COMPARISON OF THE GROWTH RATES OF THREE STRAINS OF COMMON CARP, CYPRINUS CARPIO (잉어 3품종의 성장비교)

  • Three strains of common carp, i. e. , Israeli carp, red-and-white, and golden strains, were stocked in the same pond, and their growth rates were compared with following results: From August 12 to November 21 in 1975, fingerlings of the three strains of common carp, Cyprinus carpo, each weighing about 0.5 g with total length of 2 to 3 cm, were stocked. The pond had an area of $316m^2$ with a mean water depth of 55cm, and the bottom was covered with a 20 to 30 cm thick layer of silt containing a considerable amount of decaying organic materials. Feed given was prepared with equal amounts of fish meal and polished barley, of which, in addition, $10\%$ green grass and $1\%$ table salt were mixed together when prepared into paste feed using a chopper after boiling the barley. Total protein content of the feed was $34.9\%$ in dry state with $5\%$ moisture content. Total feed given was 30.08 kg calculated in dry state to produce 20.588 kg of the common carp fingerlings, thus the feed coefficient being 1.51. By strains, the harvested Israeli carp ranged 98 to 311g each with a mean weight of $172.69g(100\%)$, red-and-white strain 15 to 318g with mean of $104.1g(60.3\%)$, and the golden strain 30 to 268g with mean of $128.7g(74.6\%)$. During the rearing season mean water temperature was $23.9^{\circ}C$ and the assumed main growth period with the water temperature above $15^{\circ}C$ was, upto the end of October, for 80 days with a mean water temperature of $23.9^{\circ}C$. Taking this main growth period as the basis for growth rate analysis, the mean daily increments, expressed as the attained body weight in times of the starting weight, become 1.075786 times (or the Israeli strain, 1.06901 times for the red-and-white strain, and 1.07185 times for the golden strain.

Effect of Dynamite Explosion Work Noise on Behavior of Israeli Carp, Cyprinus carpio in the Cage of Aquaculture (양식 향어의 행동에 미치는 발파작업 소음의 영향에 관한 연구)

  • This paper described the relationship between the behavior of the Israeli carp, Cyprinus carpio and the environmental noise level due to the dynamite explosion work. The experiment was conducted in the cage ($L10{\times}W4{\times}D4 m$) of aquaculture located at Chungjoo Lake, Chechon, in 1997. The fish trajectory was obtained by the telemetry system in which a pulsed ultrasonic pinger ($50 kHz, {\phi}16{\times}L70 mm$) attached to the fish was tracked three dimensionally, and the underwater noise levels were measured. The results of the study were as follows: 1. The underwater noise levels in the normal blasting measured at a distance of 400 m from the source of noise increased by $40 dB (re 1 {\mu}Pa)$ compared to the levels before explosion. The dominant frequency and the increased power spectrum level of the underwater noise by the explosion work were $75 to 100 Hz and 22.9 to 35.3 dB$, respectively. 2. The underwater noise levels in the test blasting measured at a distance of 350 m from the source of noise increased by average $49.5 dB (re 1 {\mu}Pa)$compared to the levels before explosion. 3. The swimming area of the fish was reduced with the time after explosion, and after more than one hour the fish represented the similar swimming area and behavior to the status of right before explosion. 4, The swimming depth layer of the fish was most of the case at the sea surface less than 1,0 m except during explosion or right after of it. But the fish swam downward when an external stimulus like the explosion noise was given to the fish. 5. The average swimming speeds of the fish before, during and after the works were about 1.2 times, 1.9 times and 1.0 times of the body length, respectively, and the speed of the fish with explosion was faster 1.6 times than the speed without of that. Consequently, the explosion noise levels measured by this study were sufficiently high to affect the fish, and the heavy shock by the explosion works could produce a considerable unfavorable effects to the fish.

Storage Stability of Blood Constituents in Fish (어류 혈액 성분의 저장 안정성)

  • An attempt was made to elucidate the stability of serum metabolites and enzyme activities in blood samples taken from rockfish(Sebastes schlegeli), Israeli carp(Cyprinus carpio) and rainbow trout Oncorhynchus mykiss) under different storing conditions. The concentrations of total protein(TP), albumin(ALB), triglyceride(TG), cholesterol(CHOL) glucose(GLC), phosphorus(P) and sodium(Na), and the activities of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in serum were investigated for 16 days at $15^{\circ}C$(room temp.) and $4^{\circ}C$ (refrigerative) condition, or frozen at $-20^{\circ}C$ for period of 30 days. Though there was a little difference between fish species, the concentrations of TP, ALB, GLC, P, Na in serum were stable at all storing temperatures, while those of TG, CHOL, ALT and AST were not stable, particularly even at the normal temperature. In general, serum components were more stable at refrigerative and frozen conditions than at room temp. storing. However, it was noticeable that the stability of CHOL in rockfish serum was found to be more unstable at $-20^{\circ}C$ than kept at $15^{\circ}C$ or$4^{\circ}C$.

Muscle Protein Compositions of Cultured Israeli Strain of Common Carp by Age (이스라엘잉어의 연령별 근육단백질 조성의 비교)

  • This experiment was carried out to compare the change of muscle protein compositions, amino acid compositions of muscle protein, and free amino acid compositions by age of the Israeli strain of common carp, Cyprinus carpio nudus. Protein compositions of the muscle were: sarcoplasmic protein $25.8-27.2\%$, myofibrillar protein $62.3-56.2\%$, residual intracellular protein $9.6-13.2\%$ and stroma $2.3-2.9\%$. In between 1 year and 3 years, there were differences as follows; myofibrillar protein in 1 year was much than 3 years, and other proteins in 3 years were much than 1 year. By SDS-polyacrylamide gel electrophoresis, sarcoplasmic protein of the samples in 1 year an 3 years were composed of 11 subunits and 10 subunits, respectively. And appeared 210,000 dalton component in 1 year but did not appeared in 3 years. Myofibrillar protein was composed of 23 subunits in both 1 year and 3 years but the differences of subunits by age were not observed. No differences were observed by age in the composition of myofibrillar protein and residual intracellular protein. Amino acid composition of muscle protein in both 1 year and 3 years were no differences to each other, but the contents of glutamic acid, aspartic acid, lysine were higher than other amino acids. The amount of total free amino acid in 1 year was much than in 3 years.

Nutrients and Energy Digestibilities of Various Feedstuffs Fed to Israeli Strain of Growing Common Carp (Cyprinus carpio) (육성용 이스라엘 잉어에 의한 원료사료의 영양소 및 에너지 소화율)

  • This study was conducted with Israeli strain of common carp to determine apparent protein, lipid and gross energy digestibility coefficients and apparent calcium and phosphorus availabilities of the following feedstuffs : steam-dried white fish meal (WFM-S), flame-dried white fish meal (WFM-F), brown fish meal (BFM), soybean meal (SBM), corn gluten meal (CGM), sesame meal (SSM), wheat flour (WFL), brewer's yeast (BY) and monocalcium phosphate (MCP). Test diets consisting of a 70:30 mixture of refoernce diet to test ingredient were utilized with chromic oxide as an external indicator. Fecal collection was made during 10 days before the end of feeding trial lasted for 27 days. The best weight gain and feed utilization were found in fish fed MCP containing test diet. Protein digestibility was high for most ingredients, ranging from 81.2% for BY to 91.9% for SBM, with the exception of SSM which was 77.6%. Lipid digestibility coefficients ranged 74% for CGM to 85.8% for two kinds of white fish meal. Digestible energy coefficients were generally high for fish meals, ranging from 77.4% for WFM-S to 81.3% for WFM-F, whereas those for plant feedstuffs were significantly (P<0.05) lower, from 64.5% for CGM to 70.6% for BY. Phosphorus availability from fish meals was variable with a low of 6.6% for WFM-F and a high of 13.6% for WFM-S. Phosphorus availability from SBM and BY was 7.3% and 57.2%, respectively. Calcium and phosphorus availabilities from MCP were 84.6% and 80.6%, respectively. These results provide more precise information concerning nutrients and energy utilization of Israeli strain of common carp and will allow ingredient substitutions in practical diet formulations based on levels of availabile protein and phosphorus.

Effect of Enteromorpha compressa on the physiological activities in carp, Cyprinus carpio (파래투여에 의한 잉어류의 생리활성에 관한 연구)

  • Physiological activities of E. compressa were examined after oral and intraperitoneal(i.p.) adminstration in young(8g) Israeli and colored carp. Hematological parameters were evaluated to test physiological respose. Anti-bacterial activity was examined by counting the number of bacterial cells in the kidney, and also by measuring the change of agglutinin titers following A. sobriae infection. There was a tendency of increase in E. compressa-fed groups in total protein, albumin and glucose levels. The most marked increase was noted in the group fed with 5% E. compressa. GPT and GOT levels were reduced with the increase of E. compressa concentration. Feeding of E. compressa did not alter haematocrit(Ht) and hemoglobin (Hb) values. The number of A. sobriae was reduced in all groups intraperitoneally treated with variable concentration of E. compressa-extract. The lowest bacterial cells were found at the group intraperitoneally treated with $30{\mu}g$ of E. compressa-extract per g of colored carp($30{\mu}g/g$), indicating that the anti-bacterial activity is maximized at this concentration. The agglutinin titers were elevated in E. compressa extract-treated groups($30{\mu}g/g$) with the maximum value of $6.0{\pm}1.1$. These results indicate that E. compressa adminstration activated physiological response, and triggered a cascade for anti-bacterial reaction.

Effect of Red ginseng extract on immune function of Israeli carp, Cyprinus carpio. (홍삼추출물이 이스라엘잉어의 면역반응에 미치는 영향)

  • Israeli carps were intraperitoneally injected with an aqueous extract of red ginseng (RG) at five different concentrations (0, 1, 10, 25 and 50 mg/kg of body weight). The fish were sacrificed after 3 days and measured the neutrophil activity and plasma components. The activity of lysozyme, Ht, TIC and NBT reduction were significantly enhanced in the concentrations of more than 10 mg/kg. Further attempts were made to measure the effects of RG on leucocyte function at five different concentrations (1-100 ${\mu}g/m{\ell}$), in vitro. The proliferation of lymphocytes co-incubated with phytohaemagglutin (PHA) and concanavalin A (Con A) was significantly increased at 10 ${\mu}g/m{\ell}$ concentration, but suppressed at the highest concentration (100 ${\mu}g/m{\ell}$). RG alone or with lipopolysaccharide (LPS) had no effect on proliferation at any concentrations tested. Migration and reactive oxygen intermediates (ROI) production of leucocytes were enhanced in all concentration higher than 10 ${\mu}g/m{\ell}$. These results suggest that the RG activates the leucocyte function and may be able to be used as a stimulant of fish innate defences.

Thermal Stability of Israeli Carp Actomyosin and Its Protection by Chemical Additives (이스라엘 잉어 Actomyosin의 열안정성과 그 보호)

  • Effects of temperature and additives on the stability of actomyosin extracted from skeletal muscle of Israeli carp, Cyprinus carpio nudus, were studied by analyzing free SH-group, ATP-sensitivity and Ca-ATPase activity. The used additives were sucrose, sorbitol, Na-glutamate and L-cysteine. Furthermore, the denaturation constant($K_D$), protective effect(${\Delta}E/M$) and the other thermo-dynamic parameters on protein denaturation are systematically discussed. The actomyosin showed $4.12{\sim}4.68 mg/ml$ in protein concentration, $2.63{\sim}2.93\%$ in ribonucleic acid to the protein, $1:2.20{\sim}2.63$ in the binding ratio of myosin and actin, $4.33{\sim}5.26\%$ in fat content, 109.78 in ATP-sonsitivity, $0.159{\sim}0.201\;{\mu}M-Pi/min/mg-protein$ in Ca-ATPase activity and $3.3{\sim}3.4M/10^5$g-protein in free SH-group content. The first-order rate plots were obtained on the decrease of Ca-ATPase activity and ATP-sensitivity with an increase in temperature, while the free SH-group was increased to $60^{\circ}C$ and decreased rapidly above the temperature. The half-life of Ca-ATPase activity on the actomyosin Ca-ATPase was 280 min at $12^{\circ}C$, 125 min at $20^{\circ}C$, 55 min at $30^{\circ}C$ and 13 min at $40^{\circ}C$, and activation energy, activation enthalpy, activation entropy and free energy of the proteins at $20^{\circ}C$ wene 5,395 cal/mole, 4,814 cal/mole, -40.42 e.u. and 17,626 cal/mole, respectively. The protective effect of the additives on the actomyosin Ca-ATPase showed that the most effective material is $3\%$ sorbitol and followed in the order of $8\%$ Na-glutamate, $1\%$ sucrose and $1\%$ L-cysteine. The actomyosin was more stable at $-30^{\circ}C$ than at $0^{\circ}C$ and $-20^{\circ}C$. and when the additives were used in the low temperature storage, $8\%$ Na-glutamate was the most effective. $3\%$ sorbitol, $1\%$ sucrose and $1\%$ L-cysteine was to become lower in the order.
