Comparision of Cation-Anion Balance in Leaves on the Stalk Positions of Young Tobacco Plant (담배식물(植物)의 엽서별(葉序別) 이온균형(均衡) 비교(比較))

  • These studies were investigated ionic balance of leaves from different stalk positions of tobacco plant (cv. NC82) grown on pots supplied with nitrate salts solution in the phytotrone. The results of studies are summerized as follows: 1. In comparision with the leaves along the stalk positions, lower position contained higher proportion of total cation and total anion including organic acid, but their concentration lessened gradually at the upper stalk. The ratio in amounts of inorganic cations and anions in leaf laminae depended on their stalk positon, being more than three times in the fifth, but becoming about 1.3 in the tenth. 2. Inorganic cations in the laminae were mostly $K^+$, $Ca^{{+}{+}}$, and $Mg^{{+}{+}}$, comprising about 80% of total with the first two regardless of their stalk positions. Among these two ions $Ca^{{+}{+}}$ tended to become higher at lower stalks, while $K^+$ was higher in younger and became lower when leaves got older and/or more number of leaves were emerged. 3. Total cations were balanced with inorganic and organic acid anions. Composition of inorganic and organic anions were different along stalk positions, ionic balance was governed by organic acid anions dominantly in older leaves, but by inorganic anions in the younger leaves. 4. The discrepancy in the balance between total cations and anions was more pronounced in the older leaves containing higher amount of cations and organic acids. It was suggested that discrepancy was due to precipitation of oxalate as Ca-salt which was not dissolved in the methylation solution of measurement employed.

  • Among the major nutrients, phosphorus is by far the least mobile and available to plants in most soil conditions. A large portion of soluble inorganic phosphate applied to soil in the form of phosphate fertilizers is immobilized rapidly and becomes unavailable to plants. To improve the plant growth and yield and to minimize P loss from soils, the ability of a few soil microorganisms converting insoluble forms into soluble forms for phosphorus is an important trait in several plant growth-promoting microorganisms belonging to the genera Bacillus and Pseudomonas and the fungi belonging to the genera Penicillium and Aspergillus in managing soil phosphorus. The principal mechanism of solubilization of mineral phosphate by phosphate solubilizing bacteria (PSB) is the release of low molecular weight organic acids such as formic, acetic, propionic, lactic, glycolic, fumaric, and succinic acids and acidic phosphatases like phytase synthesized by soil microorganisms in soil. Hydroxyl and carboxyl groups from the organic acids can chelate the cations bound to phosphate, thereby converting it into soluble forms.

Influence of Different pH Conditions and Phosphate Sources on Phosphate Solubilization by Pantoea agglomerans DSM3493

  • Pantoea agglomerans DSM3493 was isolated from green house soils collected from Chungchugnam-do province, Gongju-Gun area in South Korea and phosphate solubilization and organic acid production of the strain were assessed using three types of insoluble phosphate sources (Ca phosphate, Fe phosphate and Al phosphate) under three different pH conditions (7, 8 and 9). The highest Ca phosphate solubilization ($651{\mu}g\;mL^{-1}$) was recorded at pH 7 followed by pH 8 and 9 (428 and $424{\mu}g\;mL^{-1}$ respectively). The solubilization rate was found to be 80.4, 98.1 and $88.7{\mu}g\;mL^{-1}$ (for Fe phosphate containing medium) and 9.3, 12.1 and $29.8{\mu}g\;mL^{-1}$ (for the Al phosphate containing medium) respectively at pH 7, 8 and 9. Though increasing pH of the medium caused reduction in the rate of solubilization of Ca phosphate, solubilization of Fe and Al phosphates enhanced with increasing pH. By contrast, the highest amount of organic acid was produced with Ca phosphate while the lowest was recorded with the presence of Al phosphate. Among the organic acids, gluconic acid production was found to be the highest, followed by oxalic acid and citric acid regardless the source of phosphate. Results can thus be concluded that the production of organic acids appears to play a significant role in the inorganic phosphate solubilization.

  • Agaricus blazei Murill, a mushroom which is well known as a potent antitumor agent, contains abundant pharmaceutical substances. To identifY the major components of Agaricus blazei Murill fruiting bodies cultivated in a Korean local farm was analyzed. There were not large differences in chemical compositions of inorganic substances, carbohydrates, amino acids and fatty acids between Agaricus blazei Murill fruiting bodies cultivated in Korea and those in Japan. However, carbohydrate composition was significantly lower in Agaricus blazei Murill of Korea compared to those of Japan. Total amino acid contents were 280.75 mg% in Agaricus blazei Murill fruiting bodies. Sixteen species of amino acid were identified by high performance liquid chromatography except cysteine. Linoleic acid, the most abundant fatty acid contained in Agaricus blazei Murill fruiting bodies, was estimated to be 78.3% of total fatty acids.

Cold-Adapted and Rhizosphere-Competent Strain of Rahnella sp. with Broad-Spectrum Plant Growth-Promotion Potential

  • A phosphate-solubilizing bacterial strain isolated from Hippophae rhamnoides rhizosphere was identified as Rahnella sp. based on its phenotypic features and 16S rRNA gene sequence. The bacterial strain showed the growth characteristics of a cold-adapted psychrotroph, with the multiple plant growth-promoting traits of inorganic and organic phosphate solubilization, 1-aminocyclopropane-1-carboxylate-deaminase activity, ammonia generation, and siderophore production. The strain also produced indole-3-acetic acid, indole-3-acetaldehyde, indole-3-acetamide, indole-3-acetonitrile, indole-3-lactic acid, and indole-3-pyruvic acid in tryptophan-supplemented nutrient broth. Gluconic, citric and isocitric acids were the major organic acids detected during tricalcium phosphate solubilization. A rifampicin-resistant mutant of the strain exhibited high rhizosphere competence without disturbance to the resident microbial populations in pea rhizosphere. Seed bacterization with a charcoal-based inoculum significantly increased growth in barley, chickpea, pea, and maize under the controlled environment. Microplot testing of the inoculum at two different locations in pea also showed significant increase in growth and yield. The attributes of cold-tolerance, high rhizosphere competence, and broad-spectrum plant growth-promoting activity exhibited the potential of Rahnella sp. BIHB 783 for increasing agriculture productivity.

Current Studies on Browning Reaction Products and Acidic Polysaccharide in Korean Red Ginseng (홍삼에 함유된 갈변물질 및 산성다당체에 대한 연구현황)

  • In the browning reaction of Korean ginseng, it appears that enzymatic and non-enzymatic browning reaction occurred In initial stage of steaming fresh ginseng at low temperature, and then non-enzymatic browning reaction followed in the drying period after steaming. Browning reaction of red ginseng occurred between $60{\sim}90$ min of steaming at $100^{\circ}C$, and browning pigments of red ginseng were mostly water soluble substances. The structural characteristics of water soluble browning reaction products(WS-BRPs) isolated from Korean red ginseng were showed the presence of hydroxyl, amide carbonyl and aliphatic methane groups. From sugar analysis it was identified that L and S-1, melanoidins isolated from red ginseng, contained two kinds of sugars, glucose and xylose, and the other melanoidin S-2 contained the previous and fructose. In order to find out pertinent methods for the acceleration of browning during ginseng processing, various treatment were made on fresh ginseng with sugars, amino acids and inorganic nitrogenous compounds and the extent of browning was measured. Among sugar tested, maltose resulted in the greatest acceleration of browning followed in decreasing order by glucose and lactose, whereas pentoses, fructose, sucrose and raffinose had negligible effect. A marked browning occurred in ginseng treated with basic amino acids, while the extent of browning was not greatly increased when ginseng was treated with aliphatic amino acids, hydroxyl amino acids, or acidic amino acids. The brown color intensity gradually increased with an increase of glucose concentration far up to 0.5M. L, S-1, and S-2 were found to have an ability to donate hydrogen to DPPH, and also they had anti-oxidative activity in the experiments of hydrogen peroxide scavenging, inhibitory activity in the formation of MDA from linoleic acid, auto oxidation of ok-brain homogenates, lipid peroxidation by the enzymatic and non-enzymatic system in liver microsome fraction, and mitochondrial fraction etc. The amounts of acidic polysaccharide(AP) in red ginseng were higher than those of wild and cultured Panax quinquefolius, Panax notoginseng as well as white ginseng (Panax ginseng). In white ginseng, the AP amount is no difference in root ages or sizes, also, the AP amount of ginseng body was similar to that of rhizome, but was higher than that of leaf and epidermis. Addition of red ginseng acidic polysaccharide(RGAP) increased production of nitric oxide(NO) and tumor necrosis factor (TNF)-$\alpha$ in the rodent macrophage cultures, and treatment of RGAP in vivo stimulated tumoricidal activities of natural killer (NK) cells.

Changes in Chemical Compositions of Pumpkin(Cucurbita moschata DUCH.) Seed Sprouts (호박(Cucurbita moschata DUCH.)종실의 발아 성장 과정 중 성분 변화)

  • This study was performed for increasing the consumption and developing the function of pumpkin(Cucurbita moschata DUCH.) seed. The changes of the contents of general chemical compositions, fatty acids, amino acids, ascorbic acid and ${\beta}$-carotene during sprouting were analyzed. Also, the bitter taste, which was produced during sprouting, were purified by using thin layer chromatography and preparative high pressure liquid chromatography. The purified bitter compound was identified by mass spectrum and nuclear magnetic resonance($^1$H '||'&'||' $\^$13/C-NMR). Weight of pumpkin seed sprout was increased to 348.4% and the length of stem was dramatically increased at 8 days. In each head and stem parts of the pumpkin seed sprout, the contents of protein and lipid were decreased, however, the contents of fiber, ash and soluble inorganic nitrogen were increased. The fatty acids of the pumpkin seed sprout were mainly represented as linoleic acid, oleic acid, palmitic acid and stearic acid. During sprouting, palmitic acid was gradually increased, reversely, linoleic acid was gradually decreased. The general amino acids of head part in the pumpkin seed sprout grown at 23$^{\circ}C$ during 8 days were orderly more contained glycine, alanine, arginine, cystein and proline. Those of free amino acids were orderly more contained arginine, threonine, alanine and glutamine. The contents of L-ascorbic acid and ${\beta}$-camtene of the pumpkin seed sprout were gradually increased with increasing sprouting days. The bitter taste material of head part of the pumpkin seed sprout was detected at Rf value 0.72 on silicagel TLC plate and separuted as one peak by HPLC. The chemical structure of the puified bitter compound was identified as a cucurbitacin glycoside by MS and NMR. The content of bitter compound at 8 days was contained 42.2 mg per 1kg sprout head.

Analysis of Biological Activity by Time of Black Garlic Ripening in Seosan Yukjok Garlic and Elephant Garlic (서산육쪽·코끼리마늘의 흑마늘 숙성 시기별 생리활성 분석)

  • This study analyzed the quality characteristics of black garlic made from Seosan Yukjok Garlic and elephant garlic in Seosan, Chungnam province. Of the inorganic components, Mg content was the highest in all treatment groups, and the Ca content was high in each of the 15 day treatments. The content of K was high after 10 days aging in Yukjok garlic and after 15 days in the elephant garlic. The Fe, Na, K, and Mg content was high in Yukjok black garlic after 15 days, and Na, K, Ca, and Mg were high in the elephant black garlic aged for 15 days. The crude fat content was high in both Yukjok black garlic and elephant black garlic after 15 days. Vitamin C content was highest in both types of garlic after aging for 15 days. An analysis of four kinds of organic acids showed that citric acid was the only organic acid to appear in raw garlic of Yukjok garlic and elephant garlic. Black Yukjok garlic and elephant black garlic had a greater total amino acid content than the raw garlic of either type. However, among the tested amino acids, 13 kinds of amino acids were at their highest after five days of ripening in Yukjok black garlic, while 15 kinds of amino acids were abundant in elephant garlic after the same period. Eight kinds of amino acids were high after aging for 15 days. Through this study, it was confirmed that, in the process of making black garlic, changes in the main components of the garlic occur through different routes, and these changes vary depending on the garlic species. Therefore, this study provided basic data for the processing of Seosan's Yukjok black garlic and elephant black garlic.

Engineering and Characterization of the Isolated C-Terminal Domain of 5-Enolpyruvylshikimate-3-phosphate (EPSP) Synthase

  • 5-Enolpyruvylshikimate-3-phosphate (EPSP) synthase catalyzes the formation of EPSP and inorganic phosphate from shikimate-3-phosphate (S3P) and phosphoenolpyruvate (PEP) in the biosynthesis of aromatic amino acids. To delineate the domain-specific function, we successfully isolated the discontinuous C-terminal domain (residues 1-21, linkers, 240-427) of EPSP synthase (427 residues) by site-directed mutagenesis. The engineered C-terminal domains containing no linker (CTD), or with gly-gly ($CTD^{GG}$) and gly-ser-ser-gly ($CTD^{GSSG}$) linkers were purified and characterized as having distinct native-like secondary and tertiary structures. However, isothermal titration calorimetry (ITC), $^{15}N-HSQC$,\;and\;^{31}P-NMR$ revealed that neither its substrate nor inhibitor binds the isolated domain. The isolated domain maintained structural integrity, but did not function as the half of the full-length protein.

  • Urine contains protein and nucleic acid(urea, uric acid, creatinine, ammonia, amino acids), various organic and inorganic materials, vitamin, hormone, enzyme etc. The examination of gualitative or quantitative change of the above mentioned materials and picking up the abnormal materials are useful to diagnose diseases. The test strip for examination of urine is applied to the routine test, monitoring of medical therapy and recurrence, self monitoring, and screeing in preventive medicine. We have been using multitest strip for checking the bacterial infection(nitrite), PH, protein, glucose, ketone body, urobilinogen, bilirubin and occult blood. So it is possible to diagnose three groups of diseases as follows, abnormal metabolism of the carbohydrate, diseases of kidney and other urogenital system, diseases of hepatobiliary system and hemolytic disease causing abnormal metabolism of bile juice.
