Transformation of 'Ilmibyeo' using pCAMBIA 1300 and Microstructural Investigation of Leaves (pCAMBIA 1300 벡터를 이용한 일미벼의 형질전환 및 잎의 전자현미경적 관찰)

  • The argE gene of E.coli was introduced into #Ilmibyeo# cultivar of rice by Agrobacterium tumefaciens and a large number of transgenic plants were produced. Embryogenic calli were co-cultivated with A. tumefaciens strain AGL1 carrying the plasmid pCAMBIA1300 containing hygromycin resistance(HygR). Transgenic plants showing in vitro resistance to 50mg/L hygromycin were obtained using a selection procedure. Stable integration of argE and HPT genes into chromosomal DNA was proven by southern blot analysis and PCR analysis of genomic isolated from $T_0$ progenies. The fragments of 650 bp(HPT) were detected in transgenic rice lines. The 230 bp(argE) fragments were showed in agarose gel, and detected fragments were matched with size of argE specific primer. The microscopic feature of leaf on scanning electron microscope(SEM) revealed differences between clear and chalky in shape and arrangement of stoma but did not discriminate.

Development of transgenic disease-resistant root stock for the growth of watermelon

  • To protect the watermelon against soil-borne pathogens, we are currently producing disease-resistant transgenic root stock for the growth of watermelon, A defensin gene (J1-1) from Capsicum annum, a ACC deaminase gene from Pseudomonas syringae, a galactinol synthase (CsGolS) gene from Cucumis sativus, and a WRKY (CvWRKY2) gene from Citullus vulgaris were used as transgenes for disease resistance. The gene were transformed into a inbred line (6-2-2) of watermelon, Kong-dae watermelon and a inbred line (GO702S) of gourd, respectively, by Agrobacterium-mediated transformation. Putative transgenic plants were selected in medium containing 100mg/L kanamycin, and then integration of the genes into the genomic DNA were demonstrated by PCR analysis. Successful integration of the gene in regenerated plants was also confirmed by PCR (Figf 1), genomic Southern blot (Fig 2), RT-PCR (Fig 3), and Northern blot analysis(Fig 4). Several T1 lines having different transgene were produced, and disease resistance of the T1 lines are under estimation.

  • In pancreatic cells, NO formation is associated with increased levels of cGMP and endocrine/exocrine secretion. In the present study, the role of NO in the regulation of exocrine secretion was investigated in rat pancreatic tissues. Treatment of rat pancreatic tissue with sholecystokinin-pancreozymin (CCK-PZ) resulted in an significant increase in arginine conversion to citruline, the amount of nitrite/nitrate, the release of amylase, and the level of cGMP. Furthermore, CCK-PZ stimulated increase of amylase release and conversion of arginine to citrulline transformation were counteracted by the inhibitor of NO synthase, $N^G-nitro-L-arginine$ methyl ester. The results on the time course of CCK-PZ-induced citrulline formation within the first seconds of simulation. The kinetics of citrulline accumulation correlate well with those of cGMP rise, which further confirms the conclusion that NO mediates the response to CCK-PZ by cGMP.

  • Common aquaculture practices include the use of certain pharmaceuticals such as antibiotics in avoiding diseases and promoting a healthier growth of the culture. The aim of this study is to monitor and assess the influence of different low oxytetracycline concentrations on the transformation of nitrogen compounds under aeration condition in a lab-scale recirculating aquaculture system (RAS). Over $1mg\;L^{-1}$ dose of oxytetracycline to aquaculture had induced ammonia($NH_4-N$), nitrate($NO_3-N$), soluble COD accumulation in RAS. In addition, nitrous oxide ($N_2O$) emission from RAS was significantly reduced during the oxytetracycline dose periods. After ceasing the dose of oxytetracycline, ammonia oxidation and nitrous oxide re-emission were observed. This observation indicated that low concentrations of oxytetracycline could affect the nitrogen species in RAS. Also, the emission mechanisms of $N_2O$ may not be only dependent on nitrification process but also dependent on denitrification process in our RAS system.

Production of miraculin protein in suspension cell lines of transgenic rice using Agrobacterium (Agrobacterium을 이용한 형질전환 벼 현탁 세포주에서 miraculin 단백질의 생산)

  • To produce the miraculin protein in suspension cultures, rice (Oryza sativa L.) was transformed with Agrobacterium tumefacience EHA105 containing the miraculin AB512278 gene. The cell suspension cultures were established using cell lines selected from transgenic rice callus. The integration of the miraculin gene into the rice chromosome was confirmed using genomic PCR analysis. In addition, RT-PCR analysis indicated that the miraculin gene is expressed in the selected suspension cell lines. Thus, the recombinant miraculin was expressed in the transgenic suspension cell line, HK-2. Therefore, we have successfully developed a HK-2 line that produces miraculin. These results demonstrate that transformed cell suspension cultures can be used to produce a taste-modifying protein such as miraculin.

  • Flavonoid are large group of the polyphenolic compounds which are distinguished by an aromatic or phenolic ring structure and the phenolic compounds are induced by microbial infection, ultraviolet radiation, temperature and chemical stress. They are known for their antioxidant activity, anti-allergic, anti-inflammatory, anti-microbial and anti-cancer activities. In this study, changes in flavonoid content were investigated using heterologous chalcone isomerase (CHI) expression system. Also, phenolic compounds level was measured to examine the relation between flavonoids and phenols contents. Explants of lettuce (Lactuca sativa L.) were transformed with Agrobacterium tumefaciens LBA 4404 strain containing pFLH-CHI (derived from pPZP2Ha3) vector constructed with CHI gene from Brassica rapa. The putative transgenic plants were confirmed by genomic DNA PCR analysis. Also the transcription levels of the gene were analyzed by semi-quantitative RT-PCR with gene specific primers. The total flavonoid contents were increased at $T_0$ and $T_1$ generations over 1.4 and 4.0 fold, respectively. Total phenol contents also increased at $T_1$ generation. These results indicate that CHI gene plays an important role to regulate the accumulation of flavonoids and its component changes.

Complex Vector Current Control of Grid Connected Inverter Robust for Inductance Variation (인덕턴스 변화에 강인한 계통연계형 인버터의 복소 벡터 전류제어기)

  • This paper analyzes complex vector current control for the enhanced cross-coupling compensation in accordance with parameter variation in grid-connected inverter system, and verifies through simulation and experiment. Complex vector current control is performed in the synchronous reference frame through d-q transformation. It generates cross-coupling components with rotating nominal angular frequency. In general, cross-coupling elements are compensated by decoupling terms added to output of conventional decoupling PI controller. But, it is impossible to compensate them perfectly which transient response is especially deteriorated such as large overshoot and slow tracking, when variation of grid impedance or measurement error occurs. However, complex vector current control can improve stability and response characteristic of current control regardless of the situation as before. Decoupling controller and complex vector controller are represented through complex forms, and these controllers are analyzed by using frequency response in s-domain, respectively. It is verified that complex vector controller has more superior response characteristic than decoupling controller through MATALB, PSIM and experimental in 5kW grid-connected inverter when L filter parameter is varied from 1.1mH to increase double, 2.2mH.

Increased Thermotolerance of Transgenic Rice Plant by Introduction of Thermotolerant Gene

  • To increase thennotolerance of forage crops, transgenic rice plants as a model for transformation of monocots were generated. A cDNA encoding the chloroplast-localized small heat shock protein (small HSP) of rice, Oshsp21, was introduced into rice plants via Agrobacterium-mediated gene transfer system. Calli induced from scutella were co-cultivated with a A. tumefaciens strain EHAlOl canying a plasmid, pIGhsp21. A large number of transgenic plants were regenerated on a medium containing hygromycin. Integration of Oshsp2l gene was confirmed by PCR and Southern blot analyses with genomic DNA. Northern blot and immunoblot analyses revealed that the Oshsp21 gene was constitutively expressed and accumulated as mature protein in transgenic plants. Effects of constitutive expression of the OshspZl on thermotolerance were first probed with the chlorophyll fluorescence. Results indicate that inactivation of electron transport reactions in photosystem I1 (PSII), were mitigated by constitutive expression of the Oshsp21. These results suggest that the chloroplast small HSP plays an important role in protecting photosynthetic machinery during heat stress. (Key words : Thermotolerance, Rice, Transgenic, cDNA)

  • Volume fraction and morphology of retained austenite, tensile properties of TRIP type high strength steel sheet with Fe-C-Si-Mn-Cu chemical composition have been investigated. The retained austenite of granular, bar and film type existing in specimen was obtained after intercritical annealing and austempering. The granular type retained austenite increased with increase of intercritical annealing and austempering temperature. With increase of intercritical annealing temperature, retained austenite and carbon contents increased. Maximum contents of retained austenite was obtained by austempering at $400^{\circ}C$. The maximum tensile strength was obtained by austempering at $450^{\circ}C$ and maximum elongation was obtained at $400^{\circ}C$. T.S${\times}$E.L value increased with increase of retained austenite contents due to the elongation strongly controlled by contents of retained austenite, but tensile strength was affected with various factors such as bainitic structure etc.

  • Leaf dise explants of Solanum tuberosum cultivar. Desiree and Atlantic, were infected with a Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic and low temperature regulated gene (H28) for transformation. regenerated multiple shoots were selected on a medium containing kanamycin and carbenieillin after exposure to Agrobacterium. Both PCR analysis of NPT Ⅱ, H28 genes and northern blot analysis indicated that the genes coding for the enzyme were successfully integrated into the potato genome and could be expressed in potato plants.
