High rate sulfate reduction under acidic conditions opens possibilities for new process flow sheets that allow the selective recovery of metals from mining and metallurgical waste and process water. However, knowledge about high-rate sulfate reduction under acidic conditions is limited. This paper investigates sulfate reduction in a membrane bioreactor at a controlled pH of 5. Sulfate and formate were dosed using a pH-auxostat system while formate was converted into hydrogen, which was used for sulfate reduction. Sulfide was removed from the gas phase to prevent sulfide inhibition. This study shows a high-rate sulfate-reducing bioreactor system for the frrst time at pH 5, with a volumetric activity of 188 mmol $SO_4^{2-}$/I/d and a specific activity of 81 mmol $SO_4^{2-}$volatile suspended solids/d. The microbial community at the end of the reactor run consisted of a diverse mixed population including sulfate-reducing bacteria.
To investigate the effect of salts on the production of erythritol by Torula sp., cells were grown on the media containing various concentrations of KCl or NaCl. Cell growth and glucose consumption rates decreased when KCl or NaCl concentration increased from 0.0 to 0.5M. The production of erythritol, however, was maximal at 0.3M aCl. The erythritol concentration of 54.3g/l in the medium containing 0.3M NaCl and 200g/l glucose was obtained after 120h. The production of erythritol decreased in cultures above 0.3M NaCl or 0.4M KCl due to the inhibition of cell growth. To elucidate the effect of salts more quantitatively, KCl and NaCl concentrations were converted to osmotic pressure. As the osmotic pressure increased, the yield of erythritol from glucose increased regardless of the kinds of salts and the yield of erythritol was approximately 49% at the osmolality of 2.4Os/kg. When the osmotic pressure increased to 2.5Os/kg, the specific growth rate of cells decreased but the production rate of erythritol increased. For the effective production of erythritol, osmotic pressure must be adjusted not to inhibit markedly the growth rate of cells and to stimulate the production rate of erythritol by supplementing salt.
This study was divised to observe the inhibitory effect of growth rate of human colon cancer cells by Eucommial leaf extract, in vitro. Three species of human colon cancer cells, HRT-18, HCT-48 and HT-29, were used for the experiment. Each extract of Eucommial leaf was prepared by extraction with water, 95% alcohol, acetone, chloroform and petroleum ether, and then the inhibitory effect of each extract on the growth rate of cells was compared with control group and each other. The experimental results obtained are summarized as follows; 1. Inhibitory effects on growth rate of human colon cancer fells were strongest in the petroleum ether extract and next in the chloroform extract. 2. Inhibitory effects on the growth rate of the cancer cells by extracts of water, 95% alcohol and acetone were weaker than that of petroleum ether and chloroform. 3. Inhibitory effect of each extract on the cancer cell growth was shown most strong activity in HT-29, and was in order of HRT 18 and HCT-48. In view of the results, it could be suggested that inhibitory effects of non-polar solvent's extracts against the cancer cell growth were more stronger than that of polar solvents and the effects were indicated difference according to the species of the cells.
The purpose of this study was to investigate the nitrification characteristics of biological aerated filter (BAF) packed with ceramic media, especially focusing on nitrite build-up during nitrification. When increasing the nitrogen load above $1.63kgNH_4{^+}-N/m^3{\cdot}d$, ammonium removal efficiency decreased to less than 60% and the nitrite ratio ($NO_2{^-}-N/NO_x-N$) of higher than 75% was achieved due to the inhibitory free ammonia (FA, $NH_3-N$) concentration and oxygen limitation. FA inhibition, however, is not recommended strategy to promote nitrite build-up since FA concentration in the reactor is coupled with decreased ammonium removal efficiency. Nitrite ratio in the effluent was also affected by aeration rate and influent ammonium concentration. Ammonium oxidation was enhanced at a higher aeration rate regardless of influent ammonium concentration but, the nitrite ratio was dependent on both aeration rate and influent ammonium concentration. While a higher nitrite ratio was obtained when BAFs were fed with $50mgNH_4{^+}-N/L$ of influent, the nitrite ratio significantly decreased for a greater influent concentration of $200-300mgNH_4{^+}-N/L$. Taken together, aeration rate, influent ammonium concentration and FA concentrations kept in the BAF were found to be critical variables for nitrite accumulation in the BAF system.
The use of mouth-rinses containing sodium fluoride after tooth brushing has been proposed as a potentially useful measure for the prevention of dental caries in the practice of public health. About 700 middle school girls rinsed their mouths in the school once a week with sodium fluoride sodution after tooth-brushing. Tooth brushing was practiced everyday after meals and the rinses were carried out under the supervision of the teachers. The procedure consisted of gargling 10cc of fluoride solution for one minute and immediately followed by a second rinse with tap water. The rinsing was started in May 1976 and continued for one year. The results were as follows: 1) The inhibition of the 'D' Person rate was 76.04%, 'M' person rate was 80.79%, 'F' person rate was 4.06% and, 'D.M.F.' person rate was 76.86%. 2) the reduction of the 'D.T.' Index was 81.94%, 'M.T.' Index was 85.71%, 'F.T.' Index was 7.69% and, 'D.M.F.T.' Index was 76.83%. 3) Mouth rinsing with 0.3% NaF solution and tooth brushing are of value in the mass control of dental caries.
The effects of 6 biocides on the growth of Chlamydomonas reinhardii in pure culture were studied. For the batch culture assay of biocides, the growth rate in control tubes should be compared with in the test tubes and the effective concentration calculated on the basis of the percent decrease in growth rate at the different toxicant concentration. The concentrations at which 50% decrease in growth rate was observed are $40{\mu}g$/l for butachlor and $350{\mu}g$/l for alachlor in herbicide, $900{\mu}g$/l for phenazine-5-oxide and $3,400{\mu}g$/l for isoprothiolane in fungicide, and $3,330{\mu}g$/l for fenthion and $332,500{\mu}g$/l for trichlorfon in insecticide. The inhibitory effect on the growth of Chlamydomonas reinhardii by the treatment of various biocide concentrations was decreased in order of herbicide>fungicide>insecticide. Chlorophyll and carotenoid content per cell were increased, whereas chlorophyll a/b ratio was hardly affected by biocides. The effects of biocides on pigment content were also decreased in order of herbicide>fungicide>insecticide, which suggested the relationship between the effect of biocides on the pigment content and inhibition of growth rate.
This study aimed to develop a compact partial nitritation step by forming granules with high Ammonia-Oxidizing Bacteria (AOB) fraction using the Air-lift Granulation Reactor (AGR) and to evaluate the feasibility of treating reject water with high ammonium content by combination with the Anammox process. The partial nitritation using AGR was achieved at high nitrogen loading rate ($2.25{\pm}0.05kg\;N\;m-3\;d^{-1}$). The important factors for successful partial nitritation at high nitrogen loading rate were relatively high pH (7.5~8), resulting in high free ammonia concentration ($1{\sim}10mg\;FA\;L^{-1}$) and highly enriched AOB granules accounting for 25% of the total bacteria population in the reactor. After the establishment of stable partial nitritation, an effluent $NO_2{^-}-N/NH_4{^+}-N$ ratio of $1.2{\pm}0.05$ was achieved, which was then fed into the Anammox reactor. A high nitrogen removal rate of $2.0k\; N\;m^{-3}\;d^{-1}$ was successfully achieved in the Anammox reactor. By controlling the nitrogen loading rate at the partial nitritation using AGR, the influent concentration ratio ($NO_2{^-}-N/NH_4{^+}-N=1.2{\pm}0.05$) required for the Anammox was controlled, thereby minimizing the inhibition effect of residual nitrite.
BACKGROUND: Total petroleum hydrocarbons (TPH), which are main materials of soil contamination by oil, are a term used for any mixture of hydrocarbons. Korea Ministry of Environment established the maximum permissible level of TPH in farmland by 500 mg/kg, and reported that the TPH level of soil in 266 installation such as gas station, transport company, and military unit ranged from 1,356 to 55,117 mg/kg and were much higher than the maximum permissible level in 2011. METHODS AND RESULTS: To determine the effect of TPH on crops, we investigated the effect of gasoline, kerosene, and diesel on the germination and radicle growth of mainly consumed crops. The germination rates of control in investigated all crops ranged from 80.0-100%. The germination and radicle growth in majority of investigated crops were not inhibited even at 2,500 mg/L. However, germination in onion, leek, and green perilla and radicle growth in leek, rape, tomato, and green perilla were significantly inhibited by increasing concentrations of gasoline, kerosene and diesel treatment. Germination and radicle growth inhibition of green perilla by kerosene and diesel were the highest, the percent inhibition at the 500 mg/L were 100 and 98.6%, 100 and 88.2%, respectively. 50% inhibition of germination in green perilla by kerosene and diesel were 39.96 and 29.87 mg/L, and 50% inhibition of radicle growth were 52.76 and 177.96 mg/L, respectively. Conclusion(s): These results suggest the possibility that the maximum permissible level of TPH might to be established general level with exception by crops.
A single uniform population of specific, saturable, high affinity binding site of $[^3H]QNB$ guinuclidinyl benzilate(QNB) was identified in the rat cerebral microsomes. The Kd value(37.2 pM) for $[^3H]QNB$ calculated from the kinetically derived rate constants was in agreement with the Kd value(48.9 pM) determined by analysis of saturation isotherms at various receptor concentrations. Dimenhydrinate(DMH), histamine $H_1-blocker$, increased Kd value for $[^3H]QNB$ QNB without affecting the binding site concentrations and this effect resulted from the ability of DMH to slow $[^3H]QNB-receptor$ association. Pirenzepine inhibition curve of $[^3H]QNB$ binding was shallow(nH = 0.52) indicating the presence of two receptor subtypes with high ($M_1-site$) and low($M_2-site$) affinity for pirenzepine. Analysis of these inhibition curves yielded that 68% of the total receptor populations were of the $M_1-subtype$ and the remaining 32% of the $M_2-subtype$. Ki values for the $M_1-$ and $M_2-subtypes$ were 2.42 nM and 629.3 nM, respectively. Ki values for $H_1-blockers$ that inhibited $[^3H]QNB$ binding varied with a wide range ($0.02-2.5\;{\mu}M$). The Pseudo-Hill coefficients for inhibition of $[^3H]QNB$ binding by most of $H_1-blockers$ examined except for oxomemazine inhibition of $[^3H]QNB$ binding were close to one. The inhibition curve for oxomemazine in competition with $[^3H]QNB$ was shallow(nH = 0.74) indicating the presence of two receptor populations with different affinities for this drug. The proportion of high and low affinity was 33:67. The Ki values for oxomemazine were $0.045{\pm}0.016\;{\mu}M$ for high affinity and $1.145{\pm}0.232\;{\mu}M$ for low affinity sites. These data indicate that muscarinic receptor blocking potency of $H_1-blockers$ varies widely between different drugs and that most of $H_1-blockers$ examined are nonselective antagonist for the muscarinic receptor subtypes, whereas oxomemazine might be capable of distinguishing between subclasses of muscarinic receptor.
This study was conducted to investigate the physicochemical characteristics and biological activities of water and 30%, 50%, 70%, 100% ethanol extracts from Artemisia Argyi H. and fermented Artemisia Argyi H. The yield was the highest in the 30% ethanol extract with Argyi H. at 29.74%. Total phenol and flavonoid contents were the highest in 70% ethanol extract with Argyi H. at 72.25 mg/g and 33.34 mg/g, respectively. The antioxidant activities of all extracts were significantly increased in a dose dependent manner. The 70% ethanol extract from Argyi H. show the highest level of DPPH, ABTS radical scavenging activity and bleaching inhibition activity in ${\beta}$-carotene linoleic acid system. Tyrosinase inhibition activity was also higher in the 70% ethanol extract, and the lowest in the 100% ethanol extract with Argyi H. at 50.01% and 11.44% at $500{\mu}g/ml$ concentration, respectively. At $250{\mu}g/ml$ concentration, the xanthin oxidase inhibition activity of water extract was more than 60%, and it was higher than the extracts. These results suggest that the 70% ethanol extract of Artemisia Argyi H. has a high rate of biological activities and can be useful to develop functional food ingredients.
