An Actinomycete strain isolated from Mt. Dea-Dun had a strong antifungal activity. The culture brith produced by isolated strain showed only antifungal activity against fungi with the exception of yeast and bacteria. It was heat stable, dissolved in ehtylacetate. The concentrated antifungal agent showed cytotoxicity against HEP-2 and HeLa as tumor cell line, and showed weak cytotoxicity against VERO 36 as normal cell line. Morphological and physiological characteristics were tested with isolated strain. The spore color of isolated strain was gray. It had a short chain and produced brown colored lytic substance in yeast extract-malt agar. The cell wall of isolated strain was composed of meso-DAP, and we suggested it as genus Actinomadura. In the existing of chemical inhibitor, isolated strain grew on the condition of 0.0001% crystal violet, 0.1% phenol, 0.01% sodium azide and 10% sodium chloride. Carbon utilization of isolated strain was shown that glucose, sucrose, manitol and sodium citrate were well utilized.
In the current study, punching behavior of Reinforced concrete (RC) isolated footings was experimentally and numerically investigated. The experimental program consisted of four half-scale RC isolated footing specimens. The test matrix was proposed to show effect of footing area, reinforcement mesh ratio, adding internal longitudinal reinforcement bars and stirrups on the punching response of RC isolated footings. Footings area varied from 1200×1200 mm2 to 1500×1500 mm2 while the mesh reinforcement ratio was in the range from 0.36 to 0.45%. On the other hand, a 3D non-linear finite element model was constructed using ABAQUS/standard program and verified against the experimental program. The numerical results agreed well with the experimental records. The validated numerical model was used to study effect of concrete compressive strength; longitudinal reinforcement bars ratio and stirrups concentration along one or two directions on the ultimate load, deflection, stiffness and failure patterns of RC isolated footings. Results concluded that adding longitudinal reinforcement bars did not significantly affect the punching response of RC isolated footings even high steel ratios were used. On the contrary, as the stirrups ratio increased, the ultimate load of RC isolated footings increased. Footing with stirrups ratio of 1.5% had ultimate load equal to 1331 kN, 19.6% higher than the bare footing. Moreover, adding stirrups along two directions with lower ratio (0.5 and 0.7%) significantly enhanced the ultimate load of RC isolated footings compared to their counterparts with higher stirrups ratio (1.0 and 1.5%).
The purpose of this study is to investigate microbial contamination in water purifiers from two universities (A and B) in Chungcheong region and to evaluate about the harmfulness of the isolated bacteria to the human. The degree of microbiological contamination of six water purifiers at university A was investigated three times from July 2018 to September 2019, and nine water purifiers at university B were investigated in 2023. The isolated bacteria were biochemically identified using an API kit and Vitek-2 system, and then the bacteria were identified to the species level using MALDI-TOF MS. In addition, the possibility of human infection of the isolated bacteria was evaluated through a literature search. In July 2018 and September 2019, the number of bacteria isolated inside the faucet was below the acceptable standard for hot water, but exceed for cold water in all water purifiers. In January and September 2019, bacteria exceeding the acceptable standards were isolated nine times from the cold water of six water purifies (a total of 12 water purifiers). Bacteria identified by MALDI-TOF MS included anaerobic bacteria (Clostridium novyi, Clostridium themopalmarium etc.), Gram-positive bacilli (Microbacterium testaceum, Arthrobacter woluwensis etc.), and Gramnegative bacilli (Acinetobacter nosocomialis, Comamonas kerstersii etc.), which are difficult identify by biochemical methods. In conclusion, bacteria exceeding the acceptable standard were isolated from the cold water of most of the water purifiers. Most of the isolated bacteria were low-pathogenic bacteria from natural environment, but opportunistic bacteria that can cause infection in humans were also isolated from some water purifiers.

A new experimental system of base-isolated structures is proposed. There are two kinds of dampers usually used in the base-isolated buildings, one is a viscous-type damper and the other is an elastic-plastic hysteretic-type damper. The base-isolated structure with a viscous damper and that with an elastic-plastic hysteretic damper are compared in this paper. The viscous damper is modeled by a mini piston and the elastic-plastic hysteretic damper is modeled by a low yield-point steel. The capacity of both dampers is determined so that the dissipated energies are equivalent at a specified deformation. When the capacity of both dampers is determined according to this criterion, it is shown that the response of the base-isolated structure with the elastic-plastic hysteretic damper is larger than that with the viscous damper. This characteristic is demonstrated through the comparison of the bound of the aspect ratio. It is shown that the bound of aspect ratio for the base-isolated structure with the elastic-plastic hysteretic damper is generally smaller than that with the viscous damper. When the base-isolated structure is subjected to long-duration input, the mechanical property of the elastic-plastic hysteretic damper deteriorates and the response of the base-isolated structure including that damper becomes larger than that with the viscous damper. The effect of this change of material properties on the response of the base-isolated structure is also investigated.
The isolation, morphological study and growth characteristics of the algae were investigated from Lake Chuam. The isolated algae were applied the Agal Growth Potential test. The method of isolation and purification of the algae were used to Agar plating(AP), nutrient enrichment(NE), dilution(DI) and micro capillary technique(MC). Total isolated algae were 21 species. They were composed of Cyanophyceae, Dinophyceae, Bacillariophyceae, Euglenophyceae and Chlorophyceae. The numbers of algal strain by isolation technique were highest in dilution(21 species), and those of the rests were showed in order of NE > MC > AP. The sizes of isolated Selenastrum and Scenedesmus were $1.8\pm 1.4 \mu m$, $3.3\pm 0.9 \mu m$ in diameter and $6.4\pm 2.3 \mu m$, $13.6\pm 1.9 \mu m$ in length respectively. The morphology of isolated algae and NIES-collection strain was very similar each other, but the size was smaller isolated algae than that of NIES-collection. The optimum culture condition of isolated Selenastrum and Scenedesmus was about 30$\circ$C(25$\circ$C-35$\circ$C) in temperature and the maximum growth was appeared between 7,000 lux and 8,000 lux in the light intensity. The comparison of $\mu$(specific growth rate) on the concentration of nutrients such as nitrate and phosphate, isolated Selenastrum was appeared maximum it at 1.0 mg $NO_3-N/l$ but NIES-collection strain was showed 95% of maximum it at same nitrate concentration. Maximum g of isolated algae and NIES-collection strain in Scenedesmus onto nitrate concentration were very similar with the result of selenastrum. The specific growth rates of isolated algae and NIES-collection strain on the gradient concentration of phosphate were showed 0.72/day and 0.70/day at 0.02 mg $PO_4-P/l$ in Selenastrum but those of Scenedesmus were appeared 0.61/day and 0.57/day at same concentration $PO_4-P$.

The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 4. The contractile response of the ileum smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan extract. 6. Objectives: The aim of this study was to evaluate the response to a hepatitis B vaccination, and investigate the HBV DNA in subjects with isolated anti-HBc. Methods: 34 subjects with persistent isolated anti-HBc were included in the study. 32 subjects negative for HBsAg, anti-HBs and anti-HBc were included as a control group. They were all vaccinated with Hepaccine at 0, 1 and 2 months, and anti-HBs titers were measured 1 month after the 1st and 3rd vaccinations (1 and 3 months). The HBV-DNA was tested by polymerase chain reaction in subjects with isolated anti-HBc. Results: After the 1st & 3rd vaccinations, the anti-HBs titers$\geq$10mIU/ml were 70.6 & 70.6% in isolated anti-HBc group, and 34.4 & 81.2% in the control group, respectively. There were statistically significant differences after the 1st vaccination, but none after the 3rd, between the two groups. In the isolated anti-HBc and control groups, the primary, amnestic and no responses were 0 vs. 46.9%, 55.9 vs. 6.3% and 29.4 vs. 18.8%, respectively. The HBV DNA was not detected in all subjects with isolated anti-HBc. Conclusion: None of the subjects with isolated anti-HBc had a false positive result (primary response); therefore, they should be excluded from vaccination programs in Korea. To differentiate between immunity and occult infections, a single dose of vaccine, with a follow-up anti-HBs test, is preferable for subjects with isolated anti-HBc. An amnestic response indicates late immunity, and no response a suspect occult infection.

Pseudomonas syringae pv. actinidiae strains, the causal agents of bacterial canker on kiwifruit, were isolated from Korea and Italy in 2011. Among 87 isolates, a total of six representative strains, three from Korea and three from Italy, were identified on the basis of biochemical and physiological tests. Identities were confirmed by PCR using P. syringae pv. actinidiae-specific primers PsaF1/R2, which amplified a 280-bp DNA fragment. The strains isolated from Korea in this study displayed BOX-PCR patterns similar to those isolated from Italy but different from those isolated previously in Korea or the pathotype P. syringae pv. actinidiae strain. The effector hopA1 and hopH1 genes, which are known to be present in strains isolated recently from France and Italy, were also present in P. syringae pv. actinidiae strains, SYS1, SYS2 and SYS4, isolated from Korea in this work. However, no amplicons of the expected size were obtained from strains previously isolated from Korea and Japan. In addition, the Korean strains isolated in this work belonged to haplotype I for the cts gene identical to those strains isolated from recent outbreaks in Italy. These results suggest that P. syringae pv. actinidiae strains isolated from Korea and examined in this work are a new type of strain similar to those found from recent outbreaks in Italy. This is the first report on the occurrence of cts haplotype I strains of P. syringae pv. actinidiae affecting kiwifruit plants in Korea.
This study was conducted to monitor the antibiotics resistance of human-harmful bacteria isolated in the agricultural environment for hot peppers (Capsicum annuum) and tomato (Lycopersicon esculentum). As a result, we isolated 120 bacterial species (34 on fruits, 48 in soil, 21 in water, and 17 in manure), identified them with the 16S rRNA sequence, analyzed minimum inhibitory concentration (MIC) for 26 antibiotics using Sensititre ARIS Hi-Q system and then evaluated whether each bacterial genus acquired resistance for the tested antibiotics or not, according to the CLSI criteria. From difference in MIC between eco-friendly (EFM) and practical (PFM) cultivation farms, Klebsiella spp. isolated from EFM was resistant to ampicillin (AMP) and nalidixic acid (NAL), and that isolated from PFM was resistant to streptomycin (STR) and tetracycline (TET). Enterobacter spp. isolated from EFM was resistant to AMP and azithromycin (AZI), and that isolated from PFM was resistant to AMP, AZI, and STR. Meanwhile, Pseudomonas spp. isolated from EFM and PFM were all resistant to AMP, AZI, cefotaxime (FOT), cefoxitin (FOX), ceftriaxone (AXO), CHL, NAL, and STR. Staphylococcus spp. isolated from EFM and PFM were resistant to gentamycin (GEN), STR, and kanamycin (KAN), and in particular, that from EFM showed resistance for erythromycin (ERY). In conclusion, our study suggested that EFM lead STR antibiotics resistance for human-harmful bacteria to decrease, because only the bacteria isolated from hot pepper and tomato crop with PFM have showed resistance against STR antibiotics, regardless of bacterial genus.
This research work was aimed at assisting isolated children to adapt their school life having examined the process of their changes by reducing isolation level and isolative behavior through means of implementing group art therapy program. To accomplish this purpose of research, 36 participants were selected from children attending Class 6, 4th Grade of D Elementary School located in Dajeon Metropolitan City, to which researcher was served as homeroom teacher. And the participants were asked to complete a survey that included social and emotional isolation scale test which was designed by Heh Sung-hee(1993) and 4 children marked more than 96 points were designated as isolated children. The group art therapy program exercised in this research was the one that reorganized by research to suit the research purpose based on the precedent works, and survey was conducted total 12 times, two time a week, within 60 minutes per time, between 6 September and 24 October 2006, and researcher conducted group art therapy program directly at Class 6, 4th Grade of D Elementary School using extra hours after school. And following analyses were performed in relation to results therefrom: First, to analyze changes of isolation level of isolated children Quantitatively, the results from pre and post test concerning social and emotional isolation scale were comparatively analyzed. Second, to analyze changes of psychological state of isolated children, projective drawing tests were performed using Kinetic House-Tree-Person(K-HTP) Drawing and Kinetic School Drawing(KSD) and results therefrom were comparatively analyzed. Third, to analyze Quantitatively what changes have been occurred in the isolated children during the course of forming interrelationships with group members while participating in group art therapy, proceedings were summarized per each stage of the program and analyzed accordingly. Fourth, results from researcher's observation of what changes have been occurred in the isolative behavior of isolated children before and after implementation of group art therapy were compared accordingly. Followings are resulting conclusions. First, group art therapy program reduced isolation level of isolated children. Second, group art therapy program changed the isolated children's psychological state into a stable and positive fashion. Third, isolated children became able to experience positive interrelations as well as establishment of new human relationships with group members during the course of participating in the group art therapy program. Fourth, group art therapy program reduced isolative behaviors of the isolated children. Therefore, conclusions could have been reached that the group art therapy program implemented in this research reduced isolative behaviors of the isolated children, and changed their anxious and depressed mental state into stable and positive ones, also established positive interrelationships as well as new human relationships. And, it will be able to expect a greater effect if a program which has been associated with their family could be proceeded along with continuous guidance and teaching having developed objective test tools that will discern isolated children.
