Anatomical Studies on Patterns of Branches of Aortic Arch in Korean Native Goat (한국재래산양(韓國在來山羊) 대동맥궁(大動脈弓)의 동맥분지(動脈分枝)에 관한 해부학적(解剖學的) 연구(硏究))

  • This macroscopic observation was performed to investigate the aortic arch and their branches of the Korean native goat. Ten Korean native goats with black hairs ranging in body weight from 15 to 20kg were used. The findings were based on the latex cast of the vessels made by injection of neoprene latex. The results obtained were as follow; 1. The aortic arch of the Korean native goat gave off only one vessel, the Truncus brachiocephalicus. 2. The Truncus brachiocephalicus gave off the A. subclavia sinistra, the A. subclavia dextra and the Truncus bicaroticus. 3. The A. subclavia sinistra gave off the Truncus costocervicalis, the A. cervicalis superficialis and the A. thoracica interna, and continued as the A. axillaris sinistra. 4. The Truncus costocervicalis sinistra arose from the A. subclavia sinistra in all cases and gave off the A. scapularis dorsalis suprema and the A. cervicalis profunda, and continued as the A. vertebralis sinistra. 5. The branching pattern of the A. subclavia dextra was similar to that of the A. subclavia sinistra except the Truncus costocervicalis. The Truncus costocervicalis dextra was arisen together with the A. subclavia dextra in 8 out of 10 cases, while one of the remaining 2 cases arose as the first branch of the A. subclavia dextra and the other was given off directly from the Truncus brachiocephalicus. 6. The Truncus bicaroticus was continued from the Truncus brachiocephalicus after branching off the A. subclavia dextra in 8 out of 10 cases, but the Aa. carotidas communes sinistra and dextra were directly branched from the Truncus brachiocephalicus without forming the Truncus bicaroticus in the remaining 2 cases.

Development of Thyroid Tumors by Carcinogens and Its Expression of p21 & p53 Protein in Rats (흰쥐에서 발암물질로 유발된 갑상선 종양과 p21 및 p53 단백질의 발현)

  • Objectives: The development of thyroid tumor has a relationship with carcinogen, oncogene and tumor suppressor gene. With aminotriazole, radioactive iodine and nitrosomethylurea as carcinogens in rat, authors investigate the incidence in type of the thyroid tumors, p21 and p53 protein expression pattern by immunohistochemical stain and the relationship between the tumors and p21-p53 protein expressions. Materials and Methods: 80 experimental animals were divided into four groups; group 1(control, no carcinogen, n=20), group 2(oral administration of aminotriazole for 36 weeks, n=20), group 3(intraperitoneal injection of 131I for one time and oral administration of aminotriazole for 36 weeks, n=20), group 4(oral administration of nitrosomethylurea for 3 days and aminotriazole for 36 weeks, n=20). After 40 weeks they were sacrificed with pathologic examination and we performed immunohistochemical staining with pan-ras monoclonal antibody for p21 protein and CMI polyclonal antibody for p53 protein with paraffin-embedded specimens. Results: 1) No tumors were observed in group I, but 38.3% of nodular goiters, 11.7% of adenomas and 50.0% of carcinomas were observed in carcinogen treated groups(group 2, 3, 4). 2) The incidence of nodular goiter, adenoma and carcinoma were 70%, 20% and 10% in group 2, 40%, 15% and 45% in group 3 and 5%, 0% and 95% in group 4. 3) p21 protein was not expressed in normal thyroid tissues but was expressed in 26.1% of nodular goiters, 42.9% of adenomas and 6.7% of carcinomas. On the other hands, p53 protein was not expressed in normal thyroid tissues, nodular goiters, adenomas and in well differentiated thyroid carcinomas by immunohistochemical stain. Conclusion: The authors suggest that aminotrizole, 131I, nitrosomethylurea can be etiologic agents in the development of thyroid tumor and the p21 protein can be expressed in the early stage and in benign condition of thyroid tumor but p53 protein is not expressed in all conditions of development in rats.

Effect of the Inhibition of Platelet Activating Factor on Oxidative Lung Injury Induced by Interleukin-$1\;{\alpha}$

  • In order to know the pathogenesis of adult respiratory distress syndrome (ARDS) in association with the oxidative stress by neutrophils, the role of platelet activating factor (1-0-alkyl-2-acetyl-snglycero-3-phosphocholine, PAF) was investigated during acute lung injury induced by interleukin- $1{\alpha}$ (IL-1) in rats. An insufflation of IL-1 into the rat's trachea increased the acetyltransferase activity in the lung and the increase of PAF content was followed. As evidences of acute lung injury by neutrophilic respiratory burst, lung leak index, myeloperoxidase activity, numbers of neutrophils in the bronchoalveolar lavage fluid, neutrophilic adhesions to endothelial cells and NBT positive neutrophils were increased after IL-1 treatment. In addition, a direct instillation of PAF into the trachea caused acute lung leak and the experimental results showed a similar pattern in comparison with IL-1 induced acute lung injury. For the confirmation of oxidative stress during acute lung leak by IL-1 and PAF, a histochemical electron microscopy was performed. In IL-1 and PAF treated lungs of rats, the deposits of cerrous perhydroxide were found. To elucidate the role of PAF, an intravenous injection of PAF receptor antagonist, WEB 2086 was given immediately after IL-1 or PAF treatment. WEB 2086 decreased the production of hydrogen peroxide and the acute lung leak. In ultrastructural study, WEB 2086 mitigated the pathological changes induced by IL-1 or PAF. The nuclear factor kappa B (NFkB) was activated by PAF and this activation was inhibited by WEB 2086 almost completely. Based on these experimental results, it is suggested that the PAF produced in response to IL-1 through the remodeling pathway has the major role for acute lung injury by neutrophilic respiratory burst. In an additional experiment, we can also come to conclude that the activation of the NFkB by PAF is thought to be the fundamental mechanism to initiate the oxidative stress by neutrophils causing release of proinflammatory cytokines and activation of phospholipase $A_2$.

Expression of Calponin 3 in the Striatum Following 3-Nitropropionic Acid-induced Neurotoxicity (선조체에서 3-nitropropionic acid 투여 후 calponin 3의 발현 연구)

  • Calponin 3 is an F-actin-binding protein and plays a key role in regulating spine plasticity and synaptic activity in neurons. Unlike the other subtypes, calponin 1 and 2, which are expressed in smooth and cardiac muscle cells, calponin 3 is highly expressed in the brain. The goal of this study was to elucidate the spatiotemporal expression pattern of calponin 3 following repeated administration of 3-nitropropionic acid in mice. The repeated administration of 3-nitropropionic acid generated necrotic neuronal cell death in the striatum. Calponin 3 was up-regulated in the neuroprotective penimbral region from 1.5 days after the last injection and thereafter. Double immunofluorescence study revealed that calponin 3 was induced in GFAP-positive astrocytes. These results suggest that calponin 3 induction in the neuroprotective penumbral area following 3-nitropropionic acid intoxication may play a key role in reactive astrogliosis in the striatum.

Heat/Mass Transfer Characteristics in Impingement/Effusion Cooling System with Rectangular Fins for Combustor Liner Cooling (가스터빈 연소실 냉각을 위한 충돌제트/유출냉각기법에서 사각핀 설치에 따른 열/물질전달 특성)

  • The present study has been performed to investigate the influences of rectangular fins on heat transfer in an impingement/effusion cooling system with crossflow. To simulate the impingement/effusion cooling system with initial crossflow, two perforated plates are placed in parallel and staggered arrangements with a gap distance of 2 times of the hole diameter. The crossflow passes between the plates, and various rectangular fins are installed on the plates. Reynolds number based on the hole diameter is fixed to 10,000 and the flow rate of crossflow is changed from 0.5 to 1.5 times of that of the impinging jet. A naphthalene sublimation method is used to obtain the heat/mass transfer coefficients on the effusion plate. Also to analyze the flow characteristics, a numerical calculation is performed. When rectangular fins are installed, the flow and heat transfer pattern is changed greatly from case without fins. In the injection hole region, the jet impinges on effusion plate without deflection and wall jet spreads symmetrically. In the effusion region, the crossflow accelerates due to the decrease of cross-sectional area in the channel. Local heat/mass transfer coefficients are enhanced significantly compared to case without fins. As the blowing ratio increases, the effect of fins against the crossflow becomes more significant and then the higher average heat/mass transfer coefficients are obtained than the case without fins.

Study on the Flow Around an Elliptic Wing Using Flow Visualization Technique (유동가시화를 통한 타원형날개주위 유동연구)

  • This study deals with an investigation on the tip vortex generated by an elliptic wing with section shapes of NACA 0020. The flow structure on the wing surface is investigated by using tufts test as well as observing the cavitation pattern. The surface pressure on a foil surface is measured to complement the visualized flow field. Results show that a strong spanwise pressure gradient is a definite contributor on the formation of tip vorex, and the fluids from both sides contribute to the evolutionary process of tip vortex. On the other hand, a series of experiments are conducted to investigate the detailed structure of tip-vortex at various angles of attack. The tip-vortex formation and development are observed by producing a cavitation, and then by a laser sheet technique in conduction with a dye injection method. The shape of tip-vortex and the distance between a vortex core and the trailing vortex sheet are found to vary with the angle of attack. Overall features of tip flow are evaluated to complement the vortex model based on inviscid theory.

The Effects of Retinoic Acid on the Regenerating Limbs of the Larval Korean Newt (Hynobius leechii) (한국산의 도롱뇽(Hynobius leechii) 유생의 다리재생에 미치는 Retinoic Acid의 효과)

  • The effects of retinoic acid (RA) on the regenerating limbs of Korean newt (Hynobius leechii) larvae have been studied. Intraperitoneal injection of RA at 4 days post-amputation caused the proximalization of regenerate structures in the proximodistal (PD) axis of the limbs amputated through either the distal zeugopodium or the distal stylopodium. The mean level of proximalization (MLP) increased as a dose-dependent manner, and the MLP was also dependent on the level of amputation at a given dose of RA. At the dose of 150 $\mu$ g/g body wt., MLP's in either amputation level reached close to a peak value, and an increasing number of inhibition of regeneration or duplication in the transverse plane occurred at the dose of 200 $\mu$ g/ g body wt. and above. The changes induced by RA treatment suggest that positional values in the cells of the regenerating limbs have been modified in the three cardinal axes including PD and transverse axes in a graded mode. Furthermore, from the comparison of data in two different amputation levels, it can be speculated that the sensitivity of cells to RA effect along the PD axis might not be linear.

Expression of $\beta$-Galactosidase Gene Microinjected into Xenopus Egg During Early Development (초기발생 동안 양서류 난에 미세주입된 $\beta$-galactosidase 유전자의 발현)

  • For the effort to produce transgenic amphibians, a plasmid DNA sequence (cytoplasmic actin promoter-linked bacterial $\beta$-galactosidase gene) was microinjected into fertilized Xenopus eggs. It appeared that the injection of 20 nl solution containing 1-2 ng of DNA was not toxic, but over 4 ng was toxic to embryonic development. The translational product of $\beta$-gal gene ($\beta$-galactosidase) had enzyme activity in all three germ layers of the embryo. Expression of the injected $\beta$-gal genes was first detected at mid-gastrula stage, and the activity persisted up to stage 43 (feeding tadpole) with decreased level of retention. However, the level of the expression was various among the injected individuals as well as each experiment. That is, $\beta$-galactosidase activities did not appear in all cells, instead a localized distribution pattern. Although other possibilities could not be omitted, this mosaic distribution of gene expression seemed to arise from unequal partition of the injected DNA into each blastomere during early cleavage.

Solution-Processed Inorganic Thin Film Transistors Fabricated from Butylamine-Capped Indium-Doped Zinc Oxide Nanocrystals

  • Indium-doped zinc oxide nanocrystals (IZO NCs), capped with stearic acid (SA) of different sizes, were synthesized using a hot injection method in a noncoordinating solvent 1-octadecene (ODE). The ligand exchange process was employed to modify the surface of IZO NCs by replacing the longer-chain ligand of stearic acid with the shorter-chain ligand of butylamine (BA). It should be noted that the ligand-exchange percentage was observed to be 75%. The change of particle size, morphology, and crystal structures were obtained using a field emission scanning electron microscope (FE-SEM) and X-ray diffraction pattern results. In our study, the 5 nm and 10 nm IZO NCs capped with stearic acid (SA-IZO) were ligand-exchanged with butylamine (BA), and were then spin-coated on a thermal oxide ($SiO_2$) gate insulator to fabricate a thin film transistor (TFT) device. The films were then annealed at various temperatures: $350^{\circ}C$, $400^{\circ}C$, $500^{\circ}C$, and $600^{\circ}C$. All samples showed semiconducting behavior and exhibited n-channel TFT. Curing temperature dependent on mobility was observed. Interestingly, mobility decreases with the increasing size of NCs from 5 to 10 nm. Miller-Abrahams hopping formalism was employed to explain the hopping mechanism insight our IZO NC films. By focusing on the effect of size, different curing temperatures, electron coupling, tunneling rate, and inter-NC separation, we found that the decrease in electron mobility for larger NCs was due to smaller electronic coupling.

Dopaminergic Regulation of Gonadotropin-II Secretion in Testosterone-treated Precocious Male and Immature Rainbow Trout Oncorhynchus mykiss

  • The present work examined the role of gonadotropin-releasing hormone (GnRH) and dopaminergic drugs on the secretion of maturational gonadotropin (GTH II) in relation to testosterone m treatment. This study provides evidence that the plasma GTH II levels are increased by T treatment in precocious males, but not in the immature animal. In addition, GnRH analogue (GnRHa) alone significantly increased the plasma GTH II secretion in immature rainbow trout treated with T, as well as in T-treated and T-untreated precocious males. However, injection with either dopamine (DA) or domperidone (DOM; DA D2 receptor antagonist) alone did not alter the basal plasma GTH 11 secretion in all experimental groups. The secretion of GTH II in the T-treated precocious males was remarkably influenced by GnRHa or combination of dopaminergic drugs. Notably, the effects of dopaminergic drugs on GnRHa-induced GTH II secretion w8s prolonged by T in precocious males. In T-treated immature animals, GnRHa-induced GTH II secretion was Increased only by a dose DOM (10$\mu$g/g body n) but not by higher dose DOM (100$\mu$/g body wt). In the T-untreated immature rainbow trout, however, plasma GTH 11 secretion was not influenced by the same treatments. Therefore, these results indicate that DA may be acting indirectly by blocking the effect of GnRH on GTH II secretion in vivo. T may act to modulate the relative contribution by the stimulatory (GnRH) and inhibitory (DA) neuroendocrine factors, which would ultimately determine the pattern of GTH II secretion.
