Histopathological effects caused by formalin bath on gill and liver of Eel (Anguilla japonica) (포르말린 약욕이 뱀장어 (Anguilla japonica)의 아가미 및 간에 미치는 병리조직학적 효과)

  • Histological changes in gill and liver of eel, Anguilla japonica (average weight 96±3.6 g) were examined with formalin bath at 0~500 ppm for 1, 6 and 24 h. Hyperplasia, hypertrophy, cell fusion, desquamation and necrosis of epithelial cells at gill lamella and gill filament were observed from 6 h at 200 ppm, 1 h at 300 ppm, 1 h at 400 ppm and 1 h at 500 ppm. In the exposure of formalin 100 ppm for 24 h, epithelial cells arrangement of gill filament and gill lamella showed thinner and more regular order than the control. trophy and pyknosis of hepatocytes, congestion at sinus or central vein, degeneration of cytoplasm were observed in the liver from 24 h at 100 ppm and 200 ppm, 6 h at 300 ppm, 1 h at 400 ppm and 500 ppm. However, there were not any histological changes at liver of 100 ppm-1, 6 h, 200 ppm-1, 6 h and 300 ppm-1 h compared with the tissue of control.

Prevalence of Anisakid larvae in chum salmon Oncorhynchus keta in Korea (한국산 연어의 아니사키스형 유충의 감염현황)

  • The infestation status of anisakid type larvae was investigated in migrating chum salmon (Oncorhynchus keta), with different condition (captured area, sex, body portion) during 2006~2008. The mean infection number of anisakid larvae per individual female and male fish captured from Namdae river was $98{\pm}27$, $103{\pm}27$, respectively. The mean infection number of anisakid larvae per individual female and male fish captured from the coastal area of Yangyang was $63{\pm}18$ and $108{\pm}17$, respectively. The anisakid larvae were mainly found in abdominal muscles (85%) but only a little in the visceral portion. Two types of anisakid larvae (A. simplex, Contracaecum type) were identified but other anisakid larvae were not detected. To investigate the effect of storing temperature on the viability of anisakid larvae, the section of abdominal muscle were stored at different temperature (room temperature, $4^{\circ}C$, $-20^{\circ}C$, $-80^{\circ}C$). As a result, it was necessary to store at $-20^{\circ}C$ for more than 6 hrs to kill the larvae. The present results revealed that chum salmon caught in Korea are heavily infected with anisakid larvae, mainly in the abdominal muscle, and A. simplex was dominantly found in this study.

The Effect of Growth and Survival Rate on Feeding Rate of 3-year-old Abalone, Haliotis discus hannai rearing in Net Cage Culture (3년산 북방전복, Haliotis discus hannai의 해상가두리 내 먹이공급비율에 따른 성장 및 생존율)

  • The effect of different feeding rate on the growth and survival rate of 3-year-old abalone, Haliotis dicsus hannai were investigated in marine net cage culture. Feeding rate is determined that was fed 5, 10, 15 and 20 percentage to abalone at body weight once daily : 5 daily feeding rate (DFR), 10 DFR, 15 DFR and 20 DFR. After that, it was conducted to reared during thirteen month with two replicates in net cage culture. In the growth performance of reared abalone (initial mean shell length $73.77{\pm}11.27mm$) in net cage culture, that the absolute growth rate (ARG), daily growth rate (DGR) and specific growth rate (SGR) to the shell length and shell breadth, as well as weight gain (WG), daily weight gain (DWG) and specific weight gain (SWG) to body weight of 5 DFR were higher than those of different groups (P < 0.05). Also, survival rate of all feeding rate groups was not significant, but it was showed bellow 30 percents. Therefore, these results is showed that the daily feeding rate for natural feed of 3-year-old abalones reared in net cage culture should be to supply among five to ten percents.

Yolk absorption and growth of churn salmon, Oncorhynchus keta alevin (연어, Oncorhynchus keta전기자어의 난황흡수 및 초기성장)

  • The process of yolk absorption and the growth pattern of churn salmon, Oncorhynchus keta alevin were studied. Matured males and females of chum salmon were sampled from the Namdaechun River in the east coast of Korea, and then an artificial fertilization was carried out using their eggs and sperms. Fertilized eggs were incubated in controlled water temperature of $13^{\circ}C$ and samples of 40 alevins were randomly taken at just hatching and at every three days between 12 days alter hatching and yolk absorption. The means of total length and total weight of hatched alevins were respectively 1.97cm and 1.85g and the yolk absorption took about 33 days after hatching, based on the morphological measurement. Growth curves of total length, total weight and somatic weight were fitted well with the Gompertz growth model. However, the shrinking in yolk-sac length, yolk-sac weight, yolk-sac height and yolk-sac volume revealed a linear phenomenon until the absorption of yolk was completed. The relative growth of chum salmon alevin, such as the relationship between total length and total weight, or between total weight and yolk-sac weight, was also studied.

Effect of Water Temperature on the Embryonic Development of Slime Flounder Microstomus achne (수온이 찰가자미(Microstomus achne)의 난발생에 미치는 영향)

  • The fertilized eggs were obtained from mature adult slime flounder Microstomus achne to determine the water temperature effect on egg development. The lowest water temperature for the flounder egg development was $0.4^{\circ}C$ on average. The temperature-dependent duration from fertilization to hatching ranged 86.5 to 296.7 hours at $9\sim21^{\circ}C$ with an accelerated development at higher temperature. Agreeable hatching rates, 95.8~97.0%, were obtained at $12\sim18^{\circ}C$, while lower at both extremes, 86.9% at $9^{\circ}C$ and 9.3% at $21^{\circ}C$. The highest water temperature, $24^{\circ}C$, had the life of the fertilized eggs confined within 24 hours. Water temperature was a parameter that induced an abnormal egg development: with abnormalities of 88.3% at $21^{\circ}C$, 2.1% at $9^{\circ}C$ (P<0.05), and 0.4~0.8% at $12\sim18^{\circ}C$.

Characteristics of the Histological Structure of the Mandibular Barbels of Two Species of Catfish (Siluridae) from Korea (한국의 메기과 (Siluridae) 어류 2종의 상악 수염의 조직학적 특성)

  • Histological observations on the barbels of catfish (Siluridae) in Korea are reported. The mandibular barbel of Far Eastern catfish, Silurus asotus, has epidermis and dermis surrounding an axial rod of cartilage. The stratified epidermis embodies mucous cells, club cells, and taste buds. The mucous cells, which are thin rims of basophilic cytoplasm, and the taste buds, which are basophilic, are located at the surface and the distal portion of the epidermis, respectively. The basement membrane lies between the epidermis and dermis except in places of dermal papillae. The dermis is made up of loose connective tissue fibers enclosing nerve bundles and blood vessels. The central region of cartilage is innermost and enclosed within muscle layers. The mandibular barbel of slender catfish, S. microdorsalis, has an identical structure to those of the Far Eastern catfish however, the percentage thickness of epidermis in the upper and lower parts of the barbel and the number of taste buds in the upper and middle parts of the barbel are different from those of Far Eastern catfish. This study shows that the mandibular barbels in Far Eastern catfish and slender catfish are of a stiff and flexible type.

Cytogenetic Analysis of Three Hemibarbus Species (Cypriniformes) from Korea (한국산 누치속 어류 3종의 세포유전학적 연구)

  • Cytogenetic characteristics of three Hemibarbus species (H. labeo, H. longirostris and H. mylodon) were analyzed based on erythrocyte measurement, flow cytometric estimation of cellular DNA content, and karyological analysis. Average nuclear volumes for H. labeo, H. longirostris and H. mylodon were 22.5, 21.7 and $26.0\;{\mu}m^3$, respectively. The estimated genome sizes of those three species were not significantly different from one another, being recorded as 2.51, 2.33 and 2.35 pg/cell for H. labeo, H. longirostris and H. mylodon, respectively. Modal chromosome numbers of the three species were the same as 2n = 50. However, their karyotypes and fundamental numbers (FN) were different among species; 16M+16SM+18T/A (FN = 82) for H. labeo, 18M+16SM+16T/A (FN = 84) for H. longirostris and 18M+24SM+8T/A (FN = 92) for H. mylodon.

Starvation-induced Physiological Responses and RNA/DNA Ratios in Rock Bream, Oplegnathus fasciatus, and Olive Flounder, Paralichthys olivaceus

  • In a 12-week experiment, the rock bream, Oplegnathus fasciatus, and olive flounder, Paralichthys olivaceus, were investigated to determine the effects of starvation on their physiological parameters. The protein and DNA contents of the starved fish were significantly higher than the initial values and those of the fed fish. The RNA contents and RNA/DNA ratios of the fed fish were significantly higher than those of the other groups (P<0.05). The hematocrit, hemoglobin, red blood cells (RBC), and mean corpuscular volume (MCV) of the fed rock bream were significantly higher than at baseline (P<0.05), whereas the mean corpuscular hemoglobin concentration (MCHC) of the fed fish was lower than at baseline (P<0.05). The hematocrit, hemoglobin, RBC, and MCHC of the starved group were significantly lower than the baseline values, whereas the MCV of the starved group was significantly higher than the baseline value (P<0.05). No significant difference in alanine aminotransferase was observed between the fed fish and baseline, whereas the starved fish value was significantly higher than the baseline value (P>0.05). There were no significant differences in cortisol levels. However, the glucose level in the fed group was significantly higher than the baseline level and that in the starved group was significantly lower than the baseline level (P<0.05).

Fish Fauna and Community Structure in Lake Paldang and its Inflows (팔당호의 어류상과 군집동태)

  • Fish fauna and community structure were investigated at 2003, 2004 and 2006 in Lake Paldang and its inflows. In this survey, we collected 54 species belong to 14 families. Family Cyprinidae take 51.9% (28 species), Cobitidae and Gobiidae occupied 7.4% (each 4 species), respectively. E. erythropterus (11.8%) was dominant species and L. macrochirus (10.2%), designated as an ecosystem-invasive alien fish species by Ministry of Environment, M. yaluensis (9.1%), Z. platypus (7.7%) and R. brunneus (6.7%) were dominated in turn. H. molitrix, C. lutheri, P. altivelis, S. microdorsalis, L. costata. S. gracilis majimae, A. rivularis, P. koreanus. S. scherzeri and O. platycephala were rare species (less than 0.1% in relative abundance). In-lake area, 36 species of fishes belonging to 11 families were collected, and 48 species of 13 families were from inflows. The number of Korean endemic species were 19 species (35.2%), and 4 kinds of exotic species (7.4%), that is, C. cuvieri, H. molitrix, L. macrochirus and M. salmoides were found at this survey area. It was revealed by the analysis of fish community that diversity and richness indices were prominent at tributaries, and dominance index was high at the main body of Lake Paldang. Fish fauna showed rapid decline after dam construction which make the mid-Han river systems to lentic ecosystem, so many of meander-riffle fishes were disappeared dramatically. But after 1990s when installation of small scale wastewater treatment plants started in watershed, the water quality of small tributaries was improved. And also, expansion of the natural macrophytic vegetation of littoral zone should contribute to the development of diverse fish fauna in Lake Paldang.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.
