  • This study examines the optimal fractionation method and conditions for the isolation of endoprotease- and exopeptidase-active fractions from crude extracts of cuttlefish hepatopancreas (HP) using four fractionation methods: ammonium sulfate fractionation (ASF), polyethylene glycol fractionation (PGF), ion exchange chromatography (IEC), and gel filtration chromatography (GFC). Total endoprotease activity highest in the fraction II (concentrate of fractions 34-42; 842.60 U) of GFC, followed by fraction III (40-60% ammonium sulfate fraction; 670.25 U) of ASF, fraction I (concentrate of fractions 8-12; 436.89 U) of IEC, and fraction II (10-20% polyethylene glycol; 307.31 U) of PGF. Total exopeptidase activity of these fractions was highest in fraction II (2,704.70 U) of GFC, fraction III (2,110.50 U) of ASF, fraction III (1,605.60 U) of PGF, and fraction II (concentrate of fractions 38-44; 1,196.22 U) of IEC. These results showed that fraction II of GFC had the highest activity toward both exopeptidase and endoprotease, with exopeptidase activity being 3.21 times higher than of endoprotease. These results suggest cuttlefish HP could be used as a potential source for the extraction of exopeptidase, an enzyme capable of catalyzing the cleavage of N- and C-terminal amino acids in polypeptides, Like endoprotease, the most efficient method for separating exopeptide-active fractions was GFC.

  • S. komarovii is halophyte that grows in soil or waters of high salinity, such as in saline semi-deserts, sloughs and seashores. Traditionally, S. komarovii has been used for food and medicinal purposes in Korea. S. komarovii was extracted in 70% ethanol to measure anti-oxidative activity using DPPH and ABTS assay. The IC50 values of the S. komarovii extract against DPPH radicals and ABTS radicals were 186.10 mg/mL and 121.89 mg/mL. In addition, total polyphenol and reducing power were measured. The S. komarovii extract exhibited superior polyphenolic (22.5%) and antioxidant (28.4%) contents. Regarding cell bioactivity, MTT assay was conducted to reveal cytotoxicity of S. komarovii extract and showed the non-cytotoxicity of S. komarovii extract. Anti-inflammatory and skin whitening effects were measured at 100 ㎍/mL. Therefore, this study suggests that the S. komarovii extract can be used as a functional cosmetic product material.

  • The present study researched morphological characteristics and analyzed the genetic diversity by using RAPD in Calanthe species, native plant in Jeju, Korea. Twenty-six samples were selected by flower color, and 19 horticultural traits were investigated to study morphological characteristics. The C. discolor had the smallest leaf, the length and width of dorsal sepal, lateral sepal, petal, central lip, lateral lip, and flower stalk length were shortest and/or smallest except the spur and ovary length in Calanthe species, but those of Calanthe discolor for. sieboldii (Dence.) Ohwi (Calanthe discolor for. sieboldii) were the largest and/or biggest, and those of variants were the intermediate between C. discolor and C. discolor for. sieboldii, but spur length was the longest in C. discolor, the shortest in C. discolor for. sieboldii, and intermediate in the variants. Ovary length in C. discolor was shortest and C. discolor for. sieboldii and variants were similar with each other. The flower colors of C. discolor were brownish red, the value of CIE Lab was between 40 and 50. The flower color of C. discolor for. sieboldii was yellowish; the value of CIE Lab was between 110 and 130. And variants had various colors between 50 and 70 in the value of CIE Lab. After analyzing multiple band patterns of PCR products, 154 bands were selected as polymorphic RAPD markers. The analysis of Genetic distance of Calanthe species using RAPD showed that C. discolor and C. discolor for. sieboldii are more distant from each other than variants, and demonstrated the fact that genetic position of variants is between the other two species.

In this study, to develop torso patterns of top dresses selected as preferred designs through literature reviews and company surveys, dress form sizes suitable for standard sizes were selected and study basic patterns were made using draping techniques. The study finds characteristics of body type dimension that is changed when wearing brassiere (hereinafter, 'bra') for wedding dress through customer surveys for development of basic patterns of wedding dress. That is, wearing wedding bra was used for the purpose of increasing the bust size and we will evaluate later whether this increase helps completeness of aesthetic impression through wearing evaluation. As the result, it was found out that top torso pattern wearing wedding bra made wearers feel more comfortable and in the aspect of aesthetic impression, it provided impacts on women's beautiful silhouettes. The effects of whether a bra is used or not on changes in patterns were reviewed through the degree of polymerization of finished patterns. When a bra for dresses was worn, the waist front length increased by1.7cm, the bust circumference increased by 2.1cm, and the amount of dart increased by 1.5cm in the patterns. A new torso pattern for top dresses, which will solve problems evaluated in each of the above from the aspect of composition of pattern by a method through analysis of existing basic patterns and multi-dimensional cut patterns wearing wedding bra, will be suggested.

  • Protease inhibitors (PI) of trypsin and papain as target proteases from the roe of bastard halibut Paralichthys olivaceus were fractionated out using ammonium sulfate precipitation (A), DEAE 650M anion exchange chromatography (D), and Sephacryl S-300 gel filtration (S). The recovery percentages of the fractions with the strongest inhibitory activity for each fractionation method were 13% for the A4 fraction, 21.2% for the D3 fraction, and 21.3% for the S2 fraction, with specific inhibitory activities of the fractions toward trypsin and casein of 168, 139, and 218 U/mg, respectively, while no inhibition of papain was observed. The $IC_{50}$ for the trypsin-specific substrate $N{\alpha}$-benzoyl-$\small{L}$-arginine-p-nitroanilide (BAPNA) was 0.65, 1.55, 2.26, and 2.85 mg/mL for the A4, S2, A3, and D3 fractions, respectively. These results suggest that chromatographic fractionation methods (D and S) based on the molecular mass and charge of the protein were more effective at fractionating PI than was ammonium sulfate precipitation based on protein solubility, and that the bastard halibut roe extract acts as a serine protease inhibitor. Therefore, the PI fraction from fish roe might be useful for inhibiting proteases in foodstuffs, and could constitute an alternative food-grade inhibitor for the surimi industry.

  • Historical cost data estimating system has been introduced since 2004 in the construction industry. Based on contract prices of past projects, this estimating system estimates construction cost according to work types. The Korea Institute of Construction Technology (KICT) announces the historical cost data twice a year annually. The unit price of a small construction project is higher than large construction project due to the increase on production cost per work unit, equipment and labor cost, etc. However, the historical cost data estimating system is applied to project estimation uniformly regardless the construction project's size. This study compared and analyzed the historical cost data of large and small construction project to indicate the problem of historical cost date estimating system. The study derived that the unit price of a small construction project is 21.8% higher than that of large construction project.

  • The outer clothing of shaman that are put on while performing current gutgeori is lacking in form and composition, diversified with kinds. Above all, name of shaman clothing is not unified, accordingly, there's a big difference from traditional Gutgeori, which was definitely divided in the role of outer clothing of shaman in respect of function and role of Gutgeori. Here, this research attended a site that performed Gutgeori based on the advanced literature research data research and analyzed the state and name of outer clothing by conducting a survey and interview investigation. Survey was conducted during 2 years from 2014 to 2015 classifying Gutgeori of Seoul, Gyeongsang, Honam, Jeju, Chungcheon. Research results are as follows. First, lots of outer clothes were put on along with shamanism type in Seoul, accordingly, names were diversified. Name of outer clothing was not unified by focusing on level of divinity or behavior of Gutgeori, with no consistency being mixed. Second, name was not consistent even the outer clothing of the same type shaman along with the region, sometimes put on in different meaning, accordingly, called in different name. Combination of such name of shaman is determined to be a big reason of lack of accurate information on shaman clothing and genealogy of name of shaman clothing in shamans, as the initial research on shamanism was conducted by folklorists in advance.

  • This study finds surface color and functions of natural dyeing from Chamaecyparis obtusa leaves which help cure skin problems such as atopic dermatitis. This study also seeks to find dyeing properties and functions of natural colorants derived from Chamaecyparis obtusa and Rubia tinctorum on silk. Dyeing properties and functions are compared between silks dyed with Chamaecyparis obtusa and silks composite-dyed with Chamaecyparis obtusa and Rubia tinctorum. The comparison examines the feasibility of developing healthy and high functional fabrics and wellness of fashion merchandises. Silk dyeing with Chamaecyparis obtuse leaves yields yellow and double coloring from Chamaecyparis obtusa; however, Rubia tinctorum yields yellow/red on silks. Silks dyed with Chamaecyparis obtusa and silks composite-dyed with Chamaecyparis obtusa and Rubia tinctorum show a high level on dry cleaning, ultraviolet protection and deodorization. Especially, the dyed silks from leaves of Chamaecyparis obtusa show a 99.7% antimicrobial effect against staphylococcus aureus ATCC 653B. Silks composite-dyed with Chamaecyparis obtusa and Rubia tinctorum are better known for medicinal herb for dermatitis, and natural colorant, Rubia tinctorum does not improve significantly functions compared with silks dyed with Chamaecyparis obtusa. However, the dyeing properties improve by composite dyeing. This implies that ways to maximize effects of tie-dyeing technique could be developed.
