Isolation of a Pseudomonas sp. Capable of Utilizing 4-Nonylphenol in the Presence of Phenol

  • Enrichment techniques led to the isolation of a Pseudomonas sp. strain P2 from municipal waste-contaminated soil sample, which could utilize different isomers of a commercial mixture of 4-nonylphenol when grown in the presence of phenol. The isolate was identified as Pseudomonas sp., based on the morphological, nutritional, and biochemical characteristics and 16S rDNA sequence analysis. The ${\beta}$-ketoadipate pathway was found to be involved in the degradation of phenol by Pseudomonas sp. strain P2. Gas chromatography-mass spectrometric analysis of the culture media indicated degradation of various major isomers of 4-nonylphenol in the range of 29-50%. However, the selected ion monitoring mode of analysis of biodegraded products of 4-nonylphenol indicated the absence of any aromatic compounds other than those of the isomers of 4-nonylphenol. Moreover, Pseudomonas sp. strain P2 was incapable of utilizing various alkanes individually as sole carbon source, whereas the degradation of 4-nonylphenol was observed only when the test organism was induced with phenol, suggesting that the degradation of 4-nonylphenol was possibly initiated from the phenolic moiety of the molecule, but not from the alkyl side-chain.

Lipase Activity and Tacrolimus Production in Streptomyces clavuligerus CKD 1119 Mutant Strains

  • The effect of carbon sources on tacrolimus production by a mutant strain of Streptomyces clavuligerus CKD 1119, an isolate from soil, was examined. Among the carbohydrates and oils tested in this work, a mixed carbon source of soluble starch and com oil was the best. An analysis of the culture kinetics also showed that, in contrast to the carbohydrates, the com oil was consumed later in the antibiotic production phase, implying that the oil substrate was the principal carbon source for the biosynthesis of tacrolimus, and this was directly proven by experiments using $^{14}C$-glucose and $^{14}C$-oleate substrates. Furthermore, com oil induced the formation of lipase by the mutant strain, whereas the addition of glucose significantly repressed lipase activity. The lipase activity exhibited by the FK-506-overproducing mutants was also observed to be directly proportional to their tacrolimus yield, indicating that a high lipase activity is itself a crucial factor for tacrolimus production. A feasibility study with a 200-1 pilot-scale fermentor and the best strain (Tc-XII-15322) identified in this work revealed a high volumetric and specific productivity of about 495 mg/l and 0.34 mg/mg dry mycelium, respectively.

Thermal creep effects of aluminum alloy cladding on the irradiation-induced mechanical behavior in U-10Mo/Al monolithic fuel plates

  • Three-dimensional finite element simulations are implemented for the in-pile thermo-mechanical behavior in U-Mo/Al monolithic fuel plates with different thermal creep rates of cladding involved. The numerical results indicate that the thickness increment of fuel foil rises with the thermal creep coefficient of cladding. The maximum Mises stress of cladding is reduced by ~85% from 344 MPa on the 98.0th day when the creep coefficient of cladding increases from 0.01 to 10.0, due to its equivalent thermal creep strain enlarged by 3.5 times. When the thermal creep coefficient of Aluminum cladding increases from 0 to 1.0, the maximum mesoscale stress of fuel foil varies slightly. At the same time, the peak mesoscale normal stress of fuel foil can reach 51 MPa on the 98.0th day for the thermal creep coefficient of 10, which increases by 60.3% of that with the thermal creep un-occurred in the cladding. The maximum through-thickness creep strain components of fuel foil differ slightly for different thermal creep coefficients of cladding. The dangerous region of fuel foil becomes much closer to the heavily irradiated side when the creep coefficient of cladding becomes 10.0. The creep performance of Aluminum cladding should be optimized for the integrity of monolithic fuel plates.

  • In this paper, in order to examine cyclic hehavior characteristics and safety of underground flexible pipes for electric cables subject to cyclic vehicle load, FEM analysis and cyclic soil box test were carried out. As results of the test, it was revealed that the vertical displacement of the test was larger than that of FEM analysis because thermal effect arising from power cables made reduction of rigidity of the pipe so that large deformation of the pipe induced by the heat occured. Moreover, it was shown that the final vertical displacement under about 0.4 million times of the cyclic load test was not satisfied with elastic allowable displacement of the pipe, and long term stability of the pipe was not stable since behavior characteristics of the pipe exists plastic strain range pasted clastic strain range.

Selection and Isolation of a Mutant Yeast Strain Tolerant to Multiple Targeted Heavy Metals

  • BACKGROUND: This study was performed for selecting yeast mutants with a high tolerance for targeted metals, and determining whether yeasts strains tolerant to multiple heavy metals could be induced by sequential adaptations. METHODS AND RESULTS: A mutant yeast strain tolerant to the heavy metals cadmium (Cd), copper (Cu), nickel (Ni), and zinc (Zn) was selected by sequential elevated exposures to each metal with intermittent mutant isolation steps. A Cd-tolerant mutant was isolated by growing yeast cells in media containing $CdCl_2$ concentrations that were gradually increased to 1 mM. Then the Cd-tolerant mutant was gradually exposed to increasing levels of $CuCl_2$ in growth media until a concentration of 7 mM was reached, thus generating a strain tolerant to both Cd and Cu. In the subsequent steps, this mutant was exposed to $NiCl_2$ (up to 8 mM), and a resultant isolate was further exposed to $ZnCl_2$ (up to 60 mM), allowing the derivation of a yeast mutant that was simultaneously tolerant to Cd, Cu, Ni, and Zn. CONCLUSION: This method of inducing tolerance to multiple targeted heavy metals in yeast will be useful in the bioremediation of heavy metals.

Activation of Macrophages by Exopolysaccharide Produced by MK1 Bacterial Strain Isolated from Neungee Mushroom, Sarcodon aspratus

  • Background: The MK1 strain, a novel bacterial isolate from soft-rotten tissue of the Neungee mushroom, produces copious amounts of exopolysaccharide (EPS) in a dextrose minimal medium. This study examined the molecular characteristics and immunomodulatory activity of MK1 EPS. Methods: The EPS in the culture supernatant was purified by cold ethanol precipitation, and characterized by SDS- PAGE/silver staining and Bio-HPLC. The immunomodulatory activities of the EPS were examined using the mouse monocytic cell line, RAW 264.7 cells. Results: The molecular weights of the purified EPS were rather heterogeneous, ranging from 10.6 to 55 kDa. The EPS was composed of glucose, rhamnose, mannose, galactose, and glucosamine at an approximate molar ratio of 1.00 : 0.8 : 0.71 : 0.29 : 0.21. EPS activated the RAW cells to produce cytokines, such as TNF-${\alpha}$ and IL-$1{\beta}$, and nitric oxide (NO). EPS also induced the expression of co-stimulatory molecules, such as B7-1, B7-2 and ICAM-1, and increased the phagocytic activity. The macrophage-activating activity of EPS was not due to endotoxin contamination because the treatment of EPS with polymyin B did not reduce the macrophage-activating activity. Conclusion: The EPS produced from the MK1 strain exerts macrophage-activating activity.

Differential Induction of Pathogenesis-Related Proteins in the Compatible and Incompatible Interactions of Tomato Leaves with Xanthomonas campestris pv. vesicatoria (Xanthomonas campestris pv. vesicatoria와 토마토잎의 친화적, 불친화적 반응에서 병생성관련 단백질의 유도)

  • Inoculation with the compatible strain Ds 1 of Xanthomonas campestris pv. vesicatoria caused brownish ad water-soaked lesions, but incompatible strain Bv5-4a produced hypersensitive symptoms with local necrosis on tomato (cv. Kwangyang) leaves. Bacterial populations of the compatible strains Ds 1 propagated more greatly than the incompatible strain Bv5-4a at the frist onset, but no differences were observed 5 days after inoculation. The bacterial infection induced the synthesis and accumulation of soluble proteins in tomato leaves, especially in the incompatible interaction. Native-polyacrylamide gel electrophoresis distinguished the soluble proteins in the tomato leaves infected by the compatible or incompatible strains. A protein of low molecular weight occurred only in the incompatible interaction. Some pathogenesis-related (PR) proteins, especially the 15, 18, 23, 26 and 54 kDa proteins, were detected only in the infected tomato leaves. In the two-dimensional electrophoresis, some proteins with different molecular weights (Mr. 21∼29 kDa) and the pI 8∼9 appeared more distinctly only in the incompatible interaction. These data suggest that the de novo synthesis of some PR proteins in tomato may be significant in defense against X. c. pv. vesicatoria.

Isolation and Characterization of Burkholderia cepacia strain YJK2, Antagonistic Microorganism of Paprika Pathogens (파프리카 병원균들에 대한 길항미생물, Burkholderia cepacia strain YJK2의 분리 및 특성)

  • Although several adverse effects have been increased in recent years, synthetic agro-chemicals have been widely used to control diseases on paprika. This research was conducted to isolate and to characterize the antagonistic microorganism to control major paprika diseases, gray mold rot, fruit and stem rot, phytophthora blight, sclerotium rot, and wilt disease. Analysis of the fatty acid and analysis of the 16S rDNA gene sequence revealed that YKJ2 isolated in this research belongs to a group of Burkholderia cepacia. Specially, 16S rDNA gene sequence of YKJ2 showed 99% of sequence similarity with B. cepacia. Observation through the optical microscope revealed that YKJ2 was effective on suppression of the spore germination and the hyphal growth of pathogens. YKJ2 treatment on pathogens induced marked morphological changes like hyphal swelling and degradation of cell wall. In the case of phytophthora blight, the zoosporangium formation was restrained. On the basis of the results of this study, we propose that an antagonistic microorganism, B. cepacia, found in this study naming as "B. cepacia strain YKJ2" and has great potential as one of biological control agents against major diseases of paprika.

  • The mechanical properties of the most widely used cryogenic materials, i.e. austenitic stainless steel (ASS), aluminum alloy and invar steel, strongly depend on temperatures and strain rates. These phenomena show very complicated non-linear behaviors and cannot be expressed by general constitutive equation. In this study, an unified constitutive equation was proposed to represent the effect of temperature and strain rate on the materials. The proposed constitutive equation has been based on Tomita/Iwamoto and Bodner/Partom model for the expression of 2nd hardening due to martensite phase transformation of ASS. To simulate ductile fracture, modified Bodner/Chan damage model was additionally applied to the model and the model validity was verified by comparison of experimental and simulation results.

  • The cellabiase (${\beta}$-glucosidase) gene in a Cellulomonas sp. CS1-1 was cloned into E. coli HB101 using the vector plasmid pBR322, and the expression of the gene in E. coli studied. The chromosomal DNA of the cellulomonas was digested by seveal restriction enzymes, each of which has only one cleaving site in plasmid pBR322. The recombinant plasmid, pSB2, created with Sal I frament, was expressed for the cellobiase gene in E. coli. The recombiant plasmid was estimated to contain 6.4 Kb foreign DNA at the Sal I site of plasmid pBR322 and the inserted DNA was mapped by single and double digestion with several enzymes. E. coli HB101(pSB2) has slowly grown in a mineral liquid medium containing cellobiose as a sole carbon source. The cellobiase activity in the transformed E. coli was 132 units per liter, which is equivalent to one twenty fifth of that in doner strain Cellulomonas sp. CS1-1. The transforned cell with plasmid containing cellulase gene grow well in the LB mediuns. The synthesis of cellobiase in the strain, E. coli HB101 (pSB2), was inhibited by glucose and at high concentration of cellobiose, and induced by cellobiose at low concentration.
