Effects of HACCP Implementation on an Industry Foodservice Operation in Daegu (대구지역 사업체급식소에 대한 HACCP 적용 효과)

  • The purpose of this study was to evaluate the effects of HACCP implementation. The HACCP education was provided twice within one month to 20 employees of a contracted food service operation in Daegu. Critical control points (CCP) were determined based on food preparation processes: non-heating, preparation after heating, and heating. We evaluated the effects of HACCP implementation by checking microbiological quality, time and temperature at each of the CCPs during the receiving, preparation, cooking, and serving stages. After HACCP implementation, the biggest changes in microbiological qualities were in heated foods. At the cooking and serving stages, the microbiological qualities of heated foods improved to the standard levels. HACCP education helped employees ensure that the internal temperatures of the heated foods were kept higher than the standard (74 $^{\circ}C$) and the food holding temperature avoided the dangerous zone (5-6$0^{\circ}C$), thus lowering microbiological levels. At the serving stage, the microbiological levels of utensils also improved after HACCP education. This result strongly suggests that it is essential to educate employees in managing the temperature to treat foods safely. However, HACCP education didn't affect the microbiological levels of non-heated foods and foods prepared after heating, which continued to be higher than the standard. The reason for this was that poor microbiological quality seasonings were added to those types of foods. This indicates that seasoning factories as well as food service operations should implement HACCP to reduce hazards.

The Effects of Dietary Caloric Distribution on Insulin Secretion and Insulin Resistance in Sprague Dawley Rats (백서에서 식이내 열량 영양소의 배분이 인슐린 분비능과 인슐린 저항성에 미치는 영향)

  • The prevalence of type 2 diabetes mellitus has been rapidly increased in parallel with the westernization of eating behavior in Korea. Increased consumption of animal fat and simple sugar can be potential contributors for insulin resistance. The purpose of the study was to determine whether Western-(WD) and Korean-style(KD) diets altered insulin secretion and insulin resistance in male Sprague Dawley rats. Rats weighing 98$\pm$5g were provided by KD(77 En% of starch, 5 En% of corn oil and 13 En% of gluten plus 5 En% of casein), WD(42 En% of starch, 40 En% of butter and 18% of casein) or control diet(62 En% of starch, 20 En% of corn oil and 18% of casein) for 12 weeks. Body weights were lower in KD compared to WD. Fasting blood glucose levels were not different among diets. Insulin secretion from the beta cells was higher by 2.2$\pm$0.4 folds in WD than KD at baseline. In hyperglycemic clamp insulin secretion was higher in WD than KD and CD. Whole body glucose disposal rates referred to the state of insulin sensitivity were lowest in WD among groups. Glycogen deposits in soleus and quadriceps muscles were lowest in WD among all groups, but their triglyceride contents were highest. GLUT4 contents and glycogen synthase were lowest in WD in both muscles. In conclusions, westernization of diets needed more insulin to normalization of blood glucose levels due to increased insulin resistance. Thus, WD would lead to increased prevalence of diabetes mellitus when increased insulin resistance could not be compensated by insulin secretion in the case of elevated blood glucose levels. Effects of Branched Chain Amino Acids Added to a Diet on the Liver Regeneration in the Partial Hepatectomized Rat (분지쇄(分枝?)아미노산(酸)이 재생간(再生肝) 흰쥐에 미치는 영양학적(營養擧的) 효과(?果))

  • The effects of branched chain amino acids added to a diet on changes in the body weight with or without liver, moist and dry liver weight, protein, DNA, $^3H-thymidine$ incorporation into DNA, ana mitotic index of regenerating liver were studied in partial hepatectomized rat. Experimental diet was a 14.63% casein diet supplemented with 1.49% L-leucine, 0.90% L-isoleucine and 0.98% L-Valine, and control diet was an 18.0% casein. In both diets, 2.54% nitrogen were included. Rats fed experimental diet were significantly increased body weight with or without liver 7 days, and regenerated weight of dry liver and an index of liver regeneration 5 days after partial hepatectomy. Mitotic index, contents of protein and DNA increased in regenerating liver after partial hepatectomy was higher in experimental diet group. This results suggest that branched chain amino acids have an benefitial effect on whole body as well as liver regeneration after partial hepatectomy in rat.

  • In late December 2013, the Ebola virus emerged from West Africa. The outbreak started in Guinea and rapidly spread to Liberia and Sierra Leone. Initially, the virus is spread to the human population after contact with infected wildlife and then spread person-to-person through direct contact with body fluids such as blood, sweat, urine, semen, and breast milk. The Ebola virus infects endothelial cells, mononuclear phagocytes and hepatocytes. It causes massive damage to internal tissues and organs, such as blood vessels and the liver, and ultimately death. Most tests for the virus RNA rely on a technology called reverse-transcriptase polymerase chain reaction (RT-PCR). While this method is highly sensitive, it is also expensive, requiring skilled scientists, and delicate power supplies. The strip analytical technique (enzyme-linked immunosorbent assay or ELISA) detects antigens or antibodies to the Ebola virus. This test is cheap and does not require electricity or refrigeration. Despite ongoing efforts directed at experimental treatments and vaccine development, current medical work on the Ebola viral disease is largely limited to supportive therapy. Thus, rapid and reliable diagnoses of the Ebola virus are critically important for patient management, infections, prevention, and control measures.

Simultaneous Determination of Pesticides in Water Using a GC/MS Coupled with Micro Extraction by Packed Sorbent (MEPS-GC/MS를 이용한 농약류 동시 수질분석)

  • This study established an analytical method to simultaneously determine six organophosphorous pesticides [methyldemetone-S, diazinon, fenitrothion, parathion, phentoate, and O-ethyl O-(4-nitrophenyl) phenylphosphonothioate (EPN)] and carbaryl in water using a gas chromatography/mass spectrometry (GC/MS) system coupled with on-line micro extraction by packed sorbent (MEPS) and programmed temperature vaporizer (PTV) injector. Polystyrene divinylbenzene (PDVB) was used as a sorbent of MEPS. The effects of elution solvents, pH, elution volume and draw-eject cycles of samples on sample pretreatment process were investigated. Also, quality assurance and quality control (QA/QC) and the recovery of the pesticides in environmental samples were evaluated. The elution was performed using $30{\mu}L$ of a mixed solvent (acetone : dichloromethane = 80 : 20 (v/v)). Sample pretreatment processes were optimized with seven cycles of draw-eject of sample (1 mL) spiking an internal standard and sulfuric acid. At lower pH, the analytical sensitivity of diazinon decreased, but that of carbaryl increased. The method detection limit and the limit of quantification for this method were 0.02~0.18 and $0.08{\sim}0.59{\mu}g/L$, respectively. The method precision and accuracy were 1.5~11.5% and 83.3~129.8%, respectively, at concentrations of $0.5{\sim}5.0{\mu}g/L$. The recovery rates for all the pesticides except carbaryl in various environmental samples ranged 75.7~129.3%. The recovery rate of carbaryl in effluent sample was over 200% whereas carbaryl in drinking water, groundwater, and river water were in the acceptable range.

Treatment of Obese Women with Low Calorie Diet, Aerobic Exercise and Behavior Modificaiton (저열량균형식, 운동 및 행동수정에 의한 비만여성의 치료에 관한 연구)

  • A multidisciplinary weight control program was conducted for obese women. The major components of the program included low calorie diet therapy, exercise, behavior modification and nutritional education and counseling. Sixteen healthy volunteers in excess of body fat, above 30%, were enrolled in the group support program. But 5 person were dropped out in the 2nd week of treatment. During the 1st week of group orientation, individual cause of obesity was assessed through a computer program including survey of dietary intake, activity, eating habits and life styles. During the 5 weeks of treatment, 4.8kg of average weight loss was accomplished using a following program ; low calorie diet(1200kcal/day with all essential nutrients), low impact aerobic exercise(50~60% of $VO_{2max}$, 1 hour/day in a group, 3~5 days/week), behavior modification of individual life styles and eating habits causing obesity and nutritional education concerning nutrition, role of exercise such as brisk walking, importance of slow eating in regular meal pattern and internal motivation for weight reduction, health risk of obesity and rapid weight loss, weight recycling and yo-yo syndrome, etc. Nutritional conseling was conducted 3 times per week with checking self-records of foods, activity, emotional state and tiredness. Before and immediately after 5 weeks of treatment, blood pressure, fasting blood glucose, cholesterol and triglyceride were measured and comparied with paired t-test. After 5 weeks of treatment, body weight, body mass index, body fat and circumferences of waist, upper arm and hip were significantly decreased. Also LDL-cholesterol was significantly decreased after obesity treatment.

Bactericidal Efficacy of Non-thermal DBD Plasma on Staphylococcus aureus and Escherichia coli (비열 유전체장벽방전 플라즈마의 포도상구균 및 대장균 살균효과)

  • Objectives: The objective of this study was to examine the effect of non-thermal dielectric barrier discharge(DBD) plasma on decontamination of Staphylococcus aureus(S. aureus) and Escherichia coli(E. coli) as common pathogens. Methods: This experiment was carried out in a chamber($0.64m^3$)designed by the authors. The plasma was continuously generated by a non-thermal DBD plasma generator(Model TB-300, Shinyoung Air tech, Korea). Suspensions of S. aureus and E. coli of 0.5 McFarland standard($1.5{\times}10^8CFU/mL$) were prepared using a Densi-Check photometer(bio $M{\acute{e}}rieux$, France). The suspensions were diluted1:1000 in sterile PBS solutions(approximately$10^{4-5}CFU/mL$) and inoculated on tryptic soy agar(TSA) in Petri dishes. The Petri dishes(80mm internal diameter)were exposed to the non -thermal DBD plasma in the chamber. Results: The results showed that 95% of S. aureus colonies were killed after a six-hour exposure to the DBD plasma. In the case of E. coli, it took two hours to kill 100% of the colonies. The gram-negative E. coli had a greater reduction than the gram-positive S. aureus. This difference may be due to the structure of their cell membranes. The thickness of gram-positive bacteria is greater than that of gram-negative bacteria. The S. aureus is more resistant to DBD plasma exposures than is E. coli. It should be noted that average concentrations of ozone, a byproduct of the DBD plasma generator, were monitored throughout the experiment and the results were well below the criteria, 50 ppb, recommended by the Korean Ministry of the Environment. Thus, non-thermal DBD plasma is deemed safe for use in hospital and public facilities. Conclusions: There was evidence that non-thermal DBD plasma can effectively kill S. aureus and E. coli. The results indicate that DBD plasma technology can greatly contribute to the control of infections in hospitals and other public and private facilities.

Limnological Study on Spring-Bloom of a Green Algae, Eudorina elegans and Weirwater PulsedFlows in the Midstream (Seungchon Weir Pool) of the Yeongsan River, Korea (영산강 중류 (승촌보)의 봄철 녹조류 Eudorina elegans 대발생과 봇물 펄스방류에 대한 육수학적 고찰)

  • This study was carried out to elucidate the development of unprecedented water-bloom caused by a single species of colonial green algae Eudorina elegans in the upstream area of the Seungchon weir located in the Yeongsan River from late April to May 2013. The Yeongsan River is typically regulated system and the waterbody is seriously enriched by both external and internal sources of nutrients. Seasonal algal outbreaks were highly probable due to various potential factors, such as the excessive nutrients contained in treated wastewater, slow current, high irradiation and temperature, in diatom (winter), green algae (spring) and bluegreen algae (summer). Spring green-tide was attributed to E. elegans with level up to $1,000mg\;m^{-3}$(>$50{\times}10^4cells\;mL^{-1}$). The bloom was exploded in the initial period of the algal development and after then gradually diminished with transporting to the downstream by the intermittent rainfall, resulting in rapid expansion of the distribution range. Although the pulsed-flows by the weir manipulation was applied to control algal bloom, they were not the countermeasures to solve the underlying problem, but rather there still was a remaining problem related to the impact of pulsed-flows on the downstream. The green-tide of E. elegans in this particular region of the Yeongsan River revealed the blooming characteristics of a colonial motile microalga, and fate of vanishing away by the succeeding episodic events of mesoscale rainfall. We believe that the results of the present study contribute to limno-ecological understanding of the green-tide caused by blue-green algae in the four major rivers, Korea.

Alleviating Effects of Euphorbiae humifusae L. Extract on the Neurotoxicity Induced by Lead (납의 신경독성에 대한 지금초 추출물의 독성경감 효과)

  • This study examined the neurotoxicity induced by lead acetate (LA) on cultured C6 glioma cells and the protective effects of Euphorbiae humifusae L. (EH) extract against LA-induced cytotoxicity. In this study, LA exhibited neurotoxicity in a dose-dependent manner compared to the control, and was determined to be highly-toxic according to the toxic criteria. The $XTT_{50}$ value of LA was calculated at a concentration of $38.6{\mu}M$ after C6 glioma cells were incubated for 72 hours in the media containing $30{\sim}50{\mu}M$ of LA, respectively. In addition, LA-induced neurotoxicity was suggested to correlate with the level of oxidative stress because vitamin E, an antioxidant, increased the cell viability damaged by LA-induced cytotoxicity. The EH extract effectively prevented cell injury from LA-induced cytotoxicity via its antioxidative effects, such as inhibitory ability of lipid peroxidation, superoxide dismutase-like activity and 1,1-diphenyl-2-picrylhydrazyl-radical scavenging activity. These antioxidative effects may result in components, such as polyphenol or flavonoids including gallic acid or quercetin. In conclusion, natural resources, such as EH extracts, may be a useful putative agent for the treatment of diseases associated with oxidative stress, such as lead neurotoxicity.

The UGT1A9*22 genotype identifies a high-risk group for irinotecan toxicity among gastric cancer patients

  • Several studies have shown associations between irinotecan toxicity and UGT1A genetic variations in colorectal and lung cancer, but only limited data are available for gastric cancer patients. We evaluated the frequencies of UGT1A polymorphisms and their relationship with clinicopathologic parameters in 382 Korean gastric cancer patients. Polymorphisms of UGT1A1*6, UGT1A1*27, UGT1A1*28, UGT1A1*60, UGT1A7*2, UGT1A7*3, and UGT1A9*22 were genotyped by direct sequencing. In 98 patients treated with irinotecan-containing regimens, toxicity and response were compared according to the genotype. The UGT1A1*6 and UGT1A9*22 genotypes showed a higher prevalence in Korean gastric cancer patients, while the prevalence of the UG1A1*28 polymorphism was lower than in normal Koreans, as has been found in other studies of Asian populations. The incidence of severe diarrhea after irinotecan-containing treatment was more common in patients with the UGT1A1*6, UGT1A7*3 and UGT1A9*22 polymorphisms than in controls. The presence of the UGT1A1*6 allele also showed a significant association with grade III-IV neutropenia. Upon haplotype and diplotype analyses, almost every patient bearing the UGT1A1*6 or UGT1A7*3 variant also had the UGT1A9*22 polymorphism, and all severe manifestations of UGT1A polymorphism-associated toxicity were related to the UGT1A9*22 polymorphism. By genotyping UGT1A9*22 polymorphisms, we could identify high-risk gastric cancer patients receiving irinotecan-containing chemotherapy, who would experience severe toxicity. When treating high-risk patients with the UGT1A9*22 polymorphism, clinicians should closely monitor them for signs of toxicity such as severe diarrhea or neutropenia.
