Effects of Dietary Supplementation of Castor Aralia (Kalopanax pictus Nakai) on Performance and Fatty Acid Composition of Chicken Meat (사료 내 엄나무 첨가 급여가 육계의 생산성 및 계육의 지방산 조성에 미치는 영향)

  • The effects of dietary supplementation with Kalopanax pictus branch, leaves, and extracts on performance, proximate composition, pH, total phenol and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, cholesterol and fatty acid composition of chicken meat. Broiler chicks were fed for 5 weeks with a diet containing 5% Kalopanax pictus branch (T1), 2% Kalopanax pictus leaves (T2), and 5% Kalopanax pictus extracts (60% leaves + 40% branch) (T3). T1 and T3 resulted in much better feed conversion than T2. In proximate composition, crude fat of chicken meat was increased, but not signi-ficantly (p>0.05) in Kalopanax pictus-supplemented diets than control diet. Total phenol contents and DPPH radical scavenging activity were significantly increased (p<0.05) by the dietary-supplementation with Kalopanax pictus branch and leaves compared to the control diet. Total and high density lipoprotein cholesterols of treatment groups were higher than the control group, and control group low density lipoprotein cholesterolwas higher than treatment groups (p<0.05). Especially, T3 was significantly (p<0.05) more effective in improving cholesterol compared to other treatment groups. The treatment groups (especially T3) showed significantly higher total fatty acid and unsaturated fatty acid compositions compare to the control group.

Quantitative Determination on the Constituents of the Stem Bark and the Leaf Shoot of Kalopanax pictus by HPLC Analysis (HPLC 분석에 의한 해동피와 개두릅의 성분함량 비교)

  • To evaluate the leaf shoots and stem barks of Kalopanax pictus (Araliaceae) as an edible vegetable and a traditional medicinal drug, respectively, the compounds of syringin, liriodendrin, astragalin, quercetin, and kalopanaxsaponins were quantitatively measured by HPLC analysis. The leaf shoot exhibited low contents of syringin, liriodendrin and kalopanaxsaponins but a high chlorogenic acid content, whereas the grown leaves contained very high amounts of kalopananxsaponins. In contrast, the stem bark had very high amounts of syringin and liriodendrin and relatively low kalopanaxsaponins. In particular, the kalopanaxsaponin contents were rapidly increased with monthly variation until October but decreased from September. It was also observed that the leaf shoot contained chlorogenic acid by 30.73 mg/g and the barks showed the concentration of liriodendrin by 20.75 mg/g. These results indicate that high contents of syringin and liriodendrin in the stem bark and high contents of chlorogenic acid in the leaf shoot support scientific bases on the traditional uses of K. pictus as a medicinal drug and a functional food, respectively.

toxicology of Kalopanax pictus Extract and Hematological Effect of the Isolated Anti-Rheumatoidal Kalopanaxsaponin A on the Freunds Complete Adjuvant Reagent-Treated Rat

  • We have reported that kalopanaxsaponin A (KPS-A) Isolated from Kalopanax pictus have anti-rheumatoidal activity in the rat treated with Freunds complete adjuvant (FCA) reagent. In addition, it has been also reported that KPS-A is a potent antioxidant in the rheumatoidal rat. This research was undertaken to examine whether the saponins of KPS-A and -1 could adjust the abnormal lipid metabolisms and hematological changes in immunological diseases. KPS-A significantly inhibited the increases in both triglycerides and total proteins in addition to the decrease in total cholesterol induced by FCA reagent treatment. KPS-A treatment decreased the number of leucocytes elevated by FCA reagent treatment. Excess dose of the methanol extract produced no severe toxicity on the body weight, wet organ weights and hepatic functions. Since $LD_50$ value of K. pictus methanol extract was shown to be 4,033 ${mg/kg}$, it could be estimated to be a safe agent for anti-rheumatoidal herbal medicines.

Quality Characteristics of $Kalopanax$ $pictus$ and $Aralica$ $elata$ Shoot according to Their Salt Conditions (염장조건에 따른 엄나무와 두릅 순의 품질특성)

  • The quality characteristics of $kalopanax$ $pictus$ and Aralica elata shoots during their preservation were investigated according to the salt conditions and storage temperatures to improve the use of the shoots. The results were as follows: the pH of $kalopanax$ $pictus$ shoots duing their storage did not changes with 10% and 12% brine at $4^{\circ}C$ and with 12% brine at $10^{\circ}C$. The pH of $Aralica$ $elata$ shoots is at $4^{\circ}C$ tended to be similar to the $kalopanax$ $pictus$ shoot and decreased at the storage temperature of $10^{\circ}C$. The salinity of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to increase during their storage and did not change significantly as their storage temperature changed. The L values of $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decrease gradually during their storage, and the salt concentrations appeared high. The b value of the $kalopanax$ $pictus$ shoot tended to increase and that of the $Aralica$ $elata$ shoots, to decrease gradually. The a value tended to increase gradually but did not differ with variations in the storage temperatures and salt concentrations. The strength and the hardness of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decreased gradually during their storage and highest in 10% brine at $4^{\circ}C$, and those of $Aralica$ $elata$ shoots were highest in 8-10% brine at $4^{\circ}C$. The Chlorophyll contents of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decrease during their salt storage and did not change significantly as the temperatures and salt concentrations changed. Therefore, it can be concluded that the quality of $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots can be maintained when the are stored in 10% brine at $4^{\circ}C$.

  • In this study, we assessed the antioxidant activity and quality characteristics of cookies containing various concentrations (0, 1, 3 and 5%) of Kalopanax pictus leaf powder. Antioxidant activity was estimated by DPPH free radical scavenging activity, reducing power, and total phenolic compound content in Kalopanax pictus leaf powder and cookies. The quality characteristics of Kalopanax pictus leaf cookies were estimated as spread factor, loss rate, leavening rate, color, texture and sensory evaluations. The moisture contents, spread factor, total phenolic acid content, DPPH free radical scavenging activity and reducing power of cookies significantly increased with increasing Kalopanax pictus leaf powder content, whereas the pH of the dough, hardness, and color values of cookies significantly decreased with increasing Kalopanax pictus leaf powder content. In the sensory evaluation, the scores for color, flavor and crispiness were high in cookies with 3% Kalopanax pictus leaf powder.

  • Morphological characteristics of Kalopanox pictus Nakai were studied to examine population differentiation of this species. Based on a phenogram of using 23 morphological characteristics, differentiation of regions were distinct. Collections of 138 specimens from nine populations served as operational taxonomic unit (OTU's) were examined for phenotypic similarity and morphological variation using clustering (Ward's minimum variance method) and principal component analysis (PCA). The first three principal components were responsible for 77.0% of the total variance. Principal component 1 explained 52% of the total variance and was contributed to by the number of palmately parted, the number of pinnately lobed, and width between two lateral lobe apex.

Taxonomic Study of Poorly-known Marine Pleurostomatid Ciliates of Litonotus paracygnus and L. pictus (Ciliophora: Pleurostomatida) from Korea

  • Two poorly known and often confused pleurostomatid ciliates, Litonotus paracygnus Song, 1994 and L. pictus Gruber, 1884, were collected from the coastal waters of Yeonggeumjeong and Bongpo-port, Gangwondo in the East Sea and from the Iwon tide embankment near Ganwol-do, Chungcheongnam-do in the Yellow Sea, Korea. These species were described based on live observations, the protargol-impregnation and morphometrics of the species. Also provided are their diagnoses. The small subunit ribosomal DNA (SSU rDNA) sequences of these species were compared with previously known sequences of related species. The diagnostics of the two Litonotus species are as follows. L. paracygnus: 150-300 $\mu$m long in vivo, strongly contractile neck region, two ellipsoid macronuclei (Ma) and one micronucleus (Mi), 7 left (LSK) and 11-14 right somatic kineties (RSK), 2-4 contractile vacuoles (CV) located on the posterior end, extrusemes (Ex) distributed on the anterior region of the ventral margin only. L. pictus: about 200-600 $\mu$m long in vivo, extremely contractile, beautiful body color with rows of yellow to yellow-brownish cortical pigment granules, 12-21 Ma arranged in moniliform pattern, infrequently vermiform, 7-11 LSK and 18-26 RSK, several CV located on both margins, Ex distributed on the anterior region of the ventral margin only. In this study, this genus was firstly recorded in Korea.

  • Water relations of Kalopanax pictus leaves obtained from the P-V curve. In the upper leaves of Kalopanax pictus seedlings, the original osmotic pressure at maximum turgor was -1.44 MPa, and the osmotic pressure at incipient plasmolysis point was -1.84 MPa, and the relative water content at incipient plasmolysis point was 78.2%.

  • Several plant growth regulators were examined for on their effect on the in vitro propagation of Kalopanax pictus$(T_{HUNB}.)$ $(N_{AKAI}.)$ on WPM medium. Among the cytokinins tested, BA at $13.32\;{\mu}M$ appeared to be the most effective for multiple axillary shoot formation. Although the addition of $2.89\;{\mu}M\;GA_3$ promoted stem elongation, it produced morphologically abnormal leaves and stems. For rooting of the shoots, $4.9\;{\mu}M$ IBA seemed to be more effective than $2.69\;{\mu}M$ NAA. When the regenerated plants were transferred on artificial mixture containing vermiculite and peat-moss (1 : 1, v/v), 81% of them survived and grew normally.

Antioxidant and Cytoprotective Activity of Castor-aralia (Kalopanax pictus) Leaves

  • The present study was to investigate the in vitro antioxidant potential of hot water extract and its fractions from dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), n-butanol (n-BuOH), and water ($H_2O$) of castor-aralia (Kalopanax pictus) leaves using different antioxidant tests. Among these crude extract and fractions, EtOAc fraction exhibited higher antioxidant potency than others in 1,1-diphenyl-2-pricylhydrazyl (DPPH) free radical scavenging, reducing power assay, and reactive oxygen species (ROS) scavenging activity. However, $CH_2Cl_2$ fraction showed higher hydroxyl radical scavenging and DNA damage protective activity. This work demonstrates the potential of castor-aralia leaves as antioxidant functional food ingredients.
