Antibacterial Effect of Caesalpinia sappan Extract against Mastitis Pathogens from Dairy Cows (소목추출물의 젖소 유방염 원인균에 대한 항균효과)

  • To find alternative agents for bovine mastitis, the extracts of 30 selected herbal medicine were screened for their antibacterial activity against mastitis pathogens. The extracts of 9 herbal medicine, Terminaliae chebula, Cinnamonum loureirii, Paeonia japonica, Inula britannica, Caesalpinia sappan(C. sappan), Salix babylonica, Alpinia katsumadai, Polygonum aviculare and Coptidis japonica showed strong antibacterial activity. Among of them, C. sappan extract was found to be the most active against all bacteria in this experiment. Methanol extract of C. sappan and brazilin, the main component of C. sappan, had inhibitory activity against a variety of pathogens, however they had less of an inhibitory effect on gram negative bacteria. Minimal inhibitory concentrations of brazilin against gram positive pathogens was lower than that of methanol extract. These results showed that the extract of C. sappan has a potential as a therapeutic agent for bovine mastitis.

  • In previous study, we investigated the mechanism of suppression of inducible nitric oxide synthase (iNOS) and cyclo-oxygenase-2 (COX-2) by ergolide, sesquiterpene lactone from Inula Britannica. In this study, the suppression of iNOS and COX-2 by ergolide might be attributed to selective inhibition of NF-$\_$K/B signaling pathways.   • The volatile components of Korean and Chinese Inulae flos were identified. It is made of dried Inulae flos from the components were collected by simultaneous steam distillation extract method. Those were analyzed by combined gas chromatography(GC) and gas chromatography-mass spectrometrt(GC/MS). Sixty two components, including 10 hydrocarbons, 7 aldehydes, 18 alcohols, 3 ketones, 5 esters, 13 acids and 6 miscellaneous components were identified. Volatile components in Inulae flos were fractinated into one hydrocarbon fraction and two oxygenated hydrocarbon fractions by using silica gel column chromatography. The volatile components consisted of 16.85% hydrocarbon and 83.15% oxygenated hydrocarbons in Korean Inulae flos, 23.46% hydrocarbon and 76.53% oxygenated hydrocarbons in Chinese Inulae flos.

  • The study was conducted to investigate candidate materials as anti-inflammation agent from plant resources. Activities of 33 plant parts extracts with the final concentration of 5${\mu}g/ml$ were evaluated on the several inflammation-related markers such as the release of proinflammatoty cytokine [tumor necrosis factor-alpha (TNF-${\alpha}$) & interleukin-6 (IL-6)], nitric oxide (NO), the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and inhibitor of nuclear factor kappa-B alpha ($I{\kappa}-B{\alpha}$) in lipopolysaccharide (LPS)-treated RAW 264.7 cells. The extracts in the final concentration of 10 ${\mu}g/ml$ were also screened on peroxynitrite (ONOO$^-$) scavenging activity. Eleven extracts selected from the screening assay were verified on the inhibition activity on peroxynitrite and total reactive species oxygen (ROS) in the several concentrations. As results, Alpinia officinarum Hance (rhizome), Inula britannica var. chinensis Regel (flower), Ulmus arvifolia Jacq (trunk peel) and Aster scaber Thunb. (aerial part) showed comparatively potent anti-inflammatory activities in vitro cells or chemical level systems, and then these four plant parts should be studied on the antiinflammatory mechanism by further studies.

Screening of 94 Plant Species Showing ACE Inhibitory Activity (식물자원으로부터 Angiotensin Converting Enzyme 저해활성 탐색)

  • Angiotensin converting enzyme(ACE) belongs to the class of zinc protease and plays an important role in the regulation of blood pressure. In this experiment, we investigated the inhibitory activities of ninety four plant extracts on ACE. The extracts were prepared by water and refluxing with 70% and 100% methanol. Among the extracts, two plant extracts such as Cassia tora, Persicaria cochinchinensis Kitagawa showed more than 60% inhibitory activities, and Foeniculum vulgare Gaertner, Scutellaria baicalensis Georgl, Caragana sinica (Buchoz) Rehder, Inula britannica var. chinensis showed $45.2{\sim}49.7%$ inhibitory activities. Twenty eight plant extracts such as Hemerocallis fulva L, Camptotheca acuminata Decne, Inula britannica var. chinensis, Xanthium strumarium, Polygonatum odoratum, Phellodendron amurense Rupr, Coix lachryma-jobi var. mayuen, Prunus ansu, Hibiscus mutabilis L, Thchosanthes kirilowii, Helianthus annuus, Juglans sinensis showed $30.3{\sim}39.7%$ Inhibitory activities. These results suggest that plant extracts which contain high ACE inhibitory activities may be useful as anti-hypertension agents and to the treatment of hypertension.

Floral Characteristics of Asteraceae Flowers and Insect Pollinators in Korea (우리나라 국화과 식물의 화기구조와 방화 곤충 연구)

  • To search for the co-relationships between insect-pollinators and the plant species of Asteraceae, insects visiting in flower and the flowers in Korea, were studied from April 2010 to October 2011. The sum of flower visiting degrees are shown 38 in Lepidoptera, 38 in Diptera, 36 in Hymenoptera, and the lowest 6 in Coleoptera, respectively. 65 insect species are identified pollinators, Hymenoptera 13 species(Apidae 11 sp., Formicidae 2 sp.), Lepidoptera 29 species(Pieridae 5sp., Nymphalidae 12 sp., Satyridae 3 sp., Hesperirdae 3 sp., Lycaenidae 2 sp., Danaidae 1 sp., Moth 2 sp.), Diptera 16 species(Tachinidae 1 sp., Syrphidae 12 sp., Muscidae 1sp., Others 2 sp.), and Coleoptera 6 species(Cetoniidae 1 sp., Cermbycidae 3 sp., Chrysomelidae 1 sp., Mordellidae 1 sp.). 31 pollinator species visits the flower of $Erigeron$ $annuus$, next 15 pollinator species does the flower of $Eupatorium$ $japonicum$, and then 13 pollinator species does the flower of $Aster$ $ageratoides$. Only 2 pollinator species visit the flower of $Tephroseris$ $kirilowii$, $Ixeridium$ $dentatum$, $Inula$ $britannica$ var. $japonica$, $Carduus$ $crispus$, $Ligularia$ $fischeri$, $Ainsliaea$ $acerifolia$, $Synurus$ $deltoides$, $Cirsium$ $setidens$, $Crepidiastrum$ $enticulatum$ and $Dendranthema$ $boreale$. Pollinators of Lepidoptera visit more frequently white flower than yellow or purple one. This study found out that mutualisic relations between plants and insect pollinators is carried out in Korea.

Comparison of antioxidant activities of ethanol extracts from seven species of wild edible plants in Korea (국내 야생식용식물 7종 에탄올 추출물의 항산화 활성 비교)

  • In this study, we investigated the potential of 70% ethanol extracts from wild edible plants (Pueraria lobata sprout, Rosa multiflora sprout, Artemisia princeps leaf, Diospyros kaki leaf, Morus alba leaf, Robinia pseudoacacia flower, Inula britannica var. japonica flower), as natural antioxidants. The antioxidant contents and activities of extracts were examined using various methods. The measurements of total polyphenol content revealed that Rosa multiflora sprout extract had the highest value and total flavonoid content showed that Diospyros kaki leaf extract had the highest value. Antioxidant activities were the highest in Rosa multiflora sprout for DPPH (IC50 232.52 ㎍/mL), ABTS+ (IC50 470.10 ㎍/mL), superoxide- (IC50 431.88 ㎍/mL), nitrite (IC50 363.38 ㎍/mL) scavenging activity, and reducing power (2.47 O.D.). These results suggest that the ethanolic extract of Rosa multiflora sprout is a potential source of natural antioxidants.

Anti-ulcer effects of HT074 on HCl/EtOH induced gastric injury (염산/에탄올로 유도된 위손상 동물모델에서 HT074의 항궤양 효과)

  • Objectives : This study aimed to investigate the anti-ulcer effect of an standardized herbal extracts mixture of Inulae Flos and Paeoniae Radix (HT074) on acidified ethanol induced gastric injury and its potential mechanisms. Methods : Antioxidant activities of HT074 and its constituents were measured by DPPH (2,2-Diphenyl-1-picrylhydrazyl) and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging capacity. After the oral administration of HT074 at doses of 100, 300 mg/kg twice per day for 14 days, Gastric lesions were induced by oral administration of acidified ethanol in Sprague Dawley rats. Oxidative stress markers, such as super oxide dismutase (SOD) activity, concentrations of catalase (CAT) and glutathione (GSH) were measured in gastric mucosal tissues. Additionally, the expression of human mucin gene, Mucin 5AC (MUC5AC) mRNA in gastric mucosal tissues was measured. Results : HT074 showed dose dependent radical scavenging activities against DPPH and ABTS radicals. Oral administration of HT074 300 mg/kg for 14 consecutive days significantly decreased gastric lesions and histological damages induced by HCl/EtOH in rats. HT074 treatment significantly increased the activity of SOD (300 mg/kg) and concentration of GSH (100 and 300 mg/kg), however catalase concentration was not significantly increased. MUC5AC mRNA expression was significantly increased by HT074 100, 300 mg/kg treatment. Conclusions : HT074 protects the gastric mucosa from oxidative stress caused by acidified ethanol by increasing the activity of SOD, concentration of GSH and mucin biosynthesis. These findings suggest that HT074 could be an effective candidate for prevention and treatment of gastritis and gastric ulcer.
