Phylogenetic and pathogenic traits of YHV3 and IHHNV detected from imported frozen shrimp (수입 냉동새우에서 검출된 YHV3와 IHHNV의 계통학 및 병원성 분석)

  • Yellow head virus (YHV), Infectious hypodermal and hematopoietic necrosis (IHHNV), Taura syndrome virus (TSV), and Infectious myositis virus (IMNV) cause serious mortality to Penaeidae shrimp in the aquaculture. In this study, YHV, IHHNV, TSV, and IMNV were surveyed from imported frozen shrimps between 2019 and 2020 via molecular diagnostic assay. Among 10 shrimp groups, YHV (n=1) and IHHNV (n=4) were detected by RT-PCR and PCR, respectively. From the phylogenetic analysis based on the partial ORF 1b region of YHV, YHV was classified into YHV genotype 3 (YHV3). And IHHNVs (n=2) detected from Litopenaeus vannamei belong to infectious IHHNV type 2. Although IHHNVs (n=2) identified from Penaeus monodon showed PCR positive results (MG 831F/R primer set), the sequences of ORF 2 and 3 were not amplified, suggesting that those samples might possess type A IHHNV related sequence of P. monodon. Furthermore, in the challenge test, even though PCR-detected isolates (YHV3/type A IHHNV related sequence or infectious IHHNV type 2) were not induced mortality to L. vannamei, viral genes were amplified suggesting that the viruses in the frozen shrimp could be non-pathogenic particles which are not enough to induce mortality.

The Analysis of Sensitivity of Imported Fisheries with Regard to Non-Economic Considerations Using AHP (AHP를 이용한 수입수산물의 비경제적 민감도 분석에 관한 연구)

  • This study is to obtain the order of the sensitivity of importing marine products with regard to non - economic aspects of considerations under the free - trade. Three criteria are extracted considering various aspects of Korean fishery society. They are the socio - economic, resource - environmental, and policy - appropriateness criterion. The Analytic Hierachy Process(AHP) is used to obtain the estimates of weights among these criteria. First of all, the weights between economic and non - economic in the first hierachy of sensitivity criteria turn out to be 83.3% and 16.7%. Second, the weights among three criteria in the non - economic aspect, the second hierachy, are following that the socio - economic criterion is 19.6%, resource - environmental 31.1%, and policy - appropriateness 49.3% respectively. 47 selected fishes are evaluated based on these three non - economic criteria using 9- point scales. The over - all order of sensitivity is a croaker, hairtail, flatfish, sea breams, and so forth in descending order. The least sensitive fish turns out to be an angler. In addition to the over - all order, the sensitivity based on the three specific non - economic aspects is also investigated. The most sensitive fish in terms of the socio - economic criterion is an hairtail while the least one is a filefish. The most sensitive fish in terms of the resource - environmental criterion is an abalone while the least one is an angler. Finally, the most sensitive fish in terms of the policy - appropriateness is a mackerel while the least one is a small octopus.

Characterization of the Repetitive Sequences Present in the ORF25 Genomic Region of Megalocytiviruses from Ornamental Fishes (관상어로부터 분리한 Megalocytiviruses에서 나타나는 ORF25 유전자 부위의 반복서열 특성 분석)

  • The presence of ISKNV-like viruses in various freshwater ornamental fish species imported from Asia was confirmed by polymerase chain reaction(PCR) amplification of the ATPase(adenosine triphosphatase) gene. Interestingly, molecular analyses of the Open Reading Frame 25(ORF25) region of these isolates based on the ISKNV(Infectious spleen and kidney necrosis virus) genome revealed the presence of various repetitive sequences. ORF25 repeat sequence length had no effect on cumulative mortality of rock bream Oplegnathus fasciatus challenged with tissue homogenates of infected pearl gourami, Trichogaster leeri; silver gourami, Trichogaster microlepis; blue gourami, or Trichogaster trichopterus. All isolates induce cumulative mortalities after 12 days of infection, confirming that ORF25 polymorphism did not affect the pathogenicity of ornamental fish megalocytiviruses that cross infect rock bream, a seawater fish. Also, no statistically significant differences in spleen index or viral copy number in infected tissues was detected between isolates with varying ORF25 repeat sequence lengths. However, further studies are necessary to fully characterize the functional characteristics of these polymorphisms in megalocytivirus disease in ornamental fishes.

A Study on the Distribution Channel of Fishery Products Exported to the United States (대미 수출 수산물의 유통경로 분석 연구)

  • The United States is Korea's important trading partner, ranking third (13.7%) in exports of fishery products in Korea. The impact on exports to the United States is expected to be significant if the seafood import provisions under the MMPA are implemented in 2024 accordingly. It is expected that documents proving production information will be required for all items when exporting to the U.S. Therefore, this study attempted to examine the current status of exports and exporters in order to understand the actual conditions of major fishery products exported to the United States. Besides, there are currently no official system and procedures in Korea to prove production information for all fishery items, so we tried to suggest implications by examining distribution channels for major export items to the U.S. In this study, five items including seaweed and halibut as domestic aquaculture items, tuna and squid as deep-sea fishing items, and fish cake using imported raw materials were selected as the target items for distribution channel investigation. In addition, this study is meaningful in that it investigated the routes from production to processing, distribution, and export of each item and identified detailed distribution channel for major items exported to the U.S.

Establishment of Analytical Method for Methylmercury in Fish by Using HPLC-ICP/MS (고성능액체크로마토그래피-유도결합플라즈마 질량분석기를 이용한 어류 중 메틸수은 분석법 확립)

  • BACKGROUND: Methylmercury is analyzed by HPLC-ICP/MS because of the simplicity for sample preparation and interference. However, most of the pre-treatment methods for methylmercury need a further pH adjustment of the extracted solution and removal of organic matter for HPLC. The purpose of this study was to establish a rapid and accurate analytical method for determination of methylmercury in fish by using HPLC-ICP/MS. METHOD AND RESULTS: We conducted an experiment for pre-treatment and instrument conditions and analytical method verification. Pre-treatment condition was established with aqueous 1% L-cysteine HCl and heated at $60^{\circ}C$ in microwave for 20 min. Methylmercury in $50{\mu}L$ of filtered extract was separated by a C18 column and aqueous 0.1% L-cysteine HCl + 0.1% L-cysteine mobile phase at $25^{\circ}C$. The presence of cysteine in mobile phase and sample solution was essential to eliminate adsorption, peak tailing and memory effect problems. Correlation coefficient($r^2$) for the linearity was 0.9998. The limits of detection and quantitation for this method were 0.15 and $0.45{\mu}g/kg$ respectively. CONCLUSION: Result for analytical method verification, accuracy and repeatability of the analytes were in good agreement with the certified reference materials values of methylmercury at a 95% confidence level. The advantage of the established method is that the extracted solution can be directly injected into the HPLC column without additional processes and the memory effect of mercury in the ICP-MS can be eliminated.

Effects of Various Diets on Growth and Body Composition of Juvenile Olive Flounder, Paralichthys olivaceus (배합사료 종류가 넙치 Paralichthys olivaceus 치어의 성장 및 체성분에 미치는 영향)

  • The 7-week feeding experiment was conducted to investigate the effects of one experimental diet (ED) and five different commercial diets (CDs) on growth and body composition of juvenile olive flounder, Paralichthys olivaceus. An ED was formulated to contain 50.0% crude protein (CP) from fishmeal, casein, zein and wheat flour and 15.0% crude lipid (CL) from squid liver oil. Five CDs for seawater fish were two domestic E commercial diet (DECD) and C commercial diet (DCCD), three imported H commercial diet (IHCD), M commercial diet (IMCD) and O commercial diet (IOCD) containing 53.1~58.0% CP and 4.8~12.7% CL, respectively. Each diet was fed to triplicate groups of juvenile olive flounder initially weighing $29.1{\pm}0.8g/fish\;(mean{\pm}SD)$ in a flow-through seawater system with a water temperature of $23.4{\sim}28.0^{\circ}C$. Weight gain (WG) was significantly greatest in fish fed the IMCD; intermediate responses were observed for fish fed the DECD, DCCD, and IOCD, while the IHCD and the ED produced the lowest WG values. Feed efficiencies (FE) were similar to WG excluding fish fed the DCCD; FE was also greatest in fish fed the DCCD. Survival with no significant difference approached 100% for fish fed the all six diets in this experiment. Whole-body crude protein and ash contents were not affected excluding moisture and crude lipid by the different type of diets. Therefore, type of diets appeared to be important factor in influencing WG, FE and whole-body moisture and crude lipid of juvenile olive flounder; the best diet for juvenile olive flounder was determined to be the imported commercial M diets containing intermediate protein (55.9%) and lipid (12.7%) in natural seawater based on highest WG, and FE, respectively. This study indicates that the one commercially formulated diet containing intermediate protein and lipid used in this experiment could be a practical diet for juvenile olive flounder; these differences in growth performance between ED and CDs may be due to different dietary protein and lipid levels.

Physicochemical Properties of Fish-meat Gels Prepared from Farmed-fish (해수어를 활용한 연제품의 제조 및 물리화학적 특성)

  • Fish-meat gel is being produced mostly relying on surimi and raw materials imported from Southeast Asia and North America and present in small amount in local markets. In this study, common farmed local fishes were examined as stable and reliable sources of surimi for fish-meat gel production. For testing, five main farmed-fish of Korea, namely; Bastard halibut (Paralichthys olivaceus), Red sea bream (Pagrus major), Korean rockfish (Sebastes schlegeli), Common mulle (Mugil cephalus), and Finespotted flounder (Pleuronichthys cornutus) were used following a traditional washing process. The quality of the surimi was determined by the values of water content, whiteness index, gel strength and impurity. Accordingly, fish-meat gel and surimi quality experiments were carried out by measuring compressive and texture properties, expressible moisture content, Hunter color scale values and SDS-page protein patterns. Also gel characteristics were compared with that of FA and RA grade surimi (Alaska Pollock). Fish-meat gels were prepared by salt mincing the farmed-fish surimi with NaCl (2% w/w) and moisture adjustment to 84% by ice water adding. Prepared fish-meat paste was filled into 20-25 cm long polyvinylidene chloride casings and heated at 90℃ for 20 min. The whiteness values of fish-meat gels produced from surimi were increased by using farmed-fish and became comparable to that of FA Alaska Pollock gel. Among all tested farmed-fish, P. olivaceus and P. major exhibited better properties than RA Alaska Pollock and similar properties to FA Alaska Pollock. Therefore, current data suggests that fish farming can be an efficient and sustainable fish-meat source for fish-meat gel production in Korea.

  • Marine fish aquaculture has been introduced on the East sea since 1964. The production volume of the marine aquaculture has been increasing drastically since 1990 with the technological improvements and the introduction of new species. However, the increasing costs of feed, seedlings and medical supplies and the decreasing prices due to the increase in the imports and domestic production have decreased the profits in the aquaculture business. Futhermore, the damage from the natural disasters such as red tides and typhoons has accelerated the exit from the business. Even though managing abilities to overcome the business crisis caused by the above factors are required, business managers based on the small scale family businesses do not have enough managing abilities to deal with the difficulties in the business. On the other hand, advanced countries in the marine aquaculture such as Norway have reached the stage where the managing abilities of marine aquaculture are similar to those in the manufacturing industry. And the number of large scale aquaculture farms with developed technologies and advanced marketing strategies in those countries is increasing. Considering that the marine aquaculture in Japan under the similar fishery systems of Korea has developed the state-of-the-art management skills or lead to large scale management, it is difficult to expect the decrease in the production costs under the small scale family business in Korea and this will lead to the decreasing competitive advantage over the imported seafood. Therefore marine aquaculture in Korea needs to increase the economy of scale to acquire the competitive advantage. This study addresses the possibility of introducing the large scale aquaculture system in Korea by analysing the factors and process of leading to large scale businesses in the aquaculture in Japan.

Estimation of Consumer Value on Import Management of Seafood Obtained from IUU Fishing: Using Choice Experiment Method

  • Purpose - This study empirically analyzes the consumer value of risk management associated with illegal, unreported, and unregulated (IUU) fishing of fishery products imported to Korea. The global regulatory paradigm for IUU fishing has shifted from production-centered to market-centered. As a result, changes in the international fisheries trade environment emphasizing "transparency" and "legality" of the production process are accelerating. Therefore, changes in the management systems of fishery products entering the country are also needed. Accordingly, this study estimated the consumer value for risk management of IUU fishing, targeting major fish species imported to Korea, and derived the feasibility of introducing related policies. Design/methodology - This study used the choice experiment as an analysis model to estimate consumers' willingness to pay for the "possibility to check for IUU fishing." The choice experiment assumes that the value of a good or service is composed of separable attributes and that the sum of the part-worth of these individual attributes becomes the total value. In this study, respondents were presented with profiles comprising three attributes (country of origin, price, and possibility of checking IUU fishing) and the levels of frozen poulp squid, the subject of the analysis. The participants were asked to select their preferred profile. The marginal willingness to pay for each attribute was derived from the results of the respondents' choices using conditional logit model estimates. Findings - There is a marked difference in utility based on the preference of the country of origin of fishery products among consumers. In addition, the utility of fishery products that have undergone IUU fishing verification was observed to be higher, with the utility marked to be higher for lower prices. Originality/value - Estimating the policy value of the risk management in IUU fishing of imported fisheries products in this study is a novel attempt that has never been conducted before. Several studies have been conducted to assess the risk of IUU fishing associated with the import of fishery products internationally. However, such studies are yet to be conducted in Korea. Instead, policies and studies have focused on issues related to complying with trading partners' legal and transparent standards for exporting fishery products. This study should be the beginning of more in-depth empirical and theoretical explorations to establish order in the domestic seafood market and respond to changes in international regulations on IUU fishing.

Detection and distribution of iridoviruses in five freshwater ornamental fish species (담수관상어 5종에서의 iridoviruses 검출과 분포 분석)

  • The distribution of iridoviruses in five freshwater ornamental fishes, pearl gourami (Trichogaster leeri), dwarf gourami (Colisa lalia), silver gourami (Trichogaster microlepis), blue gourami (Trichogaster trichopterus sumatranus) and freshwater angelfish (Pterophyllum scalare), imported from Singapore was examined in 2004 and 2005. The presence of iridoviruses in 56 sample groups was determined using PCR technique and showed PCR positive in 11 sample groups. The proportion of fish infected by iridovirus was differed depending upon species; 31.8% (7/22) for pearl gourami, 18.2% (2/11) for dwarf gourami, 16.7% (1/6) for blue gourami, 9.1% (1/11) for silver gourami and 0% (0/6) for angelfish. In quantitative comparison of viral DNAs isolated from infected tissues, the DNA concentration of iridovirus in pearl gourami was higher than that in dwarf gourami. Although pearl gourami infected naturally by iridovirus showed 100% mortality in keeping experiment for 3 weeks, only 57% of those was positive in PCR. In the comparison of nucleotide sequences of the PstⅠ fragment known as the most variable genomic region, both iridoviruses isolated from pearl gourami and dwarf gourami showed identity more than 99% with infectious spleen and kidney necrosis virus (ISKNV) isolated from mandarinfish (Siniperca chuatsi).
