Single Oral Dose Toxicity Study of Aqueous Extracts of Binso-san in ICR Mice

  • Although BinSo-San(BSS), a mixed herbal formula consisted of 11 types of medicinal herbs and have been used as anti-inflammatory agent, In the present study, the acute toxicity (single oral dose toxicity) of lyophilized BSS aqueous extracts was monitored in male and female mice after oral administration according to Korea Food and Drug Administration (KFDA) Guidelines (2005-60, 2005). In order to observe the 50% lethal dose ($LD_{50}$), approximate lethal dosage (ALD), maximum tolerance dosage (MTD) and target organs, test articles were once orally administered to female and male ICR mice at dose levels of 2000, 1000, 500, 250 and 0 (control) mg/kg (body wt.) according to the recommendation of KFDA Guidelines (2005-60, 2005). The mortality and changes on body weight, clinical signs and gross observation were monitored during 14 days after dosing according to KFDA Guidelines (2005-60, 2005) with organ weight and histopathology of 12 types of principle organs. We could not find any mortality, clinical signs and changes in the body weights except for dose-independent increases of body weight and gains restricted in 1000 mg/kg of BSS extracts-dosing female group. Hypertrophic changes of lymphoid organs.thymus, spleen and popliteal lymph nodes were detectedat postmortem observation with BSS extracts dose-dependent increases of lymphoid organ weights, and hyperplasia of lymphoid cells in these all three lymphoid organs at histopathological observations. These changes are considered as results of pharmacological effects of BSS extracts or their components, immunomodulating effects, not toxicological signs. In addition, some sporadic accidental findings such as congestion spots, cyst formation in kidney, atrophy of thymus and spleen with depletion of lymphoid cells, and edematous changes of uterus with desquamation of uterus mucosa as estrus cycles were detected throughout the whole experimental groups including both male and female vehicle controls. The significant (p<0.01) increases of absolute weights of kidney and pancreas detected in BSS extracts 1000 mg/kg-treated female group are considered as secondary changes from increases of body weights. The results obtained in this study suggest that the BSS extract is non-toxic in mice and is therefore likely to be safe for clinical use. The LD50 and ALD of BSS aqueous extracts in both female and male mice were considered as over 2000 mg/kg because no mortalities were detected upto 2000 mg/kg that was the highest dose recommended by KFDA and OECD. In addition, the MTD of BSS extracts was also considered as over 2000 mg/kg because no BSS extracts-treatment related toxicological signs were detected at histopathological observation except for BSS or their component-related pharmacological effects, the immunomodulating effects detected in the present study.

Effect of Dietary Cadmium Levels on Cadmium Accumulation in Feeding Mice (식이내(食餌內) Cadmium 수준차이(水準差異)에 따른 생쥐 체내(體內)에서의 Cadmium 축적(蓄積)에 관(關)한 연구(硏究))

  • An animal experiment was performed to investigate the effects of dietary cadmium on growth rates and cadmium accumulations in internal organic tissues and blood by feeding mice with dietary $cdcl_2$ additives and/or brown rice with high cadmium content as during a 12-week feeding period. The results were as follows : Mice weights decreased with increasing levels of dietary cadmium at the end of a 9-week feeding period. The weights of mice organs in the cadmium-free feeding group were higher than in the cadmium-added groups, and the weights of mice organs did not show any significant differences among feeding groups with different levels of dietary cadmium. The concentrations of cadmium in kidney and liver were much higher than in other internal organs and blood, and the next higher concentration was in the heart. The cadmium accumulation in all internal organs and blood increased with increasing dietary cadmium levels, respectively. The ratios of cadmium accumulation in organ tissues and total blood of cadmium-added groups increased with decreasing dietary cadmium levels.

  • The purpose of this study was to investigate certain heavy metals (Hg, Pb, Cd, Cu) in 144 samples of cephalopods, to identify weekly heavy metal intakes and to evaluate potential health risks. The average concentrations in the arm, expressed in mg/kg, were: Hg 0.017 (less than the limit of detection (LOD) to 0.059), Pb 0.024 (less than the LOD to 0.092), Cd 0.030 (less than the LOD to 0.627) and Cu 2.536 (less than the LOD to 5.837). The average concentration in the internal organs, expressed in mg/kg, were: Hg 0.063 (from 0.008 to 0.543), Pb 0.579 (less than the LOD to 2.344), Cd 15.200 (from 0.654 to 75.29) and Cu 201.706 (from 2.412 to 856.4). Heavy metal concentrations were higher in the organs than in the arm. Of the four heavy metals, the ratio of internal organs to arm was highest for cadmium. The weekly intakes of Hg, Pb, Cd and Cu from Octopus minor were 0.2%, 0.08%, 0.20% and 0.00%, respectively from the arm, and 1.0%, 0.96%, 92.28% and 0.05% from the internal organs as compared to PTWI (provisional tolerable weekly intakes) established by the FAO/WHO Expert Committee for Food Safety Evaluation.

Internal Dosimetry: State of the Art and Research Needed

  • Internal dosimetry is a discipline which brings together a set of knowledge, tools and procedures for calculating the dose received after incorporation of radionuclides into the body. Several steps are necessary to calculate the committed effective dose (CED) for workers or members of the public. Each step uses the best available knowledge in the field of radionuclide biokinetics, energy deposition in organs and tissues, the efficiency of radiation to cause a stochastic effect, or in the contributions of individual organs and tissues to overall detriment from radiation. In all these fields, knowledge is abundant and supported by many works initiated several decades ago. That makes the CED a very robust quantity, representing exposure for reference persons in reference situation of exposure and to be used for optimization and assessment of compliance with dose limits. However, the CED suffers from certain limitations, accepted by the International Commission on Radiological Protection (ICRP) for reasons of simplification. Some of its limitations deserve to be overcome and the ICRP is continuously working on this. Beyond the efforts to make the CED an even more reliable and precise tool, there is an increasing demand for personalized dosimetry, particularly in the medical field. To respond to this demand, currently available tools in dosimetry can be adjusted. However, this would require coupling these efforts with a better assessment of the individual risk, which would then have to consider the physiology of the persons concerned but also their lifestyle and medical history. Dosimetry and risk assessment are closely linked and can only be developed in parallel. This paper presents the state of the art of internal dosimetry knowledge and the limitations to be overcome both to make the CED more precise and to develop other dosimetric quantities, which would make it possible to better approximate the individual dose.

Development and Evaluation of Body Surface Electric Energy Measurement System for Oriental Medical Diagnosis (한의 진단을 위한 체표에너지 측정 시스템의 개발 및 평가)

  • Objectives : It is considered not easy to diagnose the condition of acupoints or meridians. Oriental Medical instruments have rarely complied with Oriental Medical diagnostic/therapeutic theories. The aim of this study was to develop a system which measures meridian electric potential(MEP) in proportion to bio-electric charge for the purpose of Oriental Medical diagnosis. Method : To evaluate a meridian energy measurement system(MEMS), we carried out a performance test. For significance test, we conducted a clinical trial which measured the MEP of ST36(Chok-samni) on Stomach Meridian(ST). Results & Conclusions : We could evaluate the balance or the imbalance of the MEPs on the left/right ST36. Also, through the comparison of MEPs before and after meals, we could identify the correlation between the MEP and the condition of internal organs. Namely, MEP reflects the condition of acupoints/meridians and internal organs, therefore, we propose this new method for measuring and analyzing the condition of meridians and acupoints for Oriental Medical diagnosis.

  • The Present experiment was designed to investigate the effects of Soo Jeom San on the function of heart and digestive organs. And thus it was analyzed the total acidity, recovery effect, and the other various enzyme activities such as ATPase, Creatine kinase, Aspartate transaminase, and Lactate dehydrogenase. The results were obtained as follows : 1. The Total acidity decreased after Soo JeomSan administration for 6 days, however the total acidity inoreased after the drug administration for 9 days, these phenomena demonstrate that Soo Jeom San acts as a dual factor. The mechanism of decreasing the total acidity was considered to the inhibition of ATPase activity used for HCI active transport from parietal cells. 2. Soo Jeom San recovered the islets of Langerhans which was disrupted by streptozotocin. The recovery mechanism was suggested that Soo Jeom San stimulates the ${\beta}-cell$ proliferation. 3. Soo Jeom San inhibited the enzyme activities such as Creatine kinase and Aspartate transaminase, however the drug activated Lactate dehydrogenase. According to the obtained results, Soo Jeom San may be used for curing gastric ulcer and myocardiac infarction.

  • Body fluid(津液) is a general term for normal mucus in human body, including saliva, gastric juice, intestinal juice and articular fluid in joints as well as tear, running nose, sweat, etc.. The formation of Body fluid goes through two phases. First phase is digestion of food at stomach, and then evaporation and classification of energy at Triple warmer(三焦). More technically speaking, Body fluid is divided into the Jin(津) & the Aek(液). Aek is a very mild and water-like fluid, runs deep into the internal organs. Jin is a thick and sticky liquid, running shallow under the skin or in the joints of limbs. Major roles of body fluid over the body are to moisturize the internal organs, flesh, skin, etc., to soften the joints, to fill the bone marrow, and to balance Yin and Yang. This article deals with the role of body fluid and how to differentiate them, the liquid metabolism in the human body, and the formation and change of sweat, urine, tear, spit, bone marrow, etc..   • We compared and studied the results on two diagnostic machines called Yangdorack and EAV on 18 recovering paralytic patients. The results were as folIows ; 1. Patients with weak motor ability showed an average of 25% fewer units of Yangdorack and an average of 49% fewer units of EAV. 2. Deficiency/excess diagnostic results on meridian and internal organs showed 58.5% deficiency and 41.5% excess on the Yangdorack ; 67.9% of decline-degenerative activity and 32.1% of stimulus-inflarrunation activity on the EAV. 3. In the attack of meridian and internal organs in view of the five elements(五行), in rate of deficiency, decline and degeneration of the fire meridian(火經) in view of the highest rate accounts showed 19.5% on the Yangdorack and 20.4% on the EAV. 4. In comparing deficiency/excess results on the Yangdorack with results of decline-degenerative value and stimulus-inflammation value on the EAV, they both showes a 39% rate. 5. In comparing clinical syndromes on the Yangdorack and common diseases on the EAV, they showed a 72% agreement.

  • In order to investigate the effects of dietary cellulose and protein levels on chick performance, four semi-purified diets were formulated so as to contain cellulose at levels of 5% (LC) and 20% (HC) in combination with 10% (LP) and 20% (HP) protein, and fed ad libitum to 1-week-old White Leghorn male chicks for 3 weeks. There were no significant differences in feed intake, body weight gain and feed efficiency between the LC-HP and HC-HP groups. All parameters were lower in the LP groups; the HC-LP group consumed very small amount of feed and lost body weight during the experiment. The retention rates of DM, ash, nitrogen and energy were higher in the HP than the LP groups. The triglyceride concentration of carcass was lower in the HC-LP group and that of liver was higher in the LC-LP group. The carcass total cholesterol level was higher in the HC-HP group. The relative weight of most digestive organs was higher in the HP group irrespective of the cellulose level. In conclusion, the chick performance was primarily influenced by dietary protein level, and when the chicks were fed inadequate levels of protein, the low cellulose level gave a better performance than the high cellulose level.
