The cPCR technique was used to monitor rumen fermentation and attachment of Fibrobacter succinogenes to cellulose at different pH in the in vitro culture medium. The target fragments of 16S rDNA(445 bp) were amplified from genomic DNA of F. succinogenes with specific primers and internal controls(205 bp) were constructed. Cell counts were estimated from the amounts of genomic DNA, which was calculated from cPCR results. F. succinogenes in pH 6.8 and 6.2 showed apparently higher attachment than in pH 5.8 during all incubation time. There were some difference between pH 6.8 and 6.2 in the degree of attachment, but the different was not significant (P>0.05). Cellulose degradation increased in process of incubation time and the increasing rate was higher when initial pH was higher. The pH in culture medium decreased regardless of initial pH in course of incubation time. After 24 h of incubation, medium pH was dropped by 0.24, 0.58 and 0.16 units from original medium pH 6.8, 6.2 and 5.8, respectively. More gas was produced at higher initial pH in the same manner as in cellulose degradation. In summery, Initial pH of rumen culture in vitro significantly influenced cellulose digestion, gas production, pH change and bacterial attachment. Especially, low pH(5.8) resulted in much lower bacterial attachment and fiber digestion compared to higher medium pH.
This study was conducted to estimate the cultural characteristics, the production of antibiotic, and the selection of optimal media for mass culture of Bacillus licheniformis N1 isolate which was previously reported as an antagonistic bacterium to Botrytis cinerea. We investigated initial pH, temperatures and shaking speed for good cultural conditions and antibiotics production by N1 isolate. According to the results, the optimal conditions of initial pH, temperatures, and shaking speed were determined to be pH 5.0~5.5, 30~35$^{\circ}C$ and 250 rpm, respectively. Also, the optimal conditions for the antagonism by N1 isolate highly appeared in the initial pH as 5.0, and the mycelial growth inhibition was high when the substances used such as glucose or corn starch as carbon sources, and biji(soybean curd residue) flour as a nitrogen source. Furthermore, inhibitory area was significantly expanded, when 3% or 5% of corn starch was added into 5% of Biji flour as nitrogen source, were respectivley selected for mass culture of N1 isolate. Among them, 5% Biji flour medium showed higher cell density more than 10 times that in NB medium after 48 hour incubation. Therefore, the optimal medium was determined as 5% biji flour added 3~5% of corn starch for high density of cells.

Phosphate, ammonia, glucosamine, glucose, pyruvate, succinate, fumarate, malate and acetate were examined for their ability to control the heat-stable enterotoxin (ST) production in succinate salts medium or in M9 medium. The results obtained were summerized as follows. 1. When the initial phosphate concentration was adjusted to 1.0mM, ST production was decreased to 80u/ml or less. But when the initial phosphate concentration was adjusted to 64mM or 100mM, enterotoxin production was 320u/ml. 2. When the initial ammonia concentration in the medium was adjusted to 1.0mM, no ST production and cell growth were observed. But when ammonia concentration was adjusted to 10mM, 19mM, 38mM or 76mM, enterotoxin production was 320u/ml. 3. Among carbon sources, glucosamine, glucose, pyruvate, succinate, fumarate, malate and acetate, acetate supported the highest specific production (928 unit/O.D.) of heat-stable enterotoxin. From this results, we could assume that heat-stable enterotoxin production is controlled by stringent control mechanism. 4. When the pH of the succinate salts medium was kept between 6.2 to 6.5, no heat-stable enterotoxin production was observed, but when the pH of the medium was kept between pH 6.2 to 6.5, 267 unit/O.D. of heat-stable enterotoxin was produced. 5. Glucose inhibited the heat-stable enterotoxin production and the mechanism was assumed due to its capacity to lower the pH of the medium during catabolysis and its high metabolic energy.

Bacteriocin-producing lactic acid bacteria, Lactococcus sp. HY 449, was isolated from dairy products. Using response surface methodology, the various concentrations of medium compo- nents (tryptone, glucose, yeast extract, tween 80, and initial pH) were tested to find the optimum conditions for maximum bacteriocin production and growth of Lactococcus sp. HY 449. Central composite design was used to control the concentrations of medium components in the experiment. Bacteriocin production and cell growth of Lactococcus sp. HY 449 were most affected by glucose and yeast extract. Estimated optimum growth conditions of Lactococcus sp. HY 449 were as follows; tryptone 1.08%, glucose 1.129%, yeast extract 0.674%, tween 80 0.11%, and initial pH 7.19. Also estimated optimum conditions for bacteriocin production were tryptone 0.937%, glucose 1.108%, yeast extract 0.163%, tween 80 0.09%, and initial pH 6.98.
A flat-type membrane bioreactor(FMBR ) for aerobic whole cell immobilization was developed and its performance for the citric acid production was investigated using Aspergillus niger (KCTC 1232). The reactor consisted of three layers. The top layer contained flowing air for oxygen supply, the middle layer had stationary cells, and the bottom layer had flowing aqueous nutrients. The initial pH of the culture medium played an important role in citric acid production and the lower initial pH of the culture medium resulted in a higher citric acid yield. Under air and pure oxygen aerations the volumetric productivity reached 0.20 and 0.40g/Lh. Furthermore, the productivity improved with the increase of the culture medium feed rate.
The optimum conditions for the production of an antifungal antibiotic from Bacillus sp. YJ-63 were investigated. The oprimumized medium consisted of 1.5% soluble starch, 1% tryptone and 0.5% yeast extract, and temperature and initial medium pH for production were optimal at 35$^{\circ}C$ and pH 6.0, respectively. Production yield was significantly improved by shaking culture using 50 ml medium in 500 ml flasks. Under these conditions, the production of the antifungal antibiotic was growth-dependent, from 35hrs into cultivation to the stationary phase and endospore formation.

Among the bacterial strains isolated from chungkook-jang, Bacillus subtilis CH-1, Bacills circulans K-1 and Bacillus subtitis (natto) N-1, Bacillus subtitis CH-1 showed the highest productivity of biosufactant. A-medium was selected for the basal medium in the large scale production of biosurfactant, and modified to synthetic medium which containing 2% glucose, 0.3% soy peptone, and mineral salts. The surface tension was reduced to maximal value after 96 hr after fermentation, about the 43% of initial tension. Temperature and initial pH of medium was not critical factor for the biosurfactant production. The yield of crude biosurfactant was 6 g/L under the optimum condition.
A inhibitor acting on substrate proteolytic enzyme was isolated from culture broth of Streptomyces sp. SK-862, which had been isolated from soil in Wonju City, by using the colloidal agar medium. The optimum culture temperature and initial pH for the production of the protease inhibitor was 28$^{\circ}C$ and pH 8.5, respectively. The optimum culture medium was composed of 1.5% glucose, 0.5% peptone, 0.1% K2PHO4, 0.05% CaCO3 and initial pH 8.5. The inhibitor production was maximum when the strain was incubated in shaking incubator at 70 strokes for 60 hours.
The experiments were conducted to obtain culture conditions on optimal cultural conditions for propagation of a topical orchid, Haemaria discolor, through shoot tip cultrue in vitro. Survival percentage of shoot tip explant, in initial culture media was higher in H$_3$P$_4$ medium than in Murashige and Skoog's medium. Explants, cultured in H$_3$P$_4$ medium containing 1.0 mg/L kinetin, showed more shoot elongation when compared with those on other media. When the distal part of stem (pseudobulb) was longitudinally sectioned into half and cultured in H3P4 medium containing 0.1 mg/L 2ip, 0.1, or 1.0 mg/L kinetin, shoots from axillary bud were much more elongated under light condition.
A Bacillus sp. A-6 strain that produced xylanase was isolated from rice bran. The optimal temperature and pH for xylanase activity of the culture supernatant of Bacillus sp. A-6 were 40$^{\circ}C$ and pH 7, respectively. The optimal temperature and pH for xylanase production in the xylan medium were 30$^{\circ}C$ and pH 9, respectively. The optimal concentrations of oat spelt xylan and peptone for xylanase production were 0.5% and 1.5%, respectively. The best nitrogen sources for xylanase production was beef extract, but xylanase production was also supported comparably by tryptone and peptone. The bacterial growth in the optimal xylan medium reached stationary growth phase after 12 h of incubation. The xylanase production in the culture supernatant increased dramatically during the initial 12 h exponential growth phase and then remained constant at 23.8-24.5 unit/ml during the stationary growth phase. The pH of the culture medium decreased from 8.8 to 6.7 during the exponential growth phase and subsequently increased to 8.1 during the stationary growth phase. Rice bran, sorghum bran, and wheat bran as well as oat spelt xylan induced xylanase production. The xylanase production was repressed when glucose was added to the xylan-containing medium.
