Supplementation Effects of $C_{18:2}$ or $C_{18:3}$ Rich-oils on Formations of CLA and TVA, and Lipogenesis in Adipose Tissues of Sheep

  • The present study was conducted to investigate the supplementation effects of $C_{18:2}$ rich-soybean oil or $C_{18:3}$ rich-perilla oil (7% of total diet, DM basis) for 12 weeks on plasma metabolites, fatty acid profile, in vitro lipogenesis, and activities of LPL and FAS in adipose tissue of sheep. The treatments were basal diet (Control), $C_{18:2}$ rich-soybean oil supplemented diet (SO-D) and $C_{18:3}$ rich-perilla oil supplemented diet (PO-D). All the sheep were fed the diets consisting of roughage to concentrate in the ratio of 40:60 (DM basis). Oil supplemented diets (SO-D and PO-D) slightly increased contents of triglyceride (TG) and total cholesterol (TC), proportions of both cis-9 trans-11 and trans-10 cis-12 CLA and TVA, but lowered (p<0.01) those of $C_{18:0}$ compared to the control diet. No differences were observed in the contents of TG and TC and proportions of fatty acids in plasma between supplemented oils. Oil supplemented diets slightly increased the proportions of cis-9 trans-11 and trans-10 cis-12 types of CLA in subcutaneous adipose tissue of sheep compared to the control diet. The rate of lipogenesis with acetate was higher (p<0.01) for intermuscular- and subcutaneous adipose tissues than that for intramuscular adipose tissue, while that with glucose did not differ among fat locations in sheep fed SO-D. No differences were observed in the rate of lipogenesis between substrates in all fat locations. The rates of lipogenesis with glucose increased only in the intermuscular- (p<0.01) and subcutaneous adipose tissue (p<0.005) compared to those with acetate. The rates of lipogenesis with acetate were the highest in the intermuscular and intramuscular adipose tissue of the sheep fed PO-D. Oil supplemented diets slightly increased the rate of lipogenesis with glucose for all fat locations. Supplementation of oils to the diet numerically increased the fatty acid synthase activity but did not affect the lipoprotein lipase activity in subcutaneous adipose tissue.

RNA-Seq explores the functional role of the fibroblast growth factor 10 gene in bovine adipocytes differentiation

  • Objective: The present study was executed to explore the molecular mechanism of fibroblast growth factor 10 (FGF10) gene in bovine adipogenesis. Methods: The bovine FGF10 gene was overexpressed through Ad-FGF10 or inhibited through siFGF10 and their negative control (NC) in bovine adipocytes, and the multiplicity of infection, transfection efficiency, interference efficiency were evaluated through quantitative real-time polymerase chain reaction, western blotting and fluorescence microscopy. The lipid droplets, triglycerides (TG) content and the expression levels of adipogenic marker genes were measured during preadipocytes differentiation. The differentially expressed genes were explored through deep RNA sequencing. Results: The highest mRNA level was found in omasum, subcutaneous fat, and intramuscular fat. Moreover, the highest mRNA level was found in adipocytes at day 4 of differentiation. The results of red-oil o staining showed that overexpression (Ad-FGF10) of the FGF10 gene significantly (p<0.05) reduced the lipid droplets and TG content, and their down-regulation (siFGF10) increased the measurement of lipid droplets and TG in differentiated bovine adipocytes. Furthermore, the overexpression of the FGF10 gene down regulated the mRNA levels of adipogenic marker genes such as CCAAT enhancer binding protein alpha (C/EBPα), fatty acid binding protein (FABP4), peroxisome proliferator-activated receptor-γ (PPARγ), lipoprotein lipase (LPL), and Fas cell surface death receptor (FAS), similarly, down-regulation of the FGF10 gene enriched the mRNA levels of C/EBPα, PPARγ, FABP4, and LPL genes (p<0.01). Additionally, the protein levels of PPARγ and FABP4 were reduced (p<0.05) in adipocytes infected with Ad-FGF10 gene and enriched in adipocytes transfected with siFGF10. Moreover, a total of 1,774 differentially expressed genes (DEGs) including 157 up regulated and 1,617 down regulated genes were explored in adipocytes infected with Ad-FGF10 or Ad-NC through deep RNA-sequencing. The top Kyoto encyclopedia of genes and genomes pathways regulated through DEGs were the PPAR signaling pathway, cell cycle, base excision repair, DNA replication, apoptosis, and regulation of lipolysis in adipocytes. Conclusion: Therefore, we can conclude that the FGF10 gene is a negative regulator of bovine adipogenesis and could be used as a candidate gene in marker-assisted selection.

Association of GHRH, H-FABP and MYOG Polymorphisms with Economic Traits in Pigs

  • The study of candidate genes is an important tool to identify genes associated with economic traits. Three genes were selected to study the association between polymorphism and economic traits and breed of pigs. Growth hormone releasing hormone (GHRH) takes part in growth metabolism and is one of the candidate genes known to be highly associated with economic traits in pigs. Heart fatty acid-binding protein (H-FABP) is related to back fat thickness and intramuscular fat (IMF) content, and myogenin (MYOG) is associated with the amount of growth rate and lean yield in pigs. By PCR-RFLP analysis, the association between the genotypes of the three genes and the average daily gain, back fat thickness, feed conversion, body length and meat percent in 352 pigs (112 Duroc pigs, 132 Landrace pigs and 108 Yorkshire pigs) were analyzed. GHRH polymorphisms showed differences depending on breed (p<0.01) and were associated with meat percent. H-FABP polymorphisms also showed significant differences among breeds and sex (p<0.01), and were highly associated with average daily gain, feed conversion and back fat thickness (p<0.01) and even showed an association with meat percent (p<0.05). However, the MYOG gene showed no significant effect in this study. These results reconfirmed that GHRH and H-FABP are potential major genes or markers for economic traits.

Fatty Acid Profiles of Ten Muscles from High and Low Marbled (Quality Grade 1++ and 2) Hanwoo Steers

  • The aim of this research was to evaluate: 1) the fatty acid profile of ten muscles from high marbled (HM, quality grade 1++) and low marbled (LM, quality grade 2) Hanwoo carcass, 2) the relationship between the fatty acid profile and sensory traits. There were significant (p<0.001) differences in fat content and fatty acid composition among the 10 muscles obtained from HM and LM Hanwoo steers. The proportions of SFA (saturated fatty acid), MUFA (monounsaturated fatty acid) and PUFA (polyunsaturated fatty acid) were significantly (p<0.001) different among the 10 muscles due to differences in all fatty acids except eicosapentaenoic acid (EPA, C20:5n-3). The high-fat muscles had a lower n-6/n-3 ratio compared to the low-fat muscles (p<0.001). LM muscles had a significantly (p<0.05) higher proportion of SFA than HM muscles due to a higher proportion of stearic acid (C18:0). On the contrary, HM muscles had a significantly (p<0.01) higher proportion of MUFA than LM muscles due to higher oleic acid (C18:1n-9) proportion. SFA had a significant correlation with CIE a* (r=0.281; p<0.01) and drip loss (%) (r=−0.233; p<0.001). Cooking loss (%) had a significantly (p<0.05) negative correlation with PUFA (r=−0.233; p<0.05). Overall palatability was positively correlated with SFA (r=0.262; p<0.01), but negatively correlated with PUFA (r=−0.567; p<0.001). There was no significant correlation between oleic acid and any of the sensory traits (p>0.05).

Effects of prolonged photoperiod on growth performance, serum lipids and meat quality of Jinjiang cattle in winter

  • Objective: This study was conducted to investigate the potential effects of prolonged photoperiod on the serum lipids, carcass traits, and meat quality of Jinjiang cattle during winter. Methods: Thirty-four Jinjiang bulls aged between 14 and 16 months were randomly assigned to two groups that were alternatively subjected to either natural daylight +4 h supplemental light (long photoperiod, LP) or natural daylight (natural photoperiod, NP) for 96 days. The potential effects on the levels of serum lipids, carcass traits, meat quality, and genes regulating lipid metabolism in the intramuscular fat (IMF) of the cattle were evaluated. Results: Jinjiang cattle kept under LP showed significant increase in both dry matter intake and backfat thickness. the serum glucose and the plasma leptin levels were significantly reduced, while that of melatonin and insulin were observed to be increased. The crude fat contents of biceps femoris muscle and longissimus dorsi muscle were higher in LP than in NP group. In longissimus dorsi muscle, the proportions of C17:0 and C18:0 were significantly higher but that of the C16:1 was found to be significantly lower in LP group. The relative mRNA expressions in IMF of longissimus dorsi muscle, the lipid synthesis genes (proliferator-activated receptor gamma, fatty acid-binding protein) and the fatty acid synthesis genes (acetyl-coa carboxylase, fatty acid synthetase, 1-acylglycerol-3-phosphate acyltransferase) were significantly up-regulated in LP group (p<0.05); whereas the hormone-sensitive lipase and stearoyl-CoA desaturase 1 were significantly down-regulated in LP than in NP group. Conclusion: Prolonged photoperiod significantly altered the growth performance, hormonal levels, gene expression and fat deposition in Jinjiang cattle. It suggested that the LP improved the fat deposition by regulating the levels of different hormones and genes related to lipid metabolism, thereby improving the fattening of Jinjiang cattle during winter.

Effect of ACADL on the differentiation of goat subcutaneous adipocyte

  • Objective: The aim of this study was to clone the mRNA sequence of the Acyl-CoA dehydrogenase long chain (ACADL) gene of goats and explore the effect of ACADL on the differentiation of subcutaneous fat cells on this basis. Methods: We obtained the ACADL gene of goats by cloning and used quantitative real-time polymerase chain reaction (qPCR) to detect the ACADL expression patterns of different goat tissues and subcutaneous fat cells at different lipid induction stages. In addition, we transfect intramuscular and subcutaneous adipocytes separately by constructing overexpressed ACADL vectors and synthesizing Si-ACADL; finally, we observed the changes in oil red stained cell levels under the microscope, and qPCR detected changes in mRNA levels. Results: The results showed goat ACADL gene expressed in sebum fat. During adipocyte differentiation, ACADL gradually increased from 0 to 24 h of culture, and decreased. Overexpression of ACADL promoted differentiation of subcutaneous adipocytes in goat and inhibited their differentiation after interference. Conclusion: So, we infer ACADL may have an important role in positive regulating the differentiation process in goat subcutaneous adipocytes. This study will provide basic data for further study of the role of ACADL in goat subcutaneous adipocyte differentiation and lays the foundation for final elucidating of its molecular mechanisms in regulating subcutaneous fat deposition in goats.

Identification of Quantitative Traits Loci (QTL) Affecting Growth Traits in Pigs

  • Molecular genetic markers were used to detect chromosomal regions which contain economically important traits such as growth, carcass, and meat quality traits in pigs. A three generation resource population was constructed from a cross between Korean native boars and Landrace sows. A total of 240 F2 animals from intercross of F1 was produced. Phenotypic data on 17 traits, birth weight, body weights at 3, 5, 12, and 30 weeks of age, teat number, carcass weight, backfat thickness, body fat, backbone number, muscle pH, meat color, drip loss, cooking loss, water holding capacity, shear force, and intramuscular fat content were collected for F2 animals. Animals including grandparents (F0), parents (F1), and offspring (F2) were genotyped for 80 microsatellite markers covering from chromosome 1 to 10. Least squares regression interval mapping was used for quantitative trait loci (QTL) identification. Significance thresholds were determined by permutation tests. A total of 10 QTL were detected at 5% chromosome-wide significance levels for growth traits on SSCs 2, 4, 5, 6, and 8.

The Quality of Modified Atmosphere Packaged Meat from Lambs Slaughtered at 50 and 100 Days of Age

  • The aim of this study was to determine the quality of modified atmosphere (MA) packaged meat from Pomeranian rams slaughtered at 50 and 100 days of age. Determined: chemical composition, physicochemical and sensory properties of meat, and the fatty acid profile of intramuscular fat. Meat from 100-d-old lambs was characterized by a significantly higher content of dry matter, total protein and crude fat, a darker color and higher physiological maturity ($p\leq0.01$). Meat from younger lambs was marked by a better water-holding capacity ($p\leq0.05$), a lower energy value and higher concentrations of monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA). An increase in the content of dry matter, total protein and ash, and energy value was noted in MA-stored meat. Meat pH decreased over MA storage and the sensory properties deteriorated. The studied meat remained safe from microbial contamination during storage.

돼지 H-FABP 유전자의 다형성 및 경제 형질과의 연관성 구명 (A study of Association of the H-FABP RFLP with Economic Traits of Pigs)

  • 본 연구는 재래돼지와 랜드레이스를 기초축으로 이용한 F$_2$ 241두에 대해 Heart Fatty Acid- Binding Protein 유전자와 연관되어 있는 PCR- RFLP를 이용하여 그 다형성을 조사하고 돼지의 성장형질, 도체형질, 육질형질과 그 유전자형간의 연관성을 구명코자 실시하였다. H-FABP PCR-RFLP는 두 쌍의 primer에 의한 850bp와 700bp의 증폭산물을 HaeⅢ와 HinfⅠ제한효소를 사용하여 실시되었다. HaeⅢ을 이용한 PCR-RFLP 유전자형은 DD형/700+150bp, Dd형/700+400+300+ 150bp 그리고 dd형/400+300+150bp의 DNA 단편을 보였으며, Hinf1에 의한 유전자형은 HH형은 350+180+130bp, Hh형은 350+220+180+130bp, hh형/350+220+130bp의 절단된 DNA 단편을 보였다. H-FABP/HaeⅢ 유전자형 중에서 12주령 체중은 DD형에 비해 Dd와 dd형에서 유의적으로 높은 값을 보였으며(p〈0.001), 3주령 체중 (p〈0.01)과 5주령, 30주령 체중 (p〈0.05)에 경우도 Dd와 dd형에서 유의적으로 높게 관찰되었고(Table 3), 특히 ‘d’ 대립유전자가 체중과 연관성이 있음이 관찰되었다. H-FABP/HinfⅠ의 유전자형과 표현형질의 연관성을 보면 12주령, 30주령 체중 및 도체지방 과 등지방 두께에서는 hh형에 비해 HH와 Hh형에서 유의적으로 높게 나타났으며(p〈0.001), 5주령 체중과 근내지방 함량에서도 HH형에서 유의적으로 높게 관찰되었고(p〈0.05), 특히 ‘H’ 대립유전자가 체중과 도체지방, 등지방 두께 및 근내지방 함량과 연관성이 있음이 관찰되었다. 따라서 돼지성장 및 지방축적과 관련한 선발력을 높이기 위해 H- FABP PCR-RFLP(HaeⅢ & HinfⅠ)를 분자생물학적 marker로 사용할 수 있을 것으로 사료된다.

The Effect of Slaughter Season on the Fatty Acid Profile in Four Types of Fat Deposits in Crossbred Beef Bulls

  • The objective of this study was to determine the effect of slaughter season on the fatty acid profile in four types of fat deposits in crossbred (Polish Holstein Friesian Black-and-White${\times}$Limousine) beef bulls. The percentage share of fatty acids was determined by gas chromatography and were divided into the following categories of fatty acids: saturated (SFAs), unsaturated (UFAs), monounsaturated (MUFAs), polyunsaturated (PUFAs), desirable hypocholesterolemic (DFAs) and undesirable hypercholesterolemic (OFAs), n-3 and n-6. Perinephric fat was characterized by the highest SFA concentrations (59.89%), and subcutaneous fat had the highest MUFA content (50.63%). Intramuscular fat was marked by a high percentage share of PUFAs and the highest PUFA/SFA ratio. The slaughter season had a significant effect on the levels of C18:3, C20:4 ($p{\leq}0.01$) and conjugated linoleic acid ($p{\leq}0.05$). There was an interaction between the slaughter season and fat type for the content of C20:4 ($p{\leq}0.01$) and C20:5 ($p{\leq}0.05$). The results of this study show that beef from cattle slaughtered in the summer season has a higher nutritional value and more health-promoting properties.
