Bacterial Inoculant Effects on Corn Silage Fermentation and Nutrient Composition

  • The survival and effect of three new probiotic inoculants (Lactobacillus plantarum CCM 4000, L. fermentum LF2, and Enterococcus faecium CCM 4231) on the nutritive value and fermentation parameters of corn silage was studied under laboratory conditions. Whole corn plants (288.3 g/kg DM) were cut and ensiled at $21^{\circ}C$ for 105 days. The inoculants were applied at a concentration of $1.0{\times}10^{9}$ cfu/ml. Uninoculated silage was used as the control. The chopped corn was ensiled in 40 plastic jars (1 L) divided into four groups (4${\times}$10 per treatment). All corn silages had a low pH (below 3.55) and 83-85% of total silage acids comprised lactic acid after 105 days of ensiling. The probiotic inoculants in the corn silages affected corn silage characteristics in terms of significantly (p<0.05-0.001) higher pH, numerically lower crude protein content and ratio of lactic to acetic acid compared to control silage. However, the inoculants did not affect the concentration of total silage acids (acetic, propionic, lactic acids) as well as dry matter digestibility (IVDMD) of corn silages in vitro. In the corn silages with three probiotic inoculants, significantly (CCM 4231, CCM 4000) lower n-6/n-3 ratio of fatty acids was detected than in control silage. Significant decrease in the concentration of $C_{18:1}$, and significant increase in the concentration of $C_{18:2}$ and $C_{18:3}$ was mainly found in the corn silages inoculated with the strains E. faecium CCM 4231 and L. plantarum CCM 4000. At the end of ensiling, the inoculants were found at counts of less than 1.0 log10 cfu/g in corn silages.

Fermentation quality and in vitro methane production of sorghum silage prepared with cellulase and lactic acid bacteria

  • Objective: The effects of lactic acid bacteria (LAB) and cellulase enzyme on fermentation quality, microorganism population, chemical composition and in vitro gas production of sorghum silages were studied. Methods: Commercial inoculant Lactobacillus plantarum Chikuso 1 (CH), local selected strain Lactobacillus casei (L. casei) TH 14 and Acremonium cellulase (AC) were used as additives in sorghum silage preparation. Results: Prior to ensiling Sorghum contained $10^4LAB$ and $10^6cfu/g$ fresh matter coliform bacteria. The chemical compositions of sorghum was 26.6% dry matter (DM), 5.2% crude protein (CP), and 69.7% DM for neutral detergent fiber. At 30 days of fermentation after ensiling, the LAB counts increased to a dominant population; the coliform bacteria and molds decreased to below detectable level. All sorghum silages were good quality with a low pH (<3.5) and high lactic acid content (>66.9 g/kg DM). When silage was inoculated with TH14, the pH value was significantly (p<0.05) lower and the CP content significantly (p<0.05) higher compared to control, CH and AC-treatments. The ratio of in vitro methane production to total gas production and DM in TH 14 and TH 14+AC treatments were significantly (p<0.05) reduced compared with other treatments while in vitro dry matter digestibility and gas production did not differ among treatments. Conclusion: The results confirmed that L. casei TH14 could improve sorghum silage fermentation, inhibit protein degradation and decrease methane production.

Metabolite Profiling and Bioactivity of Rice Koji Fermented by Aspergillus Strains

  • In this study, the metabolite profiles of three Aspergillus strains during rice koji fermentation were compared. In the partial least squares discriminant analysis-based gas chromatography-mass spectrometry data sets, the metabolite patterns of A. oryzae (KCCM 60345) were clearly distinguished from A. kawachii (KCCM 60552) and only marginal differences were observed for A. oryzae (KCCM 60551) fermentation. In the 2 days fermentation samples, the overall metabolite levels of A. oryzae (KCCM 60345) were similar to the A. oryzae (KCCM 60551) levels and lower than the A. kawachii (KCCM 60552) levels. In addition, we identified discriminators that were mainly contributing tyrosinase inhibition (kojic acid) and antioxidant activities (pyranonigrin A) in A. oryzae (KCCM 60345) and A. kawachii (KCCM 60552) inoculated rice koji, respectively. In this study, we demonstrated that the optimal inoculant Aspergillus strains and fermentation time for functional rice koji could be determined through a metabolomics approach with bioactivity correlations.

수박 만할병균에 길항하는 Bacillus sp. YJ-3에 의한 대목용 참박 생육촉진효과 (Plant Growth-Promoting Effects of Antagonistic Bacillus sp. YJ-3 against Fusarium Wilt of Watermelon-Rootstock Gourd)

  • 여러 작물 재배지 토양에서 각종 근권미생물을 분리하여 이들 분리미생물 중 수박 만할병균인 Fusarium oxysporum f. sp. niveum(KCCM34741)에 길항하는 미생물인 YJ-19, YJ-45, YJ-3 등 12 균주를 분리하였다. 길항미생물들은 MS agar 배지 실험 및 미생물제제 첨가 상토 실험에서 모두 참박의 발아율을 향상시켰으며, 이들 중 분리주 YJ-3 균주는 참박의 발아율과 초기생육에 가장 큰 영향을 주는 미생물로 선별되었다. YJ-3 균주를 제제화하여 참박 재배 기간별로 식물체 생육실험을 행한 결과 대조구인 일반 상토 및 시판 미생물혼합 상토 보다 각각 46%와 13%정도 생육을 촉진하는 것으로 나타났다. 분리주 YJ-3 균주는 간균의 gram 양성균으로서 Bacillus sp.로 동정되었다. Bacillus sp. YJ-3은 수박 만할병균인 F. oxysporum f. sp. niveum에 대해 강한 억제효과를 나타내었고, 그외 수박 병원균인 Alternaria cucumerina, Botrytis cinerea, Colletotrichum orbiculare, Didymella bryoniae, Rhazoctonia solani 등에서도 우수한 길항력을 나타내어 본 Bacillus sp. YJ-3 균주는 수박 만할병균에 길항하는 동시에 참박의 생육을 촉진하는 미생물로 확인되었다.

Effect of Lactic Acid Bacteria Inoculation on Fermentation Characteristics of Whole Crop Barley Silage

  • An experiment was carried out to determine the homofermentative activity of Lactobacillus plantarum KCC-10 and KCC-19 on the ensiling of whole crop barley (WCB). The crude protein in the silages was slightly higher in the KCC-10 and KCC-19 treatments compared to the control, but there was no significant difference between the two inoculant-treated silages. Nutrient parameters such as acid detergent fiber, neutral detergent fiber and in vitro dry matter digestibility in L. plantarum KCC-10 and KCC-19 treated silages did not differ from those in the control silage. The lactic acid content increased in KCC-10 and KCC-19 treated silage when compared with the control silage but the contents of acetic acid and butyric acid produced in KCC-10 and KCC-19 treated silages were similar with the control silage. Further, the number of lactic acid bacteria (LAB) in KCC-10 treated silage demonstrated a significant increase when compared to the control. Especially, KCC-19 treated silage showed greater lactic acid bacterial growth potential. Other microbes such as yeast and fungi were not detected in KCC-10 and KCC-19 treated WCB silages. Hence, this study suggests that the addition of L. Plantarum KCC-10 and KCC-19 to the WCB silage can improve fermentation quality for the production of high-quality silage.

품종과 보존제의 처리가 춘계수확 연맥의 특성과 사료가치에 미치는 영향 (Effect of Cultivar and Preservative on Characteristics and Quality of Spring Harvested Oat Hay)

  • An experiment was carried out to determine effect of cultivar and preservative on the characteristics and quality of baled oat(Avena sativa L.) hay after 60 days of storage. Ihe main plots consisted of the cultivars such as 'Foothill' and 'Swan', and the subplots consisted of preservatives such as commercial propionic acid(CA), lactic bacteria inoculant(LB) and control(C0). Bale temperature of 'Foothill' oat hay maintained higher than that of 'Swan' oat hay during storage. Bale temperature of oat hay treated with CA remained lowest during the earlier storage period and no significant difference was found among preservatives during the latter storage period. Visual assessment of bale of 'Swan' oat hay was better in the evaluation of mold, odor and color than that of 'Foothill' oat hay after storage. DM content of Foothill oat hay increased markedly than that of 'Swan' oat hay through the storage period. CFU(Colony fodng unit) of total viable micmrganism of oat hay bales was not quite different between preservatives, but that of lactic bacteria of 'Swan' oat hay and 'Foothill' was different by over 16 and lo4, respectively. Total nitrogen content of 'Swan' and 'Foothill' oat hay was 1.45 and 2.22%, respectively(P<0.05) and effect of preservatives on total N content was higher in the order of LB, CA, and CO. 품종, 건조제 및 보존제의 처리가 추계수확 연맥건초의 품질변화에 미치는 영향 (Effect of Cultivar, Drying Agent and Preservative on Quality Changes of Fall Harvested Oat Hay)

  • An experiment was carried out to determine effect of cultivar, drying agent concentration and preservative on the moisture content, temperature and quality changes of fall harvested oat(Avena sativa L.) hay during field curing and storage. The main plots consisted of the cultivar such as 'Foothill' and 'Swan', the subplots consisted of drying agent concentration such as 7Skg/ha(C1), lOSkglha(C2) and control(C0) and the sub-subplots consisted of preservative such as buffered acid(BA), lactic bacteria inoculant(LB) and control(C0). Moisture contents of 'Foothill' and 'Swan' oat cultivar at harvest were 635.5 and 385.4%DW(Dry weight), respectively, but the final moisture contents of the oat cultivars were 117 and 86%DW. The moisture content of cut oat was not affeded by drying agent concentration. Core temperature of baled 'Swan' oat hay was a little higher than that of 'Foothill' and no consistent preservative effect was found. Total nitrogen content of 'Swan' oat hay was 3.08% and that of 'Foothill' oat hay was 3.45%(P<0.05). Drying agent treatment increased total nitrogen content of 'Swan'(P <0.05), but preservative had no effect. Crude fiber content of oat cultivar was increased as the curing progressed(P<0.05) and the difference of crude fiber content of oat cultivar was maintained through the whole curing period. Effect of drying agent concentration on crude fiber content was not found after preservation. Crude fiber content of 'Swan' oat hay was higher than that of 'Foothill' by 1.O% in ADF and 3.1 % in NDF(P<0.05). No preservative effect was found in crude fiber content. According to RFV of oat cultivar at harvest, forage quality of 'Foothill' and 'Swan' was Prime and Grade 1, respectively, but that of both cultivm was above Grade 2 at the final curing day. RFV of the stored 'Foothill' and 'Swan' oat hay was 122 and 114(P<0.05), respectively, and the quality of oat hay was classified as Grade 1 and 2, respectively, according to the forage quality standard assigned by AFGC.

Community of natural lactic acid bacteria and silage fermentation of corn stover and sugarcane tops in Africa

  • Objective: To effectively utilize crop by-product resources to address the shortage of animal feed during the dry season in Africa, the community of natural lactic acid bacteria (LAB) of corn stover and sugarcane tops and fermentation characteristics of silage were studied in Mozambique. Methods: Corn stover and sugarcane tops were obtained from agricultural field in Mozambique. Silage was prepared with LAB inoculant and cellulase enzyme and their fermentation quality and microbial population were analyzed. Results: Aerobic bacteria were the dominant population with 107 colony-forming unit/g of fresh matter in both crops prior to ensiling, while 104 to 107 LAB became the dominant bacteria during ensiling. Lactobacillus plantarum was more than 76.30% of total isolates which dominated silage fermentation in the LAB-treated sugarcane top silages or all corn stover silages. Fresh corn stover and sugarcane tops contain 65.05% to 76.10% neutral detergent fiber (NDF) and 6.52% to 6.77% crude protein (CP) on a dry matter basis, and these nutrients did not change greatly during ensiling. Corn stover exhibits higher LAB counts and water-soluble carbohydrates content than sugarcane top, which are naturally suited for ensiling. Meanwhile, sugarcane tops require LAB or cellulase additives for high quality of silage making. Conclusion: This study confirms that both crop by-products contain certain nutrients of CP and NDF that could be well-preserved in silage, and that they are potential roughage resources that could cover livestock feed shortages during the dry season in Africa.

A Two-Strain Mixture of Rhizobacteria Elicits Induction of Systemic Resistance Against Pseudomonas syringae and Cucumber Mosaic Virus Coupled to Promotion of Plant Growth on Arabidopsis thaliana

  • We evaluated a commercial biopreparation of plant growth-promoting rhizobacteria (PGPR) strains Bacillus subtilis GB03 and B. amyloliquefaciens IN937a formulated with the carrier chitosan (Bio Yield) for its capacity to elicit growth promotion and induced systemic resistance against infection by Cucumber Mosaic Virus (CMV) and Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana. The biopreparation promoted plant growth of Arabidopsis hormonal mutants, which included auxin, gibberellic acid, ethylene, jasmonate, salicylic acid, and brassinosteroid insensitive lines as well as each wild-type. The biopreparation protected plants against CMV based on disease severity in wild-type plants. However, virus titre was not lower in control plants and those treated with biopreparation, suggesting that the biopreparation induced tolerance rather than resistance against CMV. Interestingly, the biopreparation induced resistance against CMV in NahG plants, as evidenced by both reduced disease severity and virus titer. The biopreparation also elicited induced resistance against P. syringae pv. tomato in the wild-type but not in NahG transgenic plants, which degrade endogenous salicylic acid, indicating the involvement of salicylic acid signaling. Our results indicate that some PGPR strains can elicit plant growth promotion by mechanisms that are different from known hormonal signaling pathways. In addition, the mechanism for elicitation of induced resistance by PGPR may be pathogen-dependent. Collectively, the two-Bacilli strain mixture can be utilized as a biological inoculant for both protection of plant against bacterial and viral pathogens and enhancement of plant growth.

  • 전국(全國) 223개(個) 지역(地域)에서 수집(蒐集)한 두과작물(豆科作物)의 근류(根瘤)에서 총(總) 590주(株)의 근류균(根瘤菌)을 분리(分離)하여 숙주특이성(宿主特異性)에 따라, R. japonicum 218종(種), R. phaseoli 101종(種), R. trifolii 97종(種), R. meliloti 4종(種), R. leguminosarum 1종(種), Rhizobium species 101종(種), 미분류(未分類)된 균주(菌株) 159종(種)으로 분류(分類)하였다. 분리(分離)된 R. japonicum 218종(種)의 근류형성능(根瘤形成能)과 질소고정능(窒素固定能)을 비교(比較)하여. R. japonicum R-138, R-168, R-214 균주(菌株)를 우수균주(優秀菌株)로 하였다. 분리(分離) 선발(選拔)된 R. japonicum 중(中) 질소고정능(窒素固定能)이 우수(優秀)한 균주(菌株)의 plasmid를 확인(確認)한 결과(結果), fast-growing group 균주(菌株)들은 $1{\sim}4$개(個)의 거대(巨大) plasmid를 함유(含有)하고 있었으며 그 분자량(分子量)은 약(約) $35{\sim}300{\;}Md$ 정도(程度)이었다. 반면 대부분(大部分)의 slow-growing group 균주(菌株)들은 plasmid가 검출(檢出)되지 않았다.
