Gene Expression Analysis of Lung Injury in Rats Induced by Exposure to MMA-SS Welding Fume for 30 Days

  • The welding fume has been implicated as a causal agent in respiratory disease such as pneumoconiosis. The molecular mechanism by which welding fume induces toxicity in the lung is still unknown, but studies have focused on histological structure and indirect approach measuring the pulmonary damage markers. In the present study, gene expression profiles were analyzed in the lung of rats exposed by manual metal-arc stainless-steel (MMA-SS) welding fume for 30 days using Affymetrix GeneChip$^{(R)}$. Totally, 379 genes were identified as being either up- or down-regulated over 2-fold changes (P<0.01) in the lung of low- or high-dose group and were analyzed by using hierarchical clustering. We focused on genes involved in immune/inflammation responses were differentially regulated during lung injury induced by welding fume exposure. The information of these deregulated genes may contribute in elucidation of the inflammation mechanism during lung injury such as lung fibrosis.

Towards Quantitative Assessment of Human Exposures to Indoor Radon Pollution from Groundwater

  • A report by the national research council in the United States suggested that many lung cancer deaths each year be associated with breathing radon in indoor air. Most of the indoor radon comes directly from soil beneath the basement of foundations. Recently, radon released from groundwater is found to contribute to the total inhalation risk from indoor air. This study presents the quantitative assessment of human exposures to radon released from the groundwater into indoor air. At first, a three-compartment model is developed to describe the transfer and distribution of radon released from groundwater in a house through showering, washing clothes, and flushing toilets. Then, to estimate a daily human exposure through inhalation of such radon for an adult. a physiologically-based pharmacokinetic(PBPK) model is developed. The use of a PBPK model for the inhaled radon could provide useful information regarding the distribution of radon among the organs of the human body. Indoor exposure patterns as input to the PBPK model are a more realistic situation associated with indoor radon pollution generated from a three-compartment model describing volatilization of radon from domestic water into household air. Combining the two models for inhaled radon in indoor air can be used to estimate a quantitative human exposure through the inhalation of indoor radon for adults based on two sets of exposure scenarios. The results obtained from the present study would help increase the quantitative understanding of risk assessment issues associated with the indoor radon released from groundwater.

Visibility of Internal Target Volume of Dynamic Tumors in Free-breathing Cone-beam Computed Tomography for Image Guided Radiation Therapy

  • Respiratory-induced dynamic tumors render free-breathing cone-beam computed tomography (FBCBCT) images with motion artifacts complicating the task of quantifying the internal target volume (ITV). The purpose of this paper is to study the visibility of the revealed ITV when the imaging dose parameters, such as the kVp and mAs, are varied. The $Trilogy^{TM}$ linear accelerator with an On-Board Imaging ($OBI^{TM}$) system was used to acquire low-imaging-dose-mode (LIDM: 110 kVp, 20 mA, 20 ms/frame) and high-imaging-dose-mode (HIDM: 125 kVp, 80 mA, 25 ms/frame) FBCBCT images of a 3-cm diameter sphere (density=0.855 $g/cm^3$) moving in accordance to various sinusoidal breathing patterns, each with an unique inhalation-to-exhalation (I/E) ratio, amplitude, and period. In terms of image ITV contrast, there was a small overall average change of the ITV contrast when going from HIDM to LIDM of $6.5{\pm}5.1%$ for all breathing patterns. As for the ITV visible volume measurements, there was an insignificant difference between the ITV of both the LIDM- and HIDM-FBCBCT images with an average difference of $0.5{\pm}0.5%$, for all cases, despite the large difference in the imaging dose (approximately five-fold difference of ~0.8 and 4 cGy/scan). That indicates that the ITV visibility is not very sensitive to changes in imaging dose. However, both of the FBCBCT consistently underestimated the true ITV dimensions by up to 34.8% irrespective of the imaging dose mode due to significant motion artifacts, and thus, this imaging technique is not adequate to accurately visualize the ITV for image guidance. Due to the insignificant impact of imaging dose on ITV visibility, a plausible, alternative strategy would be to acquire more X-ray projections at the LIDM setting to allow 4DCBCT imaging to better define the ITV, and at the same time, maintain a reasonable imaging dose, i.e., comparable to a single HIDM-FBCBCT scan.

Twenty-Eight-Day Repeated Inhalation Toxicity Study of Aluminum Oxide Nanoparticles in Male Sprague-Dawley Rats

  • Aluminum oxide nanoparticles ($Al_2O_3$ NPs) are among the most widely used nanomaterials; however, relatively little information about their risk identification and assessment is available. In the present study, we aimed to investigate the potential toxicity of $Al_2O_3$ NPs following repeated inhalation exposure in male Sprague-Dawley rats. Rats were exposed to $Al_2O_3$ NPs for 28 days (5 days/week) at doses of 0, 0.2, 1, and $5mg/m^3$ using a nose-only inhalation system. During the experimental period, we evaluated the clinical signs, body weight change, hematological and serum biochemical parameters, necropsy findings, organ weight, and histopathology findings. Additionally, we analyzed the bronchoalveolar lavage fluid (BALF), including differential leukocyte counts, and aluminum contents in the major organs and blood. Aluminum contents were the highest in lung tissues and showed a dose-dependent relationship in the exposure group. Histopathology showed alveolar macrophage accumulation in the lungs of rats in the $5mg/m^3$ group during exposure and recovery. These changes tended to increase at the end of the recovery period. In the BALF analysis, total cell and neutrophil counts and lactate dehydrogenase, tumor necrosis factor-${\alpha}$, and interleukin-6 levels significantly increased in the 1 and $5mg/m^3$ groups during exposure. Under the present experimental conditions, we suggested that the no-observed-adverse-effect level of $Al_2O_3$ NPs in male rats was $1mg/m^3$, and the target organ was the lung.

  • Many reactive dyes have been used in occupational settings without knowing their toxicity and health hazard information. To investigate the toxicity of reactive dye, C.I.No. Reactive Red 195 was exposed to male and female Sprague Dawley rats by inhalation for 28 days. The rats were exposed C.I.No. Reactive Red 195 for 6 hrs per day and 5days per week. The concentrations for the inhalation exposure were 0, 10, 40 and $160mg/m^3$. After 4 weeks of exposure, rats were examined for exposure related changes through pathology, blood biochemistry and hematology. There were no dose related changes including clinical signs, body weight and relative organ weight changes, hematological and biochemical and histopathological findings. The results indicate that no observed adverse effect level (NOAEL) of 28 days inhalatrion toxicity test for C.I.No. Reactive Red 195 was $160mg/m^3$.

3D 프린팅 가동 조건 별 발생 입자크기 분포와 흡입 노출량 추정 (Size Distributions of Particulate Matter Emitted during 3D Printing and Estimates of Inhalation Exposure)

  • Objective: This study aimed to identify the size distributions of particulate matter emitted during 3D printing according to operational conditions and estimate particle inhalation exposure doses at each respiratory region. Methods: Four types of printing filaments were selected: acrylonitrile-butadiene-styrene (ABS), polylactic acid (PLA), Laywood, and nylon. A fused deposition modeling (FDM) 3D printer was used for printing. Airborne particles between 10 nm and $10{\mu}m$ were measured before, during, and after printing using real-time monitors under extruder temperatures from 215 to $290^{\circ}C$. Inhalation exposures, including inhaled and deposited doses at the respiratory regions, were estimated using a mathematical model. Results: Nanoparticles dominated among the particles emitted during printing, and more particles were emitted with higher temperatures for all materials. Under all temperature conditions, the Laywood emitted the highest particle concentration, followed by ABS, PLA, and nylon. The particle concentration peaked for the initial 10 to 20 minutes after starting operations and gradually decreased with elapsed time. Nanoparticles accounted for a large proportion of the total inhaled particles in terms of number, and about a half of the inhaled nanoparticles were estimated to be deposited in the alveolar region. In the case of the mass of inhaled and deposited dose, particles between 0.1 and $1.0{\mu}m$ made up a large proportion. Conclusion: The number of consumers using 3D printers is expected to expand, but hazardous emissions such as thermal byproducts from 3D printing are still unclear. Further studies should be conducted and appropriate control strategies considered in order to minimize human exposure.

  • 본 연구에서는 황벽나무, 두충나무, 귀룽나무 추출물 등을 포함하는 천연추출물에대해 항균활성 효과 및 안전성에 대해 검토하였다. 항균활성은 기회 병원성균인 Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli 및 Candida albicans에 대해 실험되었다. 안전성 시험으로는 세포독성실험, 단회 경구투여 독성시험, 단회 흡입투여 독성시험, 반복 흡입투여 독성시험, 안점막 자극시험이 실시되었다. 항균력효과시험 결과 추출물은 Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Candida albicans 4가지 균 모두에 대해 우수한 항균활성을 보여주었다. 천연추출물의 단회 경구투여 독성시험, 단회 흡입투여 독성시험 및 반복 흡입투여 독성시험, 안점막 자극시험의 결과 독성은 관찰되지 않았다. 따라서 황벽나무, 두충나무, 귀룽나무 추출물 등을 포함한 천연추출물은 뛰어난 안전성과 항균력이 있음을 확인하였다.

RADIOLOGICAL DOSE ASSESSMENT ACCORDING TO METHODOLOGIES FOR THE EVALUATION OF ACCIDENTAL SOURCE TERMS

  • The object of this paper is to evaluate the fission product inventories and radiological doses in a non-LOCA event, based on the U.S. NRC's regulatory methodologies recommended by the TID-14844 and the RG 1.195. For choosing a non-LOCA event, one fuel assembly was assumed to be melted by a channel blockage accident. The Hanul nuclear power reactor unit 6 and the CE $16{\times}16$ fuel assembly were selected as the computational models. The burnup cross section library for depletion calculations was produced using the TRITON module in the SCALE6.1 computer code system. Based on the recently licensed values for fuel enrichment and burnup, the source term calculation was performed using the ORIGEN-ARP module. The fission product inventories released into the environment were obtained with the assumptions of the TID-14844 and the RG 1.195. With two kinds of source terms, the radiological doses of public in normal environment reflecting realistic circumstances were evaluated by applying the average condition of meteorology, inhalation rate, and shielding factor. The statistical analysis was first carried out using consecutive three year-meteorological data measured at the Hanul site. The annual-averaged atmospheric dispersion factors were evaluated at the shortest representative distance of 1,000 m, where the residents are actually able to live from the reactor core, according to the methodology recommended by the RG 1.111. The Korean characteristic-inhalation rate and shielding factor of a building were considered for a series of dose calculations.

  • 본 연구에서는 황벽나무, 두충나무 등을 포함하는 복합수목추출물의 항균활성 효과 및 안전성에 대해 검토하였다. 항균활성은 피부상재균이며 기회 병원성균인 Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli 및 Candida albicans에 대해 disc 확산법으로 실험되었다. 안전성 시험으로는 단회 경구투여 독성시험, 단회투여 흡입독성 시험, 반복투여 흡입독성시험이 실시되었다. 항균력효과시험 결과 추출물은 Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대해 우수한 항균활성을 보여주었으나, Escherichia coli에서는 항균활성이 없었다. 복합수목추출물의 단회 경구투여 독성시험, 단회투여 흡입독성시험 및 반복투여 흡입독성시험의 결과 독성은 관찰되지 않았다. 따라서 황벽나무, 두충나무 등을 포함한 복합수목추출물은 천연 항살균제로서 상업화 가능성이 매우 높음을 알 수 있었다.

Development of Internal Dose Assessment Procedure for Workers in Industries Using Raw Materials Containing Naturally Occurring Radioactive Materials

  • Background: It is necessary to assess radiation dose to workers due to inhalation of airborne particulates containing naturally occurring radioactive materials (NORM) to ensure radiological safety required by the Natural Radiation Safety Management Act. The objective of this study is to develop an internal dose assessment procedure for workers at industries using raw materials containing natural radionuclides. Materials and Methods: The dose assessment procedure was developed based on harmonization, accuracy, and proportionality. The procedure includes determination of dose assessment necessity, preliminary dose estimation, airborne particulate sampling and characterization, and detailed assessment of radiation dose. Results and Discussion: The developed dose assessment procedure is as follows. Radioactivity concentration criteria to determine dose assessment necessity are $10Bq{\cdot}g^{-1}$ for $^{40}K$ and $1Bq{\cdot}g^{-1}$ for the other natural radionuclides. The preliminary dose estimation is performed using annual limit on intake (ALI). The estimated doses are classified into 3 groups ( < 0.1 mSv, 0.1-0.3 mSv, and > 0.3 mSv). Air sampling methods are determined based on the dose estimates. Detailed dose assessment is performed using air sampling and particulate characterization. The final dose results are classified into 4 different levels ( < 0.1 mSv, 0.1-0.3 mSv, 0.3-1 mSv, and > 1 mSv). Proper radiation protection measures are suggested according to the dose level. The developed dose assessment procedure was applied for NORM industries in Korea, including coal combustion, phosphate processing, and monazite handing facilities. Conclusion: The developed procedure provides consistent dose assessment results and contributes to the establishment of optimization of radiological protection in NORM industries.
