수종의 한약처방이 폐암세포에 미치는 영향 (Effects of Several Medicine Herb Prescriptions on Lung Carcinoma Cells)

  • Objective : In this study, the medicine herbs above were directly treated to cultured normal lung cells and lung carcinoma cells, and the effects were investigated to develope new cancer treatments with increased anti-cancer efficiency as well as decreased side-effects and to suggest more useful clinical therapies. Materials and Methods : In the experiment, WI-26 VA lung normal cell line and A-427 lung carcinoma cell line were cultured. To observe the morphological change of the treated cells, were subjected to Giemsa staining and observed under Reflected Fluorescence microscope. To examine whether cell death occurred, cells observed under Reflected Fluorescence microscope, To investigate the degree of cell death in the nucleus, cells were screened by Laser cytometry ACAS 570. Results : Samgibopye-tang(Shenqibufei-tang) stimulated not only the growth of the normal cells but also that of the carcinoma cells, Wikyeung-tang(Weijing-tang) induced morphological change such as cytoplasmic constriction in the normal cells and the carcinoma cells, but it did not show any strong inhibitory effect on the cell growth. Samso-eum(Shensu-yin) caused severe cell damage in both cell lines, Eunkyo San(Yinqiao-san) significantly damaged the nuclei and caused weak cytoplasmic constriction in both cell lines, Normal cells treated with Gilgyeung-tang(Jingeng-tang) did not show any significant morphological change while some Gilgyeung-tang(Jingeng-tang) treated carcinoma cells were observed to have a normal cell-like shape, interestingly, conclusions : As the results above, Samgibopye-tang(Shenqibufei-tang), Wikyeung-tang(Weijing-tang), and Gilgyeung-tang(Jingeng-tang) helped the growth of both cell lines, and especially Samgibopye-tang(Shenqibufei-tang) showed the best effect, However, Samso-eum(Shensuyin) and Eunkyo-san(Yinqiao-san) caused lethal damage in the normal cells and also showed strong toxicity in the carcinoma cells.

Evaluation of circulating IGF-I and IGFBP-3 as biomarkers for tumors in dogs

  • Background: Serum-based parameters are considered non-invasive biomarkers for cancer detection. In human studies, insulin-like growth factor-I and II (IGF-I and IGF-II) and insulin-like growth factor binding protein-3 (IGFBP-3) are useful as diagnostic or prognostic markers and potential therapeutic targets. Objectives: This study examined the diagnostic utility of circulating IGF-I, IGF-II, and IGFBP-3 levels in healthy dogs and dogs with tumors. Methods: The serum concentrations of these biomarkers in 86 dogs with tumors were compared with those in 30 healthy dogs using an enzyme-linked immunosorbent assay (ELISA). Results: The ELISA results showed no difference between healthy dogs and dogs with tumors in the serum IGF-II concentrations. On the other hand, there was a significant difference in the circulating IGF-I and IGFBP-3 levels between healthy dogs and dogs with tumors. The concentrations of serum IGF-I (median [interquartile range], 103.4 [59.5-175] ng/mL) in dogs with epithelial tumors were higher than those (58.4 ng/mL [43.5-79.9]) in healthy dogs. Thus, the concentrations of serum IGFBP-3 (43.4 ng/mL [33.2-57.2]) in dogs with malignant mesenchymal tumors were lower than those (60.8 ng/mL [47.6-70.5]) in healthy dogs. Conclusions: The serum IGF-I and IGFBP-3 levels can be used as diagnostic biomarkers in dogs with tumors.

Cytotoxic effects of ethanol extracts of Saussurea lappa mediated by mitochondrial apoptotic pathway

  • Saussurea lappa and Taraxacum mongolicum have been used for herbal medicinal treatments against cancers in East Asia. We performed this study to understand the molecular basis underlying the anti-tumor effects of two herbs and analyzed the effects of these medicinal herbs on proliferation and on expression of cell growth/apoptosis related molecules by using an AGS gastric cancer cell line. The treatments of Saussurea lappa dramatically reduced cell viabilities in a dose and time-dependent manner, but Taraxacum mongolicum did not. FACS analysis and Annexin V staining assay also showed that Saussurea lappa induces apoptotic cell death of AGS. Expression analyses via RT-PCR and Western blots revealed that Saussurea lappa increased expression of the p53 and its downstream effector p21/sup Waf1/, and that the both increased expression of apoptosis related Bax and cleavage of active caspase-3 protein. We also confirmed the translocation of Bax to mitochondria Collectively, our data demonstrate that Saussurea lappa induces growth inhibition and apoptosis of human gastric cancer cells, and these effects are correlated with down- and up-regulation of growth-regulating apoptotic and tumor suppressor genes, respectively.

상수도 금속관의 외면과 내면 부식속도 평가에 관한 연구 (Assessment of External and Internal Corrosion Growth Rate for Metallic Water Pipes)

  • 본 연구에서는 상수도 금속관의 관 내 외면 부식상태를 평가하고, 내 외면 부식깊이(속도)와 매설년수의 관계를 통하여 내 외면 공식성장속도, 그리고 국내의 토양특성 인자들이 관 외면부식에 미치는 영향을 평가하여 관 외면의 노후상태를 예측할 수 있는 부식속도 모델을 제안하였다. 수집된 관체의 외면 부식깊이는 평균적으로 1.38 mm, 내면은 2.13 mm이었고, 이는 수질 등 내면부식영향 인자가 토양 등 외면부식영향 인자보다 부식에 미치는 영향이 크기 때문이며, 평균 공식성장 속도도 내면이 외면보다 두 배정도 빠른 것으로 나타났다. 외면부식에 영향을 주는 각 토양과 외면부식속도와의 상관성은 높지 않았으나, 모든 토양항목을 포함하여 회귀분석을 통하여 수립된 외면부식속도 예측모델은 기존 모델과 비교하여 더 높은 상관성을 보였다.

  • Angiogenesis is a fundamental process in reproduction and wound healing. Under these condition, neovascularization is tightly regulated. Unregulated angiogenesis may lead to several angiogenic diseases, and is thought to be indispensible for solid tumor growth and metastsis. The construction of new vascular network is a multistep cascade involving basement membrane degradation, endothelial cell proliferation, endothelial cell migration, and tube formation. Newly reported anti-angiogenic agents in oriental medical field have targeted both specific and multistep stages in the angiogenic process. From recent approach in oriental medical field with several herb medicines including activating blood flow and removing blood stasis medicine(活血化瘀藥), it may be possible in the future to develope specific anti-angiogenic agents that offer a less toxic potential therapy for cancer and angiogenic disease.

Revisiting Use of Growth Factors in Myelodysplastic Syndromes

  • Myelodysplastic syndromes (MDS) represent a heterogeneous group of clonal hematologic neoplasms characterized by morphologic dysplasia, aberrant hematopoiesis and peripheral blood refractory cytopenias. MDS is recognized to be associated with an increased risk of symptomatic anemia, infectious complications and bleeding diathesis, as well as a risk of progression to acute myeloid leukemia, particularly in patients with a high IPSS score. The advent of use of hematopoietic growth factors such as granulocyte colony-stimulating factor (G-CSF) and recombinant erythropoietin (EPO) has improved symptoms in MDS patients in addition to some data that suggest there might be an improvement in survival. G-CSF is an effective therapeutic option in MDS patients, and it should be considered for the management of refractory symptomatic cytopenias. G-CSF and EPO in combination can improve outcomes in appropriate MDS patients such as those with lower-risk MDS and refractory anemia with ring sideroblasts (RARS). This article reviews use of growth factors for lower-risk MDS patients, and examines the data for G-CSF, EPO and thrombopietic growth factors (TPO) that are available or being developed as therapeutic modalities for this challenging disease.

일중음의 인체 전립선암 세포에 대한 성장억제 효과 (Cytotoxic Effect of Iljoong-eum on Human Prostate Cancer Cells)

  • Objectives : The study was intended to investigate whether Iljoong-eum (IJE) significantly affects proliferation and growth of prostate cancer cells. Materials and Methods : In vitro, after the treatment of DU-145 and PC-3 cells with IJE, we performed Sulforhodamine B (SRB) method. In vivo, a total of 8 male nude mice subcutaneously transplanted with the PC-3 cell line were divided in 2 groups. An experimental group was given IJE orally at a dose of 4.29ml/kg per day from the 8th to 31st day following tumor injection. All mice were observed for 31 days, and sacrificed by CO2 gas asphyxiation at the end of the experiment. The mean tumor volume and body weight of both groups were compared using Student's t-tests. Results : In vitro, IJE inhibited significantly proliferation and growth of DU-145 cells and PC-3 cells. In vivo, IJE inhibited significantly proliferation and growth of PC-3 cells xenografted into athymic nude mice. Conclusions : Our data has shown that IJE is effective in suppressing the growth rate of prostate cancer cells.

  • Epithelial growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) have been widely used for non-small-cell lung cancer patients. Its untoward cutaneous effects are largely well known and developed in many patients treated with EGFR TKIs. However trichomegaly of eyelash is rarely reported. Although trichomegaly is not a drug-limiting side effect, it could be troublesome of continuing the treatment because of cosmetic issue or eyeball irritation by long eyelashes. Therefore clinicians are needed to pay attention to this uncommon effect. We herein describe erlotinib induced trichomegaly of eyelashes in a woman with adenocarcinoma of the lung.

마우스 폐암 세포에서 Insulin-Like Growth Factor-I (IGF-I) 및 IGF Binding Protein (IGFBP)의 역할 (The Role of Insulin-like Growth Factor I(IGF-I), and IGF Binding Protein (IGFBP) in Mouse Lung Cancer Cells)

  • 연구배경 : 세포성장에 관여하는 여러 growth factor중 IGF-I은 IGF-IR와 결합하여 세포증식을 유발하는 mitogen으로 알려져 있다. 대부분의 정상세포 및 암세포는 IGFBPs을 분비하는데 이들은 IGF-I과 결합하여 IGF-I의 세포증식 효과를 증가 혹은 억제시킨다. 마우스 폐암세포주 (3LL)에서 IGF-I이 세포성장에 미치는 효과를 보고, 3LL 세포에서 분비되는 IGFBP을 추출 확인하고, IGFBPs이 세포 성정에 미치는 영향을 관찰하였다. 방 법 : 마우스 폐암세포주 (3LL)를 이용하여, IGF-I을 투여하여 세포성장을 MTT assay로 측정하였고, 3LL에서 분비되는 IGFBP을 추출하여 Western ligand blot 및 western immunoblot으로 확인하였다. 또한 분비된 IGFBP이 세포성장에 미치는 영향을 보기위해, anti-IGFBP antibody을 첨가하여 이의 기능을 억제하여 세포성장에 관한 기능을 관찰하였다. 결 과 : IGF-I은 serum free media에서 3LL 세포성장을 증가시켰다. 3LL 세포는 IGFBP-4를 생성하는 것을 확인하였고, anti-IGFBP-4 antibody를 첨가시 세포성장이 증가된 소견이 관찰되어 IGFBP-4는 세포증식을 억제하는 기능을 가지고 있음을 간접적으로 알 수 있었다. 결 론 : 이상의 실험 결과로 3LL 마우스 폐암세포에서 IGF-I은 세포성장을 증가시키며, 3LL에서 생성된 IGFBP-4는 세포증식을 억제하는 기능을 가지고 있다는 것을 관찰하였다. 향후 IGF-I과 IGFBPs이 암 성장에 미치는 기전과 임상적 적용에 대한 연구가 필요하리라 생각된다.

$CaF_{2}$ 결정의 annealing시 내부 bubbles의 거동 (The behaviour of the internal bubbles in $CaF_{2}$ crystals during the annealing process)

  • 결정입계를 갖는 $CaF_{2}$ 결정을 annealing할 때, 내부에 존재하는 bubbles의 거동을 결정학적으로 분석하였다. 이들 bubbles는 결정 내에서 일어나는 slip 현상처럼 $CaF_{2}$ 결정구조에 의존하여 특정한 방향으로 배향되어 있음이 확인되었다. 또한, AFM(atomic force microscope)으로 분석한 결과, 이들의 움직임은 negative grain growth에 의한 S-surface를 형성하고 있음이 관찰되었다.
