사계성 신품종 딸기 '고하'의 기내배양을 위한 배지의 적정 조건 (Appropriate in Vitro Culture Conditions of Growing Medium for New Ever-bearing Strawberry 'Goha')

  • 본 실험은 신품종 사계성 딸기 '고하'의 무병주 생산을 위해 기내배양 시 적정조건을 구명하고자 실시하였다. 식물 재료로는 생장점 배양 후 4주된 유식물체를 이용하였다. 배지 종류는 MS배지, Gamborg B5배지 및 White배지 등 3종류를 사용하였다. MS배지 농도는 1/3배, 1/2배, 1배, 2배, 3배 농도 등 5처리, sucrose 농도는 1, 3, 5 및 8% (w/v) 등 4처리를 두었다. 배지 종류별 실험 결과, MS배지의 관부직경이 2.1mm로 가장 굵었다. 생체중은 MS배지가 482mg으로 Gamborg B5배지의 394mg과 White배지의 222mg에 비해 88g, 260g 각각 더 무거웠다. MS배지 1/2배 처리가 생체 중, 건물중, D/F율 등의 생육량이 다른 농도에 비해 높았다. 초장은 sucrose 1%에서 3.6cm로 가장 길었으며, 농도가 높아질수록 점점 짧아지는 경향을 보였다. 생체중은 sucrose 3% 농도까지 증가하다가 5%이상에서는 감소하는 경향을 보였다. 따라서 사계성 딸기 '고하'의 기내배양 시 1/2 MS 배지에 sucrose 1%을 첨가하는 것이 가장 적합한 것으로 판단되었다.

In-vitro meat: a promising solution for sustainability of meat sector

  • The in-vitro meat is a novel concept in food biotechnology comprising field of tissue engineering and cellular agriculture. It involves production of edible biomass by in-vitro culture of stem cells harvested from the muscle of live animals by self-organizing or scaffolding methodology. It is considered as efficient, environmental friendly, better ensuring public safety and nutritional security, as well as ethical way of producing meat. Source of stem cells, media ingredients, supply of large size bioreactors, skilled manpower, sanitary requirements, production of products with similar sensory and textural attributes as of conventional meat, consumer acceptance, and proper set up of regulatory framework are challenges faced in commercialization and consumer acceptance of in-vitro meat. To realize any perceivable change in various socio-economic and environmental spheres, the technology should be commercialized and should be cost-effective as conventional meat and widely accepted among consumers. The new challenges of increasing demand of meat with the increasing population could be fulfill by the establishment of in-vitro meat production at large scale and its popularization. The adoption of in-vitro meat production at an industrial scale will lead to self-sufficiency in the developed world.

  • Plant regeneration has been optimized increasingly by organogenesis and somatic embryogenesis using a range of explants with tissue culture improvements focusing on factors, such as the age of the explant, genotype, media supplements and $Agrobacterium$ co-cultivation. The production of haploids and doubled haploids using microspores has accelerated the production of homozygous lines in Brassicaceae crop plants. Somatic cell fusion has facilitated the development of interspecific and intergeneric hybrids in sexually incompatible species of $Brassica$. Crop improvement using somaclonal variation has also been achieved. Transformation technologies are being exploited routinely to elucidate the gene function and contribute to the development of novel enhanced crops. The $Agrobacterium$-mediated transformation is the most widely used approach for the introduction of transgenes into Brassicaceae, and $in$ $vitro$ regeneration is a key factor in developing an efficient transformation method in plants. Although many other Brassicaceae are used as model species for improving plant regeneration and transformation systems, this paper focuses on the recent technologies used to regenerate the most important Brassicaceae crop plants.

Gene Expression of the In Vitro Fertilized or Somatic Cell Nuclear Transfer Embryos Cultured in Medium Supplemented with Different Proteins or Energy Substrates

  • Several cloned animals have been produced using somatic cell nuclear transfer (SCNT) and have interested in producing the transgenic cloned animals to date. But still its efficiency was low due to a number of reasons, such as sub-optimal culture condition, aberrant gene expression and nuclear reprogramming. The purpose of this study was to analyze gene expression pattern in in vitro fertilized (IVF) or SCNT pre-implantation embryos. IVF- or SCNT-embryos were cultured in media supplemented with different proteins (FBS and BSA) or energy sources (glucose or fructose). Blastocysts from IVF or SCNT were analyzed using semi-quantitative RT-PCR in terms of developmentor metabolic-related genes. Culture medium supplemented different proteins or energy sources had affected on the expression of developmental or metabolic genes in the SCNT blastocysts.

NCSU-23과 PZM 배양액내 첨가된 Macromolcule이 돼지 체외수정란의 발육에 미치는 영향 (Effect of Macromolecules Supplemented to NCSU-23 and PZM Culture Media on in vitro Development of Porcine Embryos)

  • 본 연구에서 배양액 내 첨가된 macromolecule이 돼지 체외수정란의 체외 발달율에 미치는 영향과 배양소적 내 첨가된 FBS가 후기배발달에 미치는 영향에 대하여 알아보았다. 이에 대한 결과를 요약 하면 다음과 같다. 1. NCSU-23을 기본 배양액으로 사용시 BSA (4mg/ml), FBS (10%), PVA (3 mg/ml)가 각각 첨가되었을 경우 배발달에 큰 차이를 보이지 않았다. 2. PZM을 기본 배양액으로 사용시 BSA (4mg/ml)가 첨가하였을 경우 2-세포 수정란 발달율과 배반포 발달율이 가장 높았으며, 이것은 NCSU-23에 비하여 PZM 배양액에는 후기배의 에너지원으로 사용하는 글루코스가 포함되어 있지 않기 때문으로 사료 된다. 본 연구를 통하여 돼지 수정란의 체외배양에 사용되는 배양액은 종류와 조성에 따라 적절한 macromolecule이 첨가되어 초기배와 후기배의 발달을 모두 촉진시킬 수 있어야 함을 알 수 있었다.

Effects of Collection Time, Culture Time and Activation Treatment of Canine Oocytes on the IVM Rates

  • These study was carried out to investigate the effects of the collection time, culture time and activation of canine oocytes on in vitro maturation rates. The activated oocytes were cultured in 10% FCS+TCM-199 media containing hormonal supplements (10 IU/ml HCG, 10 IU/ml PMSG, 10 ug/ml gonadotropin) at 5% $CO_2$, 95% air, $38^{\circ}C$. 1. IVM rate of in vitro cultured cumulus-attached oocytes recovered from ovaries that collected at follicular and luteal stages of the reproductive cycles were 11.4% and 5.7%, respectively. IVM rate of oocytes recovered from ovaries that collected at follicular stages of the reproductive cycles was significantly higher than that of luteal stage (p<0.05). 2. When IVM was carried out at different periods of 40, 48, and 70 hrs, the IVM rates of oocytes matured in vitro were 2.9%, 8.6%, 5.7%, respectively. These results indicate that the IVM time between $48{\sim}70$ hrs gives the highest maturation rate for the oocytes matured at the different stages. 3. IVM rate of oocytes matured in vitro for 10 hrs after single and combined activation treatment by ET, IP and CH and Ca+DMAP, CH+DMAP, ET+CH were $11.5{\pm}1.2%,\;10.8{\pm}1.0%,\;9.6{\pm}1.2%\;and\;12.4{\pm}1.5%,\;11.8{\pm}1.5%,\;11.2{\pm}1.4%$ respectively. This was higher than that in both single and combined stimulated groups compared to control group ($6.2{\sim}7.2%$).

체외배양 조건이 소 체외수정란의 생산에 미치는 효과 (Effects of Different Culture Conditions on In Vitro Production of Bovine Embryos)

  • This study was conducted to establish the optimal culture conditions for in vitro production of bovine embryos derived from slaughter house ovaries. Cumulus-oocyte- complexes (COCs) collected by aspiration from follicles of 2~7 mm in diameter were matured in Ham's F-10 medium supplemented with 0.01 $\mu\textrm{g}$/m1 epidermal growth factor (EGF) at 39$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$in air. After 24 hrs of culture, the oocytes were co-cultured with epididymal sperm selected off by Percoll-density gradient in TALP medium for 24 hrs. The presumptive zygotes were cultured in HECM-6 medium for 3 d post-insemination, and followed by cultured in TCM199 medium until 7 to 10d post-insemination. The cultures were compared of their cleavage and development into later stage in culture medium by additions of different protein sources (PVA, BSA and BCS) and by different embryo density. The rates of cleavage and development rates into blastocyst were not significantly (P<0.05) different among the culture media containing with BSA (75.0% and 40.5%), BCS (76.7% and 38.0%) and PVA (72.5% and 42.2%), respectively. Significantly (P<0.05) higher blastocysts rates were obtained in culturing of 30 and 40 embryos in each 50$\mu$l droplets of culture medium than in 5, 10 and 20 embryos. These results indicate that the optimal density of embryos is 30~40 embryos in a 50$\mu$l droplet of culture medium. Furthermore there is no effect of different protein sources on early embryonic development.

  • The biological control potential of entomopathogenic nematodes (EPN) can be enhanced by improved culture efficiency. Optimization of media is a key factor for improving in vitro mass production of entomopathogenic nematodes. EPN yield was dependant of complex medium concentration, of which mixture is carbohydrates, lipids, proteins, salts, and growth factors, on the growth of Heterorhabditis bacteriophora and its symbiotic bacterium Photorhabdus luminescensLipids.

랫드 전배아배양법을 이용한 2-Bromopropane의 최기형성 평가 (Teratogenicity Evaluation of 2-Bromopropane Using Rat Whole Embryo Culture)

  • Recently, we have reported that the environmental pollutant 2-bromopropane (2-BP) induces a significant embryo-fetal developmental toxicity in rats. However, the cause of developmental toxicity and the relationship between maternal and developmental toxicities could not be elucidated because the developmental toxicity of 2-BP was observed only in the presence of maternal toxicity The in vitro teratogenicity study using whole embryo culture was carried out to understand the teratogenic properties and the possible mechanism of teratogenicity induced by 2-BP in rats. Rat embryos aged 9.5 days were cultured in vitro for 48 hrs at medium concentrations of 0, 1, 3, or 10 mg/ml of 2-BP. Embryos were evaluated for growth, differentiation, and morphological alterations at the end of the culture period. At 10 mg/ml, 2-BP caused a delay in the growth and differentiation of embryos and an increase in the incidence of morphological alterations, including altered yolk sac circulation, abnormal axial rotation, craniofacial hypoplasia, open neuropore, absent optic vesicle and kinked somites. At 3 mg/ml, only a delay in the growth and differentiation of embryos was observed. There were no adverse effects on embryonic growth and development at the concentration of 1 mg/ml. The results showed that the exposure of 2-BP to rat embryos results in a developmental delay and morphological alterations at dose levels of 3 mg/ml culture media or higher and that 2-BP can induce a direct developmental toxicity in rat embryos.

인삼의 Ditylenchus destructor 선충의 인공배양 및 밀도에 영향을 주는 요인 (In Vitro Culture and Factors Affecting Population Changes of Ditylenchus destructor of Ginseng)

  • Ditylenchus destructor isolated from diseased ginseng roots was cultured on ginseng calli, fungal mycelium (Fusarium solani), carrot discs and radish sprouts. Effects of temperature, organic material and flooding on the nematode population changes were examined. D. destructor multiplied readily on the culture media except radish sprout medium, and was cultured best on the fungal culture at 2$0^{\circ}C$. Feeding of the fungal hypha and radish root hairs, molting and mating in the fungal culture medium were observed. Addition of organic materials (perilla, sesame, soybean and ginseng leaves) in soil significantly increased Aphelenchus avenae and saprophytic nematode populations, while D. destructor populations changed little and the nematode population growths were limited by the organic amendments (except sesame leaves). The nematode populations in soil including D. destructor were decreased by flooding. The results indicate that D. destructor may survive but not multiply readily in soil without host plants and that it can be effectively controlled by flooding.
