The 44 endophytic bacterial strains were isolated from surface-sterilized root of rice cultivated in seven different locations of Chungcheong province, Korea. Each isolate was introduced into rice seedlings grown gnotobiotically by inoculating scissor-cut first true leaf with cell suspensions, and the colonization capacity of each isolate in root tissue was analyzed at 7 days after inoculation. Sixteen out of 44 isolates were re-isolated from root successfully with the frequency of $10^{3-5}$ CFU/g tissue. Interestingly, seven out of 16 isolates were identified as Burkholderia species. The identity between inoculated strains and re-isolates was confirmed by genomic finger-printing and 16S rDNA sequence analysis. By a confocal laser scanning microscopic observation it was revealed that KJ001 strain, one of the sixteen isolates tagged with gfp colonized in root tissue especially around xylem. Six out of seven Burkholderia strains obtained in this study showed antagonizing activities against seven different fungal pathogens, contain nifH gene, and five of them enhanced growth of cucumber over 30%. The isolates showed no hypersensitive response on tobacco leaves and no pathogenecity in rice. From these results it was found that the endophytic Burkholderia strains will be useful in agriculture to develop a biocontrol agent or a bio-fertilizer.
Fluorescent pseudomonads have been isolated from halophytes, mesophytes, and xerophytes of Pakistan. Among these, eight isolates, GS-1, GS-3, GS-4, GS-6, GS-7, FS-2 (cactus), ARS-38 (cotton), and RP-4 (para grass), showed antifungal activity and were selected for detailed study. Based on biochemical tests and 16S rRNA gene sequences, these were identified as strains of P. chlororaphis subsp. chlororaphis and aurantiaca. Secondary metabolites of these strains were analyzed by LC-MS. Phenazine-1-carboxylic acid (PCA), 2-hydroxy-phenazine, Cyclic Lipopeptide (white line-inducing principle (WLIP)), and lahorenoic acid A were detected in variable amounts in these strains. P. aurantiaca PB-St2 was used as a reference as it is known for the production of these compounds. The phzO and PCA genes were amplified to assure that production of these compounds is not an artifact. Indole acetic acid production was confirmed and quantified by HPLC. HCN and siderophore production by all strains was observed by plate assays. These strains did not solubilize phosphate, but five strains were positive for zinc solubilization. Wheat seedlings were inoculated with these strains to observe their effect on plant growth. P. aurantiaca strains PB-St2 and GS-6 and P. chlororaphis RP-4 significantly increased both root and shoot dry weights, as compared with uninoculated plants. However, P. aurantiaca strains FS-2 and ARS-38 significantly increased root and shoot dry weights, respectively. All strains except PB-St2 and ARS-38 significantly increased the root length. This is the first report of the isolation of P. aurantiaca from cotton and cactus, P. chlororaphis from para grass, WLIP and lahorenoic acid A production by P. chlororaphis, and zinc solubilization by P. chlororaphis and P. aurantiaca.
A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Xanthomonas campestris pv. vesicatoria, PI271322, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI271322 were inoculated with race 1 and race 3 of Xanthamonas campestris pv. vesicatoria by infiltrating bacterial suspension into abaxial side of leaves. PI271322 was carrying a gene ($Bs_3$) for hypersensitive resistance to race 1 of X. c. pv. vesicatoria and also a component of non-hypersensitive resistance to race 3 inheriting in a quantitative mode. Correlation coefficient between disease indices to race 1 and race 3 of non-hypersensitive plants in $F_2$ population was highly significant, thus indicating that a component of non-hypersensitive general resistance in PI271322 acted consistently to both races. Individual plants with resistance to both race 1 and race 3 of X. c. pv. vesicatoria were selected and a breeding program for incorporation of the resistance is continued.

Diseased leaves of Job's tears were collected at six locations in Korea to isolate and identify the pathogen causing leaf blight. Conidia were brown, fusoid, and slightly curved. They were $25{\sim}46{\times}10{\sim}15\;{\mu}m$ in size, and had mostly 4 septa which were characteristic of pseudosepta. A few conidia had slightly protruding hila, whereas most had no hilum. Conidial germ tubes were produced mostly from both polar end cells (bipolar), and progressed in semiaxial direction. When seedlings of Job's tears were inoculated with conidia, long, spindle-shaped, and brown lesions were produced on leaves, and entire leaves became blighted 15 days later. Based on the aggressiveness in pathogenicity, the isolates could be divided into two groups. Considering the mycological characteristics and pathogenicity of the fungus, the pathogen causing leaf blight of Job's tears was determined to be Bipolaris coicis (Nisikado) Shoemaker.
The study was carried out to develop transformants resisting to Phyophthora blight disease in the domestic pepper cultivar Subicho. In transforming of syn600 promoter with elicitin gene using Agrobacterium (LBA4404/pBI101 syn600-syn${\alpha}$-elicitin) to cotyledons of pepper, rate of shoot formation in 'Subicho' was 11.1% in medium containing 3 mg/L zeatin and 0.05 mg/L NAA, and also 12.8% in medium containing combination of 4 mg/L zeatin and 0.05 mg/L MAA. For PCR reaction using elicitin gene primer of transformants regenerated from cotyledons, we detected a specific band of 536 bp, and also showed strong signal at position of 536 bp in accordance with NPTII gene used as probe in Southern blot. Transformants pepper shown resistance to blight fungus was inoculated to seedlings of the $T_{1}\;and\;T_{2}$ transformants by concentration (density: zoo spore $10^{3}/mL$).
In Korea, EBI fungicides which are highly effective for control of pear scab and rust but of high risk for development of resistance have been frequently sprayed by majority of pear growers. To detect any possible resistance or reduced sensitivity in the field strains of scab fungus to five EBIs, difenoconazole, fluquinconazole, flusilazole, fenarimole and hexaconazole, sensitivity tests were conducted with fungal specimens collected in Ulsan and Naju where scab usually occurs and EBIs have been intensively sprayed for many years. As the strains for which $EC_{50}$ values of the EBIs were largely shifted from those of base-line were occasionally found, the resistant or less sensitive strains were supposed to be distributed. In the activity test for the EBIs by artificial inoculation, in which EBI-treated pear leaves on the potted seedlings were inoculated with fungal spores collected in the two regions, development of resistances to EBIs were confirmed. Since the fungal spores collected at 4 and 2 orchards in Naju and Ulsan, respectively, produced much higher disease incidence on the leaves treated with hexaconazole than those on the untreated control, those fungal specimens were determined as resistant to hexaconazole. Similar results were also obtained with two specimens from Ulsan for flusilazole.

Stem blight symptom of soybean was severely developed in 2016 in Hwaseong and Yeoncheon. During the seedling period, the damping-off of seedlings and the brown or black spots of cotyledons were observed. After August, the leaves began to be yellowed, and partially browned areas on leaves began to develop. After September, microsclerotia began to form even on the surface of the stems that had exhibited water-soaking symptom. After mid-October of the harvest season, the epidermis of the stem was peeled off, resulting in the formation of a large number of microsclerotia in the cortex. The pathogens isolated from these symptoms were the best in mycelial growth at 32-35℃, and the formation of microsclerotia was the most at 20-28℃. The pathogen was identified as Macrophomina phaseolina through the morphological characteristics of the pathogen and the sequencing of the internal transcribed spacer region gene. In addition, when inoculated with a soybean stem using toothpicks cultured with the pathogen, the same symptoms as seen on the soybean field occurred. When the pathogen was re-isolated at the lesion site, the same pathogen was isolated and identified as Macrophomina phaseolina. Based on the results, the disease is reported as soybean charcoal rot.
Since many pesticides cause various health and environmental problems, alternative measures to replace them are needed, and the bacteria producing the antifungal substances can be one of them. In this study, several rhizobacteria were isolated and their antifungal activities against some important plant pathogenic fungi were examined. Pseudomonas otitidis TK1 and Paenibacillus peoriae RhAn32 inhibited the growth of Fusarium oxysporum f. sp. niveum and F. oxysporum f. sp. lycopersici by 49.8% and 45.6%, and 45.1% and 48.3%, respectively compared to those of the control. P. peoriae RhAn32 also decreased the growth of F. oxysporum f. sp. raphani by 37.5%. This growth inhibition might be due to the production of antifungal substances, such as siderophore, hydrogen cyanide and chitinase, which were produced by these rhizobacteria. P. otitidis TK1 also produced plant growth hormones indole acetic acid and indole butyric acid at $293.41{\mu}g/mg$ protein and $418.53{\mu}g/mg$ protein, respectively. When P. otitidis TK1 and B. cereus TK2 were inoculated together with F. oxysporum f. sp. lycopersici to the 4 weeks grown tomato seedlings and incubated additional 8 weeks, the stem lengths of tomato increased up to 45.7% and 55.3% and root lengths were raised to 64.9% and 60.8%, respectively than those of the control group. The wet weights increased by 118% and 182%, respectively compared to the control group.
In order to increase the persistence of plant growth promoting rhizobacteria (PGPR) in rhizpsphere soil, the growth of tomato was examined after the application of Arthrobacter woluwensis ED immobilized in alginate bead, which was known as PGPR. When tomato seedlings were treated with A. woluwensis ED of $1{\times}10^6$ cells g $soil^{-1}$ and incubated for 30 days in a plant growth chamber, the shoot length, root length, fresh weight and dry weight of the grown tomato plants treated with the suspended inoculants significantly increased by 36.2, 59, 51.1, and 37.5%, respectively compared to those of the uninoculated control. The treatment of the immobilized bacteria increased those by 42, 67.4, 62.5, and 60.4%, respectively compared to those of the uninoculated control. Therefore, the enhancement of tomato growth by the treatment of the immobilized bacteria was higher than those by the suspended inoculants. The effects of the inoculation on indigenous bacterial community and the fate of the inoculated bacteria were monitored by denaturing gradient gel electrophoresis analysis. The DNA band intensity of A. woluwensis ED in the tomato rhizosphere treated with the suspended inoculants continuously decreased after the inoculation, but the band intensity in the tomato rhizosphere soils treated with the immobilized inoculants showed the maximum at 1 week after inoculation and the decreasing rate was less than that of the suspended inoculants, which indicated the longer maintenance of the immobilized bacteria at rhizosphere soils. Therefore, encapsulation of PGPR in alginate beads may be more effective than liquid inoculant for the plant growth promotion and survival of PGPR at plant rhizosphere.
Fludioxonil is derived from the antifungal compound pyrrolnitrin produced by Pseudomonas pyrrocinia and classified as a reduced-risk fungicide by the US EPA. The efficacy of fludioxonil for the control of lettuce gray mold caused by Botrytis cinerea was evaluated under several conditions such as growth stages of host, inoculum concentrations, and amounts of potato dextrose broth (PDB) included in spore suspension of B. cinerea. At 4-leaf stage of lettuce plants, fludioxonil applied at 2 ${\mu}g$/ml was more effective for the control of gray mold than at 5- and 6-leaf stages. However, fludioxonil at more than 10 ${\mu}g$/ml provided similar control activity in all growth stages of lettuce tested. The fungicide (10 and 50 ${\mu}g$/ml) also gave excellent control of gray mold on lettuce seedlings inoculated with spore suspensions of B. cinerea ($2.5{\times}10^5$ to $2{\times}10^6$ spores/ml). But, control efficacy of fludioxonil (2 ${\mu}g$/ml) was negatively correlated with inoculum concentration. Addition of PDB in spore suspension of B. cinerea resulted in higher disease severity than non-treated control. By inoculating spore suspension including 0.5% PDB, the fungicide gave the most control activity on the disease, followed by 1% and 2% PDB. The results suggest that fludioxonil has potential to control gray mold of lettuce, but the fungicide at a concentration having moderate activity may represent low control efficacy on the disease under some conditions.
