Controlling Size and Distribution for Nano-sized Polystyrene Spheres

  • Highly monodisperse polystyrene (PS) nanospheres were fabricated by surfactant-free emulsion polymerization in water using styrene, 2,2'-azobis(2-methyl propionamidine) dihydrochloride (AIBA), and poly(vinyl pyrrolidone) (PVP). The size and distribution of the PS nanospheres were systematically investigated in terms of initiator concentration, stabilizer concentration, reaction temperature, reaction time, and reactant concentration. With increasing AIBA initiator concentration, PS particle sizes are raised proportionally, and can be controlled from 120 to 380 nm. Particle sizes were reduced with increasing PVP concentration. This decrease occurs because a high PVP concentration leads to a large number of primary nuclei in the early stage of polymerization. When the reaction temperature increased, the sizes of the PS particles decrease slightly. The particles grew quickly during the initial reaction stage (1-3 h) and the growth rate became steady-state after 6 h. The PS sizes approximately doubled when the reactant (styrene, PVP, azo-initiator) concentrations were increased by a factor of eight.

뇌졸중환자의 물리치료양상 및 기능변화에 관한 추적연구 (Follow - up Study on Functional Change and Aspect of Physical Therapy in Stroke Patients)

  • 본 연구는 뇌졸중 환자들의 퇴원 후 1개월까지의 물리치료양상 및 기능변화와 이에 관련된 요인을 알아보기 위해 1998년 1월 1일부터 5월 31일까지 약 5개월 간 부산시, 대구시, 구미시 및 안동시 등에 소재하고 있는 1개 병원에서 물리치료를 시작한 뇌졸중환자 87명을 대상으로 물리치료를 시작한 시점, 1개월 후, 퇴원 시, 그리고 퇴원 후 1개월이 지난 시점까지 추적하여 기능변화를 평가하였다. 뇌졸중환자의 물리치료 시작시점의 BI점수는 26.32였고, PS점수는 11.34이었다. 물리치료 시작시점을 기준으로 할 때 입원 1개월 후의 81점수 변화는 평균 16.61(p<0.001). 퇴원 시는 33.51(p<0.001), 퇴원 1개월 후는 43.56 만큼 호전되었다(p<0.001). PS점수도 각각 -2.1, -3.94, 그리고 -5,52만큼 호전되었다(p<0.001). 그리고 퇴원시에 비해 퇴원 후의 변화도 81점수는 10.06만큼, PS점수는 -1.51만큼 모두 유의하게 호전되었다 (p<0.001). 물리치료 시작시점과 회원시의 BI점수 변화와 유의한 관련이 있는 요인은 물리치료 시작시점의 BI점수와 연령이었는데(p<0.05), 시작시점의 BI점수가 낮고 연령이 낮을수록 기능호전이 컸다. 시작시점과 종료시점 사이의 변화는 시작시점의 BI점수. 퇴원시의 BI점수, 종교유무가 유의하게 관련이 이었다(p<0.05). 즉, 종교를 믿고 있거나 물리치료 시작시점의 BI점수가 낮고 퇴원시의 BI 점수가 높을수록 기능호전이 컸다. 퇴원 시와 1개월 후에는 퇴원시의 BI점수와 종교유무였는데, 퇴원시의 BI점수가 낮고 종교를 믿는 환자에서 기능호전이 유의하게 컸다.(p<0.05). 물리치료 시작시점과 퇴원시의 PS점수 변화와 유의한 관련이 있는 요인은 물리치료 시작시점의 PS점수였는데(p<0.05), 시작시점의 PS점수가 높을수록 기능호전이 컸다. 시작시점과 종료시점사이는 시작시점의 PS점수, 퇴원시의 PS점수, 퇴원 1개월 후에 적극적으로 물리치료를 받았는지의 여부 등이었다(p<0.05). 즉, 시작시점의 PS점수가 높고, 퇴원시의 PS점수는 낮으면서 적극적으로 물리치료를 받은 환자가 기능호전이 켰다. 퇴원 시와 1개월 후에는 퇴원시의 PS접수가 높고 적극적으로 물리치료를 받을수록 기능호전이 유의하게 컸다(p<0.05). 이상의 결과를 요약하면, 뇌졸중환자의 물리치료 효과를 BI점수로 평가하면 물리치료 시작시점 및 퇴원시의 BI점수, 연령 및 종교유무가 중요한 요인이고, PS점수로 평가하면 시작시점 및 퇴원시의 PS점수와 퇴원 1개월 후에 적극적으로 물리치료를 받았는지의 여부가 중요한 요인으로 생각된다.

Panax Saponin C의 부분화학 구조 (Partial Structure of Panax Saponin C)

  • 전보에서 저자들은 인삼의 anti-inflammatory activity를 추적하여 Panax saponin A 및 C 로 명명된 dammalene계 glycoside를 분리하였고 A에 대해서는 이미 그 화학구조를 밝혀 보고한바 있다. 본보에서는 Panax saponin C(PS-C)의 부분 화학구조를 밝혀 보고코자 한다. PS-C는 산분해하면 panaxatriol 1 mol, glucose 2 mol 및 rhamnose 1 mol을 생성하고, acetylation하면 dodeca-acetate를 형성한다. 따라서 protopanaxtriol의 20[s]-수산기는 glycoside 결합에 참여하고있다. PS-C는 6 mol의 $HIO_4$를 소모하고 permethylate에 대한 methanolysis product를 GLC로 분석한 결과 2, 3, 4-trimethoxy-methyl-rhamnoside 1 mol과 $2,\;3,\;4,\;6-tetramethoxy-{\alpha}-methyl\;glucosied\;2\;mol$이 생성되므로 PS-C 중에 존재하는 3 mol의 sugar는 oligoside 결합에 의하지 않고 monoside결합에 의하여 연결되어 있고 glucose는 ${\beta}-glycoside$ 결합을 하고 있음을 의미한다.

  • Familial form of Alzheimer's disease (FAD) is caused by mutations in presenilin-1 and presenilin-2 (PS2). PS1 and PS2 mutation are known to similar effects on the production of amyloid $\beta$ peptide (A$\beta$) and cause of cell death in the Alzheimer's brain. The importance of the alternation of calcium homeostasis in the neuronal cell death by PS1 mutation in a variety of experimental system has been demonstrated. (omitted)

  • 본 논문에서는 6~18 GHz 대역 8-비트 true time delay(TTD) 회로의 설계 및 측정결과에 대하여 기술하였다. 단위 지연 회로는 상대적으로 시간 지연 변화율이 일정한 m-유도 필터(m-derived filter)를 이용하였다. 설계한 8-비트 TTD는 2개의 single-pole double-throw(SPDT)와 7개의 double-pole double-throw(DPDT) 스위치로 구현하였으며, 인덕터를 이용하여 반사 특성을 개선하였다. 설계된 8-비트 TTD는 $0.18{\mu}m$ CMOS 공정을 이용하여 제작하였다. 측정된 TTD 회로의 시간 가변 범위는 250 ps이고, 시간 지연 해상도는 약 1 ps이다. 6~18 GHz의 동작 주파수에서 RMS 시간 지연 오차는 11 ps 미만이며, 입출력 반사 손실은 10 dB 이상이다. 공급 전압은 1.8 V이며, 소비 전력은 0.0 mW이다. 칩 면적은 $2.36{\times}1.04mm^2$이다.

Beijerinckia Indica 배양을 통한 Heteropolysaccharide-7 생산 최적화 (Optimization of Heteropolysaccharide-7 Production by Beijerinckia Indica)

  • Beijerinckia indica를 통한 Heteropolysaccharide-7(PS-7)생산 최적화를 위해 다양한 탄소원과 질소원을 첨가하여 다른 배지 조성들이 PS-7에 미치는 영향을 조사하였다. 높은 C/N ratio에서 PS-7 생산을 배양 40시간까지 효과적으로 증가시킬 수 있었으며 낮은 C/N ratio에선 탄소원으로 첨가한 glucose가 균체의 생산에 사용되었을뿐 PS-7의 생산은 낮았다. 하지만, 높은 C/N ratio에서는 배양액의 점도로 균체의 생산이 정체하는 것으로 보아 본 실험에서는 20g/l glucose와 $NH_{4}NO_3$ 7.5 mM으로 33.3의 C/N ratio가 PS-7 생산에 적절한 비율인 것으로 관찰되었다. 다양한 탄소원을 첨가한 결과, B. indica는 PS-7 생산을 위하여 탄소원으로 hexose 또는 glucose-generating 당을 효율적으로 이용할 수 있었다. Beijerinckia indica를 이용한 PS-7 생산을 위해 첨가한 다양한 탄소원 중 sucrose 농도가 최고의 PS-7(6.96 g/L)을 생산 하였다. 51 배양기를 이용한 배양에서 탄소원 20 g/L glucose, 질소원 7.5 mM $NH_{4}NO_3$이 포함된 MSM 배지내 배양된 B. indica의 세포 농도가 최고 2.5 g/L이였고, 생산된 최고 PS-7농도는 7.5 g/L(35174 cp)으로 나타났다. 질소원이 완전히 소모된 12시간 이후, 7.5 mM의 $NH_{4}NO_3$, glutamine, glutamate을 첨가한 MSM 배지에서의 B. indica 배앙 결과, 질소원이 B. indica 내에서 탄소원 대사를 조절하는 역할을 하는 것을 관찰할 수 있었으며 최종적으로 PS-7의 생산을 조절할 수 있었다.

인산가용화 사상균 Penicillium sp. PS-113의 고체배양 (Solid Culture of Phosphate-solubilizing Fungus, Penicillium sp. PS-113)

  • A fungus, Penicillium sp. PS-113, isolated from soil showed the high phosphate-solubilizing activity in patato dextrose broth-rock phosphate to produce free phosphates to the culture broth with the concentrations of 585 ppm against rock phosphate. In this medium, the optimum temperature and initial pH to solubilize rock phosphate were 30$^{\circ}C$ and pH 7.5, respectively. In order to make the mass production of the conidia from this fungus, we cultured in on various solid-based media like barley, corn, wheat, rice, rice bran, and compost. As a result, the fungus highly produced conidia ranging from 2.1 to $5.1{\times}10_9$ conidia/g${\cdot}$media on these solid media except compost-based medium, which was 0 times less than others. Effects of inoculation of the phosphate solubilizing fungus as a biofertilizer were studied in perlite-based pot cropped with Zea mays Suwon 19. Inoculation of Penicillium sp. PS-113 increased in plant height (1.4 times), plant weight (5.2~8.1 times) and root length (1.1~1.2 times) at 60-day cultivation, compared to Hogland solution either without $NH_4H_2PO_4$ or displace $NH_4H_2PO_4$ to powdered rock phosphate, a phosphorus source for plant growth.

Effect of Polygonati Sibirici Rhizoma on Cell Viability in Human Glioma Cells

  • Objectives : Although herbal medicines containing flavonoids have been reported to exert anti-tumor activities, it has not been explored whether Hwang-Jeong (Polygonati sibirici Rhizoma, PsR) exerts anti-tumor activity in human glioma. The present study was therefore undertaken to examine the effect of PsR on cell viability and to determine its underlying mechanism in A172 human glioma cells. Methods : Cell viability was estimated by MTT assay. Reactive oxygen species generation and mitochondrial membrane potential were measured by the fluorescence dyes. The phosphorylation of kinases was evaluated by western blot analysis and caspase activity was estimated using colorimetric assay kit. Results : PsR resulted in loss of cell viability in a dose- and time-dependent manner. PsR did not increase reactive oxygen species (ROS) generation and the PsR-induced cell death was also not affected by antioxidants, suggesting that ROS generation is not involved in loss of cell viability. Western blot analysis showed that PsR treatment caused rapid reduction in phosphorylation of extracellular signal-regulated kinase (ERK) without changes in p38 and Jun-NH2-terminal kinase (JNK). U0126, an inhibitor of ERK, increased the PsR-induced cell death, but inhibitors of p38 and JNK did not affect the cell death. PsR induced depolarization of mitochondrial membrane potential. Caspase activity was not stimulated by PsR and caspase inhibitors did not prevent the PsR-induced cell death. Conclusion : Taken together, these findings suggest that PsR results in human glioma cell death through caspaseindependent mechanisms involving down-regulation of ERK.

Effects of Neutral Detergent Fiber Concentration and Particle Size of the Diet on Chewing Activities of Dairy Cows

  • Six dry Holstein cows were used to evaluate the effect of dietary neutral detergent fiber (NDF) concentration and particle size (PS) on chewing activity. Treatments were arranged in a 3$\times$3 factorial design; total mixed rations contained three NDF concentrations (26, 32, 38%) and three PS (1.0, 1.5, 2.0 cm). NDF levels and particle sizes of diets were adjusted by formulating rate and cutting length of alfalfa hay and rice straw. Cows were fed twice daily at 90% of ad libitum feed intake throughout the experiment. Chewing activity was positively associated with NDF concentration, but not significantly affected by PS of diet. Eating time per unit of NDF intake was affected by PS rather than NDF concentration of diet. Time spent ruminating per unit DM or NDF intake increased with increasing NDF concentration of diet, but was not affected by PS. As the PS of diet increased, the eating time per day increased, but the rumination time decreased. In addition, as the number of rumination bolues decreased the rumination duration increased as well as the chews per bolus. The regression equation induced from relationships of NDF concentrations (NDF, %) and particle sizes (PS, cm) of diet on roughage value index (RVI, min of chewing time/kg DMI) was as follows. RVI=-19.672+1.44$\times$NDF+5.196$\times$PS, ($R^{2}$=0.81).

Effect of Lead(IV) Acetate on Procoagulant Activity in Human Red Blood Cells

  • Lead (Pb) is a ubiquitously occurring environmental heavy metal which is widely used in industry and human life. Possibly due to a global industrial expansion, recent studies have revealed the prevalent human exposure to Pb and increased risk of Pb toxicity. Once ingested by human, 95% of absorbed Pb is accumulated into erythrocytes and erythrocytes are known to be a prime target for Pb toxicity. Most of the studies were however, focused on $Pb^{2+}$ whereas the effects of $Pb^{4+}$, another major form of Pb on erythrocytes are poorly understood yet. In this study, we investigated and compared the effects of $Pb^{4+}$, $Pb^{2+}$ and other heavy metals on procoagulant activation of erythrocytes, an important factor for the participation of erythrocytes in thrombotic events in an effort to address the cardiovascular toxicity of $Pb^{4+}$. Freshly isolated erythrocytes from human were incubated with $Pb^{4+}$, $Pb^{2+}$, $Cd^{2+}$ and $Ag^+$ and the exposure of phosphatidylserine (PS), key marker for procoagulant activation was measured using flow cytometry. As a result, while $Cd^{2+}$ and $Ag^+$ did not affect PS exposure, $Pb^{4+}$ and $Pb^{2+}$ induced significantly PS exposure in a dose-dependent manner. Of a particular note, $Pb^{4+}$ induced PS exposure with a similar potency with $Pb^{2+}$. PS bearing microvesicle (MV), another important contributor to procoagulant activation was also generated by $Pb^{4+}$. These PS exposure and MV generation by $Pb^{4+}$ were well in line with the shape change of erythrocyte from normal discocytes to MV shedding echinocytes following $Pb^{4+}$ treatment. Meanwhile, nonspecific hemolysis was not observed suggesting the specificity of $Pb^{4+}$-induced PS exposure and MV generation. These results indicated that $Pb^{4+}$ could induce procoagulant activation of erythrocytes through PS exposure and MV generation, suggesting that $Pb^{4+}$ exposure might ultimately lead to increased thrombotic events.
