상엽(桑葉) 추출물의 LPS로 유도된 RAW 264.7 세포에서의 항염증 효과 (Inhibitory effect of Mori Folium ethanol extract on pro-inflammatory mediator in lipopolysaccharide - activated RAW 264.7 cells)

  • Objectives : Mori Folium is one of the traditional medicinal herb. It was commonly used for sericulture in the world and has been traditionally administered as natural therapeutic agent for the treatment of filariasis, diabetes and dropsy in East Asia. This study investigated an anti-inflammatory potential of Mori Folium ethanol extract (MFE). Methods : We examined the effects of MFE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in a murine macrophage cell line, RAW 264.7. Results : MFE inhibited production of NO and $PGE_2$ in a dose dependent manner and also decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2, interleukin (IL)-1, IL-6 and tumor necrosis factor-${\alpha}$. As a plausible molecular mechanism, increased degradation of I-${\kappa}B{\alpha}$ and phosphorylation of I-${\kappa}B{\alpha}$, NF-${\kappa}B$ and MAP kinases by LPS were partly blocked by MFE treatment. Conclusions : These results suggest that MFE has an anti-inflammatory therapeutic potential, which may result from inhibition of NF-${\kappa}B$ activation and MAPK phosphorylation, thereby decreasing the expression of pro-inflammatory genes.

Anti-inflammatory Effects of Various Mushrooms in LPS-stimulated RAW264.7 Cells

  • Mushrooms have been widely cultivated and consumed as foods and herbal medicines owing to their various biological properties. However, few studies have evaluated the anti-inflammatory effects of mushrooms. Here, we investigated the effects of mushroom extracts (MEs) on lipopolysaccharide (LPS)-induced inflammation in macrophages (RAW264.7 cells). First, we extracted MEs with either water or ethanol. Using LPS-treated RAW264.7 cells, we measured cell proliferation and NO production. Gene expression of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin (IL)-6 (IL-6), and $IL-1{\beta}$ was assessed by RT-PCR, and protein abundance of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) and phosphorylation of p65 were determined by immunoblotting. MEs prepared using both water and ethanol inhibited LPS-induced inflammation in RAW264.7 cells. Nitric oxide (NO) levels induced by LPS were reduced by treatment with MEs. Isaria japonica Yasuda water extracts and Umbilicaria esculenta (Miyoshi) Minks ethanol extracts significantly decreased the mRNA expression of inflammation-related cytokine genes including $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$. Similarly, the protein abundance of iNOS and COX-2 was also decreased. The phosphorylation of p65, a subunit of nuclear $factor-{\kappa}B$ was at least partly suppressed by MEs. This study suggests that mushrooms could be included in the diet to prevent and treat macrophage-related chronic immune diseases.

지방간에 대한 백두구 에틸아세테이트 추출물의 억제 효과 및 기전 연구 (Study of the Suppressive Effect and Its Mechanism of Amomum Cardamomum L. on Free Fatty Acid-induced Liver Steatosis)

  • Through this study, the authors investigated the anti-steatosis effects of the Amomum cardamomum ethyl acetate fraction in free fatty acids (FFAs)-induced human hepatocellular carcinoma HepG2 cells. The ethyl acetate fraction of Amomum cardamomum (ACEA) was extracted with 70% ethanol and then the extract was evaporated using a rotary evaporator prior to sequential fractionation. Human hepatocellular carcinoma were treated with different concentrations of ACEA in the presence and absence of FFAs. To demonstrate the reactive oxygen species (ROS) scavenging activity, DCFDA level was analyzed by using in vitro assay system. Cell viability, lipid accumulation, intracellular triglycerides, malondialdehyde (MDA), liver steatosis related signaling molecules and inflammatory cytokines such as interleukin (IL)-6, 8, tumor necrosis factor-alpha ($TNF-{\alpha}$) were also investigated. As results, ACEA inhibited the FFAs-induced ROS, lipid accumulation, intracellular triglycerides, and MDA in a dose dependent manner. Treatment of human hepatocellular cells with ACEA induced the phosphorylation of 5' adenosine monophosphate-activated protein kinase (AMPK) and carnitine palmitoyltransferase I (CPT1) expression using western blot analysis. ACEA also potently suppressed the FFAs-induced inflammatory cytokines including IL-6, IL-8 and $TNF-{\alpha}$. These results suggest that the ethyl acetate fraction of Amomum cardamoum extract own inhibitory effects of liver steatosis by inhibiting ROS, lipid accumulation, intracellular triglycerides, MDA through AMPK signaling and anti-inflammatory actions.

Immuno-Modulatory Effects of Bacteriocin-Producing Pediococcus pentosaceus JWS 939 in Mice

  • Pediococcus pentosaceus JWS 939 (JWS 939) is a nonpathogenic bacteriocin-producing probiotic isolated from the duck intestine. This study assessed the immunomodulatory effects of JWS 939 and compared them with those of Lactobacillus rhamnosus GG (LGG), a well-known immune enhancer. The immune-enhancing effects of JWS 939 were measured by measuring the production of nitric oxide (NO) and cytokines in C57BL/6 mouse peritoneal macrophages. In addition, to assess the immune enhancement abilities of JWS 939, in vivo, a Listeria monocytogenes challenge mice model was used. The results showed that heat-killed JWS 939 induced more NO and interleukin (IL)-$1{\beta}$ production in mouse peritoneal macrophages than in LGG, and that oral administration of viable JWS 939 in mice increased more NO, IL-$1{\beta}$, and tumor necrosis factor (TNF)-${\alpha}$ level than in LGG in serum upon L. monocytogenes challenge. In addition, mice fed with JWS 939 had a longer survival time after lethal challenge with L. monocytogenes, and these effects were stronger than those induced by LGG. Collectively, P. pentosaceus JWS 939 is a remarkable strain that, by releasing bacteriocin and enhancing host immune responses, may have potential as a duck feed additive to suppress pathogens.

Alleviation of Atopic Dermatitis through Probiotic and Mixed-probiotic Treatments in an Atopic Dermatitis Model

  • The use of lactic-acid bacteria (LAB) is effective for preventing and curing immune disorders by activating the immune system in the digestive tract and the consequent immune response in the blood. In this study, LAB and mixed LABs were used in an atopic dermatitis (AD) mouse model. Alleviation of AD was observed based on the change in cytokine level and immunohistochemical staining. An ex vivo test showed that immunoglobulin-E and interleukin (IL)-4 levels were significantly lower in all groups treated with LAB than in the group treated with only 1-chloro-2,4-dinitrobenzene. Results of an in vivo test based on the ex vivo results showed that the scratch score decreased in all groups treated with the LAB and particularly decreased in the group treated with mixed LABs. Additionally, the T helper (Th) 1 cytokines interferon-gamma and IL-12p40 were upregulated by the LAB and mixed-LABs, whereas levels of the Th2 cytokine IL-4 were downregulated in a mouse model of AD-like skin lesions. Furthermore, hematoxylin & eosin and immunohistochemical staining of the dorsal area of the mice in each group showed that AD improved in the LAB-treated groups. These results suggest that LAB and mixed LABs inhibit the development of AD in NC/Nga mice by suppressing the Th2 cell response and increasing the Th1 cell response. Our results indicate that mixed LABs are better than LAB for treating AD-like skin lesions.

다시마섭취가 정상과 당뇨쥐의 비장세포 증식에 미치는 영향 (Effect of Sea Tangle on Proliferation of Splenocytes from Normal and Diabetic Mice)

  • To investigate the effect of sea tangle on immune function in normal and diabetic states, 10-week old ICR mice were feed control(C) and sea tangle(5) diets containing 5%(w/w) cellulose and 13.6%(w/w) dry sea tangle for 4 weeks. After 4 weeks, three quarters of mice(CD and SD) were made diabetic by intramuscular injection of streptozotocin(150mg/kg bw). On the 4th day after diabetes was apparent by urinary glucose, one third of diabetic mire(CDG and SDG) were treated with glipizide(20mg/kg bw) and the other third(CDM and SDM) with metformin (500mg/kg bw) orally. Spleen weights of diabetic mice with no hypoglycemic drug treatment appeared to be higher in the sea tangle group(SD) than in control(CD), but were not different when drugs were administered. Data on splenocyte proliferation stimulated by lipopolysaccaride from Salmonella abortus equi(0.l$\mu\textrm{g}$/ml) showed that sea tangle increased mitogen response in normal mice(C group vs S group) and appeared to have the same effect in diabetic mice with or without drug treatment. Splenocyte proliferation induced by concanavalin A(0.1$\mu\textrm{g}$/ml) also showed similar results, although there were not statistically significant. Concentration of interleukin-2(IL-2) released from splenocytes of the S group seemed higher than from the C group, but the IL-2 concentrations were not different among six diabetic groups. Results of fatty acid compositions of splenocyte phospholipids showed that diabetes reduced arachidonic acid/linoleic acid ratios and that sea tangle intake and glipizide treatments increased contents of polyunsaturated fatty acids. It is concluded that dietary sea tangle has a positive effect on splenocyte proliferation under normal condition and could have the same effect under diabetic conditions. Ovalbumin으로 유발된 아토피피부염 마우스 모델에서 자감초탕(炙甘草湯)의 항알레르기 효과 (Anti-allergic Effects of Jagamcho-tang in Ovalbumin-induced Atopic Dermatitis Mouse Model)

  • Activated T helper 2 (Th2) immune function is hallmark of various allergic diseases. We investigated the anti-allergic effect of Jagamcho-tang extract(JE) on ovalbumin(OVA)-induced atopic dermatitis mice model and OVA-stimulated splenocytes isolated from the mice. Mice were intraperitoneally injected OVA/alum solution 2 times at interval of 14 days, followed by oral administration of JE for 7 days. After administration, mice were subcutaneously injected with OVA in ear. JE treatment reduced ear swelling and infiltration of inflammatory cells in ear. Serum levels of interleukin(IL)-4 and immunoglobulins, such as total-IgE and OVA-specific IgE, were decreased in JE treated group. Furthermore, JE treatment decreased OVA-induced Th2-associated cytokines like IL-4, IL-5 and IL-13 mRNA levels in splenocytes. In conclusion, JE reduced allergic immune response via IgE production and Th2 response in OVA-sensitized mice, suggesting that JE could be useful prescription for allergic diseases including atopic dermatitis.

지질다당류로 유발한 염증성 뇌손상 동물모델에 대한 황금작약탕의 억제효과 연구 (Anti-neuroinflammatory Effects of Hwanggeumjakyak-tang on Lipopolysaccharide-induced Brain Injury Model in vivo and in vitro)

  • Objectives Hwanggeumjakyak-tang (HJT) has traditionally been used to treat gastrointestinal inflammatory diseases; however, its protective effects against neuronal inflammation are still undiscovered. Methods We investigated the anti-neuroinflammatory effects of HJT water extract on lipopolysaccharide (LPS)-stimulated BV2 mouse microglia cells. BV2 cells were treated with LPS (1 ㎍/mL) 1 hour prior to the addition of HJT. We measured cell viability using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and nitrite production using the Griess assay. We performed a reverse transcription-polymerase chain reaction assay to measure messenger RNA expression of inflammatory cytokines including interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α. Western blot analysis was performed to determine protein expression of mitogen-activated protein kinases (MAPKs) and inhibitor of nuclear factor kappa B (NF-κB)α. Results HJT inhibited excessive nitrite release in LPS-stimulated BV2 cells and also significantly inhibited inflammatory cytokines such as IL-1β, IL-6, and TNF-α in LPS-stimulated BV2 cells. Moreover, HJT significantly suppressed LPS-induced MAPK and NF-κB activation and inhibited the elevation of IL-1β, IL-6, and TNF-α in the brain of LPS-injected mice. Conclusions Our study highlights the anti-neuroinflammatory effects of HJT via MAPK and NF-κB deactivation.

측두하악장애환자에서 악관절 세척술후 관절활액의 전구염증성 Cytokines의 발현 (THE APPEARENCE OF PROINFLAMMATORY CYTOKINES IN TEMPOROMANDIBULAR JOINT DISORDERS AFTER ARTHROCENTESIS AND LAVAGE)

  • The purpose of this study is that evaluate the distribution and biological roles of TNF-a, interleukin-1${\beta}$(IL-1${\beta}$), interleukin-6(IL-6) and tissue inhibitors of metalloproteinase-1(TIMP-1) in the synovial fliud of patients with non-inflammatory chronic temporomandibular joint(TMJ) disorders in relation to pain during joint movements and magnetic resonance imaging(MRI) findings. TMJ synovial fluids aspirates were obtained from 36 patients (36 joints) with chronic TMJ disorders and from 8 controls(8 joints). Patients were divided to four groups. The control group was from healthy volunteers(8 joints), group I(18 joints) was patients with anterior disc displacement with reduction, group II(5 joints) was patients with disc displacement without reduction and group III (5 joints) was osteoarthritis. The TNF-${\alpha}$, IL-1${\beta}$ and IL-6 levels in the aspirates were determined by using an enzyme-linked immunosorbent assay and the TIMP-1 level was measured by an enzyme immunoassay. Following examinations for pain during joint movements and MRI observations, these cytokines' level and frequencies of detection were compared. The level of IL-1${\beta}$was not significant different in all groups. but the level of TNF-${\alpha}$, IL-6 and TIMP-1 were significant different among groups. The level of IL-6 and TIMP-1 were correlated to pain during movement(p<0.01) and the level of TNF-a(p<0.05). Also, the level of IL-6 was correlated to the level of TIMP-1(p<0.01). Especially, The level of the TIMP-1 level was significantly correlated to the pain during movement and showed very high levle of Pearson's correlation coefficient (r=0.833)(p<0.001). The results indicated that the TNF-${\alpha}$, IL-6 and TIMP-1 levels in the TMJ aspirates of patients with chronic TMJ disorders have been raised. Especially, IL-6 and TIMP-1 were very high levels in the patients who were degraded in the TMJ. Also, TNF-${\alpha}$, IL-6 and TIMP-1 showed the significant correlation in the chronic temporomandibular joint disorders. Therefore I suggest that these cytokines were also correlated to the pain during movement in the chronic temporomandibular joint disorders.

Interleukin-1의 기관지 투여 후 나타나는 폐세척액 내 대식세포의 수적변화에 따른 Xanthine Oxidase의 활성변화 (Increase of Alveolar Macrophages Contributes to the Enhanced Xanthine Oxidase Activity in the Bronchoalveolar Lavage Fluid of Rats Given IL-1 Intratracheally)

  • 폐포강 대식세포는 사이토카인, 유해산소 대사물을 포함한 그들이 분비하는 물질들로 인해 급성 폐손상에 있어서 직접, 간접적으로 폐손상의 초기반응에 중요한 역할을 담당하는 것으로 알려져 있다. 본 연구에서는 $interleukin-1\alpha$(IL-1)로 유도된 급성 폐손상에서 폐포강 대식세포의 역할을 알아보고자 하였다. 실험군은 대조군과 IL-1투여 후 1시간, 2시간, 3시간, 4시간 그리고 5시간군으로 나누었으며, 폐포강 대식세포와 XO와의 관계를 분석하기 위해 폐세척액 내 XO의 활성도와 폐포강 대식세포, 단핵구, 그리고 호중구의 수적 변화를 측정하였다. 그리고 각 군의 미세구조 변화를 관찰하였다. 실험 결과, 폐포강 내 단핵구의 수는 IL-1투여 후 1시간군에서 대조군과 비교하여 현저히 증가되었으며 (p<0.001), 폐포강 대식세포의 수는 IL-1 투여 2시간 후에 가장 높았고, 폐세척액 내 XO의 활성도는 IL-1 투여 후 점차적으로 증가되다가 3시간 후에 현저히 증가되었다(p<0.05). 폐포강 내 호중구의 수는 IL-1투여 3시간 후부터 뚜렷이 증가되기 시작하였다. 이러한 결과로 보아 IL-1을 기관지 내로 투여한 후 유도된 급성 폐손상에서 폐포강 대식세포에서 유리된 XO는 호중구의 축적에 의한 손상보다 더 초기단계에서 폐손상을 유도하는 인자인 것으로 추정된다.
