Propagation of Cutting Method of a Rare Endemic Juniperus chinensis var. sargentii Henry in Korea (희귀수종 눈향나무(Juniperus chinensis var. sargentii Henry)의 삽목증식)

  • Juniperus chinensis var. sargentii Henry is a short and creeping evergreen shrub which reaches about 60 cm in height and only occurs in the northeast Asia and in the top of high mountains over the Korea. The Korea Forest Service protects it strictly by law since J. chinensis var. sargentii is an eccentric plant and possibly may be exterminated soon in Korea. This study was carried out to develop the propagation technique by cutting for conservation of genetic resources of J. chinensis var. sargentii. The rooting responses of branch cuttings, obtained from hard(May) and semi-hard wood shoots (August) to four growth regulators, namely, IAA, IBA, NAA and Rooton(exceptionally powder method) applied at various concentrations(0, 100, 200, 500, 1000 and 2000 $mgL^{-1}$) were examined in mixed soil media. Rooting rate showed significant difference between cutting times, among kinds and among concentrations of growth regulators. The optimum cutting time was April to May in hardwood cutting. The application of IBA 1000 $mgL^{-1}$(rooting rate : 36.4%) was most effective in callus formation and rooting of cutting. Relatively, rooting of cutting of the control taken in May was 30.4%. Root characteristics such as number, length and diameter of root were not significantly affected by kinds and concentrations of growth regulators in hardwood cutting.

  • Cytological observations in the seven varieties of Juniperus chinensis L. showed three varieties (J. chinensis var. procumbens, J. chinensis var. kaizuka and J. chinensi.s var. aureo-variegata) were tetraploid with chromosome number, 2n=44, and rest of four varieties (J. chinensis var. horizontalis, J. chinensis var. sargentii, J. chinensis var. gtobosa and J. chinensis var. aureo-globosa) to be diploid, 2n=22. Chromosome configuration and behavior in the meiosis of P.M.C. of three tetraploids appeared to be slightly irregular. These results suggest that triploid tree can be artificially produced with these specific clones.

Spatial Genetic Structure and Genetic Diversity of a Rare Endemic Juniperus chinensis var. sargentii in Mt. Halla, Korea (희귀식물인 눈향나무(Juniperus chinensis var. sargentii)의 공간분포에 따른 유전구조 및 유전적 다양성)

  • Juniperus chinensis var. sargentii Henry is a short and creeping evergreen shrub which reaches about 50㎝ in height and occurs in the northeast Asia and in high mountains over the South Korea. Its distribution is restricted, and the number of individuals are gradually decreasing. This study was conducted to estimate spatial pattern, genetic diversity and spatial genetic structure of J. chinensis var. sargentii. A total of 131 clumps were studied in the study area (40m × 60m). The spatial pattern of this population was random (Aggregation index R=1.031). In spite of the small number and the limited distribution, the level of genetic diversity (Shannon's index 1=0.463) was relatively high as compared with those of other plant species with similar ecological characteristics. ISSR genotypes of all individuals were investigated to find the genetic relationship of clumps and genets. Fifteen clumps were composed to be clones, and a total of 116 unique genotypes were composed by separate genets. Spatial autocorrelation analysis using Tanimoto distance showed that the genetic patch was established within 8m. The effect of clonal reproduction on genetic structure was almost nothing.

Molecular phylogenetic study of section Sabina (Genus Juniperus) in Korea based on chloroplast DNA matK and psbA-trnH sequences data (엽록체 DNA matK와 psbA-trnH 염기서열에 기초한 한국산 향나무절(향나무속) 식물의 분자계통학적 연구)

  • This study aims to define the phylogenetic relationship within Korean section sabina and find molecular markers which resolve the phylogenetic relationship in genus Juniperus and section sabina. cpDNA matK and psbA-trnH were used as molecular markers. The combined analyses of two genes suggested that section sabina was a clade supported by 100% BP. The relationships of [J. chinensis var. sargentii+J. davurica] clade and [J. chinensis var. chinensis+J. chinensis var. procumbens+J. chinensis var. horizontalis] clade were supported by 91% BP and 100% BP, respectively. Thus, the classification of Korean section sabina would be appropriate at follows, (1) J. chinensis var. sargentii+J. davurica, and (2) J. chinensis var. chinensis+J. chinensis var. procumbens According to the results of separate analyses, matK seems to work better resolving power to clarify the phylogenetic ambiguity in Juniperus and section sabina than psbA-trnH.

Effect of Quartz Porphyry and Hydroball Ratio on Growth of Some Wood Plant by Water Flooding Culture in Pot (목본식물의 분 담수 재배에서 맥반석 및 하이드로 볼의 비율이 식물의 생장에 미치는 영향)

  • This study aimed to select the media for water flooding culture of Pinus thunbergii Parl., Juniperus chinensis L. var. sargentii Henry, Osmanthus fragrans Lour., and Chaenomeles japonica Lindl. ex Spach planting into quartz Porphyry and Hydroball. The survival rate of Pinus thunbergii Parl. and Chaenomeles japonica Lindl. ex Spach in hydroball 80%+quartz porphyry 20% was 100%, also the ratio of Juniperus chinensis L. var. sargentii Henry and Osmanthus fragrans Lour. in hydroball 20%+quartz porphyry 80% was 100% and 80%. However survival rate of full water and decomposed granite was less than 80% in Pinus thunbergii Parl., Juniperus chinensis L. var. sargentii Henry, and Chaenomeles japonica Lindl. ex Spach and thee rate of Osmanthus fragrans Lour was less than 50%. The increasing rate of fresh weight for Pinus thunbergii Parl. and Chaenomeles japonica Lindl. ex Spach in hydroball 80%+quartz porphyry 20% was 58.6% and 15.8% which was higher than others, and the fresh weight was increased as increasing the content of hydrobol. However the fresh weight of Juniperus chinensis L. var. sargentii Henry and Osmanthus fragrans Lour. in hydroball 20%+quartz porphyry 80% was 71.4% and 59.7% which was higher than others, and the fresh weight was increased as increasing the content of quartz porphyry. The increasing rate of fresh weight of decomposed granite as control was the lowest by 32.7%, 48.0%, 33.3% and 7.0%, respectively in Pinus thunbergii Parl., Juniperus chinensis L. var. sargentii Henry, Osmanthus fragrans Lour., and Chaenomeles japonica Lindl. ex Spach. Therefore the survival rate and fresh weight was lower at water and decomposed granite. However the survival rate and increasing fresh weight of Pinus thunbergii Parl. and Chaenomeles japonica Lindl. ex Spach was higher as increasing the rate of hydrobol. And the survival rate and fresh weight of Juniperus chinensis L. var. sargentii Henry and Osmanthus fragrans Lour was higher as increasing the rate of quartz porphyry.

Genetic Variation in the Endemic Rare Tree Species, Juniperus chinensis var. sargentii HENRY (희귀(稀貴) 수종(樹種) 눈향나무 집단(集團)의 동위효소(同位酵素) 분석(分析)에 의한 유전변이(遺傳變異) 연구(硏究))

  • Genetic variation of two Juniperus chinensis var. sargentii populations in Mt. Seorak and Mt. Halla was investigated by isozyme analysis at reproducible 11 loci of 7 isozyme systems(Aat-1, Aat-2, Gdh, Idh, Lap, Mdh-1, Mdh-2, Mdh-3, 6Pgd, Pgi-1, and Pgi-2), of which 7 loci were polymorphic. The levels of genetic diversity of two populations were A=2.2, $A_e=1.61,\;P_{95}=54.5,\;H_{o}=0.179,\;H_e=0.287$(Mt. Seorak population) and A=2.1, $A_e=1.48,\;P_{95}=63.6,\;H_{o}=0.270,\;H_e=0.250$(Mt. Halla population), respectively. These values were similar to and/or somewhat higher than those observed in other Korean native conifers. Moderately low degree of genetic differentiation was observed between 2 analyzed populations ($F_{ST}=0.039$). Heterozygosity of the population in Mt. Seorak was significantly lower than expected, and much high level of inbreeding coefficient(F=0.376) was observed. Considering the limited population size and distribution range of the population, the population seemed to be influenced by inbreeding and/or random genetic drift, Consequently, Mt. Seorak population should be considered to be a more important candidate for the conservation of J. chinensis var. sargentii.

First Report on Four Novel Endophytic Fungal Species Isolated from Leaves of Juniperus chinensis var. sargentii in Korea (눈향나무의 잎에서 분리된 4종의 국내 미기록 내생균)

  • We isolated endophytic fungi from the leaves of the conifer Juniperus chinensis var. sargentii inhabiting Mt. Hallasan. The fungal isolates were identified based on phylogenetic analysis using nucleotide sequences of the internal transcribed spacer region, large subunit region of ribosomal DNA, translation elongation factor, and ${\beta}$-tubulin region. In this study, we confirmed four species of unreported endophytic fungi in Korea, Allantophomopsis lycopodina, Epicoleosporium ramularioides, Kabatina juniperi, and Seiridium podocarpi. We describe their morphological characteristics and the results of the phylogenetic analysis.

Distribution of High Mountain Plants and Species Vulnerability Against Climate Change (한반도 주요 산정의 식물종 분포와 기후변화 취약종)

  • This work aims to select the potentially vulnerable plant species against climate change at alpine and subalpine belts of Mts. Sorak, Jiri, and Halla, from central, southern, southern insular high mountains of the Korean Peninsula, respectively. The selection of global warming related vulnerable plants were performed by adapting various criteria, such as flora, endemicity, rarity, floristically specific and valuable species, species composition at mountain summits, horizontal and vertical ranges of individual species, and their distributional pattern in the Korean Peninsula. Line and quadrat field surveys along the major trails from all directions at height above 1,500 meters above sea level of Mts, Sorak, Jiri and Halla were conducted each year during spring, summer, and autumn from 2010 to 2011. Based upon above mentioned eight criteria, high level of climate change related potentially vulnerable arboral plants, such as Rhododendron aureum, Taxus caespitosa, Pinus pumila, Oplopanax elatus, Vaccinium uliginosum, and Thuja koraiensis are noticed from at subalpine belt of Mt. Sorak. Species of Abies koreana, Rhododendron tschonoskii, Oplopanax elatus, Taxus cuspidata, Picea jezoensis, and Juniperus chinensis var. sargentii belong to climate change concerned vulnerable species at subalpine belt of Mt. Jiri. High level of climate change related species vulnerability is found at alpine and subalpine belts of Mt. Halla from Diapensia lapponica var. obovata, Salix blinii, Empetrum nigrum var. japonicum, Vaccinium uliginosum, Juniperus chinensis var. sargentii, Taxus cuspidata, Rhamnus taquetii, Abies koreana, Hugeria japonica, Prunus buergeriana, and Berberis amurensis var. quelpartensis. Countermeasures to save the global warming vulnerable plants in situ are required.

Utility of Hydrophilic Polymer for Green Technology Development in Green Roofs Using Rainwater (빗물활용 옥상녹화 녹색기술 개발을 위한 친수성 중합체의 효용성)

  • Hydrophilic polymer is suitable as soil conditioners for green roofs that use rainwater, due to promotion of water retention capacity as well as enhancement of the water absorbing capacity. The objective of the present study was to investigate the effects of different levels of hydrophilic polymer concentrations (0, 0.1, 0.2, 0.4, 0.8% w/w) on the water holding capacity and growth response of 6 species in soils amended with hydrophilic polymer in 5 cm of soil thickness on green roofs. The results showed that the water holding capacity of the amended soil improved with increasing amount of applied polymer. The application of 0.8% w/w of the polymer increased the soil moisture by 87% compared to the control, and decreased slowly in green roofs during an arid period. The growth of Sedum spurium 'Dragon's blood' and Lampranthus spectabilis increased significantly and had greater than 60% relative coverage with higher hydrophilic polymer concentrations. However, Juniperus chinensis var. sargentii and Euonymus fortunei var. radicans had no significant differences upon change of hydrophilic polymer concentrations. In Carex kujuzana and Carex morrowii 'Aurea variegata', growth decreased with increase of hydrophilic polymer concentrations. 30 days after planting, Juniperus chinensis var. sargentii, Euonymus fortunei var. radicans, Carex kujuzana, and Carex morrowii 'Aurea variegata' died back due to lowest soil thickness (5 cm), but Sedum spurium 'Dragon's blood' and Lampranthus spectabilis had greater than 90% survival.
