Growth of Endothelial Cells on Microfabricated Silicon Nitride Membranes for an In Vitro Model of the Blood-brain Barrier

  • The blood-brain barrier (BBB) is composed of the brain capillaries, which are lined by endothelial cells displaying extremely tight intercellular junctions. Several attempts at creating an in vitro model of the BBB have been met with moderate success as brain capillary endothelial cells lose their barrier properties when isolated in cell culture. This may be due to a lack of recreation of the in vivo endothelial cellular environment in these models, including nearly constant contact with astrocyte foot processes. This work is motivated by the hypothesis that growing endothelial cells on one side of an ultra-thin, highly porous membrane and differentiating astrocyte or astrogliomal cells on the opposite side will lead to a higher degree of interaction between the two cell types and therefore to an improved model. Here we describe our initial efforts towards testing this hypothesis including a procedure for membrane fabrication and methods for culturing endothelial cells on these membranes. We have fabricated a 1 $\mu\textrm{m}$ thick, 2.0 $\mu\textrm{m}$ pore size, and 55% porous membrane with a very narrow pore size distribution from low-stress silicon nitride (SiN) utilizing techniques from the microelectronics industry. We have developed a base, acid, autoclave routine that prepares the membranes for cell culture both by cleaning residual fabrication chemicals from the surface and by increasing the hydrophilicity of the membranes (confirmed by contact angle measurements). Gelatin, fibronectin, and a 50/50 mixture of the two proteins were evaluated as potential basement membrane protein treatments prior to membrane cell seeding. All three treatments support adequate attachment and growth on the membranes compared to the control.

  • 테크놀로지의 발전과 밀접한 관계 속에서 이어져온 디자인의 역사는 디지털 혁명이라는 이름으로 산업혁명에 버금가는 디자인의 혁신과 새로운 역할을 요구하고 있으며, 전통적 인 삶과 생활의 모습의 변화를 이루고 있다. 이러한 시대에 공간에 대한 다양한 새로운 개념과 체계를 이해하여야 하는데 여 기에 작용하는 주요 변인요소는 디지털기기와 생활양식에 의한 물리적 요소와 디지털 적인 공간적 추상성에 근거한 감성적 요소로 나눌 수 있다. 공간 디자인에 있어 이 두 가지 요소는 연구테마의 중심이 되고 있다. 새로운 패러다임에 맞는 디자인 기법과 방법론이 합리적으로 디자인에 도입되면 디자인의 효율과 성과를 높일 수 있으며, 이러한 측면에서 공간디자인 분야에서 다양한 시도가 진행 중이다. 본 연구는 이러한 시도들을 공간 디자인(교육)에 적용하기 위한 연구로서 공간디자인에 디지털 테크놀로지를 적용하는 다양한 방식과 접근방식을 연구하여 공간 디자인의 관점에서 가상의 공간의 의미와 역할을 규정하고 그 기능적 인 상호 작용성에 대한 분석을 주목적으로 한다. 이를 통해 이미 광범위하게 활용되고 있는 사이버스페이스와 전통적인 공간 디자인의 유기적인 통합과 발전을 모색하고자 한다.

Open Channel Block of Kv3.1 Currents by Genistein, a Tyrosine Kinase Inhibitor

  • The goal of this study was to analyze the effects of genistein, a widely used tyrosine kinase inhibitor, on cloned Shaw-type $K^+$ currents, Kv3.1 which were stably expressed in Chinese hamster ovary (CHO) cells, using the whole-cell configuration of patch-clamp techniques. In whole-cell recordings, genistein at external concentrations from 10 to $100{\mu}M$ accelerated the rate of inactivation of Kv3.1 currents, thereby concentration-dependently reducing the current at the end of depolarizing pulse with an $IC_{50}$ value of $15.71{\pm}0.67{\mu}M$ and a Hill coefficient of $3.28{\pm}0.35$ (n=5). The time constant of activation at a 300 ms depolarizing test pulses from -80 mV to +40 mV was $1.01{\pm}0.04$ ms and $0.90{\pm}0.05$ ms (n=9) under control conditions and in the presence of $20{\mu}M$ genistein, respectively, indicating that the activation kinetics was not significantly modified by genistein. Genistein $(20{\mu}M)$ slowed the deactivation of the tail current elicited upon repolarization to -40 mV, thus inducing a crossover phenomenon. These results suggest that drug unbinding is required before Kv3.1 channels can close. Genistein-induced block was voltage-dependent, increasing in the voltage range $(-20\'mV{\sim}0\'mV)$ for channel opening, suggesting an open channel interaction. Genistein $(20{\mu}M)$ produced use-dependent block of Kv3.1 at a stimulation frequency of 1 Hz. The voltage dependence of steady-state inactivation of Kv3.1 was not changed by $20{\mu}M$ genistein. Our results indicate that genistein blocks directly Kv3.1 currents in concentration-, voltage-, time-dependent manners and the action of genistein on Kv3.1 is independent of tyrosine kinase inhibition.

리차드 마이어와 쿠마 켄고의 전시공간에 나타나는 빛의 표현 특징에 관한 연구 (A Study on the Lighting Characteristics Appeared Exhibition Space by Richard Meier and Kuma Kengo's Work)

  • This study aims to comparatively analyze the characteristics of light expression shown in the exhibition spaces of architects Richard Meier and Kuma Kengo, who constructed the element of invisible light into visible forms within space, using diverse techniques and components. As for the methodology therefor, it was attempted to conduct theoretical consideration of light, to experience the spaces of light by producing diverse spaces with architectural space components, that is, space, form, material, structure, and color, and to find out what space characteristics are perceived by perceivers from the interaction of the components. As for the research scope, this study intended to identify the concepts and characteristics of light and compare light in spaces, focusing on works of Richard Meier who constructed functional and mechanical spaces based on Le Corbusier's architectural language in the West and works of Kuma Kengo who claimed regionalism architecture in the East. In particular, this study intended to comparatively analyze the methods of using materials according to the filtering of light that appears different between the two architects in case that light enters into their exhibition spaces, and the effects of light production that arise from the differences, thereby pointing out the characteristics of diverse light expressions within the spaces of modern architecture. On this, this study investigated methodological differences of the two architects, who are based on East and West thoughts, in expressing light in spaces in various forms through brightness and darkness, light transmission and shadows appearing according to the nature of light, and color and materials. It is hoped that this study will provide an opportunity for promoting better understanding about other architectural cultures as well as understanding the characteristics of light in exhibition spaces.

하이브리드 코팅 시스템으로 제조된 초고경도 Cr-Si-C-N 나노복합 코팅막의 미세구조 및 기계적 특성 (Microstructure and Mechanical Properties of Superhard Cr-Si-C-N Coatings Prepared by a Hybrid Coating System)

  • Cr-Si-C-N coatings were deposited on steel substrate (SKD 11) by a hybrid system of arc ion plating (AIP) and sputtering techniques. From XRD, XPS, and HRTEM analyses, it was found that Cr-Si-C-N had a fine composite microstructure comprising nano-sized crystallites of Cr(C, N) well distributed in the amorphous phase of $Si_3N_4/SiC$ mixture. Microhardness of Cr(C, N) coatings and Cr-Si-N coatings were reported about $\~22 GPa$ and $\~35 GPa$, respectively. As the Si was incorporated into Cr(C, N) coatings, The Cr-Si-C-N coatings having a Si content of $9.2 at.\%$ showed the maximum hardness value. As increased beyond Si content of $9.2 at.\%$, the interaction between nanocrystallites and amorphous phase was gone, the hardness was reduced as dependent on amorphous phase of $Si_3N_4/SiC$. In addition, the average coefficient of Cr-Si-C-N coatings largely decreased compared with Cr(C, N) coatings.

실시간 뇌파반응을 이용한 주제관련 영상물 쇼트 자동추출기법 개발연구 (Automatic Extraction Techniques of Topic-relevant Visual Shots Using Realtime Brainwave Responses)

  • To obtain good summarization algorithms, we need first understand how people summarize videos. 'Semantic gap' refers to the gap between semantics implied in video summarization algorithms and what people actually infer from watching videos. We hypothesized that ERP responses to real time videos will show either N400 effects to topic-irrelevant shots in the 300∼500ms time-range after stimulus on-set or P600 effects to topic-relevant shots in the 500∼700ms time range. We recruited 32 participants in the EEG experiment, asking them to focus on the topic of short videos and to memorize relevant shots to the topic of the video. After analysing real time videos based on the participants' rating information, we obtained the following t-test result, showing N400 effects on PF1, F7, F3, C3, Cz, T7, and FT7 positions on the left and central hemisphere, and P600 effects on PF1, C3, Cz, and FCz on the left and central hemisphere and C4, FC4, P8, and TP8 on the right. A further 3-way MANOVA test with repeated measures of topic-relevance, hemisphere, and electrode positions showed significant interaction effects, implying that the left hemisphere at central, frontal, and pre-frontal positions were sensitive in detecting topic-relevant shots while watching real time videos.

Host-Guest Interactions Between Macrocycles and Methylsubstituted Anilinium Ions

  • The binding characteristics and analytical applications of anilinium ion complexes with 18-crown-6 were studied by polarography and NMR. First, the electrochemical reduction of the 10 species of mono and dimethylsubstituted anilinium ion complexes with 18-crown-6 as host in methanol are examined. The addition of 18-crown-6 to anilinium guest solution the polarographic waves remain well defined but shifted toward more negative potentials, indicating the complex formation. The values of formation constants, log Κ for 10 species of methylsubstituted anilinium ion complexes with 18-crown-6 varies from 2.7 to 4.8 in methanol at $25^{\circ}C$. The stability order of complexes for 18-crown-6 is anilinilum > 4-methyl > 3,4-dimethyl > 3-methyl > 3,5-dimethyl > 2,4-dimethyl > 2,5-dimethyl > 2,3-dimethyl > 2-methyl > 2,6-dimethylanilinium ion. The steric hindrance shows significant effect. Second, Proton NMR was used to elucidate their interaction characteristics. From the results of so called NMR titration techniques, the behaviors of binding sites on complexation, and the stoichiometry and stability order of complex were obtained. And the later results show the satisfactory agreement with the quantitative values obtained by polarography. Finally, the individual determinations of anilinium ion mixtures were also accomplished by addition of 18-crown-6. In some mixtures of methyl or dimethylanilinium ions the reduction peaks of differential pulse method appeared into one unresolved wave attributed to the small difference of half-wave potential, ${\Delta}E_{1/2}$. In the presence of 18-crown-6, the polarographic waves were resolved into individual maxima because of the shift toward more negative direction by the difference of selectivity of anilinium ions with 18-crown-6. It may be concluded that quantitative analysis of methylanilinium ion mixture make possible because the half-wave potential shift by the selectivity difference due to the steric hindrance between methyl group and 18-crown-6 on complexation.

나노/마이크로 패턴 PDMS를 이용한 유방암 세포의 부착에 관한 연구 (Study on the Cell Adhesion of Breast Cancer Cells using Nano/Micro Patterning PDMS)

  • Cancer cells are different from normal cells in terms of life cycle, behavior, and growth patterns. Cancer cells can migrate freely in the body through blood vessels and lymph nodes. The cancer cells easily interact with various substrates including extracellular matrix and vessels and they can differentiate in the new environment. However, it is not well known about the adhesion preference of cancer cells on the substrate and the mechanism of their interaction. In this study, we prepared the nano-, micro-patterned substrates using E-beam lithography techniques. MCF-7 cells were tested on the substrates to find out their adhesion preference. The substrates were made by polydimethylsiloxane (PDMS) with specific patterns including pillars with a diameter of 500 nm, 700 nm, $3{\mu}m$ and $5{\mu}m$. MCF-7 cells were seeded on the substrates and incubated for 24 hours. As a result, this study clearly demonstrated that the MCF-7 cells preferred 700 nm patterning.

Analyzing Correlations between Movie Characters Based on Deep Learning

  • 인간은 사회적인 동물로서, 대화로써 정보를 얻거나 사회적인 교류를 해왔다. 대화는 두 사람 이상의 작은 모임에서 서로 말을 편하게 주고받는 것으로, 한 사람이 다른 사람에게 가지는 감성에 따라 그 말의 분위기가 달라질 수 있다. 영화에서 인물들과 인물들이 펼치는 이야기는 중요한 요소로 작용하며, 인물들 간의 관계는 이야기와 인물 간의 대사를 이해하는데 꼭 필요하다. 그러나 이런 정보를 영화에서 자동으로 추출하는 방법은 아직까지 연구되지 않아서 관객들에게 제공되고 있지 못하고 있다. 따라서, 영화 속 양상을 자동으로 분석하는 모델이 필요하다. 본 논문에서는 딥 러닝 기법을 활용하여 각 영화 등장 인물들 간의 감성을 측정하여 영화 속 인물들 간의 관계를 효과적으로 분석하는 방법을 제안한다. 제안 방법은 먼저 영화 대본으로부터 주요 인물들을 추출하고, 주요 인물들 간의 대화를 효과적으로 찾는다. 그런 다음, 주요 인물들 간의 관계를 분석하기 위하여, 감성 분석을 수행하여 전체 시간 간격 내 대사의 위치에 따라 가중치를 부여하고 점수를 수집한다. 또한, 실데이터를 이용한 실험을 통하여 제안 기법이 효과적으로 영화 등장 인물들 간의 감성을 분석할 수 있음을 보인다.

Artificial neural network for predicting nuclear power plant dynamic behaviors

  • A Nuclear Power Plant (NPP) is a complex dynamic system-of-systems with highly nonlinear behaviors. In order to control the plant operation under both normal and abnormal conditions, the different systems in NPPs (e.g., the reactor core components, primary and secondary coolant systems) are usually monitored continuously, resulting in very large amounts of data. This situation makes it possible to integrate relevant qualitative and quantitative knowledge with artificial intelligence techniques to provide faster and more accurate behavior predictions, leading to more rapid decisions, based on actual NPP operation data. Data-driven models (DDM) rely on artificial intelligence to learn autonomously based on patterns in data, and they represent alternatives to physics-based models that typically require significant computational resources and might not fully represent the actual operation conditions of an NPP. In this study, a feed-forward backpropagation artificial neural network (ANN) model was trained to simulate the interaction between the reactor core and the primary and secondary coolant systems in a pressurized water reactor. The transients used for model training included perturbations in reactivity, steam valve coefficient, reactor core inlet temperature, and steam generator inlet temperature. Uncertainties of the plant physical parameters and operating conditions were also incorporated in these transients. Eight training functions were adopted during the training stage to develop the most efficient network. The developed ANN model predictions were subsequently tested successfully considering different new transients. Overall, through prompt prediction of NPP behavior under different transients, the study aims at demonstrating the potential of artificial intelligence to empower rapid emergency response planning and risk mitigation strategies.
