This study was conducted to assess the effects of the dietary supplementation of riboflavin (as a bile salt hydrolase [BSH] inhibitor) and Bacillus subtilis on growth performance and woody breast of male broilers challenged with Eimeria spp. Intestinal bacteria, including supplemented probiotics, can produce BSH enzymes that deconjugate conjugated bile salts and reduce fat digestion. A 3 × 2 × 2 (riboflavin × Bacillus subtilis × Eimeria spp. challenge) factorial arrangement of treatments in randomized complete block design was used. On d 14, birds were gavaged with 20× doses of commercial cocci vaccine (CoccivacR -B52, Merck Animal Health, Omaha, NE). Dietary treatment of riboflavin and B. subtilis did not affect body weight (BW), body weight gain (BWG), and feed conversion (FCR) d 0 to 14 and overall d 0 to 41. Eimeria spp challenge reduced BWG, feed intake (FI), and increased FCR between d 14 to 28, but increased BWG and lowered FCR between d 28 to 35. There were no effects of the Eimeria spp. challenge on the overall d 0 to 41 FCR and FI, but BWG was reduced. Eimeria spp. challenge increased the abdominal fat pad weight and slight woody breast incidences on processed birds on d 42. Dietary inclusion of B. subtilis and riboflavin at tested levels did not help birds to mitigate the negative impact of Eimeria spp. challenge to enhance the growth performance.

In this study, Rubus coreanus (R. coreanus) byproducts with high polyphenol content were fermented with R. coreanus-derived lactic acid bacteria (Lactobacillus plantarum GBL 16 and 17). Then the effect of R. coreanus-derived lactic acid bacteria fermented feed (RC-LAB fermented feed) with probiotics (Bacillus subtills, Aspergillus oryzae, Yeast) as a feed additive for pigs on the composition of intestinal microbes and the regulation of intestinal immune homeostasis was investigated. Seventy-two finishing Berkshire pigs were randomly allotted to four different treatment groups and 18 replicates. RC-LAB fermented feed with probiotics increased the genera Lactobacillus, Streptococcus, Mitsuokella, Prevotella, Bacteroides spp., Roseburia spp., and Faecalibacterium prausnitzii, which are beneficial bacteria of the digestive tract of pigs. Also, RC-LAB fermented feed with probiotics decreased the genera Clostridium, Terrisporobacter, Romboutsia, Kandleria, Megasphaera and Escherichia, which are harmful bacteria. In particular, the relative abundance of the genera Lactobacillus and Streptococcus increased by an average of 8.51% and 4.68% in the treatment groups and the classes Clostridia and genera Escherichia decreased by an average of 27.05% and 2.85% in the treatment groups. In mesenteric lymph nodes (MLN) and spleens, the mRNA expression of transcription factors and cytokines in Th1 and Treg cells increased and the mRNA expression of Th2 and Th17 transcription factors and cytokines decreased, indicating a regulatory effect on intestinal immune homeostasis. RC-LAB fermented feed regulates gut immune homeostasis by influencing the composition of beneficial and detrimental microorganisms in the gut and regulating the balance of Th1/Th2 and Th17/Treg cells.
다양한 먹이 조건에서 생활하는 아메리카동애등에(Hermetia illucens)는 장내 세균의 의존성을 가질 수 있다. 이 가설을 증명하기 위해 본 연구는 종령 유충의 소화관에 존재하는 세균을 분리, 동정하고 효소활성 및 항균 능력을 분석하였다. 종령 유충의 소화관은 몸 체장에 약 7 배의 길이를 나타냈다. 한 개체의 소화관 내 존재하는 세균 수는 $5.0{\times}10^6$ cfu로 98% 이상이 후장에 존재했다. 소화관에는 3 종류의 상이한 세균이 존재했고, 미생물 동정 장치는 이들이 각각 Morganella morganii, Providencia rettgeri 및 Bacillus halodurans로 동정하였다. 이들 소화관 세균을 16S rDNA 서열을 분석한 결과 이 외에 Proteus mirabilis, Providencia alcalifaciens, Providencia sp.를 검출하였다. 이들 장내세균은 항생제 내성을 보였고, 타 미생물의 성장을 억제하였다. 또한 섬유소, 지질, 단백질 및 탄수화물의 분해 능력을 보유하였다. 본 연구 결과들은 아메리카동애등에 소화관에 유용성이 높은 세균을 보유하고 있다고 제시하였다.
에센셜 오일은 자연에서 성장하거나 유기농법으로 재배된 방향성 식물의 잎, 줄기, 껍질, 꽃잎, 뿌리 등에서 추출한 방향유를 말하며, 천연 물질로써 항균, 항진균 등의 효과를 가진다. 본 연구에서는 리시안셔스(Eustoma grandiflorum (Raf.) Shinn.)를 이용하여 에센셜 오일을 추출하여 생활 속에 흔하게 존재하는 3종의 진균 Penicillium pinophilum, Chaetomium globosum, Aspergillus niger에 대한 항진균 활성을 측정하였다. 리시안셔스에서 추출한 에센셜 오일은 항진균 활성을 나타냈으며 특히, A. niger에 대해 높은 항진균 활성을 보였으며, 최소 억제 농도(MIC)의 결과는 A. niger에서 0.005 mg/ml로 나타났다. 추출된 에센셜 오일은 대표적인 10종의 병원균(Escherichia coli, Salmonella typhimurium, Klebsiella pneumonia, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Bacillus subtilis, Enterococcus faecalis, Vibrio paragaemolyticus)에 대한 항세균 효과를 측정하였으며, 디스크 확산법을 이용하여 측정하였다. 리시안셔스에서 추출한 에센셜 오일은 병원균 B. subtilis, L. monocytogenes, V. paragaemolyticus에서 강력한 항균 작용을 나타내었다. 이러한 결과는 리시안셔스에서 추출한 에센셜 오일이 장내 병원균에 대한 항생제로 사용 가능함을 나타내고 있다. 본 연구를 통해 리시안셔스 추출물이 의약품 및 화장품을 개발하는데 있어 잠재적인 항진균제 및 항세균제로 사용될 수 있을 것으로 사료된다.

Objective: This study investigated the effects of Bacillus subtilis CSL2 (B. subtilis CSL2) administration before Salmonella challenge on the fecal microbiota and microbial functionality of Hy-line Brown (HLB) laying hens. Methods: Fecal samples were collected from control (CON), Salmonella-infected (SAL) and Salmonella-infected, probiotic-treated (PRO) groups before and after Salmonella challenge for microbiome analysis using 16S rRNA gene pyrosequencing. Results: Infection with Salmonella led to decreased microbial diversity in hen feces; diversity was recovered with Bacillus administration. In addition, Salmonella infection triggered significant alterations in the composition of the fecal microbiota. The abundance of the phylum Firmicutes decreased while that of Proteobacteria, which includes a wide variety of pathogens, increased significantly. Bacillus administration resulted in normal levels of abundance of Firmicutes and Proteobacteria. Analysis of bacterial genera showed that Salmonella challenge decreased the population of Lactobacillus, the most abundant genus, and increased populations of Pseudomonas and Flavobacterium genera by a factor of 3 to 5. On the other hand, Bacillus administration caused the abundance of the Lactobacillus genus to recover to control levels and decreased the population of Pseudomonas significantly. Further analysis of operational taxonomic units revealed a high abundance of genes associated with two-component systems and secretion systems in the SAL group, whereas the PRO group had more genes associated with ribosomes. Conclusion: The results of this study indicate that B. subtilis CSL2 administration can modulate the microbiota in HLB laying hens, potentially acting as a probiotic to protect against Salmonella Gallinarum infection.
포자형성균은 사람과 동물용의 프로바이오틱 제제로서 사용되어져 왔다. 포자형성균의 낮은 pH에 대한 안정성과 위내 생육저해 환경에 생존능은 프로바이오틱 제제로서 매력적이다. 본 연구는, 한국 전통 대두 발효 식품의 종균인 BCNU 9028균주를 메주로부터 분리하였다. 생리학적, 생화학적 특성과 16S 리보좀 DNA 염기서열 분석 결과 BCNU 9028균주는 $Bacillus$에 속하는 것을 확인하였다. $Bacillus$ sp. BCNU 9028은 pH 2.5에서 92%의 생존률을 보였으며, 0.3% 담즙산에서도 저항성을 나타냈다. 그리고, 식품 병원성균과의 응집정도와 자가결합능에 의해 $Bacillus$ sp. BCNU 9028는 $Listeria$$monocytogenes$, $S.$$aureus$ 및 $E.$$coli$와 같은 식품 병원성균의 생물막형성과 부착을 저해할 수 있는 것을 확인하였다. BCNU 9028의 소수성 특성(63.3%)은 장관내 부착능이 우수할 것으로 나타났다. 특히, $Bacillus$ sp. BCNU 9028 균주는 그람양성 및 그람음성 병원성균 모두에 항균력을 갖는 것을 확인하였다. 이상의 결과로 $Bacillus$ sp. BCNU 9028균주가 프로바이오틱스로서 이용가능성이 있음을 제시하였다.
Ulcerative colitis (UC) is an inflammatory bowel disease (IBD) that is currently difficult to treat effectively. Both Bacillus natto (BN) and ginseng-soluble dietary fiber (GSDF) are anti-inflammatory and helps sustain the intestinal barrier. In this study, the protective effects and mechanism of the combination of B. natto JLCC513 and ginseng-soluble dietary fiber (BG) in DSS-induced UC mice were investigated. Intervention with BG worked better than taking BN or GSDF separately, as evidenced by improved disease activity index, colon length, and colon injury and significantly reduced the levels of oxidative and inflammatory factors (LPS, ILs, and TNF-α) in UC mice. Further mechanistic study revealed that BG protected the intestinal barrier integrity by maintaining the tight junction proteins (Occludin and Claudin1) and inhibited the LPS/TLR4/NF-κB pathway in UC mice. In addition, BG increased the abundance of beneficial bacteria such as Bacteroides and Turicibacter and reduced the abundance of harmful bacteria such as Allobaculum in the gut microbiota of UC mice. BG also significantly upregulated genes related to linoleic acid metabolism in the gut microbiota. These BG-induced changes in the gut microbiota of mice with UC were significantly correlated with changes in pathological indices. In conclusion, this study demonstrated that BG exerts protective effect against UC by regulating the LPS/TLR4/NF-κB pathway and the structure and metabolic function of gut microbiota. Thus, BG can be potentially used in intestinal health foods to treat UC.
Bacillus polyfermenticus SCD which is commonly called as Bisroot (equation omitted) has been appropriately used for the treatment of long-term intestinal disorder's. This strain strongly inhibited against methicillin resistance Staphylococcus aureus (MRSA) and various pathogenic microorganisms. Effects of B. polyfermentius SCD administration on death rates and egg-laying rates in two groups of hens were investigated. This strain decreased the death rates of two groups by 16.26% and 11.72%, respectively. Also this strain increased the egg-laying rates of those 2.74% and 2.66%, respectively Clinical tests of B. polyfermenticus SCD administration to healthy adults showed not adverse effects but decreased glyceride concentration from 154.52 mg/dl to 135.41 mg/dl after two week administrations.
가금티프스는 가금류에 Salmonella gallinarum이 원인균이 되어 발병하는 양계산업에 막대한 지장을 주는 질병이다. 가금티프스를 억제하기 위한 생균제 개발을 위한 목적으로써 가금티프스 원인균인 Salmonella gallinarum의 생육을 저해시킬 수 있는 길항균주를 토종닭의 내장으로부터 분리하여 생육특성과 길항물질 생산성을 조사하고, 이 균주를 분류학적으로 동정하였다. 분리된 길항균주는 Bacillus amyloliquefaciens와 98% 상동성을 나타내어 최종적으로 Bacillus amyloliquefaciens Y3로 명명하였다. 0.3% maltose, 0.2% $NH_4Cl,\; 37^{\circ}C$ pH 7에서 균생육 및 길항물질의 생산능이 가장 우수하였으나 장내 담즙에 대한 내성은 크게 나쁘지 않을 것으로 확인되어졌다. 생산된 길항물질을 추정하여 본 결과 분자량이 10,000보다 작은 저분자물질이었으며 $80^{\circ}C$에서 20분간 열처리한 후에도 80%의 활성을 유지하는 내열성 물질임을 확인할 수 있었다. 향후 선발되어진 Bacillus amyloliquefaciens Y3가 생산해내는 길항물질에 대한 연구와 개량을 통하고 장내 정착성 실험을 거쳐 우수균주로 확인되면 양계산업에 사용될 우수한 생균제로 개발이 가능할 것이라 생각된다.
정장작용, 면역체계 조절, 알레르기 저감, 콜레스테롤 경감 등 여러 기능적 효능이 알려진 프로바이오틱스 건강기능 식품이 증가하고 있다. 본 연구에서는 기능성 커피 상품개발을 위하여 커피 배합에 최적인 probiotic 균주를 선발하고자 하였다. 국내외 유통 중인 동결건조커피에서 분리한 균주 1종, 발효유스타터로 사용되는 균주 8종, Bifidobacterium 1종, Bacillus coagulans 1종으로 동결건조 후 생존율, 커피 음용 환경 생존율, 내산성 및 내담즙성을 측정하였다. Bacillus coagulans는 초기균수 $2.4{\times}10^7cfu/g$에서 동결건조 후에도 $2.0{\times}10^7cfu/g$으로 높은 생존율을 나타냈고, 커피의 음용 환경, 내산성, 내담즙성 테스트에서도 대조균인 Lactobacillus casei 균에 비하여 우수한 생존율을 나타냈다. 특히, 내산성 테스트에서는 pH 1, 120분 조건에서 Bacillus coagulans이 대조군에 비하여 3.8배 높은 생존율을 나타냈다. 이에 포자형성을 특징으로 하는 유산생성 Bacillus coagulans 균제제를 기능성 커피 배합 및 상품화에 적합한 probiotic 균주로 판단하였다.
