Effects of Some Coumarin Derivatives on the Bovine Lens Aldose Reductase Activity

  • Nine coumarin dervatives were examined for thier inhibitory effects on bovine lens aldose reductase activity (bovine-LAR). More than 50% inhibition of BLAR activities was observed in the cases of treatments with decursin, decursinol, esculin, isoimperatorin, oxypeucedanine and isobergapten at the concentration of $10^{-4}$M. Especially, BLAR activity was completely inhibited by the treatment of decursin and decursinol at this concentration. At $10^{-5}$ M, only three coumarins-decursin, decursinol and isoimperatorin, were found to have still relatively higher inhibitory effect.

A Comparative Study of between Cultivar and Wild type of Angelica dahurica in korea (한국산(韓國産) 재배백지(栽培白芷)와 야생백지(野生白芷)에 대한 비교(比較) 연구(硏究))

  • Objectives : This research was observed by examining the external and internal parts and the quantity of the surface components of the wild type and cultivar of Angelica dahurica in korea. Methods : The slice of the tested material mede by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by the microscope and examined the quantity of the surface components. Results : In the shape of the roots, the wild type had a very thin main root and 2 long roots, while the cultivar had a long root branched out from the short and thick main root making a square pyramid shape. Isoimperatorin was detected at Rt $17{\sim}18$ minutes. The wild type contained 0.56% while the cultivar contained it of 0.355%, thus the component of the wild type was about 1.5 times of that of the cultivar. Conclusions : It was thought that there might be also some differences in their genetic characteristics, etc.

Acetylcholinesterase Inhibitors from Angelica polymorpha Stem

  • Fourteen compounds were isolated from the stem of Angelica polymorpha. On the basis of spectral data, these compounds were identified as isoimperatorin (1), phellopterin (2), bergapten (3), xanthyletin (4), cnidilin (5), geijerine (6), (-)-3'-acetyl hamaudol (7), 7-demethylsuberosine (8), dehydrogeijerin (9), (-)-hamaudol (10), (+)-visamminol (11), divaricatol (12), scopoletin (13), and decursidate (14), respectively. Among them, compounds 4-6, 8, 9, 13, and 14 were isolated for the first time from A. polymorpha. Dehydrogeijerin (6) and geijerin (9) were isolated for the first time from genus Angelica. All isolates tested for inhibitory activity against acetylcholinesterae. Compounds 1 to 13 showed acetylcholinesterase inhibitory activity with $IC_{50}$ values ranging from 1.4 to $37.5{\mu}M$.

  • The MeOH extract of Notopterygium incisum showed a strong cytotoxicity against B16 murine melanoma cell line. From this extract three furanocoumarins including bergamottin, isoimperatorin, notopterol and one polyacetylenic compound (falcarindiol) together with one phenylpropanoid (caffeic acid methyl ester) and one triterpenoid (pregnenolone) were isolated. The isolated compounds were evaluated for cytotoxic activity against four kinds of cancer cell lines, e.g. P388 (murine lymphocytic leukemia), B16 (murine melanoma), A549 (human lung carcinoma) and SK-OV-3 (human ovarian cancer). Among the isolates, falcarindiol and caffeic acid methyl ester expressed a significant antiproliferative activity against all tested cell lines.

  • Angelica flaccida Kom. is a herbal plant growing wild in the marsh of valley in Korea. It has been used for sedative and analgesic as a folk medicine, but its constituents have not been clarified yet. Five known coumarins such as isoimperatorin, decursidin, (-) anomalin, decursin, decursinol and two sterols such as ${\beta}-sitosterol$ and stigmasterol were isolated from the root of this plant, Their structures were identified by the UV, IR, NMR, mass data and the physico-chemical properties.

  • Five coumarins, isoimperatorin (1), pabulenol (2), isooxypeucedanin (3), oxypeucedanin hydrate (4) and osthol (5) were isolated from the MeOH extract of Angelica genuflexa in the course of searching for anti-platelet and anti-coagulant components from plants. Pabulenol (2) was isolated from A. genuflexa for the first time. The five compounds isolated from A. genuflexa, together with decursinol angelate (6), decursin (7) and nodakenin (8) from A. gigas were evaluated for their effects on platelet aggregation and blood coagulation. Compounds 2, 5, 6 and 7 were observed to be either equally effective or 2∼4 times more inhibitory than ASA in both arachidonic acid and U46619 ($TXA_2$ mimetic) induced platelet aggregations.

  • To investigate biological active constituents from natural products, we have studied the roots of Angelica koreana Max. (Umbelliferae). Fifteen compounds were isolated from the MeOH extract by column chromatography on a silica gel. The compounds were identified as isoimperatorin, oxypeucedanin, oxypeucedanin hydrate, osthol, nodakenin, 2-hydroxy-4-methylacetophenone, cimifugin, falcarindiol, heraclenin, pabulenol, umbelliferone, demethylsuberosin, hamaudol, sec-O-glucosylhamaudol, and prim-O-glucosylcimifugin, respectivelv, by spectroscopic means. Among these, the latter eight compounds were isolated for the first time form this plant.

  • Angelica genuflexa (Umbelliferae) is a perennial herbaceous plant which has also been variously reported as A. koreana and Ostericum koreanum. The MeOH extract was reported to have strong anti-thrombotic potential in the acute thrombosis model. In our preliminary testing, the MeOH extract and one of the solvent fractions (90% MeOH fr.) were observed to have both platelet anti-aggregating and anti-coagulant effects. Five coumarins, isoimperatorin (1), pabulenol (2), isooxypeucedanin (3), oxypeucedanin hydrate (4) and osthol (5) were isolated from the MeOH extract of Angelica genuflexa in the course of searching for anti-platelet and anti-coagulant components.
