Screening Test on the Ingredient of the Genus Thalictrum Roots in Korea (한국산 Thalictrum속 식물 지하부의 성분검색)

  • This study is about the five species of the genus Thalictrum growing abundantly in South Korea. Dried roots of plants; T. aquilegifolium var. japonica, T. simplex var. affine, T. actaefolium, T. tuberiferum were compared by morphological observation and screening test on the ingredient extracted by methanol, ether etc.. The experiments by thin-layer chromatography was at tempted to find out the similarity and the difference among them. In conclusion, the author found that: 1) A special component similar to coumarin was found under UV-ray from T. actaefolium. 2) By thin-layer chromatography, they show the corresponding spot supposed to be that of berberine and magnoflorine according to the $T_{ABLE}\;IV$. Therefore it is worthwhile considering that Thalictrum species would be a substitution for Epimedium species used as a tonic. 3) Thalictrum species has resemblance of ingredient and chemotaxonomy gives more help to classify them than the morphology.

Network Pharmacology: Prediction of Astragalus Membranaceus' and Cornus Officinalis' Active Ingredients and Potential Targets to Diabetic Nephropathy (네트워크 약리학을 통한 당뇨병성 신병증에서의 황기와 산수유의 활성 성분 및 잠재 타겟 예측)

  • The purpose of this study is to predict the effects of macroscopic and integrative therapies by finding active ingredients, potential targets of Astragalus membranaceus (Am) and Cornus officinalis (Co) for diabetic nephropathy. We have constructed network pharmacology-based systematic and network methodology by system biology, chemical structure, chemogenomics. We found several active ingredients of Astragalus membranaceus (Am) and Cornus officinalis (Co) that were speculated to bind to specific receptors which had been known to have a role in the progression of diabetic nephropathy. Four components of Am and eleven components of Co could bind to iNOS; two ingredients of Am and six ingredients of Co could docking to cGB-PDE; one component of Am and nine components of Co could bind to ACE; three ingredients of Co with neprilysin; three components of Co with ET-1 receptor; four ingredients of Am and fourteen ingredients of Co with mineralocorticoid receptor; one component of Am and seven components of Co with interstitial collagenase; one ingredient of Am and ten ingredients of Co with membrane primary amine oxidase; one component of Am and four components of Co with JAK2; two ingredients of Am and one ingredient of Co with MAPK 12; one component of Am and five components of Co could docking to TGF-beta receptor type-1. From this work we could speculate that the possible mechanisms of Am and Co for diabetic nephropathy are anti-inflammatory, antioxidant and antihypertensive effects.

Applications of prescriptions including Paeoniae Radix as a main component in Dongeuibogam (동의보감(東醫寶鑑) 중(中) 백작약(白芍藥)이 주약(主藥)으로 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察))

  • This report describes 173 formulas related to which are mainly used Paeoniae Radix in Dongeuibogam. The following conclusions were induced through investigations on the formulas that are used Paeoniae Radix as a main component. 1. We found the formulas in Dongeuibogam that Paeoniae Radix is used as a main ingredient in each prescriptions. 22 times(12.4%) of them are recorded in women's disease chapter, 20 times(11.3%) are in feces chapter, 14 times(7.9%) are in uterus, 12 times(6.8%) are in wind chapter, 10 times(5.6%) are in fatigue chapter, 9 times(5.1%) are in blood chapter, which are arranged in order of frequency. 2. Formulas that utilize Paeoniae Radix as the main ingredient are used in the treatment of women's disease, diarrhea, paralysis, yin and yang-deficiency syndrome. They are also used for treating 117 different types of diseases. 3. The dosage of Paeoniae Radix in formulas is from 1 don(nearly 3.75g) to 1 nyang 5 don (nearly 56.25g), however 1 don has been taken the most for clinical application. 4. Paeoniae Radix can make various effects with other ingredient, like fulfilling yin and blood, relaxing muscles and allaying pain. And this can be found in the formulas such as Samultang(四物湯), Jakyackgamchotang(芍藥甘草湯), Sambaektang(三白湯).

Calculation of Replacement Price for Alternative Feed Ingredient in Consideration of Nutrient Content in Feed Ingredient Fed to Broiler Chickens (영양소 함량을 고려한 육계 대체 원료사료의 대체가격 계산)

  • The objective of this study was to calculate the unit price of an alternative feed ingredient for broiler chickens in consideration of the energy and nutritive contents in the feed ingredient by using a simple Excel worksheet. A corn-distiller's dried grains with solubles (corn-DDGS) was used as an alternative ingredient and corn and soybean meal as ingredients to be replaced. The net change of feed price was calculated based on the replacement values which were estimated in consideration of energy and nutrient concentration in feed ingredients used in the calculation, the price of feed ingredients and inclusion rate of the alternative ingredient. The nitrogen corrected apparent metabolizable energy(AMEn) and standardized ileal digestible AA including Lys, Met, Thr, and Trp, total Ca, and available P were employed as nutritive component to calculate the replacement values for individual feed ingredients. The equation for replacement was 1 ${\times}$ corn-DDGS + 0.0334 ${\times}$ soybean oil + 0.0182 ${\times}$ Limestone = 0.8893 ${\times}$ corn + 0.13 ${\times}$ soybean meal + 0.0004 ${\times}$ Lys + 0.0022 ${\times}$ Met + 0.0005 ${\times}$ Trp + 0.0028 ${\times}$ Thr + 0.0264 ${\times}$ dicalcium phosphate. The replacement price of corn-DDGS was calculated to be 270 won/kg when the inclusion rate was 15% and the energy and nutrient contents were considered in the calculation. In conclusion, the Excel-based ingredient price calculator may be useful to determine the economic value when an alternative feed ingredient is used in diets fed to broiler chickens.

A study on Applications of prescriptions including Rhizoma Anemarrhenae as a main component in Dongeuibogam (동의보감(東醫寶鑑) 중(中) 지모(知母)가 주약(主藥)으로 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察))

  • This report describes 68 studies related to the use of Rhizoma Anemarrhenae main blended prescription from Dongeuibogam. The following conclusion were reached through investigations on the prescriptions that use Rhizoma Anemarrhenae as a key ingredient. Prescriptions that Rhizoma Anemarrhenae was taken as a monarch drug are utilized for 21 therapeutic purposes, for example, cough, malaria, Sang Han syndromes and eye disease. In particular, 22.1% of prescriptions appear in the chapter of cough, and 13.2% of those appear in the chapter of malaria. Prescriptions that utilize Rhizoma Anemarrhenae as the main ingredient are used in the treatment of cough, asthma and malaria. Rhizoma Anemarrhenae is used in pathogenic factors such as fire, and used in pathology related to kidny system. The dosage of Rhizoma Anemarrhenae is 3bun(about 1.12 gram) to 3jeon(about 11.25 gram), however 1jeon(about 3.75 gram) has been taken the most for clinical application. Gamlihuan is the most useful base prescription which use the Rhizoma Anemarrhenae as the main ingredient.

Quantification of an active ingredient in tablets by NIR transmission measurements

  • For the quality control of tablets several parameters have to be checked. The most important one is the content of an active ingredient which has to match a narrow range around the designated content. The only useful measurement mode is transmission which provides information of the complete tablet. A measurement in diffuse reflectance would register only the surface which is useless especially in case of a coated tablet. In this work tablets for a clinical study (placebo/verum studies) with very low concentrations of the active ingredient were measured. The concentration range was 0 to 6 mg with a total weight of the tablets of 105 mg, leading to a highest concentration of the active component of 5.7% by weight. Especially the spectroscopic distinction between the placebo and the low dosage forms with 0.25 and 0.5 mg active agent requires an extraordinarily accurate sampling technique. Using the VECTOR 22/N-T in transmission mode allows the collection of the information from the complete tablets. A quantitative PLS-model with transmission spectra from the tablets described above shows that the active substance can be predicted with a RMSECV (root mean square error of cross validation) of 0.04% absolute for this special application. The results are compared with those of measurements in diffuse reflectance using different accessories.

  • Active ingredients have been around in cosmetics for a long time but have they really resulted in active cosmetic products\ulcorner In order to achieve this, the right active needs to be delivered to the right location at the right concentration for the correct period of time. And the extent (and therefore the concentration) of this delivery depends on the formulation. From a rather theoretical approach based on the polarity of the active ingredient, the stratum corneum and the oil phase, the Relative Polarity Index was established that enables the selection of a suitable emollient for ensuring skin penetration of the active ingredient. Practical examples subsequently show the validity of this approach that demonstrates that one can regulate the delivery of an active molecule (and therefore the efficacy of a cosmetic formulation) by selection and control of the emollient system. Cosmetic formulations are generally quite complex mixtures and subsequent experiments using different emulsifier systems indicated that this component of a cosmetic formulation could also have an impact on steering the active ingredient to the right layer of the skin, although it is too early to be able to derive general rules from this.

A Fast and Precise Blob Detection

  • Blob detection is an essential ingredient process in some computer applications such as intelligent visual surveillance. However, previous blob detection algorithms are still computationally heavy so that supporting real-time multi-channel intelligent visual surveillance in a workstation or even one-channel real-time visual surveillance in a embedded system using them turns out prohibitively difficult. In this paper, we propose a fast and precise blob detection algorithm for visual surveillance. Blob detection in visual surveillance goes through several processing steps: foreground mask extraction, foreground mask correction, and connected component labeling. Foreground mask correction necessary for a precise detection is usually accomplished using morphological operations like opening and closing. Morphological operations are computationally expensive and moreover, they are difficult to run in parallel with connected component labeling routine since they need much different processing from what connected component labeling does. In this paper, we first develop a fast and precise foreground mask correction method utilizing on neighbor pixel checking which is also employed in connected component labeling so that the developed foreground mask correction method can be incorporated into connected component labeling routine. Through experiments, it is verified that our proposed blob detection algorithm based on the foreground mask correction method developed in this paper shows better processing speed and more precise blob detection.

  • We composed a method of assaying Capsaicin in Kochuzang, which is the most characteristic hot seasoning or food in Korean foods, by making use of Fujida's $method^{2}$). Capsaicin, a hot ingredient of hot pepper, was isolated from the acetone extract of dried Kochuzang with paperchromatography and anlayzed quantitatively with Electrophotometry.

  • Chemometrics using near-infrared (NIR) and Raman spectroscopy have found significant uses in a variety quantitative and qualitative analyses of pharmaceutical products in complex matrixes. Most of the pharmaceutical can be measured directly with little or no sample preparation using these spectroscopic methods. During pharmaceutical manufacturing process, analytical techniques with no or less sample preparation are very critical to confirm the quality. This study showed NIR and Raman spectroscopy with principal component analysis (PCA) was very effective for the blending processing control. It is of utmost importance to evaluate critical parameters related to quality of products during pharmaceutical processing. The blending is confirmed by off-line determination of active pharmaceutical ingredient (API) by a conventional method such as high performance liquid chromatography (HPLC) and UV spectroscopy. These analytical methods are time-consuming and ineffective for real time control. This study showed the possibility for the determination of blend uniformity end-point of CR tablets with the use of both NIR and Raman spectroscopy. The samples were acquired from six positions during blending processing with U-type blender from 0 to 30 min. Using both collected NIR and Raman spectral data, principal component analysis (PCA) was used to follow the uniformity of blending and finally determine the end-point. The variation of homogeneity of six samples during blending was clearly found and blend uniformity end-point was successfully confirmed in the domains of principal component (PC) scores.
