Growth Model of Sowthistle (Ixeris dentata Nakai) Using Expolinear Function in a Closed-type Plant Production System (완전제어형 식물 생산 시스템에서 선형 지수 함수를 이용한 씀바귀의 생육 모델)

  • The objective of this study was to make growth and yield models of sowthistle (Ixeris dentata Nakai) by using an expolinear functional equation in a closed-type plant production system. The growth and yield of hydroponically-grown sowthistle were investigated under four different planting distances ($15{\times}10$, $15{\times}15$, $15{\times}20$, and $15{\times}25$ cm). Shoot dry weights per plant was the highest at $15{\times}25$ cm, but was the lowest at $15{\times}10$ cm. Shoot dry weights per area was the highest at $15{\times}15$ cm, but was the lowest at $15{\times}25$ cm. The optimum planting density and planting distance for yield of sowthistle were 44 plants/$m^2$ and $15{\times}15$ cm, respectively. Shoot dry weights per plant and per area were showed as an expolinear type functional equation. A linear relationship between shoot dry and fresh weights was observed to be linear regardless of the planting distance. Crop growth rate, relative growth rate and lost time in an expolinear functional equation showed quadratic function form. Radiation use efficiency of sowthistle was $4.3-6.1g{\cdot}MJ^{-1}$. The measured and estimated shoot dry weights showed a good agreement using days after transplanting as input data. It is concluded that the expolinear growth model can be a useful tool for quantifying the growth and yield of sowthistle in a closed-type plant production system.

Decrease of tumor necrosis factor alpha (Tnf) production by Ixeris dentata extract in RAW 264.7 macrophage cells (씀바귀 약침이 RAW 264.7 대식세포에서 tumor necrosis factor alpha 생성에 미치는 영향 연구)

  • 목 적 : 약침으로서 씀바귀의 염증 효과를 알아보기 위해 RAW 264.7 대식세포에서 Tnf 생성을 확인하였다. 방 법: 씀바귀를 RAW 264.7 대식세포에 미리 처치한 후 LPS로 염증 반응을 유도하였다. ELISA, Western blotting, RT-PCR, 그리고 EMSA을 통해 Tnf의 생성과 발현에 대한 효과를 평가하였다. 결 과 : LPS 유도된 세포에서 씀바귀 1, 5, 10, 50 ${\mu}g/ml$의 농도는 각각 23.7, 37.8, 66.4, 86.1% Tnf 생성을 억제한 것을 ELISA 통해 확인되었다. LPS로 유도된 대식세포에서 Tnf 생성은 농도에 따라 감소하였고, Western blotting, RT-PCR 분석에서는 씀바귀 5과 50 ${\mu}g/ml$가 LPS로 유도된 대식세포에서 Tnf의 mRNA와 단백질 발현을 저해 하는 것으로 관찰되었다. 또한 EMSA에서도 씀바귀 5과 50 ${\mu}g/ml$가 LPS로 유도된 Nf-kB를 감소시키는 것을 확인하였다. 고 찰 : 이런 결과는 씀바귀가 $Nf-{\kappa}B$를 저해하면서 LPS로 유발된 Tnf 생성을 감소시킨다는 것을 보여주었고, 이는 더 나아가 염증 질환에서 씀바귀가 약침으로써의 치료적 효과를 나타낼 수 있을 것으로 사료된다. 고 찰 : 이런 결과는 씀바귀가 $Nf-_{k}B$를 저해하면서 LPS로 유발된 Tnf 생성을 감소시킨다는 것을 보여주었고, 이는 더 나아가 염증 질환에서 씀바귀가 약침으로써의 치료적 효과를 나타낼 수 있을 것으로 사료된다.

Induction of Apoptosis in Human Cancer Cells with Extracts of Taraxacum coreanum, Youngia sonchifolia and Ixeris dentate (흰민들레, 고들빼기, 씀바귀 추출물의 암세포 증식 억제 효과)

  • This research examined the effects of Compositae extract on the inhibition of proliferation and apoptosis in human breast and human gastric cancer cells. Compositae extracts which is used in the experiment are Taraxacum coreanum (TC), Youngia sonchifolia (YS) and Ixeris dentata (ID). The proliferation of SK-BR-3, MDA-MB-231 and AGS cells were investigated by MTT assay. ID and YS extracts inhibited proliferation of SK-BR-3, MDA-MB-231 and AGS cells in a dose-dependent manner, but TC have barely affected. In addition, the most effective extract was ID. To assess the apoptosis of ID extract, the nuclei of human cancer cells were stained with DAPI solution respectively. Chromatin condensation, indicated apoptosis, was increased in a dose-dependent manner. We investigated change of ID extract-induced apoptosis proteins on human cancer cells by western blot analysis. The level of Bcl-2 decreased, whereas the level of Bax, cleaved-PARP increased in dose-dependent manner compared with non-treatment. Also Bax/Bcl-2 ratio, which is used in clinical indicator of apoptosis, was increased at ID extract treatment group compared with non-treatment. Moreover the Bax/Bcl-2 ratio of MDA-MB-231 cell was significantly increased as against SK-BR-3, AGS cells. These results indicated that ID extract have anti-proliferation effect better than YS or TC, and induced apoptosis in human breast cancer MDA-MB-231 cell better than human breast cancer SK-BR-3 cell, human gastric cancer. Even if further research is needed, ID can be developed as a chemopreventive or therapeutic agent of breast cancer.

Biological Characteristics and Nucleotide Relationships in Korean Tomato spotted wilt virus Isolates

  • Tomato spotted wilt virus (TSWV) was identified from seven plants at two areas, Anyang and Dangjin, in Korea. The isolates of TSWV were seven as TSWV-KATm from tomato, TSWV-KAPo from potato, TSWV-KABal from balsam, TSWV-KACTm from cherry tomato and TSWV-KAIxe from Ixeris dentata at Anyang area, and TSWV-KDSe from sesame and TSWV-KDRP from red pepper at Dangjin area. Pathogenicity of seven TSWV isolates was various on the assay plants, and could not be grouped definitely. Three isolates of TSWV-KAIxe, TSWV-KACTm and TSWV-KABal had relatively narrower host ranges among the seven isolates. Percentage of nucleotide substitution in nucleotide sequences encoding nucleocapsid protein (NCP) was 1.2-1.7% among seven TSWV isolates and TSWV-KP. Korean TSWV isolates were divided into three groups by nucleotide homology or amino acid compositions. From the analysis of nucleotide sequences of Korean TSWV isolates compared with those of TSWV reported from other 5 countries including Japan, the Korean seven isolates of TSWV was grouped with German TSWV (D13926). No Korean TSWV isolates were grouped with those from The Netherlands, Brazil and USA.

Cytotoxicity of Some Natural Products in Human Cancer Cells (수종의 한국산 야생식물에서의 항암효과 검색)

  • The objective of this reseach is to find new antitumoral substances from natural products. Several of natural products have been used as food that were isolated into hexane(Hex.) and/or ethylacetate(EtOAc) extracts. we have tested cytotoxicities of these plants against human solid tumor cells. The cytotoxic activity of these plants were tested using Sulforhodamin B(SRB) assay. Hexane extracts of Chrysanthemum sinense, Allium tubersum. Beta vulgaris, Ixeris dentata have revealed cytotoxicities against five human solid tumor cells, and its cytotoxicities of each cell line were $10-100\;{\mu}l/ml$ ED50 (Effective dose that cause 50% inhibition of cell growth in vitro)

Screening of Domestic Plants with Antibacterial Activity (국내 자생식물의 항균활성)

  • To select new useful plants with antibacterial activity, ninety five sample of eighty different species of wild plants were collected, and extracted with methanol. Antibacterial activity of the methanol extracts was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Vibrio parahaemolyticus. The methanol extracts from Artemisia capillaris, Hemistepta lyrata, Youngia japonica, Prunella vulgaris, Lamium amplexicaule and Juniperus chinensis was effective against all bacterial strains tested, and eight methanol extracts including Ixeris dentata, Gnaphalium affine, Chelidonium majus and Spiraea prunifolia exhibited the antibacterial activity against at least 3 bacterial strains. Methanol extracts from leaf of Syringa vulgaris, Drava nemorosa and clove of Erythronium japonicum showed a selective antibacterial activity against two gram negative bacteria, V. parahaemolyticus, and B. subtilis, respectively. With investigations on antibacterial activity against a certain bacterial strains tested, metahnol extracts from clove of Erythronium japonicum, Spiraea prunifolia, leaf and twig of Camelia japonica, and Drava nemorosa showed strongest activities against B. subtilis, S. aureus, E. coli, and V. parahaemolyticus, respectively. Nine methanol extracts based on the results were successively fractionated with n-hexane, chloroform, ethyl acetate and water portions, which were examined antibacterial activity against B. subtilis and V. parahaemolyticus. Among the all fractions tested, chloroform fractions of Hemistepta lyrata showed strongest antibacterial activity against both B. subtilis (17mm) and V. parahaemolyticus (29 mm). Chloroform fractions of Youngia japonica, n-hexane fractions of Artemisia capillaris, Iexeris dentata and Prunella vulgaris, and ethyl acetate fraction of leaf and twig of Camelia japonica showed relatively a strong antibacterial activity. On the other hand, Juniperus chinensis and Equisetum arvense was distributed to all fractions except for water fraction.

Ecological Studies on the Vegetation of Pyeong-il Island (평일도 식물상에 관한 생태학적 연구)

  • The vegetation of Pyeongil island was ecologically investigated four times from July, 1979 to May, 1981. The vegetation of this island consisted of 80 families, 172 genera, 173 species, 21 varieties and 4 forma. Evergreen, broad-leaved trees comprised a total of 11 species including Eurya japonica Thunb. and Trachelostermum asiaticum var. intermedium Nakai. The distribution of evergreen, broad-leaved trees was very spares in this island as a whole. It may also be considered that evergreen, broad-leaved trees, such as Camellia japonica Linne (30cm-35cm in diameter at breast height), Pittosporum fobira Aiton, Eurya japonica Thunb. and Trachelostermum asiaticum var. intermedium Nakai which might have been dominant species on this island, have undergone artificial succession caused by the planting of Pinus thunbergii Parl. dominant species of today. This was due to human interferences for fuel, building, grazing and farmland use. Especially, the vegetation of this island seems to have been severely destroyed by high population density, farming rate and by rapid increase in the area of cultivated land. However, the forest of Pinus thunbergii Parlatore at Wolsong-ri is worthy of conservation. The five species of seaside plants (Ixeris dentata (Thunb.) Nakai, Rosa rugosa Thunb., Carex laticeps Clarke, Calystergia soldanella Reom. et Schult. and Phelloterus littoradis) were distributed in the vicinity of sand beaches leading to Wolsong-ri, Dongbaeg-ri and Sadong-ri.

Studios on the Host Range of Cucumber Mosaic Virus in Korea (한국에서의 오이모자익 바이러스의 기주범위에 관한 연구)

  • Various plant species, as many as 145 species in 43 families, were tested for susceptibility to the ordinary strain of the cucumber nosaic virus for two years from 1973 to 1974. Inoculations were made by mechanical method using carborundum. Plants in 71 species belonging to 27 families were infected. Of these species, systemic mosaic developed on the new leaves of plants in 57 species belonging to 24 families. Twenty-four species of plants, previously not reported as hosts of the CMV, were found to be infected in this experiment. These are Stellaria aquatica, Achyrauthes japonica, Agerratum houstonianum, Centipeda minima, Gillardia pulchella, Henisteptalyrate, Ixeris dentata, Saussurea uchiyamana, Brassica campestris, Lepidiumapetalum, Lobelia chinensis, Chenopodium bryoniaefolium, Carex neofilipes, Acalypha austalis, Amphicarpaea edgeworthii, Lotus corniculatus var japonicus, Phaseolus angularis, Sedum aizoom var heterodontum, Mosla punctulata, Perilla frutescens var japonica, Teucrium japonicum,. Linum usitatissimum, Mazus japonicus, Verbena hybrida. Twenty-three species reported to be susceptible by previous workers, but negative results were obtained in our experiment with Allium cepa, Celosia cristat, Daucus carota var. sativa, Artemisia asiatica, Callistenphus chinensis, Erigeron canadensis, Helianthusannuus, Tagetes eracta, Impatiens balsamina, Raphanus sativus, Ipomea batatas, Glycine max, Phaseolus vulgaris, Lilium longifolium, Papaver gomniferum, Sorghum vulgare, Triticum aestivum, Zea mays, Rumex coreanus, Potulaca grandiflora.

Effects of Aqueous Medicinal Herb Extracts and Aqueous Fermented Extracts on Alcohol-Metabolizing Enzyme Activities (약용식물의 열수추출물과 적정 조성추출물 및 그 발효물이 알콜대사 효소활성에 미치는 영향)

  • To develop an effective anti-hangover product, hot-water extracts of 25 medicinal herbs were screened for inhibition or activation of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH), and 12 herbs were selected for further study. Chosen medicinal herb extracts(CMHEs) were fermented by Lactobacillus delbruechii subspecies lactis for 10 days at $35^{\circ}C$ after saccharification with nuruk(malt inoculated by 5 types of microbs) for 72 hours at $35^{\circ}C$ and both CMHEs and fermented CMHEs(FCMHEs) were explored for anti-hangover effects in vitro. We found significant ADH inhibition by hot-water extracts of Pueraria thunbergiana, Hovenia dulcis Thunb, Lycium chinense, Glycyrrhiza uralensis, Acanthopanax sessiliflorus, Liriope platyphylla, and Ixeris dentata, and significant ALDH activation by extracts of Acanthopanax sessiliflorus, Lycium chinense, Ixeris dentata, and Polypori umbellati of the Polyporaceae. The ADH effects on CMHE and FCMHE were -20.22% and -62.63% of control values, and the ALDH effects 173.20% and 280.17%, respectively. In rats given 20%(v/v) alcohol(15 mL/kg), FCMHEs significantly decreased blood acetaldehyde concentrations on 3 hours after ethanol administration, in a dose-dependent manner(p<0.05). Notably, blood acetaldehyde concentrations were markedly reduced in animals given FCMHEs(400 mg/kg) compared to levels seen in rats receiving CADB(commercial alcohol detoxification beverage). Thus, anti-hangover effects were promoted by fermentation of certain medicinal herb extracts.

Development of an Environmental Friend Pellet Coating Oil for Replacement of Antibiotics for Ruminant Aninmal (천연물질을 이용한 반추동물 항생제 대체용 친환경 펠렛코팅오일의 개발)

  • This study investigated the effect of coated feed using mixed oil on rumen fermentation characteristics. Two experiments were conducted based on materials that were mixed. First, cashew nut and soybean oils were mixed with white mineral oil. And second, different plant extracts were mixed with white mineral oil. At first experiment, inclusion levels of mixed oil on diet (0.03%, 0.1%, 0.5%) were applied as variables. A coated diet was fermented with rumen inoculum according to in vitro rumen fermentation and its parameters were investigated. In the result of first experiment, no negative effects on rumen pH were found. Significantly decreased dry matter digestibility was detected at 0.5% treatment (P<0.05). Total gas productions in control and 0.03% were significantly greater than those of others (P<0.05). Significantly reduced methane productions were found in all treatments compared to the control (P<0.05). Inclusion of mixed oil did not affect on ammonia production. Total volatile fatty acid production was also not influenced by coating with mixed oils. Rumen fermentation parameters were greatly changed according to introduced plant extracts at second experiment. The significantly lowest and greatest ammonia productions were found at treatments with Ixeris dentata and Plantago asiatica, respectively (P<0.05). The significantly greatest acetate and propionate productions were detected at treatments with Crucuma longa and Zizyphus vulgaris 1, respectively (P<0.05). All treatments, except Chrysanthemum idicum, Euyale ferox seed, Moringa leaf and fruit and Zizyphus vulgaris 1, showed significantly increased total volatile fatty acid production compared to the control (P<0.05). Only Paeonia lactiflora showed significantly lesser gas production than the control (P<0.05). In methane production, Ceramium, Zizyphus, Paeonia, Agrimonia, Torilis, Mugwort, Foeniculum, Euphorbia, Taraxacum, Artemisia, Momordica, Curcuma and Moringa reduced methane significantly compared to the control (P<0.05).
