Inhibitory effects of Maduryung(Aristolochiae Fructus) on alcohol, acetaminophen and galactosamine induced hepatitis in rats (마두령(馬兜鈴)의 흰쥐 간염(肝炎) 억제(抑制) 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • Maduryung Extract, one of herbal medicine was tested for inhibitory effect to alcohol, acetaminophen and d-galactosamine induced hepatitis in rats. The results were as follows. 1. Increaced serum GOT, GPT levels by alcohol were significantly decreased by Maduryung extract.(p<0.01) 2. Maduryung extract decreaced GOT value in acetaminophen induced hepatitis and this effect may be due to increace of GSH level in liver tissue.(p<0.05) 3. Repeat administration of Maduryung extract showed inhibitory effect on s-GOT, s-GPT levels in d-galactosamine induced hepatitis. (p<0.05) According to the above results, it seems that Maduryung could be use as drug for alcohol or drug induced hepatitis treatment.

Radiosensitizing and Topoisomerase I Inhibitory Effects of Aloe vera, Formitella fraxinea, and Ulmus davidiana Extracts

  • Ulmus davidiana, Formitella fraxinea, and Aloe vera extracts were detected to have inhibitory effects against topoisomerase I at treatment of $5{\mu}g$. Ulmus davidiana and Aloe vera extracts were found to show inhibitory effect similar to camptothecin, Formitella fraxinea extract was found to have weak activity. We also found the potential use of those extracts as a radiation sensitizer. Radiosensitizing effect at combination treatment was increased more than 2 times at single treatment of radiation, Ulmus davidiana or Formitella fraxinea extracts. Ulmus davidiana and Formitella fraxinea extracts were found to have significant radiosensitizing effect on test tumor cell line. But, Aloe vera extract was not detected to have activity as a radiosensitizer. Ulmus davidiana and Formitella fraxinea extracts are potent radiosensitizers on tumor cell lines and should be considered for further study of active compounds.

  • The inhibitory effect of ferulic acid and related phenolic compounds on the growth of normal cell lines and can­cer cell line was evaluated by the MTT and XTT methods. Ferulic acid decreased the cell viability of human skin melanoma cells by the MTT method and the cell adhesion activity of human oral epithelioid carcinoma cells by the XTT method. These results suggest that ferulic acid has a potential anticancer activity.

  • This study was devised to observe the inhibitory effect of 7 kinds of the acidic polysaccharide fraction(PGI, PGa, PG3, PGa, PGs, PG6 and PG7) from Korean white ginseng on a lipolytic action of Toxohormone-L. Toxohormone-L is a lipolytic factor, found in ascites fluid of .sarcoma -180 bearing mice and of patients with hepatoma. A substace that inhibited the lipolytic action of Toxohormone-L was isolated from white ginseng powder. This substance was an acidic polysaccharides. In vitro test showed that the inhibitory effect of PGs fraction of the lipolysis by Toxohormone-L was highest percent among other treatments at concentration of 50, 10, 200, 500 and 1,000 Ug/ml of reaction mixture. And total inhibitory activity(units) of PGI and PG4 was highest among other treatments at the same concentration and that of 10 Ug/ml.

  • Extracts of 24 edible vegetables were tested concerning their action on in vitro inhibition on dopamine ${\beta}$-hydroxylase (DBH) and monoamine oxidase (MAO). All vegetables were purchased in Korean market and their common names were kept. Radish sprouts, ‘kkoch-na-mul’, ‘chong-gyong-chae’, ragwort, applemint showed strong DBH inhibitory effect when tyramine and crude bovine adrenal DBH were used as substrate and enzyme, respectively. ‘Cham-chwi’(Aster scaber), kale, ‘cham-na-mul’(Pimpinella brachycarpa), leek were found to have MAO-A inhibitory effect with serotonin and crude rat brain MAO-A. Lettuce, ‘chong-gyong-chae’, radish sprouts, beet leaves were found to have MAO-B inhibitory effect with benzyl amine and crude rat liver MAO-B.

  • The present study was carried out to estimate the comparative effects of hydroponic heavy metals (Al, Cd, Hg, and Pb) on ATP content in plant leaves grown with Hoagland solution under green house condition. The two plants, kidneybean(Phaseolus vulgaris L.) and buckwheat(Fagopyrum esculentum M nch), showed similar inhibitory effect of heavy metals on ATP content in order of Hg, Cd, Pb, and Al. But the overall inhibitory effect was greater in kidneybean than in buckwheat. The affinity of heavy metals, in vitro, toward the enzyme (luciferin-luciferase) is in order of Hg, Al, Cd, and Pb, similar to that toward ATP. The results showed that the inhibitory effect of heavy metals on ATP hydrolysis is mainly due to the coordination of heavy metals with enzyme than ATP.

20(S)-ginsenoside Rg3 inhibits glycoprotein IIb/IIIa activation in human platelets

  • The Panax ginseng Mayer is used in conventional medicine in Asia owing to its preventing effects on thrombosis, hypertension, atherosclerosis, vasorelaxation and myocardial infarction. Because platelets are crucial mediators of cardiovascular diseases, many studies have investigated its functions. The previous study showed the antiplatelet effects of crude ginseng fraction and two of its components, ginsenoside Rg3 (20S and 20R). In addition, ginsenoside Rg3-enriched fraction shows an inhibitory effect on collagen-activated rat platelets. However, the mechanism underlying this effect remains unclear. Thus, I investigated the inhibitory action of ginsenoside Rg3 (20S, G-Rg3) on the regulation of signaling molecules involved in ${\alpha}IIb/{\beta}_3$ activation. I found that G-Rg3, in a cyclic AMP dependent manner, inhibited thrombin-induced activation of human platelets and affinity of fibrinogen and fibronectin with ${\alpha}IIb/{\beta}_3$. Thus, in the present study, G-Rg3 showed an inhibitory effect on glycoprotein IIb/IIIa (${\alpha}IIb/{\beta}_3$) activation, suggesting its potential use for preventing platelet-mediated thrombotic disease.

  • Treatability and maximum no inhibitory effect concentration of benzene were measured in the synthetic wastewater medium by the activated sludge in the continuous activated sludge reactor. The maximum no inhibitory effect concentration was 1, 600mg per liter. Benzene concentration over 500mg per liter inhibited the growth of microorganims by the measurment of E/BOD, and the treatability of benzene at the maximum no inhibitory effect concentration was over 95%.

  • Antioxidative and cytoprotective effects of tectorigenin and glycitein isolated from the pueraria thunbergiana and its derivative, genistein, were determined. Among these three compounds, tectorigenin and glycitein bearing 6-methoxyl groups in both isoflavones showed significant free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase (XOD) generating superoxide anion radical. Tectorigenin only showed a slight inhibitory effect on XOD. We further studied the inhibitory effects of these isoflavones on the lipid peroxidation in rat liver microsomes induced by enzymatic and non-enzymatic methods. Each of them exhibited inhibitory effect on both ascorbic $acid/Fe^{2+}-{\;}and{\;}ADP/NADPH/Fe^{+3}-induced$ lipid peroxidation. Moreover, tectorigenin exhibited the highest protection of hydrogen peroxide damage on HepG2 and Vero cells among the three isoflavones, in the cytoprotective assay. It was suggested that the pattern of antioxidative and cytoprotective effect of isoflavones could be crucially by the aromatic substitution of oxygen-containing groups.

In vitro Screening of UVA Phototoxicity Inhibitors using the Natural Products (In vitro 실험법에 의한 천연물 중의 UVA 광독성 억제제 검색)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.
