To clarify resistance mechanisms of lincosamide antibiotics, it was examined whether lincosamide antibiotics was able to induce high resistance to macrolide and lincosamide antibiotics against EMR-1 strain of Bacillus species. And it was also examined whether the inducible resistance was plasmid-mediated or chromosome-mediated. This strain was identified as Bacillus licheniformis by its morphological and physiological characteristics. The subinhibitory concentrations of lincomycin and clindamycin induced high resistance in the strain to lincosamide antibiotics, but not to macrolide antibiotics. The inducible resistance was not eliminated by treating the strain with ethidium bromide, and plasmid was not identified by the alkaline lysis method of plasmid preparation. These results indicate, therefore, that the inducible resistance to macrolide and lincosamide antibiotics in the strain may be chromosome-mediated, not plasmid-mediated.
연구배경 : IOS 혹은 FOT는 외부의 loudspeaker에서 발생하는 기류의 진동을 자발호흡에 부가시킴으로써 발생하는 압력의 변화를 분석하여 폐기능을 평가하는 방법으로, 일반적인 폐기능검사 방법들과는 달리 노력성 호기방법이 필요치 않고 자발성 상시호흡에서 검사가 가능하다는 장점이 있다. 또한 microprocessor 의 도입으로 Fourier 변환법에 의한 복잡한 신호 분석이 가능해 지면서 주파수에 따른 호흡기계의 impedance를 실측부분 (Rrs : resistance) 과 추정부분 (Xrs : reactance)의 두요소로 보다 깊이 파악할 수 있게 되었다. IOS의 임상적 이용은 소아의 폐기능검사, 역학적 연구, 소기도질환의 확인, 유발검사 등에 장점이 있다고 알려져 있다. 그러나 흡연자에서 발생하는 소기도 폐쇄 또는 조기 기도폐쇄를 확인하는데 있어서 IOS의 역할에 대한 연구 보고는 논란이 있어 왔다. 이에 본 연구에서는 흡연자에서의 조기 기도폐쇄를 측정하는 방법으로서의 IOS가 갖는 임상적 유용성을 검증해보고자 하였다. 방 법 : 종합검진센타를 방문한 남자중에서 노력성 호기성 폐활량 측정 검사상 정상 소견을 보인 356명($40.6{\pm}9.7$세)을 대상으로 IOS(Jaeger company : Version 4.1)을 시행하여 흡연자군 (234명 : $40.2{\pm}9.3$세)과 비흡연자군(122명 : $41.5{\pm}10.6$세)으로 구분하여 양군간에 두가지 검사볍 지표들에 대한 유의성을 검증하였다. 결 과 : Spirometry 지표인 FVC, FVC%pred, FEV1, FEV1%pred, FEV1/FVC, PEF, PEF%pred, FEF25~75%, FEF25~75%pred, FEF50%, FEF50%pred 등은 모두 흡연자군과 비흡연자군간에 유의한 차이가 관찰되지 않았다. IOS 지표중 total resistance(흡연자군 : 비 흡연자군=$2.58{\pm}1.71$ : $2.22{\pm}1.20$), peripheral resistance($1.47{\pm}0.10$ : $1.25{\pm}0.62$), bronchial compliance($0.47{\pm}0.16$ : $0.44{\pm}0.12$) 등은 양군간에 유의한(p<0.05) 차이가 관찰되었으나 central resistance와 lung compliance는 유의한 차이가 관찰되지 않았다(단위는 resistance=hPa/l/s ; compliance=1/hPa). 주파수에 따른(5, 10, 15, 20, 25, 30, 35Hz) Rrs은 양군간에 유의한 차이를 관찰할 수 없었고, Xrs는 낮은 주파수, 즉 X5($-0.76{\pm}0.48$ : $-0.62{\pm}0.28$) 와X10($-0.15{\pm}0.33$ : $-0.06{\pm}0.19$)에서 유의한 차이를 관찰할 수 있었다(단위는 hPa/l/s). 결론 : IOS은 흉연자와 비흡연자의 조기폐쇄를 확인할 수 있는 검사법이라고 판단되며, total resistance, peripheral resistance, bronchial compliance, 그리고 5Hz와 10Hz 의 낮은 주파수에서의 Xrs가 유용한 지표라고 판단된다.
The purpose of this study was to examine the influence of conditions for quenching and/or tempering on the corrosion and hydrogen diffusion behavior of ultra-strong automotive steel in terms of the localized plastic strain related to the dislocation density, and the precipitation of iron carbide. In this study, a range of analytical and experimental methods were deployed, such as field emission-scanning electron microscopy, electron back scatter diffraction, electrochemical permeation technique, slow-strain rate test (SSRT), and electrochemical polarization test. The results showed that the hydrogen diffusion parameters involving the diffusion kinetics and hydrogen solubility, obtained from the permeation experiment, could not be directly indicative of the resistance to hydrogen embrittlement (HE) occurring under the condition with low hydrogen concentration. The SSRT results showed that the partitioning process, leading to decrease in localized plastic strain and dislocation density in the sample, results in a high resistance to HE-induced by aqueous corrosion. Conversely, coarse iron carbide, precipitated during heat treatment, weakened the long-term corrosion resistance. This can also be a controlling factor for the development of ultra-strong steel with superior corrosion and HE resistance.
Selected isolates of Pseudomonas fluorescens (Pf4-92 and PfRsC5) and P. aeruginosa (PaRsG18 and PaRsG27) were examined for growth promotion and induced systemic resistance against Fusarium wilt of chickpea. Significant increase in plant height was observed in Pseudomonas treated plants. However, plant growth was inhibited when isolates of Pseudomonas were used in combination with Fusarium oxysporum f. sp. ciceri (FocRs1). It was also observed that the Pseudomonas spp. was colonized in root of chickpea and significantly suppressed the disease in greenhouse condition. Rock wool bioassay technique was used to study the effect of iron availability on the induction of systemic resistance to Fusarium wilt of chickpea mediated by the Pseudomonas spp. All the isolates of Pseudomonas spp. showed greater disease control in the induced systemic resistance (ISR) bioassay when iron availability in the nutrient solution was low. High performance liquid chromatography (HPLC) analysis indicated that an the bacterial isolates produced more salicylic acid (SA) at low iron ($10\;{\mu}M$ EDDHA) than high iron availability ($10\;{\mu}Fe^{3+}$ EDDHA). Except PaRsG27, all the three isolates produced more pseudobactin at low iron than high iron availability.
Chemoresistance is a major barrier to successful cisplatin-based chemotherapy for epithelial ovarian cancer (EOC), and emerging evidences suggest that microRNAs (miRNAs) are involved in the resistance. In this study, it was indicated that miR-363 downregulation was significantly correlated with EOC carcinogenesis and cisplatin resistance. Moreover, miR-363 overexpression could resensitise cisplatin-resistant EOC cells to cisplatin treatment both in vitro and in vivo. In addition, data revealed that EMT inducer Snail was significantly upregulated in cisplatin-resistant EOC cell lines and EOC patients and was a functional target of miR-363 in EOC cells. Furthermore, snail overexpression could significantly attenuate miR-363-suppressed cisplatin resistance of EOC cells, suggesting that miR-363-regulated cisplatin resistance is mediated by snail-induced EMT in EOC cells. Taken together, findings suggest that miR-363 may be a biomarker for predicting responsiveness to cisplatin-based chemotherapy and a potential therapeutic target in EOC.

The non-pathogenic, non-glycopeptide-producing actinomycete Streptomyces coelicolor carries a cluster of seven genes (vanSRJKHAX) that confers inducible, high-level resistance to vancomycin. The van genes are organised into four transcription units, vanRS, vanJ, vanK and vanHAX, and these transcripts are induced by vancomycin in a vanR-dependent manner. vanHAX are orthologuous to genes found in vancomycin resistant enterococci that encode enzymes predicted to reprogramme peptidoglycan biosynthesis such that cell wall precursors terminate in D-Ala-D-Lac, rather than D-Ala-D-Ala. vanR and vanS encode a two-component signal transduction system that mediates transcriptional induction of the seven van genes. vanJ and vanK are novel genes that have no counterpart in previously characterised vancomycin-resistance clusters from pathogens. VanK is essential for vancomycin resistance in S. coelicolor and it is required for adding Gly branch to stem peptides terminating D-Ala-D-Lac. Because VanK can recognise D-Lac-containing precursors but the constitutively expressed femX enzyme, encoded elsewhere on the chromosome, cannot recognize D-Lac-containing precursors as a substrate, vancomycin-induced expression of VanHAX in a vanK mutant is lethal. Further, femX null mutants are viable in the presence of glycopeptide antibiotics but die in their absence. Bioassay using vanJp-neo fusion reporter system also showed that all identified inducers for van genes expression were glycopeptide antibiotics, but teicoplanin, a membrane-anchored glycopeptide, failed to act as an inducer.
This study was carried out to investigate the effects of wood vinegar on growth and resistance of peppers. It was observed that heights and dried weights of the peppers treated with diluted wood vinegar were increased, especially 1:500 diluted wood vinegar was the most effective. The Wood vinegar also showed antibacterial activity against Xanthomonas axonopodis pv. vesicatoria directly. The growth of X. axonopodis pv. vesicatoria was completely inhibited when incubated for 12 hours at $30^{\circ}C$ with non diluted wood vinegar. The peppers applied wood vinegar did not show induced systemic resistance after injecting X. axonopodis pv. vesicatoria.
The plant growth-promoting rhizobacterium Ochrobactrum lupini KUDC1013 elicited induced systemic resistance (ISR) in tobacco against soft rot disease caused by Pectobacterium carotovorum subsp. carotovorum. We investigated of its factors involved in ISR elicitation. To characterize the ISR determinants, KUDC1013 cell suspension, heat-treated cells, supernatant from a culture medium, crude bacterial lipopolysaccharide (LPS) and flagella were tested for their ISR activities. Both LPS and flagella from KUDC1013 were effective in ISR elicitation. Crude cell free supernatant elicited ISR and factors with the highest ISR activity were retained in the n-butanol fraction. Analysis of the ISR-active fraction revealed the metabolites, phenylacetic acid (PAA), 1-hexadecene and linoleic acid (LA), as elicitors of ISR. Treatment of tobacco with these compounds significantly decreased the soft rot disease symptoms. This is the first report on the ISR determinants by plant growth-promoting rhizobacteria (PGPR) KUDC1013 and identifying PAA, 1-hexadecene and LA as ISR-related compounds. This study shows that KUDC1013 has a great potential as biological control agent because of its multiple factors involved in induction of systemic resistance against phytopathogens.
In this study, a new spot welding with electric resistance heated dies is suggested for the spot welding of non-ferrous metal plates for drawing and concurrent spot welding. This welding method involves two heating processes such as heating by conduction of electric resistance heated dies and heating by resistance between contacted surfaces of two plates by welding current induced to copper dies for the fusion of contacted metal. This welding process has welding variables such as current induced in heated and copper dies, the inner diameters of heated dies, and edge shape of the copper dies. Experimental conditions for each current should be established to get successful welding strength. The welding strength could reach to the desired value in industrial fields under the following conditions of contact diameters of heated dies in this case of overlapped aluminum5052 plates with 0.3-mm thickness: inner and outer diameters of 5.0 and 16mm, respectively; diameter of copper dies, 5.0mm; heating current, 6.8kA in heated steel dies; welding current, 8.6 kA in copper dies.
In these days, the piezoresistive material has been applied to various sensors in order to measure the change of physical quantities. But the relationship between the sensitivity of a sensor and the position and size of piezoresistor has rarely been studied. Therefore, this paper was focused on the effect of residual stress induced in piezoresistor on the distribution of resistance change ratio and supposed the feasible position of piezoresistor. The resulting are following; The tensile residual stress in the vicinity of piezoresistor decreased the value of resistance change ratio and could not effect on all the area of diaphragm but local area around the piezoresistor. Also, the piezoresistor in the diaphragm type pressure sensor with boss should fabricate in the edge of boss in order to increase the sensitivity of pressure sensor.
