Significance of a Highly Specific and Sensitive Enzyme Linked Immunosorbent Assay on Evaluation of Environmental Toxicant-Mediated Allergic Responses

  • Enhancement of antigen-specific IgE is a hallmark of allergic hyperresponsiveness, therefore it is necessary to adopt or develop a highly sensitive and specific assay for determination of allergen-specific IgE levels in vivo. In this presentation, we introduce an ELISA (enzyme linked immunosorbent assay) system developed to measure the levels of chicken egg ovalbumin (OVA)-specific IgE in serum. The ELISA method uses a commercially available purified rat anti-mouse IgE as a capture Ab and biotinylated OVA as a detection reagent. Avidin-peroxidase with its substrate is used for color development resulting in optical density measurement at 405 nm. The ELISA system produces a highly sensitive dose-response relation-ship between optical density levels and the dilution titer of the OVA-IgE standard serum but no cross-reaction with unrelated IgE or IgG. It is believed that the system is an Efficient tool to delineate an adjuvant effect of environmental pollutants on development of asthmatic and atopic responses.

  • The vast number of compounds which have been introduced into our environment in recent years is far beyond our capacity to assess for carcinogenicity using expensive, life-time carcinogenicity studies in rodents. Several in vitro short term assays which are very rapid and inexpensive have therefore been applied as mass screening methods. However, increasing evidence has indicated that mutagenicity results do not always correlate with long-term carcinogenicity. Therefore, the existence of discrepancies dictates the necessity of suitable in vivo rapid assay systems which could bridge the gap between mutagenicity and conventional long-term testing.

  • 이광자 현미경(two-photon microscopy)은 다양한 생명현상을 살아있는 조직의 깊은 곳에서 관찰할 수 있다는 장점 때문에 차세대 영상기술로 발전하고 있다. 이에 따라 다양한 목적에 사용할 수 있는 이광자 센서의 개발이 활발하게 진행 중이다. 이 총설에서는 금속이온 이광자 센서에 관한 최근의 연구 결과를 소개하고자 한다.

Radiolabeled Benzamide Derivatives for Development of Malignant Melanoma Imaging Agents

  • Malignant melanoma has an aggressive nature and high metastatic potential that result in one of the highest cancer mortality rates. Over the past three decades, primary and metastatic melanoma incidence has rapidly increased. The recent advances in diagnostic technology have shown promise, but there is still an enormous need for specific detection methods to diagnose malignant melanoma. Positron emission tomography can visualize a particular biomarker of malignant melanoma and promise a noninvasive image of micrometastases. However, the development of PET radiopharmaceuticals remains necessary for diagnosing malignant melanoma by using positron emission tomography. In this review, the history and a general overview of PET radionuclide labeled benzamide derivatives, including their radiosynthesis, in vivo characterization, and evaluation, are provided as imaging agents for malignant melanoma.

양성자 선량 분포 검증을 위한 즉발감마선 분포측정 장치 최적화 연구 (Study on Optimization of Detection System of Prompt Gamma Distribution for Proton Dose Verification)

  • 양성자 치료에서 치료의 목표를 달성하고 환자의 안전을 제고하기 위해 인체 내 양성자 빔의 분포를 확인하는 것이 중요하다. 양성자 선량분포와 밀접한 관계가 있는 즉발감마선의 2차원 분포 측정을 위하여 본 연구팀에서는 다수의 CsI(Tl) 섬광체가 1차원 종형으로 배열된 검출기 배열과 집속장치 및 다채널 신호처리 장치로 이루어진 측정장치를 개발하고 있다. 이에 본 연구에서 몬테칼로 기반의 MCNPX 코드를 이용하여 최적화된 측정 장치를 설계하고자 하였다. 즉발감마선을 효과적으로 측정하기 위해 CsI(Tl) 섬광체의 크기를 $6{\times}6{\times}50mm^3$로 결정하였으며, 배경감마선의 영향을 최소화하고 빔의 진행방향에서 수직방향으로 발생하는 즉발감마선만 측정하기 위해 집속장치의 구멍 크기는 면적 $6{\times}6mm^2$, 길이 150 mm로 최적화되었다. 150 MeV 양성자 빔에 대한 성능 예측 전산모사연구를 수행한 결과, 본 연구에서 최적화된 측정 장치를 통해 즉발감마선 2차원 분포를 측정할 수 있었으며, 1 mm 오차범위에서 양성자 빔의 비정을 결정할 수 있었다. 이를 바탕으로 현재 다채널의 신호처리 장치를 개발하고 있으며 실제 양성자 빔을 이용한 즉발감마선 분포측정을 통해 측정 장치의 성능을 검증할 것이다.

$^{32}P$-Postlabelling 방법을 이용한 미토마이신 C에 의하여 형성된 DNA adduct의 검출 (Detection of DNA Adduct Formed by Mitomycin C by $^{32}P$-Postlabelling)

  • Mitomycin C(MMC) has been used as an anticancer drug and behaves as an alkylating agent forming covalent cross-link between complementary strands of double strand DNA. The purpose of this research was to determine number of DNA adducts, formed in vivo by Mitomycin C, in mouse organs. DNAs from liver, lung, brain and pancreas were isolated and used for $^{32}P$-postlabelling. The labeled nucleotides were separated by 2D-TLC and subjected to autoradiography. Numbers of MMC-DNA adducts were 9,9,5,4 in liver, pancreas, lung and brain, respectively.

  • To diagnose vascular diseases from the viewpoint of hemodynamics, detailed quantitative hemodynamic information of related blood flows with a high spatial resolution of tens micrometer and a high temporal resolution in the order of millisecond were required. For investigating the hemodynamic phenomena of vascular circulatory diseases, a new diagnosing technique combining a medical radiography and PIV method was newly developed. This technique consisted of a medical X-ray tube, an X-ray CCD camera, a shutter module for double pulse-type X-ray source, and a synchronizer. Through several preliminary tests, the feasibility of the angiographic PIV technique was verified. For in-vivo measurements to real blood flows, we developed tracer microcapsules which were optimized to this system by encapsulating a contrast material of iodine with a matrix material of PVA (polyvinylpyrrolidone). In near future, the angiographic PIV technique would be useful for diagnosing hemodynamic phenomena of vascular diseases and for their early detection.

  • $^{19}$ F imaging of mice was carried out. For $^{19}$ F imaging, 5-flouro-uracil (5-FU) was injected into a mouse and in vivo detection of the catabolism of 5-FU to a-fluoro-P-alanine (FBAL) was carried out. The chemical shift selective (CHESS) imaging technique was employed. The 19F spectra and images give temporal and spatial information of the metabolism for 5-FU in mice.

  • Endotoxin has been detected by the Limulus amoebocyte lysate (LAL) test. However, aluminum hydroxide used as an adjuvant and adsorbent for the recombinant protein antigen is known to increase efficacy of lipopolysaccharide vaccine in vivo thus interfering endotoxin test. The aim of this study is to determine effect of aluminum hydroxide on the LAL test using the hepatitis B vaccine as a model and to optimize the LAL test condition not to be interfered by aluminum gydroxide.   • 합성화학물질들이 환경중에 유입되어 인체에는 물론 환경생태계에 많은 영향을 미치고 있어 이들의 유해성 검증은 매우 중요한 일이라 할 수 있다. 실제 산업체에서 사용되는 수많은 화학물질들의 유전적 손상 유발유무는 더욱이 중요한 일이라 할 수 있다. 이에 산업체 공정과정에서 널리 사용되는 것으로 알려진 8종의 합성화학물질에 대해 마우스의 골수세포를 이용한 in vivo 소핵시험을 수행하여, 소핵형성 유발유무를 관찰하였다. 양성대조군으로 사용된 mitomycin는 음성대조군과 비교시 유의하게 소핵을 유발하는 반면, 비교적 마우스에서 높은 50% 치사량을 보이는 phenylisocyanate, m-aminochlorobenzene 및 2-chloro-4-nitroaniline 등의 합성물질들을 비롯한 나머지 5종의 물질들은 본 실험결과 통계적으로 유의하게 소핵을 유발하지 않는 것을 관찰 할 수 있었다.
