Thermal denaturation analysis of protein

  • Near infrared (NIR) spectroscopy is a powerful technique for non-destructive analysis that can be obtained in a wide range of environments. Recently, NIR measurements have been utilized as probe for quantitative analysis in agricultural, industrial, and medical sciences. In addition, it is also possible to make practical application on NIR for molecular structural analysis. In this work, Fourier transform near infrared (FT-NIR) measurements were carried out to utilize extensively in the relative amounts of different secondary structures were employed, such as Iysozyme, concanavalin A, silk fibroin and so on. Several broad NIR bands due to the protein absorption were observed between 4000 and $5000\;^{-1}$. In order to obtain more structural information from these featureless bands, second derivative and Fourier-self-deconvolution procedures were performed. Significant band separation was observed near the feature at $4610\;^{-1}$ ,. Particularly the peak intensity at $4525\;^{-1}$ shows a characteristic change with thermal denaturation of fibroin. The structural information can be also obtained by mid-IR and CD spectral. Correlation of NIR spectra with protein structure is discussed.

  • In this paper, the novel approach for the fault diagnosis of the bearing equipped with rotational mechanical facilities was studied. As research works, by applying the ball bearing used extensively in many industrial fields, experiments were conducted in order to propose the new prognostic method about the condition monitoring for the rotational bodies based on the condition analysis of infrared thermography. Also, by using the vibration spectrum analysis, the real time monitoring was performed. As results, it was confirmed that infrared thermography method could be adapted into monitor and diagnose the fault for bearing by evaluating quantitatively and qualitatively the temperature characteristics according to the condition of the ball bearing.

Quantitative Analysis of Indomethacin by the Portable Near-Infrared (NIR) System (근적외분광분석법을 이용한 인도메타신의 정량분석)

  • Near-infrared (NIR) system was used to determine rapidly and simply indomethacin in buffer solution for a dissolution test of tablets and capsules. Indomethacin standards were prepared ranging from 10 to 50 ppm using the mixture of phosphate buffer (pH 7.2) and water (1 : 4). The near-infrared (NIR) transmittance spectra of indomethacin standard solutions were collected by using a quartz cell in 1 mm and 2 mm pathlength. Partial least square regression (PLSR) was explored to develop calibration models over the spectral range 1100∼1700 nm. The model using 1 mm quartz cell was better than that using 2 mm quartz cell. The PLSR models developed gave standard error of prediction (SEP) of 0.858 ppm. In order to validate the developed calibration model, routine analysis was performed using another standard solutions. The NIR routine analysis showed good correlation with actual values. Standard error of prediction (SEP) is 1.414 ppm for 7 indomethacin samples in routine analysis and its error was permeable in the regulation of Korean Pharmacopoeia (VII). These results show the potential use of the real time monitoring for indomethacin during a dissolution test.

Infrared Reflector Design using the Phase Field Method for Infrared Stealth Effect (적외선 피탐지를 위한 페이즈 필드법 기반의 적외선 반사층 설계)

  • In this paper, infrared reflector design targeting infrared stealth effect is presented using structural optimization based on the phase field method. The analysis model was determined to accomplish the design that an incident infrared wave was reflected to a desired direction. The design process was to maximize the objective value at the measuring domain located in a target region and the design objective was set to the Poynting vector value which represents the energy flux. Optimization results were obtained according to the variation of some parameter values related to the phase field method. The model with a maximum objective value was selected as the final optimal model. The optimal model was modified to eliminate the gray scale using the cut-off method and it confirmed improved performance. In addition, to check the desired effect in the middle wave infrared range(MWIR), the analysis was performed by changing the input wavelength. The finite element analysis and optimization process were performed by using the commercial package COMSOL combined with the Matlab programming.

Systematic Review and Meta-Analyses of Diagnostic Accuracy of Infrared Thermometer when Identifying Fever in Children (아동에서의 적외선 체온 측정의 진단정확도 연구 : 체계적 문헌 고찰과 메타분석)

  • Purpose: Infrared thermometers are increasingly used as a convenient, non-invasive assessment method for febrile children. However, the diagnostic accuracy of the infrared thermometer for children has been questioned, particularly in relation to sensitivity and specificity. The aim of this study was to evaluate diagnostic accuracy of infrared thermometers in febrile children. Methods: Articles published between 1966 and 2012 from periodicals indexed in the Ovid Medline, Embase, CINAHL, Cochrane, KoreaMed, NDSL, KERIS and other databases were selected, using the following keywords: 'infrared thermometer'. The QUADAS-II was applied to assess the internal validity of the diagnostic studies. Selected studies were analyzed using meta-analysis with MetaDisc 1.4. Results: Nineteen diagnostic studies with high methodological quality, involving 4,304 children, were included. The results of meta-analysis showed that the pooled sensitivity, specificity and AUC (Area Under the Curve) of infrared tympanic thermometers in children over 1 year were 0.80 (95% CI 0.78, 0.81), 0.94 (95% CI 0.93, 0.95) and 0.95 respectively. However the diagnostic accuracy of infrared tympanic thermometers in children with hyperthermia was low. Conclusion: The diagnostic accuracy of infrared tympanic thermometer was similar to axillary and rectal thermometers indicating a need for further research to substantiate these findings in children with hyperthermia.

  • This paper presents the method for analyzing surface temperature of Electric incoming Apparatus. For the experiment, the surface temperature of electric power apparatus was measured and analyzed by using a infrared thermal imaging system. Surface Discharges(SD) have very complex characteristics of discharge patterns, therefore it requires the development of precise analysis methods. recently, studies on infrared thermal imaging system are carried out to analyze temperature distribution of power equipments through condition diagnosis and to diagnose the degradation of power equipments. The changes in suface temperature was measured by using the infrared thermal imaging system under hot line condition. The system was set up based on the diagnostic method of the electric incoming apparatus.

  • Estimation of sugar and reducing sugar content in molasses is very important task in sugar refineries. Conventional methods of determination of sugar content in molasses samples are highly time consuming and employ hazardous chemicals. Due to the physical properties of molasses, probability of error in conventional analytical techniques is high. These methods have proven to be inefficient for a process control in any sugar industry. Hence development of a rapid, inexpensive, physical and also accurate method for sugar determination in molasses will be highly useful. Near Infrared spectroscopy is being widely used worldwide as an analytical technique in food industry. The technique offers the advantage of being non-destructive and rapid. The present paper highlights the potential of near infrared reflectance spectroscopy as a rapid and automated analytical technique for determination of sugar and reducing sugar content in molasses. A number of molasses samples were collected during and after the sugar season from Havana Sugar Industry, Havana. The samples were chosen so as to obtain a wide range of concentration of sugar and reducing sugars. This was done in order to achieve a good calibration curve with widely spread data points. These samples were scanned in the region of 1100 - 2500 nm in diffuse reflectance mode. An indigenous ELICO NIR spectrophotometer, modified according to the requirements of sugar industry was used for this purpose. Each sample was also analyzed simultaneously by standard chemical methods. Chemical values were taken as reference for near infrared analysis. In order to obtain the most accurate calibration for the set of samples, various mathematical treatments were employed. Partial Least Square method was found to be most suitable for the analysis. A comparison is made between the actual values (chemical values) and the predicted values (NIR values). The actual values agree very well with the predicted values showing the accuracy of the technique. The validity of the technique is checked by predicting the concentration of sugar in unknown molasses samples using the calibration curve. The present investigation assesses the feasibility of the technique for on-line monitoring of sugars present in molasses in sugar industries.

Near Infrared Reflectance Spectroscopy for Non-Invasive Measuring of Internal Quality of Apple Fruit

  • In this study, we investigated the feasibility of non-destructive determination of internal quality factors of Fuji apple fruit using near infrared(NIR) reflectance spectroscopy and developed the calibration models. As the reference methods, refractometer, titration and texture analyzer for sugar content, acidity and firmness were used, respectively. Samples were scanned from 1100∼2500nm with InfraAlyzer 500C spectrometer and SESAME software was used for data analysis. A multiple linear regression(MLR) analysis was performed to develop the calibrations. The correlation coefficient(R) and standard error of prediction(SEP) were as follows; 0.91, 0.41$^{\circ}$Brix for sugar content, 0.90, 0.04% for acidity and 0.84, 0.094 kg for firmness, respectively. This study shows that NIR spectroscopy can be used to evaluate the sugar content acidity and firmness of apple fruit with acceptable accuracy.

Fatigue performance assessment of welded joints using the infrared thermography

  • Taking the superficial temperature increment as the major fatigue damage indicator, the infrared thermography was used to predict fatigue parameters (fatigue strength and S-N curve) of welded joints subjected to fatigue loading with a high mean stress, showing good predictions. The fatigue damage status, related to safety evaluation, was tightly correlated with the temperature field evolution of the hot-spot zone on the specimen surface. An energetic damage model, based on the energy accumulation, was developed to evaluate the residual fatigue life of the welded specimens undergoing cyclic loading, and a good agreement was presented. It is concluded that the infrared thermography can not only well predict the fatigue behavior of welded joints, but also can play an important role in health detection of structures subjected to mechanical loading.

  • In quantitative analysis of sulfonamide drugs by means of infrared Spectrophotometry, the author tried to analyse Sulfamethoxydiazine and Sulfamethoxypyridazine in their Pharmaceutical Preparations. By the survey of infrared absorption spectra of Sulfamethoxydiazine and Sulfamethoxypyridazine. We found quantitative properties at the peaks of wave number 1150 cm$^{-1}$ (Sulfamethoxydiazine) and 1090 cm$^{-1}$ (Sulfamethoxypyridazine) in the spectra. These peaks obey Beer's Law in a range of 50-150 ug for Sulfamethoxydiazine and 70-350 ${\mu}$g for Sulfamethoxypyridazine in 300 mg KBr disc. By this method, Sulfamethoxydiazine and Sulfamethoxypyridazine could be analyzed less than 2% error.
