Studies on the Effects of Herbal medicines on the Fetus during Pregnancy (II) - Mutagenesis and chromosomal aberration of herbal medicines - (한약이 임신중 태아에 미치는 영향(II) -한약이 돌연변이원성과 염색체이상에 미치는 효과-)

  • Oriental herbal medicines were examined for mutagenicity in the reverse mutation test on Salmonella typimurium T A98/100 and chromosomal aberration test on cultured mammalian cells (Chinese hamster cell lines). The reverse mutation test was performed by a plate incorporation method with and without a metabolic activation system (S9 mix). The tested herbal medicines did not significantly increase revertible colonies on any of the test strains with and without a metabolic activation system (S9 mix) at concentrations of 1 mg/ml. In the chromosomal aberration test, most tested herbal medicines did not significantly increase the number of aberrant cells on any of the test strains with a metabolic activation system (S9 mix) at concentrations of 1 mg/ml, compared with the vehicle control. However. Ansu Semen significantly increased the number of aberrant cells without a metabolic activation system (S9 mix). Paeoniae Radix. Hoelen, Aurantii nobilis Pericarpium, Cnidii Rhizoma, Angeliacae gigantis Radix, Perillae Herba and Moutan Cortex Radicis slightly increased revertible colonies on any of the test strains with a metabolic activation system (S9 mix), These results indicate that most herbal medicines might be carefully used in obstetrics and gynecology, although they do not have the potent mutagenic potential under the present test conditions.

Growth Comparison of Juvenile Abalone, Haliotis discus hannai in Different Culture Systems in the West Coast of Korea (서해안에서 사육시스템 종류별 참전복 치패의 성장 비교)

  • Comparisons of growth and survival rate in different culture systems for juvenile abalone were determined in the West Coast of South Korea from June, 2000 to April, 2001. Daily growth rate reached at the maximum by $148.6{\pm}48.96\;{\mu}m/day$ with a stagnant system and at the minimum by $95.3{\pm}21.45\;{\mu}m/day$ with a sprinkler culture system. Survival rate of the juvenile abalone was significantly higher than that in sprinkler culture with a rotary culture (P<0.05). The highest survival rate was 50.0% in the rotary culture and 38.3% was observed in the stagnant culture system. The results from this study indicate that effective survival and growth rate could be predicted in the rotary culture system.

Advanced tube formation assay using human endothelial colony forming cells for in vitro evaluation of angiogenesis

  • The tube formation assay is a widely used in vitro experiment model to evaluate angiogenic properties by measuring the formation of tubular structures from vascular endothelial cells (ECs). In vitro experimental results are crucial when considered the advisability of moving forward to in vivo studies. Thus, the additional attentions to the in vitro assay is necessary to improve the quality of the pre-clinical data, leading to better decision-making for successful drug discovery. In this study, we improved the tube formation assay system in three aspects. First, we used human endothelial colony forming cells (ECFCs), which are endothelial precursors that have a robust proliferative capacity and more defined angiogenic characteristics compared to mature ECs. Second, we utilized a real-time cell recorder to track the progression of tube formation for 48 hours. Third, to minimize analysis error due to the limited observation area, we used image-stitching software to increase the microscope field of view to a $2{\times}2$ stitched area from the $4{\times}$ object lens. Our advanced tube formation assay system successfully demonstrated the time-dependent dynamic progression of tube formation in the presence and absence of VEGF and FGF-2. Vatalanib, VEGF inhibitor, was tested by our assay system. Of note, $IC_{50}$ values of vatalanib was different at each observation time point. Collectively, these results indicate that our advanced tube formation assay system replicates the dynamic progression of tube formation in response to angiogenic modulators. Therefore, this new system provides a sensitive and versatile assay model for evaluating pro- or anti-angiogenic drugs.

  • In this paper, we implemented a speech emotion recognition system that can distinguish human emotional states from recorded speech captured by a single microphone and classify them into four categories: neutrality, happiness, sadness and anger. In general, a speech recorded with a microphone contains background noises due to the speaker environment and the microphone characteristic, which can result in serious system performance degradation. In order to minimize the effect of these noises and to improve the system performance, a MA(Moving Average) filter with a relatively simple structure and low computational complexity was adopted. Then a SFS(Sequential Forward Selection) feature optimization method was implemented to further improve and stabilize the system performance. For speech emotion classification, a SVM pattern classifier is used. The experimental results indicate the emotional classification performance around 65% in the computer simulation and 62% on the ARM platform.

  • An electronic services which are based on Internet/Intranet business transactions are available to e-market places and get the broader business concepts. Component-based e-commerce technology is a recent trend towards resolving the e-commerce chanange at both system and ap-plication levels. The basic capabilities of component based systems should include the plug and play features at various granularities, interoper-ability across networks and mobility in various networking environments. E-business application developers are attempting to more towards web-service as a mechanism for developing component-based web-applications. Traditional process and development models are inadequately architectured to meet the rapidly evolving needs for the future of scalable web services. In this thesis, we focus specifically on the issues of e-business system architecture based on web service for establishing e-business system. We specifies and identifies design pattern for applying e-business domain in the context of multiple entities. We investigate prototype and frameworks to develope components for e-business application based suggested process. We present a worked example to demonstrate the behavior of Customer Authentication System(CAS) development with component and recommend process. Final]Y, we indicate and view on future directions in component-based development in the context of electronic business.

  • Recently great loss of life and property is occurring because of fire, natural disaster, earth quake, tsunami and so on. People spend 80~90% of their time indoor environment like office, supermarket, campus. Therefore Indoor navigation and guidelines system became so essential for most of all. Incase of emergency we must be careful earlier, in such a cases 5G kind of new technology may also cannot work. So immediate action and quick routing notification for guidelines and protection is the most. Considering this issue We proposed indoor evacuating guidance system based on microcontroller Wi-Fi board for Indoor APP using mobile. Focusing various kind of technology like, ok google, voice search APP we purposed node architecture based system. When we listen fire alarm while living inside the room. Then to be safe we connect with server and start Arduino UNO+IoT ESP8266 Wi-Fi shield version1-IoT module to store data in MySQL DB server. We make application to escape out from the building up-to the three exits giving information from source point to destination. Our program can send information to the users emergency location and situations. For this when the user get sound or vibration in their mobile device it indicate fire out near by. At that time we update message from Arduino to DB server for the fixed current position inside the building which give routing signal for that fire out location by changing values from 0 to 1. We have user in point 10 where user is near by. Later we detect Wi-Fi signal form Nodemcu as room of each floor and try to connect with user. Main purpose of this paper is to save life of people in short time and find out the shortest path up to nearest exits in the time of emergencies and rescue them.

Comparison of Job Satisfaction and Nursing Performance between Nurses on Fixed Nights and Nurses on Three shifts, and Nurses Understanding of Fixed Night Shift System (밤번고정근무 간호사와 3교대근무 간호사의 직무만족도, 간호업무수행정도 비교 및 밤번고정근무제도에 대한 인식)

  • Purpose: The purpose of this study was to compare job satisfaction and nursing performance of nurses on fixed night shifts (FNS) and nurses working three shifts (W3S), to identify the environment of night work and recognition of the viability of the FNS system. Methods: The research was conducted with 106 nurses on FNS and 257 on W3S. The nurses had worked for 1 to 11 years at A hospital, Seoul. Results: Job satisfaction was significantly higher (t=-3.51, p< .001) for nurses on FNS. However, no significant difference was found for nursing performance (t=-1.019, p=.309) between the two groups. Both groups of nurses were mostly satisfied. Subjective fatigue scores (0 to 10 points) during the night for nurses on FNS (6.02) were lower than for W3S nurses (7.28) A high percentage (88.7%) of nurses on FNS indicated a willingness to further participate in FNS. Conclusion: Results indicate that the demonstration FNS system showed its effectiveness. Therefore, efforts are needed to enhance and revitalize FNS and to upgrade the system to control patient transfer time from emergency departments to wards, and to provide nurses needed resources for emergency or intensive care.

Dechlorination of High Concentrations of Tetrachloroethylene Using a Fixed-bed Reactor

  • We evaluated the properties of a fixed-bed column reactor for high-concentration tetrachloroethylene (PCE) removal. The anaerobic bacterium Clostridium bifermentans DPH-1 was able to dechlorinate PCE to cis-1,2-dichloroethylene (cDCE) via trichloroethylene (TCE) at high rates in the monoculture biofilm of an upflow fixed-bed column reactor. The first-order reaction rate of C. bifermentans DPH-1 was relatively high at $0.006\;mg\;protein^{-1}{\cdot}l{\cdot}h^{-1}$, and comparable to rates obtained by others. When we gradually raised the influent PCE concentration from $30\;{\mu}M$ to $905\;{\mu}M$, the degree of PCE dechlorination rose to over 99% during the operation period of 2,000 h. In order to maintain efficiency of transformation of PCE in this reactor system, more than 6 h hydraulic retention time (HRT) is required. The maximum volumetric dechlorination rate of PCE was determined to be $1,100\;{\mu}mol{\cdot}d^{-1}l$ of reactor $volume^{-1}$, which is relatively high compared to rates reported previously. The results of this study indicate that the PCE removal performance of this fixed-bed reactor immobilized mono-culture is comparable to that of a fixed-bed reactor mixture culture system. Furthermore, our system has the major advantage of a rapid (5 days) start-up time for the reactor. The flow characteristics of this reactor are intermediate between those of the plug-flow and complete-mix systems. Biotransformation of PCE into innocuous compounds is desirable; however, unfortunately cDCE, which is itself toxic, was the main product of PCE dechlorination in this reactor system. In order to establish a system for complete detoxification of PCE, co-immobilization of C. bifermentans DPH-1 with other bacteria that degrade cDCE aerobically or anaerobically to ethene or ethane may be effective.

Coordinated Control Strategy and Optimization of Composite Energy Storage System Considering Technical and Economic Characteristics

  • Control strategy and corresponding parameters have significant impacts on the overall technical and economic characteristics of composite energy storage systems (CESS). A better control strategy and optimized control parameters can be used to improve the economic and technical characteristics of CESS, and determine the maximum power and stored energy capacity of CESS. A novel coordinated control strategy is proposed considering the coordination of various energy storage systems in CESS. To describe the degree of coordination, a new index, i.e. state of charge coordinated response margin of supercapacitor energy storage system, is presented. Based on the proposed control strategy and index, an optimization model was formulated to minimize the total equivalent cost in a given period for two purposes. The one is to obtain optimal control parameters of an existing CESS, and the other is to obtain the integrated optimal results of control parameters, maximum power and stored energy capacity for CESS in a given period. Case studies indicate that the developed index, control strategy and optimization model can be extensively applied to optimize the economic and technical characteristics of CESS. In addition, impacts of control parameters are discussed in detail.

Development of ELISA System for Screening of Specific Binding Inhibitors for Src Homology (SH)2 Domain and Phosphotyrosine Interactions

  • In the present study, an in vitro ELISA system to assess the interaction between Src homology (SH)2 domains and phosphotyrosine that contain peptides was established using purified GST-conjugated SH2 proteins and synthetic biotinylated phosphotyrosine that contain oligopeptides. The SH2 domains bound the relevant phosphopeptides that were immobilized in the streptavidin-coated microtiter plate in a highly specific and dose-dependent manner. The epidermal growth factor receptor (EGFR)-, T antigen (T Ag)-, and platelet-derived growth factor receptor (PDGFR)-derived phosphopeptides interacted with the growth factor receptor binding protein (Grb)2/SH2, Lck/SH2, and phosphatidyl inositol 3-kinase (PI3K) p85/SH2, respectively. No cross-reactions were observed. Competitive inhibition experiments showed that a short phosphopeptide of only four amino acids was long enough to determine the binding specificity. Optimal concentrations of the GST-SH2 fusion protein and phosphopeptide in this new ELISA system for screening the binding blockers were chosen at 2nM and 500nM, respectively. When two candidate compounds were tested in our ELISA system, they specifically inhibited the Lck/SH2 and/or p85/SH2 binding to the relevant phosphopeptides. Our results indicate that this ELISA system could be used as an easy screening method for the discovery of specific binding blockers of protein-protein interactions via SH2 domains.
