Isolation and Characterization of Siderophore-Producing Bacteria with Various Plant Growth-Promoting Abilities as a Potential Biocontrol Agent (잠재적 미생물 농약으로서 다양한 식물성장 촉진 활성을 가진 siderophore 생산 세균의 분리와 특성)

  • To develop eco-friendly microbial inoculants, siderophore-producing bacteria were isolated and identified, and their production characteristics and plant growth-promoting abilities were investigated. A strain S21 was isolated from rhizosphere of Korean perilla (Perilla frutescens) and identified as Enterobacter amnigenus by phenotypic properties and 16S rRNA gene sequencing. The highest siderophore production was obtained in a medium containing 0.5% fructose, 0.1% urea, 0.5% K2HPO4 and 0.1% succinic acid. By using this improved medium, siderophore production increased by 2.5 times compared to that of basal medium. The strain S21 showed insoluble phosphate solubilizing, ammonification and antifungal activities, and also produced hydrolytic enzymes (protease and lipase), indoleacetic acid and 1-aminocyclopropane-1-carboxylate deaminase. Our data suggest that E. amnigenus S21 is a potential candidate that can be used as eco-friendly biocontrol agent and biofertilizer.

Effects of Maesil Extract on the Quality Characteristics of Yellow Layer Cake(I) - Rheology Characteristics of Cake Batter - (매실 엑기스 첨가가 Yellow Layer Cake의 품질 특성에 미치는 영향(I) - 케이크 반죽의 리올로지 특성 -)

  • This study was conducted in order to investigate the effects of maesil extract on medium and cake flour using the falling number and RVA tests, as well as farinography, and alveography. The maesil extract were added on the medium and cake flour bases, 3% and 5% each. The falling numbers of the medium and cake flour with maesil extract were increased due to alteration of the protein in the flour. Analysis of the RVA characteristics showed that the addition of maesil extract did not have a significant effect on the initial pasting temperature. Peak viscosity, holding strength, final viscosity, and the breakdown and setback values of the medium flour were not consistent, but those of the cake flour decreased. Farinography demonstrated that the addition of maesil extract decreased water absorption and improved the stability of the medium and cake flour. The characteristics of the alveogram showed that the addition of maesil extract increased the extensibility and swelling index. Based on the experimental data, the processing rheology may be good when using maesil extract to make yellow layer cake.

Improved in vitro Regeneration of Potato (Solanum tuberosum cv. Superior) Transformed by Agrobacterium Expressing $\beta-Glucuronidase$

  • In order to enhance the system of potato transformation and further regeneration, potato was transformed using the Agrobacterium tumefaciens harboring $\beta$-glucuronidase (GUS) gene. We found that a series fo modified medium ttained 100% shoot regeneration within 5 weeks after the preincubated explants on stage I medium were infected with Agrobacterium. Callus appeared at the cut edges of stem segments on stage II medium, mainly at the basal parts. Some explants started to form shoots after two to three weeks on stage III medium containing kanamycin (50 mg/L). When transferred to MS medium containing 200 mg/L kanamycin, 81% of the transformed shoots formed roots at the cut edge of the plantlets. In contrast, untrasformed shoots never rooted and became yellowish after few weeks under the same conditions. Southern and northern analysis indicated in vitro shoot regeneration on the callus derived from the potato explants, which were incubated with Agrobacteria. The regeneration cycle was shortened after the transformatin and finally the transformation efficiency was highly enhanced.

  • Actinobacillus pleuropneumoniae (APP) is a causative agent of porcine pleuropneumonia. Therefore, the development of an effective vaccine for APP is necessary. Here, we optimized the culture medium and conditions to enhance the production yields of Apx toxins in APP serotype 1, 2, and 5 cultures. The use of Mycoplasma Broth Base (PPLO) medium improved both the quantity and quality of the harvested Apx toxins compared with Columbia Broth medium. Calcium chloride (CaCl2) was first demonstrated as a stimulation factor for the production of Apx toxins in APP serotype 2 cultures. Cultivation of APP serotype 2 in PPLO medium supplemented with 10 ㎍/ml of nicotinamide adenine dinucleotide (NAD) and 20 mM CaCl2 yielded the highest levels of Apx toxins. These findings suggest that the optimization of the culture medium and conditions increases the concentration of Apx toxins in the supernatants of APP serotype 1, 2, and 5 cultures and may be applied for the development of vaccines against APP infection.

Rheological Characteristics of Flour Batters in the Presence of Mugwort Powder (쑥 분말을 첨가한 소맥분의 리올로지 특성에 관한 연구)

  • This study was conducted to investigate the effects of mugwort powder on two types of flour batters, i.e., medium and cake, using the Falling Number test, RVA test, alveogram, and farimogram. The mugwort powder was added at 3% and 5% on the medium and cake flour bases, respectively. The Falling Numbers of the medium and cake flour batters with mugwort powder were increased due to the alteration of the protein in the flour. The characteristics of the alveogram showed that the addition of mugwort powder increased the extensibility and the swelling index. Farinogram demonstrated that the addition of mugwort powder decreased the water absorption and improved the stability of the medium and cake flour batters.

High-frequency Plant Regeneration from Mature Seed-derived Callus Cultures of Orchardgrass (오차드그래스 성숙종자로부터 캘러스 유도 및 고효율 식물체 재분화)

  • In an effort to optimize tissue culture conditions for genetic transformation of orchardgrass (Dactylis glomerata L.), an efficient and high-frequency plant regeneration system from seed-derived calli was established. Embryogenic calli induced on MS medium containing 3mg/L 2,4-D and 0.1mg/L BA had significantly improved regeneration ability. Plant regeneration rate was 92% when embryogenic calli were cultured on N6 medium supplemented with 1mg/L 2,4-D and 3mg/L BA. Among three kinds of medium, MS and N6 medium were optimal for embryogenic callus induction and plant regeneration, respectively. Ho difference in callus induction frequency was observed among four cultivars of orchardgrass, however, "Roughrider" cultivar showed higher regenerability with the frequency of 61%. Addition of maltose to the regeneration medium as a carbon source dramatically increased regeneration frequency up to 69%. A short tissue culture period and high-frequency regeneration system would be beneficial for molecular breeding of orchardgrass through genetic transformation.

Micromagnetic Computer Simulation of Ultra-high density Recording with the Use of a Planar-type Head

  • A computer simulation, utilizing the Landau-Lifshitz-Gilbert equation, of ultra-high- density recording on continuous longitudinal media is carried out. The two important features of this work are the use of a planar-type head, which enables a high write field of 14183 Oe ts be generated at the center of the recording medium, and the media with very high coercivities up to 13010 Oe. From a systematic investigation, it is found that the optimum write field is higher than the medium coercivity by only 3400 Oe over a wide coercivity range. This new finding allows one to write an a medium with a very high coercivity by using a planar-type head. It is demonstrated that a reasonably good bit pattern with a bit density of 605 kfci is generated on the medium with a coercivity of l1720 Oe, and, combined with a high track pitch density of 100 ktpi, a recording density of 60 Gb/in$^2$can be obtained in a single layer medium. With an improved write- head designs even a higher recording density of 75 Gb/in$^2$may be possible since comparison of the results for the bit pattern from the present head profile and the ideal Lindholm profile indicates an increase in the track pitch density of about 27%. Even at this density, the thermal stability parameter (KV/kT) at room temperature is high enough (60) to provide ample room for thermal stability.

  • Insoluble phosphate-solubilizing bacteria are routinely screened by a plate assay method using Pikovskaya agar and a modified Pikovskaya medium. A modified Pikovskaya medium to improve the clarity of the yellow-colored halo has not necessarity improved the plate assay. Colonies of phosphate-solubilizing bacteria tested could be redily selected after 48 h of incubation by green-colored colony formation on plate in which bromcresol green(BCG) was included. Among them, two bacterial strains did not produce distinct yellow halos after 48 h of incubation. We suggest that the green colony formation on plate medium containing BCG is advantageous ofr rapid isolating phosphate-solubilizing bacteria directly from soil.

  • Effects of growth conditions on the growth of Zoogloea ramigera and the production of zooglan were investigated. The production of zooglan was greatly reduced in the phosphate-limiting medium. $NH_4Cl$ and ${(NH_4)}_2SO_4$ improved cell growth when they were used as a nitrogen source. The medium containing 45 g/L of glucose and 27 g/L of $NaNO_3$ resulted in the highest production of zooglan at 18.5 g/L.
