Characterization and Electron Microscopic Visualization of the Plasmid and the Gene Expression in the Escherichia coli (E. coli의 유전자 발현 및 플라스미드 DNA 구조의 특성에 관한 연구)

  • Characterization and electron microscopic visualization of the plasmid and the gene expression of Escherichia coli were carried out. Transcriptional units of active structural genes were observed after lysis of Escherichia coli cells. The ribosomes attached to the E. coli genome on mRNA molecule as polyribosomes. From this gradient of polyribosome length, we estimated location of mRNA synthesis initiation site. In this experiment, a granule is ofen present which may correspond to a RNA polymerase at the promoter site. pOX1, pOX7, pOX7A, $pOX7{\Delta}1$, pSTP36, pSTP21, pBR322, and pJH12 were visualized by way of electron microscope, and their estimated sizes were determined to be $5.70{\pm}0.08{\mu}m,\;2.15{\pm}0.10{\mu}m,\;2.14{\pm}0.12{\mu}m,\;7.39{\pm}0.08{\mu}m,\;4.03{\pm}0.04{\mu}m,\;1.50{\pm}0.03{\mu}m\;and\;1.25{\pm}0.09{\mu}m$ respectively. One micrometer of measured length corresponded to about 3.0 Kb. Mica-press adsorption method that allows selectivs visualization of the plasmid DNA released in situ from the bacterial cell is rapid and useful for visualization of plasmids. The released plasmid DNA was adsorbed preferently on mica in a divalent cation-free solution. Miller chromatin-spreading method was useful to observe the plasmid and transcripts. BAC method and cytochrome C monolayer were useful to observe the plasmid DNA. Our ability to visualize ultrastructural aspects of the expression of E. coli has given us a unique tool with which to study the regulation the level of an individual gene.

Sensitivity analysis of tunnel stability with a consideration of an excavation damaged zone (암반손상대를 고려한 터널 안정성 민감도 분석)

  • An Excavation Damaged Zone (EDZ), in which rock properties are permanently changed due to blasting impact or stress redistribution, can influence the behavior and stability of structures. In this study, the mechanical stability of an underground opening was simulated by using FLAC, which is a two-dimensional modeling code, with a consideration of EDZ. A sensitivity analysis was also carried out with fractional factorial design. From the modeling, it was found that the behavior and the stability of an underground tunnel are strongly dependent on the existence of the EDZ. The sensitivity analysis showed that the key parameters affecting the factor of safety around the tunnel are in-situ stress ratio, depth, cohesion, reduction ratio, internal friction angle, and height and width of the tunnel. It is necessary to consider the EDZ, which can significantly affect mechanical stability in tunnel design.

  • With their wide geographical distribution, unconventional resources are continuously compared against conventional resources, but their development is expanding because TRRs (Technical Recoverable Resources) are similar to conventional resources. In particular, there is active development of unconventional gas resources such as shale gas, tight gas, CBM (coalbed methane) and gas hydrate. However, it is difficult to calculate the material properties of unconventional resources, especially the gas content, with current geophysical logging technology. Additionally, some overseas companies have monopolies on related equipment and materials. Therefore, this study developed a reservoir PCS (Pressure Core Sampler). It can collect core samples without gaseous loss by maintaining high pressure from the moment the core is sampled and record pressure and temperature in real time. Successful performance testing was also carried out for official verification of the manufactured PCS. The reservoir PCS will contribute to the acquisition of geophysical well logging data as well as accurate and reliable cores.

  • Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using FISH (fluorescence in situ hybridization) technique were carried out in Nicotiana plumbaginifolia. Somatic chromosome number of N. plumbaginifolia was 2n=20 and kyarotype was composed of three pairs of metacentric chromosomes (1,2 and 7) and seven pairs of submetacentric chromosomes (3, 4, 5, 6, 8, 9 and 10). Length of chromosomes was ranged from 2.29 to 4.50 ${\mu}{\textrm}{m}$. Chromosome 1 and 2 were identified as satellite chromosomes. In FISH experiment, one pair of 5S rDNA signals was detected on the centromeric region of chromosome 2 and one pair of 45S rDNA signals was detected on the terminal region of chromosome 1.

Hydrological Variability of Lake Chad using Satellite Gravimetry, Altimetry and Global Hydrological Models

  • Sustainable water resource management requires the assessment of hydrological variability in response to climate fluctuations and anthropogenic activities. Determining quantitative estimates of water balance and total basin discharge are of utmost importance to understand the variations within a basin. Hard-to-reach areas with few infrastructures, coupled with lengthy administrative procedures makes in-situ data collection and water management processes very difficult and unreliable. In this study, the hydrological behavior of Lake Chad whose extent, extreme climatic and environmental conditions make it difficult to collect field observations was examined. During a 10 year period [January 2003 to December 2013], dataset from space-borne and global hydrological models observations were analyzed. Terrestial water storage (TWS) data retrieved from Gravity Recovery and Climate Experiment (GRACE), lake level variations from Satellite altimetry, water fluxes and soil moisture from Global Land Data Assimilation System (GLDAS) were used for this study. Furthermore, we combined altimetry lake volume with TWS over the lake drainage basin to estimate groundwater and soil moisture variations. This will be validated with groundwater estimates from WaterGAP Global Hydrology Model (WGHM) outputs. TWS showed similar variation patterns Lake water level as expected. The TWS in the basin area is governed by the lake's surface water. As expected, rainfall from GLDAS precedes GRACE TWS with a phase lag of about 1 month. Estimates of groundwater and soil moisture content volume changes derived by combining altimetric Lake Volume with TWS over the drainage basin are ongoing. Results obtained shall be compared with WaterGap Hydrology Model (WGHM) groundwater estimate outputs.

Genetic Variation of some Patches of Eleutherococcus senticosus (Rupr. & Maxim) Maxim. in Korea (국내(國內) 가시오갈피 군락(群落)의 유전변이(遺傳變異) 분석(分析))

  • The aim of this study was to described the genetic structure of Eleutherococcus senticosus in Korea. We investigated 10 patches, which are eight Korean patches and two foreign patches come from Russia and China growing at Korean habitat, using ISSR(inter-simple sequence repeats) markers. In ISSR PCR, the overall percentage of polymorphic ISSR amplicons was 76% and the mean number of amplicons per ISSR primer was 11.5, which were higher than the RAPD results for the some cultivars collected in Korea(Kim et al., 1998) ; 57% and 5.7, respectively. So ISSR markers provide more powerful tool than RAPD markers for the investigation of genetic variation in E. senticosus. There are relatively high genetic variation among patches as 62.8%, but low variation within eight Korean patches. Such pattern of genetic variation, which is not ordinary in other tree species, may be result from the narrow and limited habitats and the asexual reproduction of this species at the natural stands in Korea. Although the small sample size in this study seemed to be resulted in the high genetic variation among patches, the overall genetic interpretation of this study might not be much affected on the basis of the characteristics of the distribution and the reproduction system of E. senticosus. Analysis of genetic distance between all pairs of the patches did not reveal any trends with regard to geographic distance, which was confirmed by the results obtained from AMOVA(analysis of molecular variance) and PCA(principal component analysis). These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of patches with small number of individuals per patch is more effective for the ex situ conservation and for maintaining the genetic diversity of E. senticosus in Korea than smaller patches with large number of individuals.

  • Within the framework of DECOVALEX-2019 Task D, full-scale engineered barriers experiment (FEBEX) at Grimsel Test Site was numerically simulated to investigate an applicability of implemented Barcelona basic model (BBM) into TOUGH2-MP/FLAC3D simulator, which was developed for the prediction of the coupled thermo-hydro-mechanical behavior of bentonite buffer. And the calculated heater power, temperature, relative humidity, total stress, saturation, water content and dry density were compared with in situ data monitored in the various sections. In general, the calculated heater power and temperature provided a fairly good agreement with experimental observations, however, the difference between power of heater #1 and that of heater #2 could not captured in the numerical analysis. It is necessary to consider lamprophyre with low thermal conductivity around heater #1 and non-simplified installation progresses of bentonite blocks in the tunnel for better modeling results. The evolutions and distributions of relative humidity were well reproduced, but hydraulic model needs to be modified because the re-saturation process was relatively fast near the heaters. In case of stress evolutions due to the thermal and hydraulic expansions, the computed stress was in good agreement with the data. But, the stress is slightly higher than the measured in situ data at the early stage of the operation, because gap between rock mass and bentonite blocks have not been considered in the numerical simulations. The calculated distribution of saturation, water content, and dry density along the radial distance showed good agreement with the observations after the first and final dismantling. The calculated dry density near the center of the FEBEX tunnel and heaters were overestimated compared with the observations. As a result, the saturation and water content were underestimated with the measurements. Therefore, numerical model of permeability is needed to modify for the production of better numerical results. It will be possible to produce the better analysis results and more realistically predict the coupled THM behavior in the bentonite blocks by performing the additional studies and modifying the numerical model based on the results of this study.

A Pilot Scale Biostimulation Study for In-situ Remediation of Organic Pollutants and PAHs contained in Coastal Sediment (연안 해역퇴적물에 함유된 유기오염물과 PAHs의 현장정화를 위한 파일럿 규모의 생물활성촉진연구)

  • A pilot scale biostimulation experiment was performed under field conditions to degrade organic pollutants and PAHs (Polycyclic Aromatic Compounds) in contaminated coastal sediment. During the experiment, the seawater temperature around the sediment varied from $16.5^{\circ}C$ to $21^{\circ}C$, while the sediment pH was stable at 8.4-8.5. The experiment was conducted over a one year period with a control group and a sample group with a applied biostimulant composed of acetate, sulfate, and nitrate. Chemical oxygen demand decreased 39% in the control group and 79% in the sample group. Volatile solids were reduced from 15 to 7 g/kg in the control group and 2.5 g/kg in the sample group. Out of the 2-, 3- ,4- ,5-, and 6-rings of the 16 vital PAHs, the compound including naphthalene (2-ring), took 2 months to degrade completely, while the degradation efficiency was 55.6% in the control group during the same period. In the case of the 3-ring and 4-ring PAHs, complete degradation in the sample and 46% - 100% degradation in control was observed after one year of the experiment. The 5-ring and 6-ring PAHs were degraded to about 77%-100% in the sample group and 26%-87% in the control group during the one year of the experiment. The study results show that biostimulation is a very effective method to improve the degradation rate of organic pollutants and PAHs (Polycyclic Aromatic Compounds) in contaminated coastal sediment.

Rubblization of Thick Concrete Pavement (두꺼운 콘크리트포장의 원위치파쇄 기층화공법)

  • A popular alternative to extend the life of aged pavement is asphalt overlay. However, it has a very serious and inherent shortcoming in deterring a reflection crack. Although joint-rehabilitation and stress-relief techniques have been applied to deter such reflection cracks in aged pavement, the techniques had a limited success only in slowing down the progress of a reflection crack. Rubblization technique rubblizes the concrete pavement slab in situ and uses the rubblized slab as the base material. Then, pavement overlay is applied to finish off the rehabilitation of aged pavement. This rubblization technique has the advantage of solving the problem of reflection cracking completely. When rubblization technique is applied, the upper layer of aged concrete pavement is rubblized between 40mm-70mm in depth. However, the lower layer is typically rubblized more than 100mm in depth. Nevertheless, it is difficult to turn the entire concrete pavement of more than 30cm in depth into rubblized aggregate of appropriate size. Thus, a simulation experiment was carried out to find the appropriate rubblized depth, which avoids the reflection cracking and still maintains the function of subbase, by varying the depth of rubblized depth in loom increments of 0cm, 10cm, and 20cm. The result indicated the optimum rubblized depth was 10cm (Lee, 2006). Additionally, a small rubblizinge equipment was developed in order to derive the rubblization technique appropriate for thick concrete pavement. This equipment was tested out on an experimental pavement, which was constructed with the same standard and specification for the road in actual use, by varying its rubblizing head shape and energy as well as the effective area of rubblization. This experiment led to a prototype equipment for rubblization of thick concrete pavement. The prototype was put into use on a highway, undergoing a test construction and monitoring afterwards. This entire process was necessary for the validation of the proposed rubblization technique.

The evaluation on the biological safety of diagnostic ultrasound using radiation-induced apoptosis in the external granular layer of mouse cerebellum (마우스 소뇌과립층의 apoptosis를 지표로 한 진단용 초음파의 안전성 검증)

  • We have studied, by a nonisotopic in situ end-labeling(ISEL) technique, frequency of apoptosis in the external granular layer(EGL) of the cerebellum of immature mice by ${\gamma}$-rays irradiation from $^{60}Co$ or diagnostic ultrasound exposure. The total number of normal cells and cells showing morphological features of apoptosis were counted. The frequency of apoptotic cells was expressed as a percentage of the total number of cells in EGL. The extent of changes following 200 cGy(1090 cGy/min) was studied at 2, 4, 6, 8, 12, or 24 hours after exposure. The maximal frequency was found 6~8 hours after exposure. The immature mice that received 18, 36, 54, 108, 198, 396 cGy of ${\gamma}$-rays or diagnostic ultrasound(7.5MHz, 4.2mW, $I_{SPTA}=7.9mW/cm^2$, $I_{SPTA}=114.3W/cm^2$) for 10 or 30 minutes were examined 6 hours after irradiation. Measurements performed after ${\gamma}$-ray irradiation showed a dose-related increase in apoptotic cells in each of the mice studied. The dose-response curves were analyzed by a linear-quadratic model ; frequency of apoptotic cell in the EGL was y = $(0.1349{\pm}0.01175)D$+$(-0.0001522{\pm}0.0000334)D^2$+0.048($r^2$ = 0.981, D = dose in cGy). In the experiment of ultrasound exposure, the frequency of apoptotic cell was $0.106{\pm}0.130$(10 minutes exposure) and $0.167{\pm}0.220$(30 minutes exposure). We estimated the relative dose of the yield from the experiment with ultrasound by substituting the yield from ultrasound exposure into the curve from the ${\gamma}$-irradiation. The relative dose of ultrasound exposure compared with ${\gamma}$-irradiation were 0.432 cGy(10 minutes exposure) and 0.885 cGy(30 minutes exposure). We have found that there is no evidence to indicate that diagnostic ultrasound involves a significant risk.
