Effect of KiFAY on Performance, Insulin-like Growth Factor-1, and Thyroid Hormones in Broilers

  • A comparative study was performed to investigate the efficacy of KiFAY as a feed additive on performance parameters, thyroid, and pancreatic hormone levels in broilers. Ninety birds (Vencobb 400) were randomly divided into three groups viz., Control (no DL-methionine supplementation), Treatment1 (containing added DL-methionine) and Treatment 2 (containing KiFAY and without DL-methionine supplementation). The performance parameters (weekly body weight, body weight gain, feed intake, and feed consumption ratio) were recorded and calculated during the whole study of 4 weeks. Analyses of insulin and insulin-like growth factor (IGF 1), triiodothyronine (T3), thyroxine (T4) and thyroid stimulating hormone (TSH) were performed at the end of the study. The results show that birds on supplementation of KiFAY performed significantly (p<0.001) better than other treatments. The weekly body weight, body weight gain, feed in-take and feed consumption ratio improved in KiFAY treated birds. The study found an increase in insulin and IGF1 levels (p<0.001) in KiFAY compared with the other treatments. Serum T3, T4, and TSH levels in the Treatment 2 were higher than other treatments (p<0.001). The KiFAY supplementation was able to improve performance with associated responses at a hormonal level in broilers.

Insulin-like Growth Factor-1 (IGF-1) Gene Expression Is Enhanced under Hypothermia but Depressed under Additional Ischemic Stimulus

  • There are several studies that show hypothermia improves cellular ischemia damages on experimental and clinical bases. However, its exact molecular mechanisms are unclear. In this study, we demonstrate that hypothermia induced insulin-like growth factor 1 (IGF1) gene expression, and its expression was dramatically decreased under ischemic insults. It was also demonstrated that hypothermia activated endoplasmic reticulum (ER) stress sensors especially both the phosphorylation of $eIF2{\alpha}$ (eukaryotic translation initiation factor 2 alpha) and ATF6 (activating transcription factor-6) proteolytic cleavage. However, the factors of apoptosis and autophagy were not associated with hypothermia. We suggest that hypothermia-treated IGF1 gene expression after ischemia may show a good possibility for the development of treatments and diagnostic methods in cerebral ischemic damages.

Daily Profiles of Blood Insulin, Insulin-Like Growth Factor-I, Thyroxine and Triiodothyronine in Ewes Under Three Levels of Feed Intake

  • Daily variation in the serum concentrations of insulin and insulin-like growth factor-I and in the plasma concentrations of thyroxine and triiodothyronine were evaluated in ewes fed 30%, 100% and 200% of theoretical maintenance energy requirements. The single daily meal has had significant effects (p<0.05) on almost all profiles. In general, serum or plasma hormone concentrations have increased after the meal, in particular at the two higher levels of energy intake. In the group submitted to the lowest level of energy intake, the consequences of the meal on circulating levels were almost imperceptible. The effects of feeding levels on serum or plasma concentrations have widely varied among hormones, not showing any objective pattern or relationship. Because these variations may affect the interpretation of these blood indicators, knowledge of daily profiles and of the effect of feed level must be considered. In order to maximize the diagnostic value of those indicators, the most suitable times for blood collection seem to be 16 h after the meal and (or) just before the meal. The collection 16 h after the meal apparently allows the characterization of a relatively steady metabolic state, intermediate between the close effects of food intake and the final phase of the intensification of body reserves mobilization. The collection just before the meal will give a good indication of the level of activity of those mobilization mechanisms.

The roles of PKC-δ on the regulation of insulin-like growth factor(IGF)-I and insulin-Like growth factor binding protein-3 secretion by all-trans retinoic acid in MCF-7 cell (MCF-7 cell에서 all-trans retinoic acid에 의한 insulin-like growth factor-I와 insulin-like growth factor binding protein-3 분비조절에 있어서 PKC-δ의 역할)

  • All-trans retinoic acid (AtRA) induces growth inhibition and apoptosis in a variety of tumer cells, including MCF-7 cells. Insulin-like growth factors (IGFs) system has been reported to be associated with the development of cancer. Although MCF-7 cell with AtRA is to be the major stimulus for the cell growth and apoptosis, the mechanism of insulin-like growth factor-I (IGF-I)/insulin-like growth factor binding protein-3 (IGFBP-3) system remains to be elucidated. Thus, this study was conducted to the effect of AtRA on the gene expression and level of IGF-I and IGFBP-3. In addition, we investigated the involvement of PKC-${\delta}$ on the IGF-I and IGFBP-3 secretion in MCF-7 cell. AtRA(${\geq}10^{-7}M$) decreased the IGF-1 secretion and mRNA expressions, but increased IGFBP-3 secretion and mRNA expressions in MCF-7 cells. Especially, the treatment of AtRA at 72 hours caused a significant reduction in the IGF-I secretion and mRNA expressions but increment in IGFBP-3 secretion and mRNA expressions (p < 0.05). $10^{-7}M$ AtRA activated PKC-${\delta}$ that is one among PKC-$\iota$, ${\alpha}$, ${\lambda}$ and ${\delta}$ in MCF-7 cell. Rotllerin, a PKC-${\delta}$ inhibitor, blocked AtRA-induced inhibition of the IGF-I and mRNA expressions, and increase of lGFBP-3 and mRNA expressions in MCF-7 cell. Together, AtRA inhibited the IGF-I secretion and mRNA expressions, but increased IGFBP-3 secretion and mRNA expressions in MCF-7 cell. Furthermore, AtRA-induced alteration of IGF-I, IGFBP-3 secretion, and the gene expressions were mediated via PKC-${\delta}$ activity.

  • Insulin and insulin-like growth factor-1 (IGF-1) are hormones that play an important role in the physiological regulation of metabolism, growth, and longevity in vertebrates. Likewise, insulin-like peptides (ILPs), which are structurally similar to insulin and IGF-1, are crucial in insect physiology. In this review, we present an integrated summary of insect ILPs and their receptor signaling, which has been shown to be comparable to insulin and IGF-1 receptor signaling in vertebrates based on genetic studies of Drosophila melanogaster. Additionally, we review the control of ILP synthesis and secretion in the brain in response to nutrition, as well as the ILPs' physiological role in insect metabolism. Moreover, we discuss the contribution of ILPs to growth, development, reproduction, and diapause. Finally, we consider the possibility of targeting ILP receptor signaling in pest management.

Effects of Growth Factors and Gut Regulatory Peptides on Glucose Uptake in HC 11 Mouse Mammary Epithelial Cells

  • The large and rapid changes of glucose utilization in lactating mammary tissue in response to changes in nutritional state must be largely related by external signal of insulin. This also must be related with the quantity and composition of the diet in vivo. To characterize the mode of growth factors and gut regulatory peptides with insulin, in vitro experiment was conducted with HC11 cells. All the growth factor alone and the combinations of growth factors significantly (p<0.05) increased in glucose uptake. Insulin, EGF and IGF-1 exhibited a stimulation of glucose uptake for at least 24 h. Furthermore, the highest (p<0.05) synergistic effect was shown in EGF plus IGF-1 and the second synergistic effect in insulin plus EGF while no synergistic effect was found between insulin and IGF-1. However, the gut regulatory peptides neither potentiated nor inhibited the action of insulin on glucose uptake. Although growth factors did not modulates glucose uptake via increasing the rate of translation of the GLUT1 protein, RT-PCR analysis indicated that the growth factors significantly (p<0.05) increased the expression of GLUT1. The growth factors are therefore shown to be capable of modulating glucose uptake by transcription level with insulin in HC 11 cells.

  • 골감소증 환자의 혈청중에 존재하는 IGFBP-5의 존재와 변화를 검토한 결과, 골감소즈 환자에서는 정상 대조군에 비해 IGFBP-5가 감소하였는데 이 변화는 IGFBP-5의 분해효소가 작용하기 때문인 것으로 나타났다. IGFBP-5에 작용하는 단백질분해효소 저해제중 metallo계인 EDTA 및 1,10-phenanthroline과 seriner계인 aprotinin, heparin, heparin cofactor 2(HC2), heparine+HC2가 IGFBP-5에 대해 저해효과가 크므로 metalloprotease이면서 serine protease의 성질을 가지는 효소들이 IGFBP-5에 작용하였다. IGF-I과 IGF-II 그리고 insulin은 효소 활성에 아무런 영향이 없었다. IGFBP-5의 zymography에서 정상인과 골감소증 환제어서 180 kDa 크기의 band가 나타났고, gelatin zymography에서 정상 대조군의 경우 66 kDa과 97 kDa 정도의 band가 확인되었고 골감소증 환자의 경우는 69 kDa의 band가 확인되었다.

Effect of Insulin, Transferrin and Platelet-Derived Growth Factor Supplemented to Synthetic Oviduct Fluid Medium on In Vitro Development of Bovine Embryos Matured and Fertilized In Vitro (합성난관배양액에 첨가된 Insulin, Transferrin 및 Platelet-Derived Growth Factor (PDGF)가 소 수정란의 체외발육에 미치는 영향)

  • In vitro development of bovine embryos is affected by many factors such as energy substrates, amino acids, and some growth factors. It has been reported that mRNA of insulin, PDGF and their receptors are detected in cow embryos, and that some chelating agents such as EDTA and transferrin have beneficial role on mouse and bovine embryos. The author hypothesized that insulin, transferrin arid PDGF added to a culture medium increase in vitro development of bovine embryos by chelating toxic substance(s) or increasing cell growth and metabolism. Immature oocytes from slaughtered ovaries of Holstein cows and heifers were matured for 24 hours in a TCM199 containing 10% fetal calf serum, FSH, LH and estradiol with granulosa cells in vitro. Matured oocytes were coincubated with sperm for 30 hours in a modified Tyrode's medium (IVF). Embryos cleaved to 2- to 4-cell at 30 hours after IVF were selected and cultured in a 30-$\mu$l drop of a synthetic oviduct fluid medium (SOFM) containing 0.8% BSA, Minimum Essential Medium essential and non-essential amino acids, and insulin, transferrin or PDGF for 9 days. Supplementation of a SOFM with insulin, and /or transferrin did not increase develop-mental rate to expanding and hatching blastocyst of 2- to 4-cell bovine embryos compared with control. The highest developmental rate to hatching blastocyst was shown when PDGF was added at the concentration of 10 ng /ml among the supplementing doses tested in the present study (p<0.05). Addition of PDGF without insulin to a SOFM could not increase embrye development, but combined addition of PDGF with insulin significantly increased (p<0.05) embryo development to hatching blastocyst (50%) compared with control (38%). In conclusion, insulin and PDGF supplemented to a SOFM may act synergistically and have beneficial effect on in vitro development of 2- to 4-cell bovine embryos matured and fertilized in vitro.

Metabolic Regulation of Insulin-like Growth Factor-1 Expression (쥐의 insulin-like growth tractor리 유전자 발현의 대사조절기전에 관안 연구)

  • The present study was aimed at investigating the metabolic regulation of insulin-like growth factor-I(IGF-I) expression in fasting animals. The expression of IGF-I gene was determined by a solution hybridization/RNase protection assay using total RNA from control, 4d-fasting, and 2d-fasting-refed rats. The levels of IGF-I transcripts were reduced in 4d-fasting than in control by decreasing its transcriptional rate, which was measured through nuclear nun-on assay. DNase I footprinting, which was performed using nuclear extracts from fasting rat, demonstrated protein binding to a sequence that extended from +179 to +210 (termed region B). These data suggest that the expression of IGF-I is transcriptionally regulated through DNA-liver enriched protein binding in a sequence which is located downstream from major transcription initiation site of IGF-I gene.
