Potentimetric Biosensor for Detection of L-Malate and D-Isocitrate Employing ${CO_{3}}^2-$ -Selective Electrode and Enzyme Immobilization in Flow Injection Analysis

  • Ion-selective eleltrodes(ISEs) are simple electrodechemical devices for the direct measurement of ions in the samples. A novel potentiometric biosensor for the determination of L-Malate or D-isocitrate has been developed by using CO2-3 -ISE-FIA system was composed of a pump, an injector, a malic enzyme or isocitric dehydrogenase enzyme reactor, a CO2-3 -ISE, a pH/mV meter, and an integrater. The various factors, such as buffer capacity types of plstericizer and polymer, were optimized for the CO2-3 selectivity. In this novel CO2-3 --ISE-FIA system, the potential difference due to the amount of CO2-3 produced from each enzyme reaction was proportional to the amount of L-malate or D-isocitrate.

Possible Application of Artificial Insemination Buffer for Increasing Production Efficiency of Female Cow Offspring

  • The present research was carried out to evaluate the possibility of increasing female offspring production ratios using artificial insemination buffer (AIB) before artificial insemination (AI). In this experiment, we optimized AIB composition, made an AIB gun and analyze factors affecting AI non-return rate after AIB treatment. The AIB was made with the base of Tris-buffer supplemented with L-arginine and several other chemicals that might reduce the motility of male sperm compared to the female counterpart, therefore, increasing the possibility of fertilization by female sperm. AIB must be deposited into $2^{nd}$ to $4^{th}$ cervix by AIB gun. After 15 min of AIB deposition, frozen semen was deposited into the same place. A total of 348 cattle were inseminated with AIB insemination, and there were no significant differences between AIB and traditional AI non-return rates (56.8% vs. 55.7%). The AI non-return rate in AIB group, however, differed significantly among 7 Hanwoo farms. The parturition numbers ($1^{st}$ to $7^{th}$) of cows did not affect AIB AI rate. The proportion of AIB AI success rates was significantly higher in Hanwoo cows than in dairy cows (61.0% vs. 48.7%), but the average AI success rate did not differ significantly between AIB and conventional AI (56.8% vs. 55.7%). The female offspring production rate in $2^{nd}$ to $4^{th}$ cervix deposition place was significantly higher than that in the uterus body (77.7% vs. 59.6%, p<0.05). The injection volume of AIB in 5 and 10 ml was significantly higher than that in 2 ml (77.7%, 78.7% vs. 51.8%, p<0.05), but there were no differences in AIB injection volume between 5 and 10 ml. The best exposure time of AIB in the cervix was 10 to 15 min rather than 5 min (79.2%, 77.2% vs. 52.6%, p<0.05). AIB therefore needs to have an exposure time of at least over 10 min for a higher production rate of female offspring. In conclusion, AIB could be used in AI industry to increase the female offspring ratio and AIB AI can increase the AI success rate.

  • In this paper, we describe a versatile use of fullerene(C60) as a charge transporting material for organic light-emitting diodes. The use of fullerene as a buffer layer for an anode, a doping material for hole transport layer, and an electron transport layer was investigated. Fullerene improved the hole injection from an anode to a hole transport layer by lowering the interfacial energy barrier and enhanced the lifetime of the device as a doping material for a hole transport layer. In addition, it was also effective as an electron transporting material to get low driving voltage in the device.

  • The decreased activities of liver enzymes relating to carbohydrate metabolism such as glucose- 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of streptozotocin injected rats were significantly modified by the intraperitoneal injection of ginseng saponin mixture and/or purified ginsenosides. However, several enzymes such as pyruvate kinase, malic enzyme and glycogen phosphorylase were not modified appreciably by the saponin administration, suggesting that the effect of ginseng saponin might be depend upon individual enzymes. Examination of liver enzymes by liver professing technique using perfusion buffer containing saponin (10-3%) showed that the ginseng saponin might stimulate insulin biosynthesis as well as the related enzyme activities.

Failure and Phase Transformation Mechanism of Multi-Layered Nitride Coating for Liquid Metal Injection Casting Mold

  • Ti-Al-Si target and Cr-Si target are sputtered alternately to develop a multi-layered nitride coating on a steel mold to improve die-casting lifetime. Prior to the multi-layer deposition, a CrN layer is developed as a buffer layer on the mold to suppress the diffusion of reactive elements and enhance the cohesive strength of the multi-layer deposition. Approximately 50 nm CrSiN and TiAlSiN layers are deposited layer by layer, and form about three ㎛-thickness of multi-layered coating. From the observation of the uncoated and coated steel molds after the acceleration experiment of liquid metal injection casting, the uncoated mold is severely eroded by the adhesion of molten metallic glass. On the other hand, the multi-layer coating on the mold prevents element diffusion from the metallic glass and mold erosion during the experiment. The multi-layer structure of the coating transforms the nano-composite structured coating during the acceleration test. Since the nano-composite structure disrupts element diffusion to molten metallic glass, despite microstructure changes, the coating is not eroded by the 1,050 ℃ molten metallic glass.

Capillary electrophoresis 및 근적외선분광분석기를 이용한 황기의 원산지 판별 (Discrimination of Geographical Origin for Astragalus Root (Astragalus membranaceus) by Capillary Electrophoresis and Near-Infrared Spectroscopy)

  • 외국농산물의 국내 유입증가와 이에 따른 신속한 원산지 판별법 확립이 요구되는 가운데 최근 수입이 급증한 품목 중 하나인 황기를 선택, CE 및 NIRS를 이용하여 분석조건을 확립하고 원산지판별에의 적용 가능성을 검토하였다. CE를 이용하여 분석 시 추출은 methanol: 0.1M phosphate buffer(pH 2.5)(3:7)를 사용하였으며 uncoated fused silica capillary$(50\;{\mu}m\;I.D.{\times}27cm)$를 이용하여 $45^{\circ}C$, 14 kV로 분석, 200 nm에서 검출하였다. 분석 buffer는 0.1 M phosphate buffer(pH 2.5)에 20% methoxy ethanol과 40 mM HSA를 첨가하여 사용하였으며, 8초간 pressure injection 하였다. Peak의 재현성을 증대시키기 위하여 시료 주입 전 분석 buffer를 1분간 분석 시와 같은 방향으로(F) 흘려주고 0.1 M phosphoric acid와 1 M sodium hydroxide는 각각 4분, 5분간 반대방향으로(R) 홀려주었다. 증류수를 다시 1분간 흘려주고(R) 분석 buffer로 2분간 평형화(F) 시킨 후 시료를 주입하였다. 이상의 조건으로 국내산(97점)과 수입(113점) 황기를 분석한 결과 전체 peak의 양상은 유사하였으나 약 $11{\sim}13$분에 용출되는 2개의 peak(peak am-1, am-2)의 면적 비율에서 차이가 나타나 국산은 peak am-2가 peak am-1의 약 4배인 반면, 수입 산은 10배로 나타나 원산지 판별이 가능하였으며, 약 80%의 편별율을 나타내었다. NIRS는 국산 및 수입산 황기 raw 스펙트럼의 2차 미분 스펙트럼 R값이 0.915로 비교적 안정된 값을 얻을 수 있었고, SEP는 약 14.3%로 나타났다. 이를 국산과 수입산 황기에 적용 시 전체 판별율이 약 97%로 비교적 높은 판별율을 보였다. 또한 NIRS로 판별이 불가능한 시료가 CE로는 판별이 가능하여 이 두 기기를 함께 사용 시 상호보완하여 신속 정확한 원산지 판별법의 개발 가능성이 시사되었다.

Effects of injection of hydrolysis plasma protein solution on the antioxidant properties in porcine M. Longissimus Lumborum

  • Background: Plasma protein hydrolysates have been shown to possess antioxidant activity. However, no report has yet to examine the antioxidant effects of injection of plasma protein hydrolysates on meat quality. Therefore, in this study, the effects of injection of hydrolysis plasma protein solution on meat quality and storability were investigated in porcine M. longissimus lumborum. Methods: Twelve pigs were randomly selected at a commercial slaughter plant and harvested. Dissected loins were injected with one of five solutions: C- control (untreated), T1- 10 mM phosphate buffer solution (PBS), T2- 10 mM PBS with 0.01 % butylated hydroxytoluene, T3- 10 mM PBS with 5 % plasma proteins, and T4- 10 mM PBS with 5 % hydrolysis plasma proteins. Results: T3 and T4 induced greater reduction in protein content of the loin muscle than other treatments. T2 resulted in the lowest pH as well as highest cooking loss. After a storage period of 3-7 days, both lightness and redness of meat were unaffected by all injection treatments. However, yellowness was significantly elevated by treatment with T4 relative to the control. T4 also resulted in the lowest shear force (a measure of meat toughness), suggesting improvement of texture or tenderness. Further, T4 resulted in the most stable TBARS values during storage, indicating that this treatment might retard rancidity in meat. Conclusion: Injection of porcine M. longissimus lumborum with hydrolysis plasma protein solution could improve overall pork quality, including tenderness and storability.

다양한 실험조건에 따른 경주 벤토나이트 완충재 블록의 팽윤 거동 해석 (Swelling behavior Simulation Study of KJ-II Bentonite Buffer Blocks under Various Experimental Conditions)

  • 본 연구에서는 완충재 블록의 팽윤 거동 특성을 파악하고자 COMSOL Multiphysics의 비선형 탄성모델을 활용하여 완충재 팽윤압 측정실험에 대한 수치해석을 수행하였다. 수치해석에서는 실험 조건과 동일하게 사방 구속조건 및 물 주입압을 경계조건으로 설정하였으며, 실험값과 수치해석 결과를 비교하여 수치해석 모델을 검증하였다. 이후 해당 수치해석 모델을 활용하여 비구속 조건에서의 팽윤변형, 건조밀도별 팽윤압, 그리고 완충재의 기하학 형태에 따른 팽윤압을 모사하였다. 해석결과, 모델은 포화 과정에 따른 팽윤변형 현상과 건조밀도가 높아질수록 팽윤압이 증가하는 현상을 적절히 모사하였다. 또한, 완충재 기하학 형태에 따른 팽윤압 모사 결과에서는 팽윤압이 증가하는 속도가 원통형 시료보다 U자형 시료에서 훨씬 빠르게 나타났고, 포화 과정에 따라 U자형 시료의 내부 모서리에서 선제적으로 응력이 발현되는 것으로 분석되었다. 다만, 완충재의 실제 거동을 보다 정확하게 모사하기 위해서는 비선형 탄소성 모델을 적용하여 해석모델의 수준을 고도화해야 할 것으로 판단된다.

Role of a ZnO buffer layer for the formation of epitaxial NiO films

  • NiO는 니켈 공공과 침입형 산소 이온에 의한 비화학적양론 특성 때문에 자발적으로 p-형 반도체 특성을 나타내는 것으로 알려져 있다. NiO는 3.7 eV 의 넓은 밴드갭을 가지고 있어 투명소자를 위한 hole injection layer 나 hole transport layer로 사용하기 위한 연구가 많이 이루어지고 있다. 또한, 안정적인 p-형 반도체 특성은 n-형 산화물 반도체와의 접합을 통해 복합소자의 구현이 용이하기 때문에, ZnO 등과의 접합을 통한 소자 구현이 가능하다.[1] 하지만, 기존의 많은 연구에서는 내부의 결함이 많이 존재하는 다결정 박막을 사용하였기 때문에, 전하의 이동에 제한이 발생해, 충분한 소자 특성을 나타내지 못하였다. 최근 Dutta의 연구에 의하면, 결정질 사파이어 기판위에 박막을 성장할 경우 [111] 방향으로 우선 배향성을 가진 NiO 박막을 얻을 수 있다고 알려져 있다.[2] 본 실험에서는 NiO 박막을 이용한 PN 접합소자 구현을 위해 사파이어 위에 p-NiO 박막을 에피택셜하게 성장한 후 구조적 특성을 분석하였으며, n-ZnO 박막을 그 위에 성장하여 소자를 제작하였다. 그 결과 ZnO 또한 에피택셜한 성장을 하는 것을 확인할 수 있었다. 성장순서에 따른 PN 접합구조 특성을 확인하기 위해 사파이어 위에 ZnO 를 성장시킨 후 NiO 를 성장시킨 결과 NiO 박막의 우선성장 방향이 [100]으로 변하는 것을 확인할 수 있었다.
