In our search for monoamine oxidase (MAO) inhibitors from natural resources, we found that the methanol extract of the roots of Sophora flavescens showed an inhibitory effect on mouse brain monoamine oxidase (MAO). Bioactivity-guided isolation of the extract yielded two known flavonoids, formononetin (1) and kushenol F (2), as active compounds along with three inactive compounds, oxymatrine (3), trifolirhizin (4), and ${\beta}$-sitosterol (5). Formononetin (1) and kushenol F (2) showed significant inhibitory effects on MAO in a dose-dependent manner with $IC_{50}$ values of 13.2 and $69.9\;{\mu}M$, respectively. Formononetin (1) showed a slightly more potent inhibitory effect against MAO-B ($IC_{50}:\;11.0\;{\mu}M$) than MAO-A ($IC_{50}:\;21.2\;{\mu}M$). Kushenol F (2) also preferentially inhibited the MAO-B activity than MAO-A activity with the $IC_{50}$ values of 63.1 and $103.7\;{\mu}M$, respectively.
The extract of Ginkgo biloba leaves was measured for inhibitory activity against xanthine oxidase. Aceton extract of G. biloba leaves showed strong inhibitory activity. Inhibitory activities of the fractionated extract were in the order of water > ethyl acetate fractions. Two fractions exhibiting strong inhibitory activities ware further purified via repeated silica gel, Amberlite IRN-78, Polyclar AT, and Sephadex LH-20 column chromatographies. Active components were isolated and identified through $^1$H-NMR and $^{13}$ C-NMR. The compounds were characterized as kaempferol 3-Ο-$\alpha$-(6$^{"′}$-p-coumaroylglucosyl-$\beta$-1,4-rhamnoside), and quercertin 3-Ο-$\alpha$-(6$^{′}$-p-coumaroylglucosyl-$\beta$-1,4-rhamnoside).

This study evaluated tyrosinase, elastase inhibitory, and antioxidant activities of fermented Rhododendron micranthum Turcz. extract using a lactic acid bacterium, Lactobacillus rhamnosus. The optimum conditions for fermentation of R. micranthum Turcz. extract were $37^{\circ}C$ and 3% R. micranthum Turcz. extract for 3 days based on the bacterial cell number, total phenolic compounds, DPPH radical scavenging activity, and tyrosinase and elastase inhibitory activity. After culture for 3 days using 3% R. micranthum Turcz. extract, the cell mass of L. rhamnosus reached $5.7{\times}10^9CFU/ml$. The results indicated that R. micranthum Turcz. extract can be used for industrial lactic acid bacteria culture. After fermentation under optimum conditions, the total content of phenolic compounds of the fermented R. micranthum Turcz. extract was 157 GAE mg/ml, and the $IC_{50s}$ of DPPH radical scavenging activity, ABTS radical scavenging activity, and tyrosinase and elastase inhibitory effects were 78.8, 79.8, 329.1, and $449.5{\mu}g/ml$, respectively. The fermented R. micranthum Turcz. extract exhibited 1.2-, 1.3-, 1.5-, 2.4-, and 5.6-fold higher total content of phenolic compounds, DPPH radical scavenging activity, ABTS radical scavenging activity, and tyrosinase and elastase inhibitory effects than the nonfermented R. micranthum Turcz. extract. These results indicated that fermented R. micranthum Turcz. extract using L. rhamnosus can be used for developing new natural functional ingredients for the health food or cosmetic industry.

We investigated ${\alpha}$-amylase, ${\alpha}$-glucosidase and lipase inhibitory activity of extracts collected from wild plants in Gangwon-do. 90 wild plants were collected and their water and ethanol extracts were obtained. Results of measuring ${\alpha}$-amylase inhibitory activity indicated more than 80% of activity inhibition in 10 mg/mL concentration for ethanol extracts of three plants and water extracts of two plants. For ${\alpha}$-glucosidase inhibitory activity, ethanol extracts of thirteen plants and water extracts of three plants showed more than 80% of activity inhibition in 10 mg/mL concentration. In the experiment of inhibiting lipase activity, ethanol extracts of seven plants and water extracts of one plants showed above 80% of activity inhibition in 10 mg/mL concentration. These results suggest that the selected extracts could be potentially used as a resource of bioactive materials for health functional foods.
We investigated several biological activities using the ethanol extract and its fractions from Pimpinella komarovii leaves to evaluate the usefulness of its extract as a functional biomaterial. The ethanol extract showed antioxidant activities, such as DPPH scavenging activity $(IC_{50}=231.8{\mu}g/m{\ell})$. superoxide scavenging activity $(IC_{50}=23.6{\mu}g/m{\ell})$, and xanthine oxidase inhibitory activity $(IC_{50}=587.8{\mu}g/m{\ell})$. Its EtOAc fraction showed the strongest antioxidant activities among several fractions. The inhibitory effect of ethanol extract on tyrosinase activity was higher than water fraction. When $50{\mu}g/m{\ell}$ of EtOAc fraction was applied, the inhibition ratio of tyrosinase activity was much higher (42%) than that of melasolv. The EtOAc fraction also showed higher inhibitory effect on melanogenesis in Melan-a cells. The n-hexane and EtOAc fractions dose-dependently inhibited the NO production in a RAW 264.7 cells. These results suggest that extract of Pimpinella komarovii could be used as functional biomaterial in developing a skin whitening agent having the antioxidant activity.
Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

In order to evaluate anti-wrinkle activity of Carex humilis extract, free radical scavenging activity, elastase inhibitory activity and reduction of expression Matrix metalloproteinase-1 (MMP-1) mRNA and MMP-1 protein were investigated. The roots of Carex humilis were extracted with 95 % ethanol and successively partitioned with organic solvents with increasing polarity of the solvents. Each fraction of organic solvent were investigated by using free radical scavenging activity and elastase inhibitory activity test. Among them, EtOAc fraction showed antioxidant activity ($SC_{50}$=4.89 ${\mu}g/mL$) and elastase inhibitory activity ($IC_{50}$=23.5 ${\mu}g/mL$). EtOAc fraction was developed on silica gel by open-column chromatography and consecutively re-developed on C18 resin by prep-HPLC to give ${\alpha}$-viniferin as a major component, which was confirmed by spectrometric analysis. In the assay on expression of MMP-1 mRNA by RT-PCR and protein by western-blot, EtOAc layer (10 ~ 100 ${\mu}g/mL$) was reduced about 50 ~ 60 %, 50 ~ 65 % respectively and ${\alpha}$-viniferin (0.5 ~ 2 ${\mu}g/mL$) was inhibited about 60 ~ 75 %, 55 ~ 65 % respectively in human fibroblast. Therefore, our findings suggest that EtOAc layer of Carex humilis containing ${\alpha}$-viniferin can be useful as an active ingredient for cosmeceuticals of anti-wrinkle effects.
The phenolics of Sambucus sieboldiana var. pendula leaves for functional resources were examined on inhibitory activity against biological enzyme and anti-microbial activity. The amount of phenolic compounds were 11.60±0.18 and 12.43±0.07 mg/g by water and 50% ethanol extraction, respectively. The antioxidative activity of phenolic in extracts was higher than solids. The inhibition activities on angiotensin converting enzyme were 92.08 and 78.33% at 200 ㎍/mL phenolic concentration in water and ethanol extracts, respectively. The xanthine oxidase inhibitory activity were 100% at 200 ㎍/mL phenolic in water and ethanol extracts, respectively. These result was higher than 70.37% of allopurinol as positive control at 200 ㎍/mL. The inhibitory activity against hyaluronidase were each 25.35 and 43.38% in water and ethanol extracts. The water extract from S. sieboldiana var. pendula leaves showed antibacterial activity on the Propionebacterium acnes, Staphylococcus aureus and Streptococcus mutans in water extract and S. mutans in ethanol extract. This result suggests that phenolic from S. sieboldiana var. pendula leaves are suitable as functional foods with anti-hypertension, anti-gout, anti-inflammation and anti-microorganism activities.
To investigate the inhibitory effect of soybean and Korean traditional fermented soybean products on the chemically induced mutagenesis, we extracted soybean, Kanjang, Doenjang, Kochujang, and Chongkukjang with water, methanol and hexane. Inhibitory effect of the extracts was assayed by the SOS chromotest using Escherichia coli PQ37 as a test strain. 4-nitroquinoline-1-oxide(4NQO), N-methyl-N'-nitro-N-nitrosoquanidine(MNNG), and aflatoxin B$_{1}$(AFB$_{1}$) were used as mutagens. Methanol extracts showed relatively higher inhibitory effect than water and hexane extracts. Methanol extracts of soybean, Doenjang, Kochujang, and Chonhkukjang showed inhibitory effect of 68.4, 96.3, 17.5, and 100.9% against MNNG, and 28.6, 109.1, 41.3, and 101.8% against AFB$_{1}$, respectively. Doenjang methanol extract showed inhibitory effect of 51.0, 96.3, and 109.1% against 4NQO, MNNG, and AFB$_{1}$, respectively. Methanol extract of Doenjang showed dose-dependent inhibitory effect against 4NQO, MNNG, and AFB$_{1}$. Inhibitory effect of heat-treated Doenjang and Chongkukjang methanol extracts on the mutagenecity of MNNG and AFB$_{1}$ was remained over 95% of the inhibitory effect of heat-untreated extracts, demonstrating the heat stability of the potent antimutagenic activity.
Objectives : Methicillin-resistant Staphylococcus aureus (MRSA) CCARM 3251 and S. aureusKCTC 1928 have been known to be resistant to many kinds of antibiotics. The extract of Glycyrrhizae Radix showed antibacterial activity against MRSA and antibiotics-resistant S. aureus. Methods : We examined the effects of the water-soluble extract and the methanol-soluble extract of Glycyrrhizae Radix on MRSA and antibiotic-resistant S. aureus. The methanolic extract was further fractionated with organic solvents such as hexane, chloroform, and ethyl acetate in that order. Results and Conclusions : The methanol-soluble extract of Glycyrrhizae Radix showed relatively high antibacterial activity against MRSA and antibiotic-resistant S. aureus. However, the water-soluble extract of Glycyrrhizae Radix showed no antibacterial activity against MRSA and antibiotic-resistant S. aureus. Among the fractions tested, the chloroform fraction showed the highest antibacterial activity against MRSA and antibiotic-resistant S. aureus. The methanol-soluble extract of Glycyrrhizae Radix minimal inhibitory concentrations (MICs) against MRSA and antibiotics-resistant S. aureus were $5{\;}mg/m{\ell}$ in both. The methanol-soluble extract of Glycyrrhizae Radix was separated using thin-layer chromatography and detected with UV -detector. Further study should be carried out to identify which effects cell growth inhibition of MRSA and antibiotics-resistant S. aureus.
