마렉병 바이러스 강독주의 실험 접종에 의해 유발된 닭 피부병변에 침윤한 림프구 표현형의 변화 (Phenotypical changes of lymphocyte subsets infiltrated in the skin lesions induced experimentally by very virulent strain of Marek's disease virus in chickens)

  • Marek's disease virus (MDV) can cause skin lesions including inflammatory to tumorous. The phenotypical changes of lymphocytes infiltrating in the skin lesions induced by MDV were not clear. Therefore, the skin biopsies taken at weekly intervals for 8 weeks from the same specific-pathogen free chickens inoculated with Md/5 MDV were examined to analysis the phenotypical changes of lymphocytes. Histologically skin lesions progressed from initial inflammatory to late tumorous. Sequentially CD4+ T lymphocytes increased gradually in number from initial skin lesions and were major composition cells in the tumor lesions. Regardless of inflammatory or tumor lesions, CD8+ T cells and ${\gamma}{\delta}$ T cells infiltrated particularly in the dermis and subcutaneous on which MDV was actively replicated in the feather follicle epithelium(FFE). In addition, IgG bearing B lymphocytes in considerable number infiltrated in the dermis and subcutaneous tissues. From these results, the development of MDV-induced skin lesions was inflammatory following tumorous. In addition, each CD8+, ${\gamma}{\delta}$ and CD4+ T cells and B cell might act to protect MDV replication in the FFE or tumor cells which turned on lytic cycle.

구강내 만성염증병소에서 보이는 형질세포육아종의 면역조직화학적 관찰 (Immunohistochemical Observation of Plasma Cell Granuloma in Intraoral Chronic Inflammatory Lesions)

  • Purpose: Chronic inflammatory gingival lesions occur as pyogenic granulomas or non-specific chronic suppurative lesions. Methods: Of the 59 chronic inflammatory gingival lesions examined, plasma cell granuloma (n=14), which showed an intense antibody-mediated immune reaction with the increased infiltration of plasma cells, was observed as a pseudotumor-like gingival overgrowth and myofibroblastic or fibrohistiocytitc proliferation of stromal cells with a heavy collection of plasma cells. The levels of CD3, CD20, CD31, CD68, RANKL, cathepsin G, cathepsin K, lysozyme, TNF${\alpha}$, MMP-2, and MMP-9 in the 14 cases of gingival plasma cell granuloma with immunohistochemical detection were measured to determine the pathogenetic progresses of the plasma cell granuloma compared to the common pyogenic granuloma (n=45) in the gingiva. Results: The gingival lesions of the plasma cell granuloma could be divided into three histological types, plasma cell predominant type (PPT, n=8), mixed inflammatory cell type (MICT, n=2), and sclerosed fibrosis type (SFT, n=4). The PPT showed a condensed infiltration of plasma cells into the perivascular spaces of the granulomatous lesion with frequent formation of Russel's body in their cytoplasm. The MICT showed the concomitant infiltration of many macrophages together with plasma cells, resulting in the diffuse destruction of stromal fibrous tissue. The SFT showed granulomatous lesions replaced gradually by thick collagenous fibrous tissue, resembling an inflammatory pseudotumor. The SFT expressed strongly the lymphocytic markers, CD3 and CD20, and the macrophage/monocyte markers, CD31 and CD68, but showed reduced expression of common inflammatory markers, TNF${\alpha}$, cathepsin G, lysozyme, MMP-2, and MMP-9, as well as the reduced expression of osteoclastogenic markers, RANKL and cathepsin K. Conclusion: These results suggest that a gingival plasma cell granuloma shows variable gene expression for cell-mediated immunity and stromal tissue degeneration, undergoing sclerotic fibrosis with a persistent inflammatory reaction.

The Beneficial Effect of Avocado on Skin Inflammation in a Mouse Model of AD-like Skin Lesions

  • Avocado, superfood, contains a variety of essential nutrients and phytochemicals. The purpose of this study was to explore whether avocado could modulate skin inflammation in vivo. We elucidated the pharmacological effects of avocado on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD)-like skin lesions in mice. Additionally, we investigated the anti-inflammatory activity of avocado and its underlying mechanism including its effect on the expression levels of inflammatory-related genes and nuclear factor-κB (NF-κB) in DNCB-induced AD-like skin lesions. The findings of this study demonstrate that avocado attenuated AD-clinical symptoms including itching, eczematous, erythema and dryness and histamine levels in mice. Moreover, avocado suppressed both inflammatory cytokines expression as well as NF-κB and caspase-1 activation in AD-like skin lesions in mice. Taken together, these results demonstrate that avocado may be a potential candidate for treating skin inflammatory diseases like AD.

Predictors of Small Bowel Transit Time for Capsule Endoscopy in Children with Inflammatory Bowel Disease

  • Purpose: The development of assistive devices has allowed for the performance of capsule endoscopy in children. Anticipating the capsule's transit time could affect the efficacy of the investigation and potentially minimize the fasting period. This study determined the predictors of small bowel transit time for small-bowel capsule endoscopy in children and adolescents with inflammatory bowel disease. Methods: We retrospectively examined children and adolescents with inflammatory bowel disease who underwent capsule endoscopy by the age 18 at a Japanese tertiary care children's hospital. Small bowel transit time predictors were analyzed using multiple regression with explanatory variables. Results: Overall, 92 patients, aged 1-17 years, with inflammatory bowel disease (63 Crohn's disease and 29 ulcerative colitis cases) were examined for factors affecting small bowel transit time. In the simple regression analysis, diagnosis, age, height, weight, serum albumin, general anesthesia, and small intestine lesions were significantly associated with small bowel transit time. In the multiple regression analyses, serum albumin (partial regression coefficient: -58.9, p=0.008), general anesthesia (partial regression coefficient: 127, p<0.001), and small intestine lesions (partial regression coefficient: 30.1, p=0.037) showed significant associations with small bowel transit time. Conclusion: Hypoalbuminemia, the use of general anesthesia for endoscopic delivery of the capsule, and small intestine lesions appeared to be predictors of prolonged small bowel transit time in children and adolescents with inflammatory bowel disease. Expecting the finishing time may improve examination with a fasting period reduction, which benefits both patients and caregivers.

6-Shogaol reduces progression of experimental endometriosis in vivo and in vitro via regulation of VGEF and inhibition of COX-2 and PGE2-mediated inflammatory responses

  • Endometriosis (EM) is one of the most common gynaecological disorder affecting women in their reproductive age. Mechanisms involved in the pathogenesis of EM remains poorly understood, however inflammatory responses have been reported to be significantly involved. The efficacy of 6-shogaol on proliferation of endometriotic lesions and inflammatory pathways in experimentally-induced EM model was explored in this study. EM was stimulated in Sprague-Dawley rats by implantation of autologous endometrium onto the peritoneum abdominal wall. Separate groups were treated with 6-shogaol (50, 100 or 150 mg/kg b.wt/day) via oral gavage for one month period. Gestrinone (GTN) group received GTN (0.5 mg/kg/day) as positive control. Five weeks after implantation, the spherical volume of ecto-uterine tissues was determined. Treatment with 6-shogaol significantly reduced the implant size. Histological analysis reported atrophy and regression of the lesions. 6-shogaol administration effectively down-regulated $NF-{\kappa}B$ signaling, VEGF and VEGFR-2 (Flk-1) expression in the endometriotic lesions. Excess production of $IL-1{\beta}$ and IL-6 (pro-inflammatory cytokines), PGE2 and nitric oxide (NO) were reduced. Overall, the results of the study reveal the efficacy of 6-shogaol against endometriosis via effectively suppressing proliferation of the lesions and modulating angiogenesis and $COX-2/NF-{\kappa}B$-mediated inflammatory cascades.

  • Objectives NJ is being used to treat inflammatory diseases in Korea. In this study, we attempted to evaluate the effects of NJ on atopic dermatitis (AD)-like lesions and mast cell-mediated allergy inflammation in vivo and in vitro. Methods and Results We investigated to ascertain the pharmacological effects of NJ on 2,4-dinitrofluorobenzene (DNFB)-induced allergic reactions under in vivo conditions. Additionally, to find possible explanations for the anti-inflammatory mechanisms of NJ, we evaluated the regulatory effects of NJ on the level of inflammatory mediators in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). Conclusions NJ inhibited the production of the inflammatory cytokines (IgE, IL-6, IL-8 and TNF-${\alpha}$) significantly in vivo and in vitro.

Reduced Expression of Limd1 in Ulcerative Oral Epithelium Associated with Tobacco and Areca Nut

  • Purpose: The aim of this study was to cast light on initiating molecular events associated with the development of premalignant oral lesions induced by tobacco and/or areca nut. Method: Immunohistochemical analyses of cell cycle regulatory proteins (LIMD1, RBSP3, p16, RB, phosphorylated RB, p53), EGFR and SH3GL2 (EGFR associated protein) were performed with inflammatory/ulcerative epithelium and adjacent hyperplastic/mild dysplastic lesions. Results: No change in expression of the proteins was seen in inflammatory epithelium. Reduced nuclear expression of LIMD1 was evident in ulcerative epithelium. In hyperplastic lesions, reduced expression of RBSP3, p16, SH3GL2 and overexpression of p-RB and EGFR were apparent. Reduced nuclear expression of p53 was observed in mild dysplastic lesions. Conclusion: Our data suggest that inactivation of LIMD1 in ulcerative epithelium might predispose the tissues to alterations of other cell cycle regulatory and EGFR signaling proteins needed for the development of premalignant oral lesions.

도축 한우의 복강내 지방괴사증 발생과 병리조직학적 연구 (An abattoir survey and pathologic study of abdominal fat necrosis on Korean indigenous cattle)

  • The bovine fat necrosis is often seen as an incidental lesion in the adipose tissues of the abdominal cavity. Most of affected animals, however, have been detected at the slaughter house or routine necropsy. The purpose of this study was to examine the occurrence and distribution of abdominal fat necrosis in Korean native cattle, and its pathologic features. Postmortem inspection at an Chonju abattoir during a three-month period in 1997 detected at necrosis lesions in 67(6.6%) of the 1,012 animals received for slaughter. The occurrence was mainly in alimentary tracts and perirenal. Both sexes were affected, but the lesion were predominantly occurred in female. Gross lesions were white or yellowish in color and formed hard lumps irregular in shape ranging from small nodules to large solid masses. On the cut surface, lesions were occasionally seen chalky calcified granules and some of the lesions contained oil-like fluid. The surface of irregular shaped masses constricted kidneys and intestinal loops. Microscopically, the masses were encapsulated by thickened fibrous tissue, which infiltrated deeply and divided them into many irregular lobules. Initial lesions presented mild inflammatory cell and fibrous proliferation. It became fibroplasia in progressive lesions and resembled chronic lesion, eventhough no grossly apparent inflammatory reaction. The hard consistency of masses, due to fibroplasia, can compress the intestines, urinary organs, and reproductive organs.

다발성 낭종성 병변을 보인 급성 파종성 뇌척수염 - 증례보고 - (Acute Disseminated Encephalomyelitis(ADEM) Presenting as Multiple Cystic Lesions - A Case Report -)

  • Acute disseminated encephalomyelitis(ADEM) is an uncommon immune-mediated inflammatory demyelinating disorder that typically affects the white matter of the central nervous system. Radiologic findings of acute disseminated encephalomyelitis are not pathognomomic. The differential diagnosis is always difficult. Occasionally, the clinical features, radiological and histopathological findings of patients with acute disseminated encephalomyelitis mimic the brain tumor or other space occupying lesions. The authors report a 6-year-old girl who presented with right hemiparesis two days after nausea and vomiting. Brain MRI of the patient revealed non-enhanced multiple cystic lesions in subcortical white matter of both cerebral hemisphere with prominent edema. One of the cystic lesions was resected to differentiate with metastatic tumor or inflammatory disease such as abscess and confirmed as the acute disseminated encephalomyelitis via various immunohistochemical stains. Pertinent literature is reviewed with discussion on this uncommon ADEM associated with multiple cystic lesions.

Ga-67 골주사와 X선사진을 이용한 실험적 하악골 염증성 병변의 활성도 평가 (Ga-G7 SCINTIGRAPHIC AND RADIOGRAPHIC EVALUATION OF THE ACTIVITY OF EXPERIMENTALLY INDUCED MANDIBULAR INFLAMMATORY LESION)

  • The purpose of this study was to evaluate the usefulness of Ga-67 scintigram in predicting the disappearance of inflammatory activity in mandibular lesions of dogs. Inflammatory bony lesions were produced artificially by the injection of sclerosing solution and bacterial suspension through the distal root canals of the lower 4th premolars in 10 dogs. The serial Ga-67 scintigrams and periapical radiograms were obtained. After 3 to 6 weeks duration of antibiotic therapy, the experimental animals were killed, and the mandibles were dissected. Bacteriologic culture and antibiotic sensitivity test were done. 1. The following results were obtained. The negative Ga-67 scan has 100% predictive value, and is a strong evidence of the resolution of inflammatory activity. In 2 animals of positive Ga-67 scan, I animal had negative culture. In 14 cases of positive Ga-67 scan, radiographically there was increase in the size of lesion in 9 cases, no change in 5 cases. In 8 cases of negative Ga-67 scan, radiographically there was increase in the size of lesion in I case, no change in 6 cases and decrease in 1 case.
