A strain of Acremonium strictum, the mycoparasite of Botrytis cinerea, showed strong antifungal activities both in vitro and in vivo against several phytopathogenic fungi. An antifungal substance was purified from the liquid cultures of A. strictum and identified as verlamelin by instrumental analyses. Verlamelin exhibited in vitro antifungal activity against some phytopathogenic fungi such as Magnaporthe grisea, Bipolaris maydis, and Botrytis cinerea, while it was net active against all the bacteria tested. In viva, verlamelin exhibited strong protective and curative activities, particularly against barley powdery mildew. At 100 μg/ml, it inhibited the development of barley powdery mildew with control values of more than 90% in 7-day protective and 2-day curative applications. This is the first report on the production of verlamelin by Acremonium species.
Antifungal activities of crude extractum of Nanshancha Seed Cake (NSC), to inactivate postharvest pathogens were investigated. Highest inhibitory rate was found against C. musae, C. gloeosporioides and C. papaya P.Henn, which was much stronger than that by tea saponin. Compared to tea saponin, effects of NSC extractum was relatively weak and similar on C. gloeosporioides Penzig and P. italicum. In an in vivo study, best controlling effects by NSC extractum was found with banana anthracnose disease development, which showed no inhibitory effects by tea saponin. NSC extractum controlled in vitro C. musae growth through directly inhibiting germination rate and germ tube elongation, and causing distortation, rupture and indentation of C. musae mycelium. In banana fruit subject to C. musae inoculation, higher PAL, POD, GLU and CHT activity was observed in banana fruit treated with crude NSC extractum than that of water control fruits. Current study proved the best controlling effects of crude NSC extractum in C. musae in vitro and in vivo development, which through direct inhibition of C. musae growth and increasing defense system of the banana fruit.
Fourteen Bacillus thuringiensis isolates having both insecticidal activity and in vitro antifungal activity were selected and tested for in vivo antifungal activity against tomato late blight, wheat leaf rust, tomato gray mold, and barley powdery mildew in growth chambers. All the isolates represented more than 70% disease control efficacy against at least one of four plant diseases. Specifically, 12 isolates exhibited strong control activity against barley powdery mildew. Under glasshouse conditions, four (50-02, 52-08, 52-16, and 52-18) of the isolates also displayed potent control efficacy against cucumber powdery mildew. To our knowledge, this is the first report of B. thuringiensis isolates that have disease control efficacy against powdery mildew of barley and cucumber as well as insecticidal activity.

In order to develop a new microbial fungicide for the control of vegetable diseases using endophytic bacteria, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth media, their antifungal activities were screened by in vivo bioassays against Botrytis cinerea(tomato gray mold), Pythium ultimum(cucumber damping-off), Phytopkhora infestans(tomato late blight), Colletotrichum orbiculare(cucumber anthracnose), and Blumeria graminis f. sp. hordei(barley powdery mildew). As the results of screening, 38 bacterial strains showed potent antifungal activities against at least one of 5 plant pathogens. A bacterial strain EB072 displayed potent disease control activities against 3 plant diseases. Among the bacterial strains with a potent antifungal activity against cucunlber anthracnose, three bacterial strains, EB054, EB151 and EB215, also displayed a potent in vitro antifungal activity against C. acutatum, a fungal agent causing pepper anthracnose. A bacterial strain EB215 obtained from roots of cucumber was identified as Burkholderia cepacia based on its physiological and biochemical characteristics and 165 rRNA gene sequence. An antifungal substance was isolated from the liquid cultures of B. cepacia EB215 strain by ethyl acetate partitioning, repeated silica gel column chromatography, and invitro bioassay, Its structural determination is in progress by various instrumental analyses.
Methanol extracts of fresh materials of 57 plants were screened for in vivo antifungal activity against Magna-porthe grisea, Corticium sasaki, Botrytis cinerea, Phyto-phthora infestans, Puccinia recondita, and Blumeria graminis f. sp. hordei. Among them, seven plant extracts showed disease-control efficacy of more than 90% against at least one of six plant diseases. None of the plant extracts was highly active against tomato gray mold. The methanol extracts of Chloranthus japonicus (roots) (CjR) and Paulownia coreana (stems) (PcS) displayed the highest antifungal activity; the CjR extract controlled the development of rice blast, rice sheath blight, and wheat leaf rust more than 90%, and tomato gray mold and tomato late blight more than 80%. The PcS extract displayed control values of more than 90 % against rice blast, wheat leaf rust, and barley powdery mildew and more than 80% against tomato gray mold. The extract of PcS also had a curative activity against rice sheath blight and that of CjR had a little curative activity against rice blast. On the other hand, the extract of Rumex acetocella roots reduced specifically the development of barley powdery mildew. Further studies on the characterization of antifungal substances in antifungal plant extracts are underway and their disease-control efficacy should be examined under greenhouse and field conditions.
Antifungal activities of 43 different plant oils were evaluated against different phytopathogenic fungi. Thyme oil showed highest antifungal activity among the tested oils. The major of thyme oil were found to be thymol, carvacrol, bomeol, p-cymene and linalool. Thymol and carvacrol were found to be responsible for thyme's antifungal activity. The spore germination assay was conducted on Alternaria mail and Botrytis cinerea. Thymol and carvacrol strongly inhibited spore germination in the fungi test. In addition, thymol and carvacrol showed a curative effectiveness to gray mold disease on cucumber crop. The antifungal activities of alkylphenol and alkylaniline compounds, which has similar molecular structure to that of thymol or cavacrol, were also tested. It was found that alkylphenol compounds also show higher inhibition to spore germination. Thus, thymol, carvacrol and alkylphenol compounds can be used an potent antifungal agents.
6-[(N-3,4-Dibromophenyl)amino]-7-chloro-5,8-quinolinedione(FCK13) was tested for antifungal activities. The MIC values were determined by the two-fold dilution method. The therapeutic potential of RCK13 had been assessed in comparison with ketoconazole and fluconazole against systemic infections with candida albicans in normal mice. RCK13 had ED50,0.80$\pm$0.21 mg/kg but ketoconazole had ED50, 8.00$\pm$0.73 mg/kg respectively. And administered RCK13 at the ED50 for 14 days improved survival rates as well as ketoconazole. Acute oral toxicity studies of RCK13 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK13 were low and LD50 values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK13 had been evaluated. RCK13 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK13 with in vivo mouse micronucleus assay. RCK13 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK13 has no genotoxic potential under these experimental conditions.

Taiwan, formerly known as Formosa, located on tropical and subtropical climate zones with abundant biological resources. According to the latest version of the Flora of Taiwan, there are 4339 species of vascular plants including 1054 endemic species. First, Taiwania (Taiwania cryptomerioides), named after its native island of Taiwan, have been isolated more than 500 secondary metabolites, including lignans, terpenoids, steroids, and flavonoids. Several of the metabolites are reported to have antibacterial, antifungal, antimite, antitermite and antitumor activities. In order to investigate plant secondary metabolic diversity toward industrial applications, we established deep transcriptome resources for non-model plants and fungi to produce terpenoid metabolites of economic importance. Second, many plants of Lauraceae have been utilized in folk medicine for their exciting bioactivities. The twigs and leaves from 27 tree species of Lauraceae grown in Taiwan were performed to evaluate potential bioactivity. The leaves of Cinnamomum osmophloeum are traditionally used in folk medicines, and many biological activities have been identified, such as antibacterial, antifungal, antitermite, antidiabetic, antihyperuricemia, antiinflammatory, and antioxidant activities. However, C. osmophloeum has nine chemotypes with various secondary metabolite profiles. In order to efficiently produce active compounds, we established the genetic markers to identify the chemotype plants. Finally, Cinnamomum kanehirae is the host of the medicinal mushroom Antrodia cinnamomea. Several in vivo and in vitro studies indicated that A. cinnamomea possesses a diverse range of biological activities. Because of the potential pharmacological application, we established the transformation system to enhance the triterpenoid contents production.

Chitosan (deacetylated form of chitin) possesses strong antibacterial activities such as antimicrobial effect, antifungal effect and the induction of plant defense response. Chitosan itself, however, has high molecular weight and viscosity as well as water-insolubility, These natures may restrict applications in various fields, especially in in vivo system. While the hydrolysates of chitosan, chitooligosaccharides (COS) are not only lower in the molecula. weight and viscosity, but also water-soluble. Thus, they would be expected more efficient absorption in vivo. Besides several documents have been reported antibacterial activities of COS against microorganisms (Kendra et al., 1989; Uchida et al., 1989). (omitted)
In vivo antifungal activity of 44 fungicides consisting of 3 clubroot fungicides, 7 Oomycetes fungicides, 7 botriticides, 7 blasticides, 9 sterol biosynthesis inhibitors, and 11 broad spectrum fungicides were investigated against Plamodiophora brassicae, the causal agent of clubroot disease in Chinese cabbage. When fluazinam, flusulfamide and cyazofamid, commercial fungicide to control clubroot of Chinese cabbage in Korea, were applied to infested soil, club formations by P. brassicae were strongly inhibited at pot (35 $cm^2$) per 0.63 mg. Ethaboxam and cymoxanil, Oomycetes fungicides, completely controlled Chinese cabbage clubroot at 5 mg/pot, but cymoxanil represented sever phytotoxicity. Besides, dichlofluanid and procymidone of botriticides effectively controlled the development of Chinese cabbage clubroot at 2.5 mg/pot. Chlorothalonil, quintozene and trichlamide, broad spectrum fungicides, showed disease-control efficacy of 85%, 100% and 100% at 2.5 mg/pot, respectively. Most of sterol biosynthesis inhibitors displayed the strong antifungal activity against P. brassicae on cabbage seedlings and plant growth -retarding activity. From these results, 7 fungicides were selected and further tested in vivo antifungal activity against P. brassicae in glasshouse. Among them, ethaboxam showed the most antifungal activity against P. brassicae on cabbage seedlings, followed by fenarimol, procymidone, nuarimol and chlorothalonil.
