Estimation of Ecological Carrying Capacity for Oyster Culture by Ecological Indicator in Geoje-Hansan Bay (생태지표를 이용한 거제한산만 굴양식장의 생태학적 수용능력 산정)

  • The importance of aquafarming is increasing all over the world, however the coastal environment in the semi-closed inner bay has been aggravated due to the long term production and the high stocking density. For the sustainable aquafarming, there is a requirement for a eco-friendly fishery management by the estimation of ecological carrying capacity. The model development and application is still in the initial step, because it has to consider the whole ecosystem and all culture activities. As an alternative, there is a requirement for ecological indicator to assess the ecological performance. This study tried the estimation of ecological carrying capacity using ecological indicator. The production and the facility of the oyster farms was 4,935M/T, $49ind./m^3$ in Geoje-Hansan Bay(2008). Filtration pressure indicator was 0.203 which could provide a guidance on the present level of culture development. According to the environmental characteristics and the present oyster farms in Geoje-Hansan Bay, the newly assessed filtration pressure for the acceptable ecological carrying capacity was 0.102. Consequently, ecological carrying capacity in Geoje-Hansan Bay was 2,480M/T, $25ind./m^3$ and this represents the level of culture that can be introduced into Geoje-Hansan Bay without leading to significant changes to ecological process, species, populations or communities. Our study utilized the ecological indicator to estimate ecological carrying capacity of oyster farming for sustainable productivity and this could be the scientific basis for the eco-friendly fishery management.

Qualitative PCR Detection of Stack Gene GM Rice (LS28 X Cry1Ac) Developed in Korea (국내개발 stack gene GM 벼(LS28 X Cry1Ac)에 대한 정성 PCR 분석)

  • For the development of qualitative PCR detection method of genetically modified (CM) rice, rice species-specific gene, OsCc-1 (rice cytochrome c gene), was selected as suitable far use as an endogenous gene in rice. The primer pair OsCytC-5'/3'with 111 bp amplicon was used for PCR amplification of the rice endogenous gene, OsCc-1 and no amplified product was observed from 8 different crops as templates. Qualitative PCR method was carried out with stack traits of L528$\times$CryIAc1 GM rice developed in Korea. For the qualitative PCRs, some primer pairs were designed with a construct-specific and event-specific type based on T-DNA and junction sequences of T-DNA in GM rice. Actck-5'/3' amplifying between actin promoter and OsCK1 gene introduced in LS28 gave rise to an amplicon 306 bp; also, CrLB-5'/3' from CryIAcl and CKRB-5'/3'amplifying the junction region of T-DNA and genome sequence from LS28 as event-specific primers gave rise to an amplicon 142 bp and 91 bp, respectively. These primer pairs for the detection of event-specific targets not produced PCR amplicons on non-CM rice and various crops in contrast to event lines. Therefore, in this study we verified that event-specific primers were effective to specifically detect stack trait lines and demonstrated that this method presented could be provided with the detection-method data for risk assessment analysis of GM rice to be commercialized.

Adaptation Test of Scotch Pine (Pinus sylvestris L.) in Korea -Thirty-six-year-old Growth Performance of Twenty-two Provenances- (구주소나무 적응성검정 시험 -22개 산지 36년생 결과-)

  • This study was conducted to select superior provenances of Scots pine (Pinus sylvestris L.) well adapted to Korean environment for timber production. In 1976, twenty-two provenances of Scots pine were introduced from Sweden and the seeds were sown in seed beds in March. After one year, the seedlings were transplanted to nursery beds. The resulting 1-1 seedlings of 22 provenances were planted at Whaseong in 1978. Randomized complete block design with 3 replications were used for test plantation. Each provenance was planted with 20-tree row plot in each block and at a spacing of $1.8{\times}1.8m$. The growth performance of each provenance was monitored up to 33-years after planting. There were significant differences among provenances in volume growth. F3001 provenance showed the best volume growth of 33-years after planting ($0.160m^3$), which was 2.2 times greater than that of the lowest provenance W2027 ($0.072m^3$). The ranking of provenances was stabilized after 14 years. Comparing to reference tree species, Japanese red pine (Pinus densiflora), all Scots pine provenances showed poor growth performance. In other words, volume growth of Japanese red pine at age 28 and 33 were 2.1 and 3.3 times greater than that of Scots pine, respectively. Moreover, survival rate of Scots pine was lower than that of Japanese red pine. Based on these results, it was suggested that Scots pine was not suitable to Korean environments. The cause of maladaptation of Scots pine and the implications of introduction breeding were discussed.

Effects of Brassica rapa SHI-RELATED SEQUENCE overexpression on petunia growth and development (배추 SHI-RELATED SEQUENCE 유전자 발현이 페튜니아 생장 발달에 미치는 영향)

  • SHI-RELATED SEQUENCE (SRS) genes are plant-specific transcription factors that contain a zinc-binding RING finger motif, which play a critical role in plant growth and development. Among Brassica rapa SRS genes, BrSRS7 and BrLRP1 genes, isolated from shoot apical regions are important regulators of plant growth and development. In order to explore the function of BrSRS genes in horticultural plant growth and development, two constructs containing BrSRS7 and BrLRP1 under the control of a cauliflower mosaic virus 35S promoter were introduced into petunia by Agrobacterium-mediated transformation. The resulting transgenic plants were dwarf and compact plants with reduced plant height and diameter. Additionally, these transgenic plants had upward-curled leaves of narrow width and short internodes. Interestingly, the flower shapes of petunia were different among transgenic plants harboring different kinds of SRS genes. These phenotypes were stably inherited through generations $T_2$ and $T_3$. Semi-quantitative RT-PCR analyses of transgenic plants revealed that BrSRS7 and BrLRP1 regulate expression of gibberellin (GA)- and auxinrelated genes, PtAGL15- and PtIAMT1-related, involved in shoot morphogenesis. These results indicate that the overexpression of BrSRS7 and BrLRP1 genes suppressed the growth and development of petunia by regulating expression of GA- and auxin-related genes. From these data, we deduce that BrSRS7 and BrLRP1 genes play an important role in the regulation of plant growth and development in petunia. These findings suggest that transformation with the BrSRS genes can be applied to other species as a tool for growth retardation and modification of plant forms.

Removal of As(III) in Contaminated Groundwater Using Iron and Manganese Oxide-Coated Materials (철/망간 산화물 피복제를 이용한 오염지하수에서의 As(III)제거)

  • Permeable reactive barrier using iron oxide coated sand is one of effective technologies for As(V) contaminated groundwater. However, this method is restricted to As(III), because As(III) species tends to be more weakly bound to adsorbent. In order to overcome the limitation of iron oxide coated sand application to As(III) contaminated groundwater, manganese oxide materials as promoter of As(III) removal were combined to the conventional technology in this study. For combined use of iron oxide coated sand and manganese oxide coated sand, two kinds of removal methods, sequential removal method and simultaneous removal method, were introduced. Both methods showed similar removal efficiency over $85\%$ for 6 hrs. However, the sequential method converted the As contaminated water to acid state (pH 4.5), on the contrary, the simultaneous method maintained neutral state (pH 6.0). Therefore, simultaneous As removal method was ascertained as a suitable treatment technology of As contaminated water. Moreover, for more effective As(III) remediation technique, polypropylene textile which has the characteristics of high surface area, low specific gravity and flexibility was applied as alternative material of sand. The combined use of coated polypropylenes by simultaneous method showed much more prominent and rapid remediation efficiency over $99\%$ after 6 hrs; besides, it has practical advantages in replacement or disposal of adsorbent for simple conventional removal device.

Determination of Grades and Design Strengths of Machine Graded Lumber in Korea (국내 기계등급구조재의 등급구분체계 및 기준설계값 결정방법 연구)

  • Based on comparative studies on standards and grading procedures of machine graded lumber in Korea and other countries, this study proposed a procedure of determining the grade classification and design strengths of domestic machine graded lumber. Differences between machine stress rated lumber and E-rated laminations were detailed in order to clarify the need for the procedure improvement. To this improvement the use of average MOE requirement for grading was introduced instead of the fixed minimum MOE requirement which is currently used in the Korean standards. It was found that the fixed minimum MOE requirement method was easier for an inspector to grade but, less efficient as a strength predictor than the average MOE requirement method. The advantage of average MOE requirement method is statistically MOR-MOE regression-based MOR prediction and highly efficient in quality control though it requires a computer-aided operation system in an initial setup. A major weakness of the current Korean grading system was found that different strength characteristics depending on wood species were not reflected on the grade classification and the tabulated allowable design stress. The proposed procedures were developed taking advantages of respective merits of both methods and based on MOR-MOE regression analysis. Through this procedure, the grades of machine stress rated lumber should be revised to become interchangeable with E-rated lamination, which would be beneficial to the cost competitiveness of domestic machine graded lumber and glued laminated timber industry.

Development of Prevotella nigrescens ATCC $33563^T$-Specific PCR Primers (Prevotella nigrescens ATCC $33563^T$ 균주-특이 중합효소연쇄반응 프라이머 개발)

  • A Pn10 DNA probe was introduced as a Prevotella nigrescens ATCC $33563^T$-specific DNA probe. In that study, the specificity of the Pn10 was tested with only type or reference strains of 5 oral bacterial species. The purpose of this study is to evaluate the specificity of the Pn10 using the wild type strains of P. nigrescens and is to develop the P. nigrescens ATCC $33563^T$-specific PCR primers based on the nucleotide sequence of the Pn10. The specificity of the Pn10 DNA probe was determined by Southern blot analysis. The nucleotide sequence of Pn10 DNA probes was determined by chain termination method. The PCR primers were designed based on the nucleotide sequence of cloned DNA fragment. The data showed that Pn10 DNA probe were hybridized with the genomic DNAs from P. nigrescens ATCC $33563^T$ and KB6. The Pn10 homologous region, KB6-Pn10, of P. nigrescens KB6 was cloned by PCR and sequenced. The Pn10 and KB6-Pn10 DNA fragments were consisted of 1,875 bp and 1,873 bp, respectively. The percent identity of the two was 98.8% and the divergence of them was 0.6%. The two primer sets (Pn10-F-AC/ Pn10-R-AC and Pn10-F-A/ Pn10-R-A), designed base on the nucleotide sequences of Pn10 DNA probe, were specific to the P. nigrescens ATCC $33563^T$. The two PCR primer sets could detect as little as 4 pg of genomic DNA of P. nigrescens ATCC $33563^T$. These results indicate that the two PCR primer sets have proven useful for the identification of P. nigrescens ATCC $33563^T$, especially with regard to the maintenance of the strain.

Carbon Budget Evaluated in Two Urban Parks of Seoul (서울의 두 도시 근린공원에서 평가된 탄소수지)

  • This study was carried out to assess the carbon budget of two urban parks and one natural park and to prepare the plan for improving the ecological functions of the park including carbon sink. Net Ecosystem Production (NEP) of those study sites was calculated from the relationship between Net Primary Production (NPP) and soil respiration of each study site. To understand carbon budget of the whole area designated as the park, carbon budget of the urban park was analyzed by classifying the vegetated and the non-vegetated zones. NEP of the Nohae and the Sanggye parks calculated by reflecting areal size that the non-vegetated zones occupy were shown in -1.0 and $0.6\;ton\;C\;ha^{-1}yr^{-1}$, respectively. On the other hand, NEP of Mt. Bulam natural park as a reference site was in $2.3\;ton\;C\;ha^{-1}yr^{-1}$. Based on the result, the Nohae park was assessed as carbon source rather than carbon sink. On the other hand, the Sanggye park was classified as carbon sink but the role was poor compared with natural park. The result is, first of all, due to lower NPP of the vegetation introduced for the parks compared with natural vegetation. The other reason is due to wide arrangement of non-vegetated zone. To solve those problems and thereby to create the urban park with high ecological quality, selection of plant species suitable for the ecological characteristic of the park, their arrangement imitated natural vegetation, and ecological zoning were recommended.

Effect of Species and Seed Mixture on Productivity and Botanical Composition in Oversown Hilly Pasture, the Central Northern Area of Korea (중북부 산지초지에서 국내육성 목초 이용 불경운 초지조성 혼파조합이 목초 생산성 및 식생구성에 미치는 영향)

  • This study was conducted to find out the effects of seed mixture using new domestic grass varieties orchardgrass 'Onnuri' and tall fescue 'Greenmaster' on botanical composition and dry matter productivity from 2015 to 2018 in oversown hilly pasture, the central northern area of Korea. This experiment was consisted of 4 mixed grasses (Treatment 1 : tall fescue oriented mixture with imported varieties; Treatment 2 : orchardgrass oriented mixture with imported varieties; Treatment 3 : orchardgrass oriented mixture with domestic varieties, Treatment 4 : tall fescue oriented mixture with domestic varieties). Evaluation of seasonal changes in botanical composition of pasture showed that orchardgrass was the highest in all treatments, ranging from 46 to 89% and Kentucky bluegrass was the second highest in all treatments. Treatment 3 (7,633 kg ha-1) and Treatment 4 (7,570 kg ha-1), mixed grasses using domestic varieties, were showed the highest dry matter yield than Treatment 1(6,950 kg ha-1) and Treatment 2 (6,934 kg ha-1), which were mixed grasses using introduced varieties. Therefore, these results showed that the orchard grass oriented grasses mixture with domestic varieties was good for grassland vegetation and productivity in oversown hilly pasture, the central northern area of Korea.

Pulmonary Oxalosis Caused by Aspergillus Niger Infection (Aspergillus Niger 감염에 의한 폐옥살산염 1예)

  • The Aspergillus species produces metabolic products that play a significant role in the destructive processes in the lung. We experienced a case of chronic necrotizing pulmonary aspergillosis caused by an Aspergillus niger infection, which contained numerous calcium oxalate crystals in the necrotic lung tissue. A 46-year-old man, who had a history of pulmonary tuberculosis, presented with high fever, intermittent hemoptysis and pulmonary infiltrations with a cavity indicated by the chest radiograph. Despite being treated with several antibiotics and anti-tuberculosis regimens, the high fever continued. The sputum cultures yielded A. niger repeatedly, and intravenous amphotericin B was then introduced. The pathological specimen obtained by a transbronchial lung biopsy revealed numerous calcium oxalate crystals in a background of acute inflammatory exudates with no identification of the organism. Intravenous amphotericin B was continued at a total dose of 1600 mg, and at that time he was afebrile, although the intermittent hemoptysis continued. On the $63^{rd}$ hospital day, a massive hemoptysis (about 800 mL) developed, which could not be controlled despite embolizing the left bronchial artery. He died of respiratory failure the next day. It is believed that the oxalic acid produced by A. niger was the main cause of the patient's pulmonary injury and the ensuing massive hemoptysis.
