Comparison of the fecal microbiota with high- and low performance race horses

  • Exercise plays an important role in regulating energy homeostasis, which affects the diversity of the intestinal microbial community in humans and animals. To the best of the authors' knowledge, few studies have reported the associations between horse gut microbiota along with their predicted metabolic activities and the athletic ability of Jeju horses and Thoroughbreds living in Korea. This study was conducted to investigate the association between the gut microbiota and athletic performance in horses. This study sequenced the V3 and V4 hypervariable regions of the partial 16S rRNA genes obtained from racehorse fecal samples and compared the fecal microbiota between high- and low-performance Jeju horses and Thoroughbreds. Forty-nine fecal samples were divided into four groups: high-performance Jeju horses (HJ, n = 13), low-performance Jeju horses (LJ, n = 17), high-performance Thoroughbreds (HT, n = 9), and low-performance Thoroughbreds (LT, n = 10). The high-performance horse groups had a higher diversity of the bacterial community than the low-performance horse groups. Two common functional metabolic activities of the hindgut microbiota (i.e., tryptophan and succinate syntheses) were observed between the low-performance horse groups, indicating dysbiosis of gut microbiota and fatigue from exercise. On the other hand, high-performance horse groups showed enriched production of polyamines, butyrate, and vitamin K. The racing performance may be associated with the composition of the intestinal microbiota of Jeju horses and Thoroughbreds in Korea.

16S rDNA 염기서열 분석을 통한 제주연안 소라(Turbo cornutus) 장내세균 다양성 조사 (Analysis of Intestinal Microbial Communities of Topshell (Turbo cornutus) fromCoast of Jeju Island, Korea by 16S rDNA Sequence Analysis)

  • 본 논문은 제주연안에서 채집한 소라(Turbo cornutus)의 장내세균을 분리하고 군집의 다양성을 조사하였다. 표준배지를 사용하여 순수 정체 배양 결과 MA agar에서 가장 많은 군집을 나타났다. 일반 배양 CFU값은 평균 1.8×105, 혐기 배양 CFU값은 평균 0.4×10으로 보다 적게 나타났다. 기존 표준균주와 16S rDNA sequence 유사도 비교 분석 결과 크게 4문 12과 26속 67종으로 분류되었다. 표준균주와의 상동성 범위는 93-100%로 나타났다. 소라장내세균 군집은 크게 Proteobacteria 39% (α-proteobacteria; Phyllobacteriaceae (1), Rhodobacteraceae (8) / γ-proteobacteria; Alteromonadaecae (1), Shewanellaceae (4), Vibrionaceae(12))로 가장 우점하였고, Firmicutes 34% (Bacillaceae (21), Paenibacillaceae (2)), Actinobacteria 21% (Cellulomonadaceae (1), Mycobacteriaceae (6), Nocardiaceae (4), Streptomycetacea (3)), Bacteroidetes 6% (Flavobacteriaceae (4))로 각각 나타났다. Bacillus sp., Vibrio sp.이 가장 우점 하였으며, 그 외 대부분의 분리 균주는 해양 유래 관련 균주로 나타났다. 이전 보고된 제주 연안에 서식하는 해양동물 장내세균군과 비슷한 양상을 보였다. 분리된 일부 균주가 단당류(Cellulose), 다당류(alginate, agar)분해능을 갖고 있는 것으로 나타났는데, 대부분이 해조류 유래 세균으로 소라의 섭이가 장내세균군과 관련됨을 알 수 있었다. 동시에 probiotics 기능을 갖고 있는 일부 균주도 분리되었다.

Effects of multi-strain probiotic supplementation on intestinal microbiota, tight junctions, and inflammation in young broiler chickens challenged with Salmonella enterica subsp. enterica

  • Objective: This study assessed the effects of probiotics on cecal microbiota, gene expression of intestinal tight junction proteins, and immune response in the cecal tonsil of broiler chickens challenged with Salmonella enterica subsp. enterica. Methods: One-day-old broiler chickens (n = 240) were randomly allocated to four treatments: negative control (Cont), multi-strain probiotic-treated group (Pro), Salmonella-infected group (Sal), and multi-strain probiotic-treated and Salmonella-infected group (ProSal). All chickens except those in the Cont and Pro groups were gavaged with 1×108 cfu/mL of S. enterica subsp. enterica 4 days after hatching. Results: Our results indicated that body weight, weight gain, and feed conversion ratio of birds were significantly reduced (p<0.05) by Salmonella challenge. Chickens challenged with Salmonella decreased cecal microbial diversity. Chickens in the Sal group exhibited abundant Proteobacteria than those in the Cont, Pro, and ProSal groups. Salmonella infection downregulated gene expression of Occludin, zonula occludens-1 (ZO1), and Mucin 2 in the jejunum and Occludin and Claudin in the ileum. Moreover, the Sal group increased gene expression of interferon-γ (IFN-γ), interleukin-6 (IL-6), IL-1β, and lipopolysaccharide-induced tumor necrosis factor-alpha factor (LITAF) and reduced levels of transforming growth factor-β4 and IL-10 compared with the other groups (p<0.05). However, chickens receiving probiotic diets increased Lactobacillaceae abundance and reduced Enterobacteriaceae abundance in the ceca. Moreover, supplementation with probiotics increased the mRNA expression of Occludin, ZO1, and Mucin 2 in the ileum (p<0.05). In addition, probiotic supplementation downregulated the mRNA levels of IFN-γ (p<0.05) and LITAF (p = 0.075) and upregulated IL-10 (p = 0.084) expression in the cecal tonsil. Conclusion: The administration of multi-strain probiotics modulated intestinal microbiota, gene expression of tight junction proteins, and immunomodulatory activity in broiler chickens.

Gut microbiota derived from fecal microbiota transplantation enhances body weight of Mimas squabs

  • Objective: Compared to Mimas pigeons, Shiqi pigeons exhibit greater tolerance to coarse feeding because of their abundant gut microbiota. Here, to investigate the potential of utilizing intestinal flora derived from Shiqi pigeons, the intestinal flora and body indices of Mimas squabs were evaluated after fecal microbiota transplantation (FMT) from donors. Methods: A total of 90 one-day-old squabs were randomly divided into the control group (CON), the low-concentration group (LC) and the high-concentration group (HC): gavaged with 200 μL of bacterial solution at concentrations of 0, 0.1, and 0.2 g/15 mL, respectively. Results: The results suggested that FMT improved the body weight of Mimas squabs in the HC and LC groups (p<0.01), and 0.1 g/15 mL was the optimal dose during FMT. After 16S rRNA sequencing was performed, compared to those in the CON group, the abundance levels of microflora, especially Lactobacillus, Muribaculaceae, and Megasphaera (p<0.05), in the FMT-treated groups were markedly greater. Random forest analysis indicated that the main functions of key microbes involve pathways associated with metabolism, further illustrating their important role in the host body. Conclusion: FMT has been determined to be a viable method for augmenting the weight and intestinal microbiota of squabs, representing a unique avenue for enhancing the economic feasibility of squab breeding.

분변 미생물군집 프로젝트 (Toward The Fecal Microbiome Project)

  • 차세대 염기서열 분석(next generation sequencing, NGS) 기술의 발전으로 16S rRNA의 염기서열 분석이 미생물 군집 분석의 주된 방법으로 사용되고 있다. 인간의 건강과 질병에 관여하는 미생물들을 밝혀내기 위한 인간 미생물군집 프로젝트(human microbiome project, HMP)가 최근에 완료되었다. HMP는 세균에 의해 발생하는 여러 질병들의 특성들을 밝혀내었고, 특히 장에 서식하는 세균들에 대해 많은 연구가 수행되었다. 비록 인간의 장내 세균들에 대한 연구는 잘 수행되어왔지만, 다른 가축의 장내 세균에 대한 연구는 거의 이루어지지 않았다. 본 연구에서는 가축의 분변 미생물 다양성에 관해 조사하였고, 분변미생물 생태연구의 중요성을 제시 할 것이다. 한국에서의 분변 미생물 군집 프로젝트(fecal microbiome project) 시작을 본 연구논문을 통해 보고하고자 한다.

Effects of Antibiotics on the Uterine Microbial Community of Mice

  • The gut microbiota is involved in the maintenance of physiological homeostasis and is now recognized as a regulator of many diseases. Although germ-free mouse models are the standard for microbiome studies, mice with antibiotic-induced sterile intestines are often chosen as a fast and inexpensive alternative. Pathophysiological changes in the gut microbiome have been demonstrated, but there are no reports so far on how such alterations affect the bacterial composition of the uterus. Here we examined changes in uterine microbiota as a result of gut microbiome disruption in an antibiotics-based sterile-uterus mouse model. Sterility was induced in 6-week-old female mice by administration of a combination of antibiotics, and amplicons of a bacteria marker gene (16S rRNA) were sequenced to decipher bacterial community structures in the uterus. At the phylum-level, Proteobacteria, Firmicutes, and Actinobacteria were found to be dominant, while Ralstonia, Escherichia, and Prauserella were the major genera. Quantitative comparisons of the microbial contents of an antibiotic-fed and a control group revealed that the treatment resulted in the reduction of bacterial population density. Although there was no significant difference in bacterial community structures between the two animal groups, β-diversity analysis showed a converged profile of uterus microbiotain the germ-free model. These findings suggest that the induction of sterility does not result in changes in the levels of specific taxa but in a reduction of individual variations in the mouse uterus microbiota, accompanied by a decrease in overall bacterial population density.

Administration of antibiotics contributes to cholestasis in pediatric patients with intestinal failure via the alteration of FXR signaling

  • The link between antibiotic treatment and IF-associated liver disease (IFALD) is unclear. Here, we study the effect of antibiotic treatment on bile acid (BA) metabolism and investigate the involved mechanisms. The results showed that pediatric IF patients with cholestasis had a significantly lower abundance of BA-biotransforming bacteria than patients without cholestasis. In addition, the BA composition was altered in the serum, feces, and liver of pediatric IF patients with cholestasis, as reflected by the increased proportion of primary BAs. In the ileum, farnesoid X receptor (FXR) expression was reduced in patients with cholestasis. Correspondingly, the serum FGF19 levels decreased significantly in patients with cholestasis. In the liver, the expression of the rate-limiting enzyme in bile salt synthesis, cytochrome P450 7a1 (CYP7A1), increased noticeably in IF patients with cholestasis. In mice, we showed that oral antibiotics (gentamicin, GM or vancomycin, VCM) reduced colonic microbial diversity, with a decrease in both Gram-negative bacteria (GM affected Eubacterium and Bacteroides) and Gram-positive bacteria (VCM affected Clostridium, Bifidobacterium and Lactobacillus). Concomitantly, treatment with GM or VCM decreased secondary BAs in the colonic contents, with a simultaneous increase in primary BAs in plasma. Moreover, the changes in the colonic BA profile especially that of tauro-beta-muricholic acid ($T{\beta}MCA$), were predominantly associated with the inhibition of the FXR and further altered BA synthesis and transport. In conclusion, the administration of antibiotics significantly decreased the intestinal microbiota diversity and subsequently altered the BA composition. The alterations in BA composition contributed to cholestasis in IF patients by regulating FXR signaling.

Protective effects of Bacillus subtilis against Salmonella infection in the microbiome of Hy-Line Brown layers

  • Objective: This study investigated the effects of Bacillus subtilis CSL2 (B. subtilis CSL2) administration before Salmonella challenge on the fecal microbiota and microbial functionality of Hy-line Brown (HLB) laying hens. Methods: Fecal samples were collected from control (CON), Salmonella-infected (SAL) and Salmonella-infected, probiotic-treated (PRO) groups before and after Salmonella challenge for microbiome analysis using 16S rRNA gene pyrosequencing. Results: Infection with Salmonella led to decreased microbial diversity in hen feces; diversity was recovered with Bacillus administration. In addition, Salmonella infection triggered significant alterations in the composition of the fecal microbiota. The abundance of the phylum Firmicutes decreased while that of Proteobacteria, which includes a wide variety of pathogens, increased significantly. Bacillus administration resulted in normal levels of abundance of Firmicutes and Proteobacteria. Analysis of bacterial genera showed that Salmonella challenge decreased the population of Lactobacillus, the most abundant genus, and increased populations of Pseudomonas and Flavobacterium genera by a factor of 3 to 5. On the other hand, Bacillus administration caused the abundance of the Lactobacillus genus to recover to control levels and decreased the population of Pseudomonas significantly. Further analysis of operational taxonomic units revealed a high abundance of genes associated with two-component systems and secretion systems in the SAL group, whereas the PRO group had more genes associated with ribosomes. Conclusion: The results of this study indicate that B. subtilis CSL2 administration can modulate the microbiota in HLB laying hens, potentially acting as a probiotic to protect against Salmonella Gallinarum infection.

16S rDNA 염기서열 분석에 의한 옥돔(Branchiostegus japonicus)의 장내미생물 군집의 다양성 조사 (Research on the Diversity of Intestinal Microbial Communities of Red tilefish (Branchiostegus japonicus) by 16S rDNA Sequence Analysis)

  • 본 연구는 제주연안에서 채집한 옥돔(Branchiostegus japonicus)의 장내기관으로부터 장내미생물의 분리하여 다양성을 조사하였다. 1차로 1.5% BHIA, MA, TSA 및 R2A agar 배지상 순수분리한 결과, 1.5% BHIA에서 가장 많은 colony개수를 나타냈다. 호기성, 혐기배양은 평균 $1.7{\times}10^6CFU/g^{-1}$, $1.1{\times}10^5CFU/g^{-1}$로 나타났으며 총 147개의 순수 colony가 분리되었다. 16S rDNA염기서열 분석결과 58속 74종으로 나타났으며 기본균주와 95-100%의 유사도를 나타내었다. 크게 5문(Phylum)으로 나뉘었으며, 주요 계통군으로 Proteobacteria 문이 50%로 Moraxellaceae, Rhodobacteraceae, Shewanellae, Halomondaceae, Enterobacteriaceae, Vibrionaceae, Hahellaceae, Pseudomonadaceae, Erythrobacteraceae 총 9과 35속 35종으로 우점도가 제일 높게 나타났다. Actinobacteria문은 24%, Microbacteriaceae, Intrasporangiaceae, Dietziaceae, Dermabacteraceae, Dermacoccaceae, Nocardiodaceae, Brevibacteriaceae, Propionobacteriacea 총 8과 11속 17종, Frimicutes문은 16%, Bacillaceae, Staphylcoccaceae, Planococcaceae, Streptococcaceae, Paenibacillaceae, Clostridiaceae 총 6과 8속 17종, bacteroidetes문은 6%, Cyclobacteriaceae, Flavobacteriaceae 총 2과 3속 4종을, Deinococcus-thermus문은 4%로 Deinococcaceae 단일 1과 1속 1종으로 나타났다.

Microbial Community of Healthy Thai Vegetarians and Non-Vegetarians, Their Core Gut Microbiota, and Pathogen Risk

  • Pyrosequencing analysis of intestinal microflora from healthy Thai vegetarians and non-vegetarians exhibited 893 OTUs covering 189 species. The strong species indicators of vegetarians and non-vegetarians were Prevotella copri and Bacteroides vulgatus as well as bacteria close to Escherichia hermanii with % relative abundance of 16.9 and 4.5-4.7, respectively. Core gut microbiota of the vegetarian and non-vegetarian groups consisted of 11 and 20 different bacterial species, respectively, belonging to Actinobacteria, Firmicutes, and Proteobacteria commonly found in both groups. Two species, Faecalibacterium prausnitzii and Gemmiger formicilis, had a prevalence of 100% in both groups. Three species, Clostridium nexile, Eubacterium eligens, and P. copri, showed up in most vegetarians, whereas more diversity of Collinsella aerofaciens, Ruminococcus torques, various species of Bacteroides, Parabacteroides, Escherichia, and different species of Clostridium and Eubacterium were found in most non-vegetarians. Considering the correlation of personal characters, consumption behavior, and microbial groups, the age of non-vegetarians showed a strong positive correlation coefficient of 0.54 (p = 0.001) to Bacteroides uniformis but exhibited a moderate one to Alistipes finegoldii and B. vulgatus. Only a positive moderate correlation of body mass index and Parabacteroides distasonis appeared. Based on the significant abundance of potential pathogens, the microbiota of the non-vegetarian group showed an abundance of potential pathogen varieties of Bilophila wadsworthia, Escherichia coli, and E. hermannii, whereas that of the vegetarian group served for only Klebsiella pneumoniae. These results implied that the microbiota of vegetarians with high abundance of P. copri and low potential pathogen variety would be a way to maintain good health in Thais.
