N-Acetyl-D-Glucosamine Kinase Interacts with Dynein-Lis1-NudE1 Complex and Regulates Cell Division

  • N-acetyl-D-glucosamine kinase (GlcNAc kinase or NAGK) primarily catalyzes phosphoryl transfer to GlcNAc during amino sugar metabolism. Recently, it was shown NAGK interacts with dynein light chain roadblock type 1 (DYNLRB1) and upregulates axo-dendritic growth, which is an enzyme activity-independent, non-canonical structural role. The authors examined the distributions of NAGK and NAGK-dynein complexes during the cell cycle in HEK293T cells. NAGK was expressed throughout different stages of cell division and immunocytochemistry (ICC) showed NAGK was localized at nuclear envelope, spindle microtubules (MTs), and kinetochores (KTs). A proximity ligation assay (PLA) for NAGK and DYNLRB1 revealed NAGK-dynein complex on nuclear envelopes in prophase cells and on chromosomes in metaphase cells. NAGK-DYNLRB1 PLA followed by Lis1/NudE1 immunostaining showed NAGK-dynein complexes were colocalized with Lis1 and NudE1 signals, and PLA for NAGK-Lis1 showed similar signal patterns, suggesting a functional link between NAGK and dynein-Lis1 complex. Subsequently, NAGK-dynein complexes were found in KTs and on nuclear membranes where KTs were marked with CENP-B ICC and nuclear membrane with lamin ICC. Furthermore, knockdown of NAGK by small hairpin (sh) RNA was found to delay cell division. These results indicate that the NAGK-dynein interaction with the involvements of Lis1 and NudE1 plays an important role in prophase nuclear envelope breakdown (NEB) and metaphase MT-KT attachment during eukaryotic cell division.

The effects of temperature on the growth rate and nitrogen content of invasive Gracilaria vermiculophylla and native Gracilaria tikvahiae from Long Island Sound, USA

  • The red alga Gracilaria vermiculophylla, a species native to the waters of Korea and Japan, has invaded marine coastal areas of Europe and the Americas, thriving in conditions that differ from those of its native habitat. In recent years, G. vermiculophylla has been discovered in the Long Island Sound (LIS) estuary growing alongside the native congener Gracilaria tikvahiae. The goal of this study was to determine whether the two strains of G. vermiculophylla from different regions of the world have evolved genetic differences (i.e., ecotypic differentiation) or if the physiological performance of the strains simply reflects phenotypic plasticity. Two strains of G. vermiculophylla (isolated in Korea and LIS) and a strain of the LIS native G. tikvahiae were grown for four weeks under temperatures ranging from 20 to $34^{\circ}C$ using a temperature gradient table (all other environmental conditions were kept constant). At the end of each week, wet weight of each sample was recorded, and thalli were reduced to the original stocking density of $1gL^{-1}$ (excess biomass was preserved for tissue carbon and nitrogen analysis). Generally, the growth rates of Korean G. vermiculophylla > LIS G. vermiculophylla > G. tikvahiae. After one week of growth G. tikvahiae grew 9.1, 12.0, 9.4, and 0.2% $d^{-1}$, at temperatures of 20, 24, 29, and $34^{\circ}C$, respectively, while G. vermiculophylla (LIS) grew 6.6, 6.2, 5.7, and 3.6% $d^{-1}$. G. vermiculophylla (Korea) grew 15.4, 22.9, 23.2, and 10.1% $d^{-1}$, much higher than the two strains currently inhabiting the LIS. On average, the LIS G. vermiculophylla strain contained 4-5% DW N, while the Korean strain and G. tikvahiae had more modest levels of 2-3% N DW. However, tissue N content declined as temperature increased in LIS and Korean G. vermiculophylla. The non-native haplotype may have evolved genetic differences resulting in lower growth capacity while concentrating significantly more nitrogen, giving the non-native a competitive advantage.

  • 최근 기상 이변으로 인해 홍수, 가뭄 등과 같은 수재해가 빈번히 발생되고 있다. 이에 수재해예방 등의 안전 기술과 관련된 관심이 증가되고 있다. 수재해 예방을 위해서는 먼저 홍수, 가뭄 등과 같은 수재해 감시가 필요하며, 이를 위해 위성, 레이더 등으로 관측된 자료를 활용한 수문인자 정보는 매우 중요하다. 미국 NASA의 LIS(Land Information System)는 위성 및 지상관측 자료를 활용하여 홍수, 가뭄, 기상, 산사태, 농업 등의 정보를 생산할 수 있는 프레임워크이다. LIS는 크게 지표면 모델 및 자료동화를 위한 변수들의 전처리 과정(LDT), 지표면 모델을 활용한 분석 및 자료동화 과정(LDAS) 및 분석된 자료의 검보정(LVT) 과정으로 구성되어 있다. LIS에서 산출 가능한 인자는 Energy Balance, Water Balance, Surface/Subsurface State, Evaporation, Hydrologic, Cold Season Processes, Compared Data, Carbon 등 9개로 분류되며 약 78개의 인자를 산출한다. 홍수, 가뭄 등과 같은 수재해 감시를 위한 수문인자는 강수량, 증발산량, 토양수분, 지표면 온도 등을 비롯한 여러 가지 인자들이 필요하다. LIS는 주로 미국, 캐나다 등 평활한 지역에 활용되어 공간해상도는 약 10km(0.1deg) 이하로 자료를 산출한다. 산악 지형이 대부분인 한국 지형에 적용하기에는 자료의 정확성이 낮아 10km 이상의 공간해상도 자료가 필요하며, 한국형 수재해정보 플랫폼에서 홍수, 가뭄 등의 기초자료로 사용하기 위한 수문인자의 선정이 필요하다. 이에 본 연구에서 NASA LIS를 통해 산출 가능한 인자를 정리하고 한국형 수재해 정보플랫폼에 활용 가능한 수문인자의 항목을 조사하였다. 홍수, 가뭄 등 수재해 분석에 필요한 기초자료는 강우량, 유출량, 잠재적 증발산량, 식생의 증산량, 토양수분, 표면온도, 알베도 등의 수문인자이며, NASA LIS에서 이와 같은 수문인자 산출이 가능하다. NASA와 국제공동 연구중인 한국형 물순환분석 프레임워크(K-LIS(안)) 개발을 통해 한국 지형에 적합한 홍수 및 가뭄 등의 수재해 감시 평가 예측이 가능할 것이며, K-LIS에서 산출되는 고해상도의 수문인자들을 수재해 정보 웹 포털의 정보 제공 서비스를 통하여 손쉽게 접근 가능할 것으로 사료된다.

The 2D Measurement of Soot Diameter and Number Density in a Diesel Engine Using Laser Induced Methods

  • It is necessary to diagnose accurately the characteristics of soot formation and oxidation in a diesel engine. Whereas past measurement techniques for soot concentration give limited information for soot, laser-based two-dimensional imaging diagnostics have a potential to provide temporally and spatially superior resolved measurements of the soot distribution. The technique using laser sheet beam has been applied to an optically accessible diesel engine for the quantitative measurement of soot. The results provided the information for reduction of soot from the diesel engine. Both LIS (Laser Induced Scattering) and LII (Laser Induced Incandescence) techniques were used simultaneously in this study. The images of LIS and LII showed the quantitative distribution of the soot concentration in the diesel engine. In this study, several results were obtained by the simultaneous measurements of LIS and LII technique. The diameter and number density of soot in combustion chamber of the test engine were obtained from ATDC 20 degree to 110 degree. The soot diameter increased about 37% between ATDC 20 degree and 110 degree. The number density of soot, however, decreased significantly between ATDC 40 degree and 70 degree.

  • The new millennium with mystifying convergence of IT has opened new challenges to LIS profession. This has resulted to continuous skill development and lifelong learning, for capacity building in consonant with contemporary changes. The LIS profession in India has created several mechanisms to organize and conduct several skills development programmes through which professionals are continuously enabled to update their professional knowledge and skills, in practice and teaching. There is a good organizational infrastructure and identified areas related to the recent developmental trends and as such LIS professionals and educators have facilities for updating their knowledge at veritable levels and intervals. The paper has surveyed programmes conducted by different organizations at different levels and highlights the aspects of the organizational infrastructure and the emergent areas of applications in LIS teaching, research and professional practices in India. The selection of areas and subjects for various skill development programmes suggest the competencies acquired by the LIS professionals from India, and the empowerment of their professional knowledge can be of immense value to its neighbouring countries. The paper highlights the knowledge and competencies of the LIS professionals in India and the broad-based facilities available here which can be a model for those countries which are seeking to utilize the LIS human resources from this part of the sub-continent. The paper in this circumstance surveyed on the institutional infrastructure for skill developmental opportunities available in India which would meet such requirements within and to the countries outside in the South Asian region seeking its collaboration in such manpower development work.

A Study on Behavioral Traits of Library and Information Science Students in South India

  • Human behaviour normally depends on the environment of the incident and the time of its occurrence. The behaviour of people depends on many factors and these behaviour traits are an important aspect in the Library and Information Science (LIS) field. Hence in this paper an attempt has been made to examine the behaviour traits of LIS students in South India. Out of 400 questionnaires distributed 367 have responded and the response rate is 91.75%. In this survey three aspects comprising student behaviour have been analysed such as Work Environment, Natural Environment, and Social Environment. In the case of Work Environment the respondents were grouped as Workaholic, Impatience, Achievement oriented, Rash nature, and Punctuality. Further, in respect to Natural environment, the respondents are grouped as Complacent, Patience, Easygoing, and Relaxed. Last, the respondents were grouped in the Social Environment as Balancing nature, Magnanimity, Naturalistic, Assertive nature, Dependency, Lucrative, Lonely nature, and Time Based personality. Finally the authors conclude that LIS students need to possess these qualities and behaviours to work in different environments.

Research Publishing by Library and Information Science Scholars in Pakistan: A Bibliometric Analysis

  • Scholarly communication plays a significant role in the development and dissemination of research outputs in library and information science (LIS). This study presents findings from a survey which examines the key attributes that characterize the publishing by Pakistani LIS scholars, i.e. academics and professionals, in national journals. A pilot-tested, electronic questionnaire was used to collect the data from the target population. 104 respondents (or 69.3% of target) provided feedback on areas such as number of articles published, number of citations, and the nature of any collaboration with other authors. The findings of this survey revealed that, among the various designated regions of Pakistan, the Punjab region was the most highly represented. In articles published in national journals, there was a clear preference among all respondents to collaborate with at least one other author. The citation metrics for LIS articles in national journals were relatively low (30.22%), which aligns with Scimago’s Journal and Country Rankings. The uptake of social scholarly networks mirrors international trends. Respondents were asked to score factors which could impact negatively on their ability to undertake research and/or publish the results. The study recommends that concerned stakeholders work together, as appropriate, to address concerns. In addition, it recommends that further research be undertaken to define patterns of Pakistani co-authorship in the social sciences.

DDC in DSpace: Integration of Multi-lingual Subject Access System in Institutional Digital Repositories

  • The paper discusses the nature of Knowledge Organization Systems (KOSs) and shows how these can support digital library users. It demonstrates processes related to integration of KOS like the Dewey Decimal Classification, $22^{nd}$ edition (DDC22) in DSpace software (http://www.dspace.org/) for organizing and retrieving (browsing and searching) scholarly objects. An attempt has been made to use the DDC22 available in Bengali language and highlights the required mechanisms for system-level integration. It may help a repository administrator to build an IDR (Institutional Digital Repository) integrated with SKOS-enabled multilingual subject access systems for supporting subject descriptors based indexing (DC.Subject metadata element), structured navigation (browsing) and efficient searching.

  • Local languages based information storage and retrieval system is essential for any online digital repository system. This paper reports the development of an interface in Bengali that allows users not only browsing and searching Indic-script based documents but also allows administrator performing various system level operations. This paper briefly describes the origin and key characteristics of Indic-scripts along with their encoding in Unicode standard with special reference to Bengali language. It also demonstrates the development processes of Bengal-script based information representation and retrieval (IRR) system viz. BURA (Burdwan University Research Archive) using different open standard and open source software (OSS) including different factors essential for building such successful Indic-script based multilingual digital libraries. The suggested strategies may help digital library developers to design an appropriate multi-script based information access services in any other Indic-script based languages.
