Emission Detection of Mercuric Ions in Aqueous Media Based-on Dehybridization of DNA Duplexes

  • To quantify the presence of mercuric ions in aqueous solution, double-stranded DNA (dsDNA) of poly(dT) was employed using a light switch compound, $Ru(phen)_2(dppz)^{2+}$ (1) which is reported to intercalate into dsDNA of a right-handed B-form. Addition of mercuric ions induced the dehybridization of poly(dT)${\cdot}$poly(dA) duplexes to form a hairpin structure of poly(dT) at room temperature and the metal-to-ligand charge transfer emission derived from the intercalation of 1 was reduced due to the dehybridization of dsDNA. As the concentration of $Hg^{2+}$ was increased, the emission of 1 progressively decreased. This label-free emission method had a detection limit of 0.2 nM. Other metal ions, such as $K^+$, $Ag^+$, $Ca^{2+}$, $Mg^{2+}$, $Zn^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ni^{2+}$, $Cu^{2+}$, $Cd^{2+}$, $Cr^{3+}$, $Fe^{3+}$, had no significant effect on reducing emission. This emission method can differentiate matched and mismatched poly(dT) sequences based on the emission intensity of dsDNA.

  • Most data used for urban planning is aggregated by administrative district. Thus, a fundamental limit to analysing the changes of micro-geographical units exists. The object of this study is to estimate spatial development patterns based on characteristic city indicators. Gangnam, an area that was analysed, was divided into hexagon polygons. The development density and characteristic city indicators were input into each polygon. Moreover, this study analysed the influence of characteristic city indicators on development density using multinomial regression analysis. According to the results, distance between a polygon and both a road and a bus stop led to a decrease of development density in the polygon. However, distance between a polygon and a river led to an increase of development intensity. The method of this analysis and the results can be used to disaggregate the zonal data in the urban planing area.

  • The purpose of this study is to evaluate the developed dose verification program for in vivo dosimetry based on transit dose in radiotherapy. Five intensity modulated radiotherapy (IMRT) plans of lung cancer patients were used in the irradiation of a homogeneous solid water phantom and anthropomorphic phantom. Transit dose distribution was measured using electronic portal imaging device (EPID) and used for the calculation of in vivo dose in patient. The average passing rate compared with treatment planning system based on a gamma index with a 3% dose and a 3 mm distance-to-dose agreement tolerance limit was 95% for the in vivo dose with the homogeneous phantom, but was reduced to 81.8% for the in vivo dose with the anthropomorphic phantom. This feasibility study suggested that transit dose-based in vivo dosimetry can provide information about the actual dose delivery to patients in the treatment room.

Wnt/$\beta$-catenin/Tcf Signaling Induces the Transcription of a Tumor Suppressor Axin2, a Negative Regulator of the Signaling Pathway

  • Axin2/Conductin/Axil and its ortholog Axin are negative regulators of the Wnt signaling pathway, which promote the phosphorylation and degradation of ${\beta}$-catenin. While Axin is expressed ubiquitously, Axin2 mRNA was seen in a restricted pattern during mouse embryogenesis and organogenesis. Because many sites of Axin2 expression overlapped with those of several Wnt genes, we tested whether Axin2 was induced by Wnt signaling. Endogenous Axin2 mRNA and protein expression could be rapidly induced by activation of the Wnt pathway, and Axin2 reporter constructs, containing a 5.6 kb DNA fragment including the promoter and first intron, were also induced. This genomic region contains eight Tcf/LEF consensus binding sites, five of which are located within longer, highly conserved non-coding sequences. The mutation or deletion of these Tcf/LEF sites greatly diminished induction by ${\beta}$-catenin, and mutation of the Tcf/LEF site T2 abolished protein binding in an electrophoretic mobility-shift assay. These results strongly suggest that Axin2 is a direct target of the Wnt pathway, mediated through Tcf/LEF factors. The 5.6 kb genomic sequence was sufficient to direct the tissue specific expression of d2EGFP in transgenic embryos, consistent with a role for the Tcf/LEF sites and surrounding conserved sequences in the in vivo expression pattern of Axin2. Our results suggest that Axin2 participates in a negative feedback loop, which could serve to limit the duration or intensity of a Wnt-initiated signal.

  • A new open-loop signal processing technique of digital phase tracking is known to have a Potential to solve the problems in the open-loop processor such as limited dynamic range, dependence on the optical intensity fluctuations, and dependence on gain fluctuations of signal path. But new problems with digital phase tracking must be solved before it can be a useful signal processing method. In this paper, barriers to the success of the digital phase tracking such as harmonics content, phase difference, amplitude variations of the phase modulation(PM) signal, bandwidth limit of the signal path, and the implementation of the mixer, are pointed out and their effects on the performance of the signal processor are analyzed to calculate the requirements of the signal processor for $1{\mu}rad$-grade FOG.

Determination of Glucose in Whole Blood by Chemiluminescence Method (화학발광법에 의한 전혈 중의 당 정량)

  • A method for the determination of glucose in human whole blood by chemiluminescence method using a stopped flow injection system has been studied. The method is based on the differences in the chemiluminescence intensities of luminol due to the different amounts of hydrogen peroxide produced from the glucose oxidase catalyzed reaction. The enzyme reactor was prepared by immobilization of glucose oxidase on aminopropyl glass beads and the chemiluminescence from a flow cell was measured by means of an optical fiber bundle. In order to obtain the optimum experimental conditions, effects of pH for the chemiluminogenic solution and enzyme reactor, flow rate and temperature on the chemiluminescence intensity were investigated. The calibration curve obtained under optimum experimental conditions was linear over the range from $1.0{\times}10^{-1}$ mM to 7.0 mM and the detection limit was $6.0{\times}10^{-2}$ mM. The proposed method was applied to the determination of glucose in whole human blood sample and the results were compared with those obtained by an official method. The present method was also evaluated by the results of recovery experiments.

  • Methods for mixing variable types of steel fibers have been developed recently to suppress outbreak of crack or to control the width of crack and improve the load resistible capacity at the same time. On the other hand, uncertainty by complex nature of destruction dynamics of steel fiber reinforced concrete(SFRC) is included. In this study, analysis of reliance considering uncertainty of SFRC beam is done. For this, intensity limit state model was proposed. Moreover, characteristic values about almost every kinds of probability variables were collected and presented according to home and foreign references. Process of improving uncertainty from the result of experiments by Bayseian updating method is also proposed on the purpose of offering better statistical characteristic values with more data in the new future. Nonlinear distortion compensation of LD and optical intensity modulator using predistorter (전치왜곡기를 이용한 LD 및 광변조기의 비선형 왜곡 보상)

  • Nonlinear distortion introduced by the optical transmitter using a direct modulation of laser diode(LD) and an indirect modulation of optical modulator can limit considerably the performance of analog optical link system such as subcarrier multiplexing system. In this paper, we have compensated the nonlinear distortion of LD and optical modulator using electrical predistorter. In the case of LD, two-tone experiments using 877.5 MHz and 882.5 MHz have shown reduction of 19 dB in third-order intermodulation product(IMP$_3$). And in the case of optical modulator, we have achieved nearly 21 dB reduction in IMP3 through two-tone experiments using 1.8975 GHz and 1.9025 GHz. The spurious free dynamic ranges of LD and optical modulator are also improved by 5 dB/Hz$^{2}$ 3/ and 7 dB/Hz$^{2}$ 3/, respectively.

Spectrofluorimetric Determination of Pb ion in Aqueous Media Using an Optical Sensor (광센서를 이용한 수용액 중 납이온의 형광분광법적 정량)

  • A method to determine lead ion in aqueous media using an optical sensor loaded on a fluorescent optode membrane incoporating a metal ion-selective ionophore, a proton-selective chromoionophore and lipophilic anionic sites has been studied. The effects of pH and thickness of membrane on the fluorescence intensity were investigated. The effects of foreign ions such as $Na^+$, $K^+$, $Mn^{2+}$ and $Zn^{2+}$ on the determination of lead ion were also studied. The linear range in the calibration curve for the determination of lead ion was found to be 5.0${\times}10^-7$ to 5.0${\times}$$10^-3$M and the correlation coefficient in this range was -0.99107 under the optimal experimental conditions. The relative standard deviation of the blank signals was 3.0% and the detection limit of lead ion was 5.0${\times}$$10^-9$M.

Detecting Chaotic Motions of a Piecewise-Linear System in the Noisy Fields by Mean Poincare Maps (평균 포인케어맵을 이용한 Noisy Field에서의 chaos거동의 검출방법)

  • The method to distinguish chaotic attractors in the perturbed response behaviors of a piecewise-linear system under combined regular and external randomness is provided and examined. In the noisy fields such as the ocean environment, excitation forces induced by wind, waves and currents contain a finite degree of randomness. Under external random perturbations, the system responses are disturbed, and consequently chaotic signatures in the response attractors are not distinguishable, but rather look just random-like. Mean Poincare map can be utilized to identify such chaotic responses veiled due to the random noise by averaging the noise effect out of the perturbed responses. In this study, the procedure to create mean Poincare map combined with the direct numerical simulations is provided and examined. It is found that mean Poincare maps can successfully distinguish chaotic attractors under stochastic excitations, and also can give the information of limit value of noise intensity with which the chaos signature in system responses vanishes.
