Chryseobacterium strain KJ1R5 was studied to enhance its biocontrol activity against Phytophthora capsid. Chryseobacterium strain KJ1R5 has previously been shown to control Phytophthora blight of pepper (Capsicum annuum L.). Strain KJ1R5 could utilize carbon sources such as L-arabinose, D-cellobiose, ${\beta}-lactose$ and D-galactose well. P. capsici could utilize D-glucose well, showing the absorbencies ranged from 0.577 to 0.767 at 600nm. When 2% L-arabinose, which could only be utilized by the bio-control strain KJ1R5, was amended into the bacterial suspension, the efficacy of biological control increased. Among the amendments of various carbon sources into bacterial suspension, L-arabinose and D-(+)-glucose significantly enhanced biological control activity, resulting in a reduction of disease incidence to 6.9%, compared to 21.9% for the strain KJ1R5 alone and 81.3% for P. capsici inoculation alone, indicating that amendment with specific carbon sources could increase the biological control activity.

Characterization of Plasmids from Bifidobacterium sp.

  • Ten strains of plasmid-harboring Bifidobacterium sp. were isolated from the feces of adults and children, and named as Bifidobacterium sp. GE1-GE8, ST, and SH5. These plasmids were categorized into three homologous groups (pKJ50-homologous, pKJ36-homologous, and non-homologous groups) according to Southern hybridization patterns using the formerly characterized bifidobacterial plasmids, pKJ50 and pKJ36, as probes. nine strains harboring the plasmids were shown to accumulate single-stranded DNA as a replication intermediate, based on the S1 nuclease treatment and Southern hybridization. These results suggest that the strains replicate by a rolling circle mechanism. Minimal inhibitory concentrations (MIC) of the isolated bifidobacteria against several antibiotics were determined. Two strains, GE2 and GE3, showed relatively high MiC values against tetracycline ($793.6{\mu}g/ml$) and erythromycin ($153.6{\mu}g/ml$), respectively. The tetracycline resistance of GE2 disappeared when the resident plasmid of GE2 was cured by ethidium bromide. These results show that pKJ36-homologous and pKJ50-homologous plasmids are prevalent among various Bifidobacterium strains and some Bifidobacterium plasmids appear to code for antibiotic resistance.

Gluconacetobacter persimmonensis KJ145를 이용한 Bacterial Cellulose 및 초산발효에 미치는 Ethanol의 영향 (Effect of Ethanol on the Production of Cellulose and Acetic Acid by Gluconacetobacter persimmonensis KJ145)

  • G. persimonensis-KJ145를 차용하여 BC 생산과 식초생산에 미치는 에탄올의 영향을 조사하였다. 그 결과 사과쥬스배지에 에탄올을 2%(v/v) 첨가하였을 때 BC의 생산성이 가장 좋았으며, 7%(v/v)를 첨가하였을 때 총산의 함량이 가장 높았다. BC와 식초를 동시 생산하기 위해서는 에탄올을 5%(v/v) 첨가하는 것이 가장 좋을 것으로 생각되며 발효과정 중의 식초 품질 변화에 대한 보다 자세한 연구가 요구되었다. BC와 식초 생산에 가장 효과적인 질소원은 CSL인 것으로 조사되었고 최적 농도는 15%(w/v)이었다. 배양시간의 영향을 조사한 결과 14일간 배양하는 것이 가장 많은 양의 BC를 생산하였으며, 최적조건에서 생산되는 BC 생산량은 7.55 g/L이었다.

Penicillium sp. KJ 81에 의한 Erythritol 생산 최적 배양 조건 (Optimization of Cell Culture Condition for Erythritol Production by Penicillium sp. KJ8l)

  • Erythritol은 저 칼로리 건강 감미 물질로서 우리는 이미 자연계로부터 폭넓은 탐색을 통하여 Penicillium sp. KJ8l을 분리, 동정하였다. 본 연구에서는 Penicillium sp. KJ8l의 erythritol 생산을 위한 최적 배양 조건을 검토하였다. 이 균주는 기질로서 glucose, sucrose, fructose, mannose, lactose, maltose 그리고 galactose를 이용하여 erythritol을 생산할수 있었으나 mannitol, arabinose, sorbitol 그리고 xylose를 이용하여서는 erythritol을 생산할 수 없었다. 또한 sucrose를 기질로 사용하였을 때 가장 높은 erythritol 생산성을 보였다. Penicillium sp. KJ8l은 배지의 sucrose 농도가 60%까지 erythritol을 생산할 수 있었으나, 30% sucrose농도에서 erythritol 생산성이 가장 좋았다. 또한 배지내 yeast extract의 농도가 0.5%일 때 erythritol 생산성 이 가장 좋았다. 본 균주는 30% sucrose와 0.5% yeast extract 배지에 0.5% ammonium sulfate를 첨가해 주었을 때 erythritol의 생산성이 향상되었다. Penicillium sp. KJ8l은 5ι jar. fermentor에서 30% sucrose, 0.5% yeast extract, 0.5% ($(NH_{4})_{2}SO_{4}$, 0.1% $KH_{2}PO_{4}$, 0.01% $MgCl_{2}$ 조성의 배지에서 1 vvm으로 산소를 공급해 주면서 200 rpm으로$37^{\circ}C$에서 11 일간 배양하였을 때 최고의 erythritol 생산을 보였다. 이 때 erythritol 생산 수율은 28.2g/l이었다.

자외선 B파 조사가 느타리버섯의 이화학적 특성에 미치는 영향 (Effects of UV-B Irradiation on the Physicochemical Characteristics of Oyster Mushrooms (Pleurotus ostreatus))

  • The aim of this study was to investigate the effects of UV-B irradiation on the physicochemical characteristics of oyster mushrooms (Pleurotus ostreatus). Vitamin $D_2$ concentration, weight loss rate, color value, total plate counts and consumer acceptability of irradiated oyster mushrooms were measured. UV-B irradiation at doses of $0\;kj/m^2$, $20\;kj/m^2$ and $40\;kj/m^2$, significantly increased the vitamin $D_2$ concentrations from $0\;{\mu}g/g$ dry weight (control) to $85.87\;{\mu}g/g$ dw and $116.28\;{\mu}g/g$ dw, respectively, at 5% level. Rate of weight loss was also significantly increased from 0% (control) to 2.21% and 4.31% at $20\;kj/m^2$ and $40\;kj/m^2$ UV-B irradiation, respectively, at 5% level. Although there was no significant difference between the UV-B irradiated groups, total plate counts were significantly decreased from $1.0{\times}10^5$ (control) to $9.4{\times}10^3$ and $1.9{\times}10^3$at $20\;kj/m^2$ and $40\;kj/m^2$ of UV-B irradiation, respectively, at 5% level. There was no significant difference in L values and consumer acceptability between the groups. Therefore, UV-B irradiated oyster mushrooms could be used as health promoting ingredients for many foods.

KJ27을 복용한 슬관절염(膝關節炎) 환자 30예(例)에 대한 임상적 연구 (Case Studies on 30 Patients with knee Arthritis treated with KJ27)

  • Osteoarthritis is a disease that has considerable effect on both individuals and economy. The number of osteoarthritis patients are gradually increasing as people get to enjoy longevity. This study was conducted to examine 30 osteoarthritis patients. Following is the report which shows the effectiveness of KJ27 based on statistical data analysis. Among 5 who have reported that they were not able to walk or use their knees at all, 80% of them were completely recovered. Among 18 who have stated that it was very hard for them to walk for more than a kilometer, 66.7% were completely recovered. Among 7 who have reported that it was little bit difficult for them to walk, 71.4% were completely recovered. Among 25 out of 30 who were very unsatisfied with former treatments, 70% of them stated that they were very satisfied with KJ27 treatments, and 30% of them stated that they were satisfied with it. Among 5 who had not had any type of osteoarthritis treatments, 100% have stated that they were very satisfied with the treatment. 18 patients who were under this treatment for a month were completely recovered; 10 patients who were under this treatment for 2 months were completely recovered; and finally 2 patients who were under this treatment for 3 months and longer were completely recovered. It is not clearly understood whether KJ27 will eradicate the entire symptoms of osteoarthritis, yet. Numerous case studies and experiments should be conducted to improve and apply KJ27.

Gluconacetobacter persimmonis sp. nov., Isolated from Korean Traditional Persimmon Vinegar

  • Screening was performed to isolate cellulose-producing microorganisms from the Korean traditional fermented persimmon vinegar. The resulting strain, KJ $145^{T}$, was then taxonomically investigated by phenotypic characterization, particularly chemotaxonomic, and by phylogenetic inference based on a 16S rDNA sequence analysis including other related taxa. Strain KJ $145^{T}$ was found to grow rapidly and form pale white colonies with smooth to rough surfaces on a GYC agar. Strain KJ $145^T$ also produced acetate from ethanol, and was tolerable to 10% ethanol in SM medium. In a static culture, a thick cellulose pellicle was produced, and in GYC broth, the strain grew at temperatures ranging from 28 to $40^\circ{C}$ with an optimum pH of 4.0. The genomic DNA G+C content of strain KJ $145^T$ was 61.9 mol%, and the predominant ubiquinone was Q 10 as the major quinone and Q9 as the minor quinone. The major cellular fatty acids were $C_{16:0}$ and the sum in feature 7 ($C_{18:1}$ w9c, w12t and/or w7c). A 16S rRNA-targeted oligonucleotide probe specific for strain KJ $145^T$was constructed, and the phylogenetic position of the new species was derived from a 16S rDNA-based tree. When comparing the 16S rDNA nucleotide sequences, strain KJ $145^T$ was found to be most closely related to G. hansenii LMG $1527^T$ (99.2%), although KJ $145^T$ was still distinct from G. hansenii LMG $l527^T$ and G. xylinus LMG $1515^T$ in certain phenotypic characteristics. Therefore, on the basis of 16S rDNA sequences and taxonomic characteristics, it is proposed that strain KJ $145^T$ should be placed in the genus Gluconacetobacter as a new species, Gluconacetobacter persimmonis sp. nov., under the type-strain KJ $145^T$ (=KCTC =$10175BP^T$=KCCM=$10354^T$).

  • 식물병 또는 비생물적 스트레스를 감소시키는 것으로 보고된 세균 4종(HS1, H30-3, KJ40 와 BC42)이 Colletotrichum orbiculare 에 의한 오이 탄저병, Acidovorax citrulli에 의한 과실썩음병, Fusarium oxysporum에 의한 오이 덩굴쪼김병에 대한 병 억제 효과가 있는지 검정하였다. HS1, H30-3, KJ40 와 BC42를 토양에 관주처리 할 경우 유도저항성에 의해 오이 탄저병이 감소하였고, KJ40와 BC42는 A. citrulli의 발병을, BC42는 오이 덩굴쪼김병을 억제하였다. 따라서, KJ40은건조피해저감뿐만아니라유도저항성에의 한 오이 탄저병 억제 및 A. citrulli에 의한 병발생 감소효과를 가지며, BC42는 오이 탄저병, A. citrulli와 F. oxysporum에 병을 모두 억제하는 것으로 보아 넓은 범위의 적용 범위를 갖는 생물적 방제제 자원으로 활용할 수 있을 것이다.

Bacterial Cellulose 생산균 KJ-1으로 부터 uv와 NTG mutant들의 cellulose 생산수율의 증가

  • 본 연구에서는 BC 생산균주인 KJ-1을 UV와 NTG mutagenesis를 통하여 초기에 주입된 glucose를 효과적으로 이용하여 cellulose 수율이 향상된 균주로 계량하였다. Mutant들 중 수율이 2배정도 향상한 균주에서는 cellulose를 비롯한 gluconic acid, 2-keto gluconic aicd, 5-keto gluconic acid의 비율이 parent strain과 차이를 보였고, 시간에 따른 acid 농도 분포를 살펴보면 수율이 증가한 원인 중에 mutant가 parent strain보다 합성한 acid를 효율적으로 이용하고 있다고 추론 할 수 있었다.

  • The amino acid threonine was produced from glycine and ethanol in a reaction mixture using cell free extract of the methylotrophic bacterium isolated from soil and identified as mellthylo-bacterium sp. KJ29. Although the isolate could grow on carbon source other than methanol, only the cell free extract from the cells grown on methanol produced threonine. Methanol dehydrogenase (MDH) activity was present only in the cells grown on methanol when compared to the cells grown on heterotrophic substrates.
