Photosystem I and II Reaction Centers in a New Type Oxygenic Photosynthesis of Acaryochloris marina Based on Chlorophyll d: Studies of Delayed Fluorescence and Triplet State ESR

  • A cyanobacteria-like organism Aaryochloris marina performs oxygenic photosynthesis with near far-red light by the use of chlorophyll d. Reaction center chlorophyll (Chl) of Photosystem (PS) II of A. marina was studied by analysis of millisecond-delayed fluorescence. Delayed fluorescence is emitted by Chi d indicating efficient energy transfer between antenna Chi d molecules and the unknown primary electron donor of PS II. P740 a reaction center Chl of PS I of A. marina is shown to give a dimer type cation, and triplet state with a D value of 245xlO$\^$-4/ cm$\^$-l/ in contrast to the 280-290 xlO$\^$-4/cm$\^$-l/ values of P700 suggesting triplet spins interacting at a 5% larger distance in P740 than in P700.

Prediction of Listeria monocytogenes Growth Kinetics in Sausages Formulated with Antimicrobials as a Function of Temperature and Concentrations

  • This study was conducted to develop a model to describe the effect of antimicrobials [potassium sorbate (PS), potassium lactate (PL), and combined PL and sodium diacetate (SDA, PLSDA)] on the growth parameters of Listeria monocytogenes such as specific growth rate (SGR) and lag phase periods (LT) in air-dried raw sausages as a function of storage temperature (4, 10, 16, and $25^{\circ}C$). Results showed that the SGR of L monocytogenes was dependent on the storage temperature and level of antimicrobials used. The most effective treatment was the 4% PLSDA, followed by the 2% PLSDA and 4% PL and 0.2% PS exhibited the least antimicrobial effect. Increased growth rates were observed with increasing storage temperatures from 4 to $25^{\circ}C$. The growth data were fitted with a Gompertz equation to determine the SGR and LT of the L. monocytogenes. Six polynomial models were developed for the SGR and LT to evaluate the effect of PS (0.1, 0.2%) and PL (2,4%) alone and PLSDA (2, 4%) on the growth kinetics of L. monocytogenes from 4 to $25^{\circ}C$.

Cloning, Expression, and Purification of Recombinant Uricase Enzyme from Pseudomonas aeruginosa Ps43 Using Escherichia coli

  • Uricase is an important microbial enzyme that can be used in the clinical treatment of gout, hyperuricemia, and tumor lysis syndrome. A total of 127 clinical isolates of Pseudomonas aeruginosa were tested for uricase production. A Pseudomonas strain named Ps43 showed the highest level of native uricase enzyme expression. The open reading frame of the uricase enzyme was amplified from Ps43 and cloned into the expression vector pRSET-B. Uricase was expressed using E. coli BL21 (DE3). The ORF was sequenced and assigned GenBank Accession No. KJ718888. The nucleotide sequence analysis was identical to the coding sequence of uricase gene puuDof P. aeruginosa PAO1. We report the successful expression of P. aeruginosa uricase in Escherichia coli. E. coli showed an induced protein with a molecular mass of about 58 kDa that was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blotting. We also established efficient protein purification using the Ni-Sepharose column with activity of the purified enzyme of 2.16 IU and a 2-fold increase in the specific activity of the pure enzyme compared with the crude enzyme.

Growth mechanism of three dimensionally structured TiO2 thin film for gas sensors (가스 감응용 3차원 구조체 TiO2 박막 성장기구)

  • Polystyrene (PS) microspheres were used to good advantage as a template material to prepare macroporous $TiO_2$ thin films. This is enabled to run the thermal decomposition of the PS without the collapsing of the 3-D macroporous framework during the calcination step. $TiO_2$ thin films were deposited onto the colloidal templated substrates at room temperature by RF sputtering, and then samples were thermally treated at $450^{\circ}C$ for 40.min in air to remove the organic colloidal template and induce crystallization of the $TiO_2$ film. The macroporous $TiO_2$ thin film exhibited a quasi-ordered partially hexagonal close-packed structure. Burst holes, estimated to be formed during PS thermal decomposition, are seen as the hemisphere walls. the inner as well as the outer surfaces of the hollow hemispheres formed by the method of thermal decomposition can be easily accessed by the diffusing gas species. As a consequence, the active surface area interacting with the gas species is expected to be enlarged about by a factor of fourth as large as compared to that of a planar films. Also the thickness at neighboring hemisphere could be controlled a few nm thickness. If the acceptor density becomes as large that depletion width reaches those thickness, the device is in the pinch off-situation and a strong resistance change should be observed.

Effect of the aeration rate and agitation speed on heteropolysaccharide-7 production by Beijerinckia indica

  • Effect of aeration rate and agitation speed on cell growth and the production of heteropolysaccharide-7 (PS-7) by Beijerinckia indica was investigated. Aeration rate and agitation speed in a 7L bioreactor ranged from 0.5 to 1.5 vvm and from 300 to 500 rpm, respectively. Higher agitation speed with an aeration rate of 0.5 vvm in the bioreactor resulted in maintenance of higher concentration of dissolved oxygen in the medium, which enhanced the production of PS-7. In this study with a 7L bioreactor, maximal production of PS-7 was 11.0 g/L and its conversion rate from 2% (w/v) glucose was 0.55 when the aeration rate and agitation speed were 1.0 vvm and 500 rpm, respectively. Proper aeration rate and agitation speed might enhance the production of PS-7 as well as reduce the time to reach maximal production.

Characterization of Surface Modified Polysulfone Membranes with Various Fluorine Chemicals

  • The surface of polysulfone membranes has been modified using the fluorine chemicals, ITFE (2-iodo-1,1,1-trifluoro-ethane F.W.=209.94) and PFPI (1H,1H-pentafluoro-n-propyl iodide F.W.=259.95), and PFI (1H,1H,2H,2H-perfluorohexyl iodide, F.W.=373.99) based on Friedel-Crafts reaction mechanism with varying reaction temperatures, reaction time, and catalysis types. The resulting membranes were characterized through mainly the contact angle measurement and pure water permeability. The smaller reactant shows the larger contact angles. FeBr$_3$ catalyst is more effective than AlCl$_3$. Typically, the PS film treated with ITFE at $25^{\circ}C$ under FeBr$_3$ catalyst showed the contact angle 78.5$^{\circ}C$ which indicated 10% over the value of unreacted PS films. More than 50% of pure water flux 8.0 g/$m^2$hr, reduced at reaction time 10 min relative to the original flux, 3.49 g/$m^2$hr.

  • Purpose: The purpose of this study was to clarify longitudinally the professional socialization (PS), self-esteem (SE1), and self-efficacy (SE2) over time in nursing students. Methods: Using a structured questionnaire, data were collected from the same nursing students in 2008, 2009, and 2010 years. Descriptive statistics and one-way ANOVA test with the SAS package were used for the data analysis. Results: The differences in the PS, SE1, and SE2 over time were not significant in nursing students. However, the differences in decision making over time were significant in nursing students. Conclusion: These results suggest that SE1 and SE2 continuously affected professional socialization over time. Therefore, it is necessary to devise a continuing and long-term strategy, which strengthens SE1 and SE2 in order to improve professional socialization.

  • Polyurethane prepolymers were prepared from poly (carbonate diol), isophrone diisocyanate and dimethylol propionic acid. Then, aniline terminated waterborne polyurethane dispersion (ATPUD) was synthesized by capping the NCO group of the prepolymer with aniline monomer. Subsequently, ATPUD and waterborne polyurethane dispersion (PUD), respectively, were blended with conducting polymer, poly (3,4-ethylenedioxythiophene)/polystyrene sulfonate [PEDOT/PSS], to yield antistatic coating solutions, and the mixture was coated on the polycarbonate substrates. At adequate addition amounts of PEDOT/PSS less than or equal to 2.5 g, the surface resistances ($1.0{\times}10^{11}{\sim}2.5{\times}10^8{\Omega}/cm^2$) of coating films from ATPUD showed better electronic conductivities than those ($5.0{\times}10^{11}{\sim}6.3{\times}10^9{\Omega}/cm^2$) from PUD. However, at excess amount of PEDOT/PSS of 3.0 g, the surface resistance from ATPUD showed similar electronic conductivity with that from PUD.

Inhibition of Foodborne Pathogens on Polystyrene, Sausage Casings, and Smoked Salmon Using Nonthermal Plasma Treatments (비열 플라즈마 처리를 이용한 polystyrene, 소시지 케이싱, 그리고 훈제연어에서의 식중독균 저해)

  • The effects of nonthermal plasma treatments against Salmonella Typhimurium, Escherichia coli O157:H7, and Listeria monocytogenes living on polystyrene (PS), sausage casings, and smoked salmon were investigated. Inoculated PS, casings, and salmon were treated with nonthermal plasma generated with helium (5 L/min) or with both helium (5 L/min) and oxygen (100 mL/min) at 60 Hz and 30 kV/cm for 2, 5, or 10 min. S. Typhimurium exhibited the highest sensitivity to the helium-used treatment. The greatest reduction (3.9${\pm}$0.8 log$CFU/cm^2$) was observed with L. monocytogenes on PS after the treatment with the mixed gas for 5 min. The treatment with the mixed gas inhibited L. monocytogenes on casings and salmon by 0.5${\pm}$0.3 log$CFU/cm^2$ and 1.0${\pm}$0.3 log CFU/g, respectively. Different treatment times did not result in different reductions of L. monocytogenes on both casings and salmon. The types of treatment gas and material of contamination need to be considered for evaluating the antimicrobial effects of nonthermal plasma treatments.

Mode Analysis and Modal Delay Measurement of a Few-Mode Fiber by Using Optical Frequency Domain Reflectometry

  • A novel mode analysis method and differential mode delay measurement technique for a multimode optical fiber based on optical frequency domain reflectometry has been proposed for the first time. We have used a conventional OFDR with a tunable external cavity laser and a Michelson interferometer. A few-mode optical multimode fiber was prepared to test our proposed measurement technique. The differential mode delay (DMD) of the sample fiber was measured to be 16.58 ps/m with a resolution of 1.5 ps/m. We have also compared the OFDR measurement results with those obtained using a traditional time-domain measurement method.
